[A method of fluorescent immunoenzyme analysis in the complex evaluation of viral and rickettsial vaccines].

With the use of the unified indirect solid-phase fluorescent enzyme immunoassay the combined evaluation of the antigenic and immunogenic properties of experimental whole-virion inactivated virus vaccines against Venezuelan and eastern equine encephalomyelitides, as well as of dried chemical typhus vaccine, has been made; their safety was determined indirectly by the content of ovalbumin. The protective role of antibodies evaluated by the solid-phase fluorescent enzyme immunoassay in typhus immunity has been shown.

[A comparative evaluation of an immunoenzyme analytical method using a fluorogenic or chromogenic substrate in determining the Marburg virus antigen].

A comparative evaluation of the developed variants of indirect solid-phase enzyme immunoassay using fluorogenic and chromogenic substrates for the determination of Marburg virus antigen was carried out. The resolving capacity of this method was 3.8 x 10(-9) g of protein for the former and 3.1 x 10(-8) g of protein for the latter substrate. Cross titrations demonstrated the lack of common antigens with Ebola virus.

[Development and study of the properties of immunoglobulin against Ebola fever].

Immunoglobulin to Ebola fever has been for the first time prepared from hyperimmune equine blood sera by alcohol fractionation after Cohn. Preclinical study of the physicochemical and immunobiological properties of immunoglobulin showed that it protects up to 100% Papio hamadryas infected intramuscularly at doses of 110 to 29 LD50 Ebola virus. Scheme for the use of Ebola immunoglobulin has been experimentally validated.

[An experimental study of the immunogenic and protective properties of Coxiella burnetii strain M-44 and a live enteral vaccine against Q fever].

High effectiveness of C.burnetii strain M-44 was confirmed in experimental studies on subcutaneously immunized guinea pigs ¿correction of pig] after their intratracheal infection with the virulent culture of the infective agent. The study revealed that the dose necessary for the enteral immunization of guinea pigs and monkeys was 10(4.8) -- 10(5.0) times less than that needed for the oral immunization of the animals. Live enteral vaccine obtained on the basis of this strain possessed sufficiently pronounced immunogenic and protective properties, protecting 75% of immunized monkeys from subcutaneous infection with C.burnetii virulent culture.

[Development of the immunoenzyme test-system for detection of Ebola virus antigen].

An enzyme immunoassay system has been developed for the detection of Ebola virus antigen. It permits a highly accurate and sensitive rapid detection of the antigen. Optimal dilutions of specific immunoglobulin (1:500, corresponding to protein concentration of 50 micrograms/ml) and conjugate were found. The resolving capacity of the new test system is 1.9 x 10(-7) g protein.

[Development and study of immunoglobulins against Lassa fever].

The authors validate the use of horses as producers of immune antiserum to Lassa virus. Specific immunoglobulin with at least 1:512 titer of virus neutralizing antibodies to Lassa virus was obtained from the serum of immunized horses by Kohn's alcohol method. The resultant preparation does not differ from the heterologous commercial immunoglobulins. Preclinical studies of immunoglobulin against Lassa virus demonstrated its safety and high specific activity. The strategy of treating with the immunoglobulin alone and in combination with virasol has been experimentally validated.

A horse may serve the producer of immune antiserum to Lassa virus. Specific immunoglobulin with at least 1:512 titer of virus-neutralizing antibodies to Lassa fever was obtained by alcohol sedimentation after Cohn from the blood serum of immunized horses. The preparation does not differ from heterologous commercial immunoglobulins. Preclinical studies of immunoglobulin to Lassa fever demonstrated its safety and a high specific activity. The agent can be injected both alone and in combination with virasole.
