Modifications in the phagocytosis of human neutrophils induced by vinblastine and cytochalasin B: the effects of lithium.

The effects of lithium on the microtubular and microfilament systems of neutrophil granulocytes is described. The phagocytic capacity of human neutrophils was reduced or inhibited by using vinblastine or cytochalasin B. Lithium at concentrations ranging from 0.5 to 2.0 mEq/l proved to be capable of antagonising the action of vinblastine, whereas the action of cytochalasin B was found to be unaffected by lithium. Based on the antagonising effect of lithium on vinblastine-induced inhibition of phagocytosis, it is suggested that lithium acts--at therapeutically achievable levels--on the microtubular system of neutrophils, possibly through modulation of cyclic AMP levels.

Role of autorosette forming cells in antibody synthesis in vitro: suppressive activity of ARFC in humoral immune response.

The role of autologous rosette forming cells (ARFC) in humoral immune responses was studied using an in vitro system. While depletion of ARFCs from PBL resulted in a significant increase of either total IgG or anti-TT IgG, addition of these cells to the system decreased the production of immunoglobulin to a level comparable to that of unfractionated PBL. The majority of the ARFCs reacted with anti-Leu2a and anti-Leu8. In contrast, the majority of non-ARFCs reacted with Leu3a and only 10% with Leu8 monoclonal antibodies. Stimulation of unfractionated PBL with concanavalin A (ConA) resulted in an increase of the ARFC population. ConA stimulation also increased the number of cells reactive with anti-Leu2 and/or anti-Leu8. The autorosette population had a higher purine nucleoside phosphorylase (PNP) content than the non-ARFC population. Although the ARFC suppressed synthesis of antibody by B cell in vitro when they were mixed with either autologous or allogeneic B cells, a marked proliferation of non-B cells was evident. We conclude that at least two different subpopulations of T cells are capable of forming rosettes with autologous red blood cells.

Effects of lithium carbonate on leukocyte functions in chronic benign neutropenia.

The effects of lithium carbonate on leukocyte functions in a case of chronic benign neutropenia are presented. Lithium was able to induce leukocytosis and to bring about increases in chemotaxis, marrow granulocyte reserve test and phagocytosis. After lithium interruption, leukocyte functions returned to initial values. Some hypotheses are advanced to account for lithium action.

Lonidamine plus adriamycin versus adriamycin alone in the adjuvant treatment of recurrent papillary carcinomas of the urinary bladder.

This paper reports the interim results of 25 patients included in a double-blind study in which the combination of Lonidamine plus adriamycin is compared to adriamycin alone in the adjuvant treatment of papillary carcinomas of the urinary bladder. Although it is impossible to draw any conclusion at this time on the therapeutic activity of this combination, tolerance was judged satisfactory.

Methisoprinol restoration of leukocyte chemotaxis inhibition by blocking factors in immunoproliferative diseases.

The ability of methisoprinol to antagonize the serum-inhibiting activity present in the sera of patients affected by immunoproliferative diseases (Hodgkin's disease and multiple myeloma) was investigated. Using a chemotaxis test, it was found that whereas donor leukocytes preincubated in patient sera exhibited impaired chemotaxis, the presence of methisoprinol (500 micrograms/ml) during the preincubation caused a significant recovery of this leukocyte function. Since the results exclude the assumption of a non-specific stimulation of cell motility, two hypotheses appeared possible for the action of methisoprinol: either an antagonistic activity against the blocking factor at the receptor or the metabolic level, or a direct inactivation of this factor. The clinical implications, with regard to the prophylaxis of infections and a better response to chemotherapy, are discussed.

Immunomodulators and enzymes of purine metabolism in human lymphocytes.

The enzymes ADA and PNP were evaluated in lymphocytic subpopulations in peripheral blood obtained from healthy subjects, elderly subjects and patients with immunoproliferative diseases. Some similar assessments were performed on lymphoid cells from cord blood. Preliminary studies indicate that Thymostimulin can in some cases correct enzymic defects.

[Behavior of some enzymes of nucleoside catabolism in circulating lymphocytes during paraproteinemia].

The circulating lymphocytes of 16 normal subjects and of 18 patients with paraproteinemia have been characterized by measuring the levels of ADA, AMPA, PNPase, CDA. The results obtained reveal a highly significant increase in PNPase of subjects affected by paraproteinemia as compared to that of normal subjects (p less than 0.005). The fact that the increased enzymatic levels are found mainly in patients not affected by Bence-Jones proteinuria seems to indicate that the evolution of the paraproteinemia disease is in some way related to the intralymphocyte levels of PNPase.

Blood lymphocytes in chronic lymphocytic leukaemia and Hodgkin's disease: Immunological features and enzymes of nucleoside metabolism.

Purified lymphocytes from venous blood of sixteen healthy adult subjects, eight patients with chronic lymphocytic leukaemia (CLL) and ten with Hodgkin's disease (HD), were examined for adenosine deaminase (ADA), cytidine deaminase (CDA), purine nucleoside phosphorylase (PNPase) and adenosine monophosphate deaminese (AMPA), after thawing and homogenization. The same cells were examined for the capacity to form E rosettes and to respond to phytohaemagglutinin (PHA) stimulation. In CLL a significant reduction (P less than 0-001) of AMPA, PNPase and ADA activities was observed without variation of CDA. In contrast in HD PNPase, AMPA and CDA were increased (P less than 0-01) while ADA was in the normal range. The E-rosette forming cells were significantly reduced in both diseases and the capacity to respond to PHA-stimulation was strongly impaired in CLL. By this experimental approach it seems possible to demonstrate different states of functional activity of the lymphocytic cells in two diseases characterized by reduced T-cell-mediated immunity.
