Electron microscopy detection and characterization of viral particles in dog stools.

A number of apparently normal dog stool samples, randomly collected on the sidewalks of Paris were examined by Electron Microscopy. The study revealed the presence of viral particles in 27 cases. Morphological criteria lead to the characterization of rotavirus in 2 specimen, coronavirus in 7 and parvovirus in 5. Rotavirus particles appeared always alone while coronavirus and parvovirus particles were present together in 5 cases. Similar particles have been implicated in animal and human gastroenteritis. The importance of their presence in canine dejections is discussed in view of pollution by dog stools of urban and suburban areas.

[Determination of IgM antibodies by complement-fixation after serum fractionation by sucrose density gradient in children and adults with evidence of Mycoplasma pneumoniae infection (author's transl)].

Sera from 45 patients with serologic evidence of Mycoplasma pneumoniae infection were fractionated by sucrose gradient centrifugation. The distribution of complement-fixing (CF) antibodies for Mycoplasma pneumoniae within the 19S and the 7S immunoglobulins was determined all patients below 20 years of age and 44% of older patients had 19S specific antibodies. A predominance of 19S relative to 7S CF antibodies was observed in 40% of cases below 20 years and in 18% of cases above 20 years. The meaning of these data is discussed.

[Comparative study of anti EBV antibodies in non-Hodgkin's lymphomams and angio-immunoblastic lymphadenopathies (author's transl)].

Antibody titers to Epstein-Barr virus (EBV)--related antigens (viral capsid antigen : VCA; early antigen : EA; and EBV associated nuclear antigen : EBNA) were determined in the sera of 86 patients and 150 matched control subjects. The patients belonged to four histological groups : diffuse and nodular non-hodgkin's lymphomas angio-immunoblastic lymphadenopathies and apparented syndroms. The incidence of antibodies to other herpes-viruses (cytomégalovirus, herpes simplex virus, and varicella zoster virus) was compared. There was a significantly higher incidence of anti VCA and anti EA titers in some patients, not associated with an increase in titres of antibodies to other herpes viruses.

[A method for the preparation of a Chlamydiae group-specific antigen on hela-229 cells infected with a strain of "Chlamydia trachomatis" for use in the complement fixation test (author's transl)].

A simple method for the preparation of a potent group-specific antigen on HeLa-229 cells infected with MRC-1 (LB) (TRIC/GB/MRC-1 Gf) strain of Chlamydia trachomatis is outlined. HeLa-229 cells are infected (MRC-1 strain, 102 inclusion-forming units per cell) with centrifugation at 4,000 g for 1 h in flat-bottomed vials. The cells are removed by brief trypsinization with 0.25% trypsin and put into 75 cm2 culture flasks (12 x 106 cells by flask) in BHK-21 medium supplemented with foetal bovine serum. The flasks are incubated for 5 days at 37 degrees C. The destroyed cell monolayer and the supernatant are centrifuged at 100,000 g for 1 h. The pellet is collected and resuspended in PBS and subjected to ultrasonic vibration for 30 min. This antigen may be used for detection of complement-fixing antibodies in lymphogranuloma venereum and ornithosis.

Cytomegalovirus infection in malignant blood diseases:clinical and laboratory data in 29 patients.

Twenty-nine patients treated for malignant blood diseases developped CMV infection. Their clinical and laboratory features were studied. The results indicated that this infection apparently did not influence the prognisis of the underlying disease. The main hematological feature was pancytopenia. The data of viremia suggested active infection. A marked increase of CMV CF antibodies were observed in 27/29 patients, and the peak titers of 42% of our cases were greater than 1:1024. Homogenous Ig were detected in 7/29 patient's serum.

Rapid detection of human cytomegalovirus viraemia from small volumes of heparinized whole blood.

Rapid detection of human cytomegalovirus (HCMV) from blood was performed in parallel using inoculation into human fibroblastic cells of both 500 microliters of whole blood and of buffy coat derived from 9.5 ml of blood. Of the 46 samples tested, 20 were positive for HCMV, 18 when the buffy coat was inoculated (90%) and 17 (85%) when whole blood was used. Fifteen samples were positive by both techniques. Four samples gave discordant results: in three cases, viraemia was detected only by buffy coat assay and in one case only by whole blood assay. These results suggest that HCMV isolation from blood should be performed by inoculating whole blood into cell cultures if only small blood volumes can be collected.

AIDS-Related Opportunistic Infections

[Bacteriological, parasitological and virological study of the digestive flora in alpha-chain disease].

Intestinal flora was explored in twelve patients affected with alpha-chain disease at different stages (stage A: 2 cases; stage B: 6 cases; stage C: 4 cases). Bacterial overgrowth in the jejunum was observed in 11 cases, but intestinal flora was diverse and no one species was always present; although a 3-month oral antibiotic treatment induced complete remission in one patient (stage A) it was not possible to demonstrate any pathogenic bacterial species. Intestinal lambliasis was present in 40 p. 100 of cases. Virologic studies were negative. At stages A and B of the disease, antibiotic treatment was able to improve malabsorption and/or plasma protein digestive losses in 62 p. 100 of cases; this effect seemed related to the reduction of the bacterial flora and to giardiasis eradication.
