Generalized demodicosis associated with diabetes mellitus in two cats.

Generalized demodicosis with concurrent diabetes mellitus was diagnosed in 2 cats. Both cats had generalized alopecia and a bilateral brown aural exudate. Mites were seen microscopically in skin scrapings and ear swab specimens. Histologic examination of the skin showed the follicular orientation of the mites. Abnormal results on oral dexamethasone suppression tests, clinical findings, and bilateral adrenal hyperplasia found on necropsy of one of the cats suggested concurrent hyperadrenocorticism.

Localization of immunoglobulins and complement by the peroxidase antiperoxidase method in autoimmune and non-autoimmune canine dermatopathies.

Formalin-fixed, paraffin embedded skin biopsy specimens from 44 dogs with various dermatopathies were examined for the presence of immunoglobulins (IgG, IgM, IgA) and complement (C3) by the peroxidase antiperoxidase method (PAP). Final diagnoses of the skin diseases were determined by clinical findings, fungal and bacterial cultures, skin scrapings, serum endocrinologic tests, and histologic features of cutaneous biopsies. The dermatopathies included examples of pyoderma (folliculitis/furunculosis), demodecosis, sarcoptic mange, dermatophytosis, endocrine dermatopathy, and autoimmune skin disease (AISD). In the latter category, 7 cases of pemphigus foliaceus, 1 pemphigus vulgaris, 4 discoid lupus erythematosus (DLE) and 4 examples of indeterminate AISD were examined. Immunoglobulins, C3, or both, were localized in tissue sections from AISD (15/16), pyoderma (7/11), demodecosis (4/8) sarcoptic mange (3/3), and dermatomycosis (2/3). Endocrine dermatopathy biopsies were consistently negative for Ig and C3 (0/3). The pattern of localization was most often intercellular (31/44 positive biopsies) with basement membrane immunoreactivity in 4 cases of DLE, and 1 case of indeterminate AISD. There was no consistent correlation between histologic features of hematoxylin and eosin-stained biopsies and the presence of Ig and C3. Clinical outcome was similar in both Ig and C3 positive and negative cases of non-AISD dermatitis. The high percentage (95%) of positive results in AISD biopsies indicates the usefulness and sensitivity of the PAP method for the localization of Ig and C3. Because of the high percentage of Ig localization in pyoderma (73%), biopsy specimens with extensive inflammatory skin disease are not suitable for diagnosis of AISD. The PAP method is useful in the diagnosis of AISD in biopsy specimens with minimal inflammation. Caution is warranted in the interpretation of immunoreactivity independent of clinical and histologic information.

Idiopathic periadnexal multinodular granulomatous dermatitis in twenty-two dogs.

A distinctive periadnexal multinodular granulomatous dermatitis of unknown cause was diagnosed over a 6-year period in 22 dogs (13 males and nine females) aged 8 months to 10 years, purebred as well as mixed-breed. Nodules, usually more than two, started on the face and were often unaccompanied by signs of irritation. All dogs were otherwise healthy except a Viszla with bilateral anterior uveitis and a Basset hound with nasal congestion and sneezing. Microscopically, cutaneous lesions occurred in a periadnexal pattern, replaced sebaceous glands, coalesced, and extended into the subcutis and in more advanced cases through the panniculus carnosus. Infectious agents were not demonstrated by aerobic, anaerobic, or fungus cultures or by light or electron microscopy, nor were ectoparasites seen. Immunofluorescence and peroxidase-antiperoxidase techniques gave negative results for intraepidermal and basement membrane deposits of IgA, IgG, IgM, and C3. Lesions were unresponsive to a variety of antibiotics; they regressed spontaneously in two untreated dogs and rapidly in dogs given only corticosteroid therapy. Continuous low-dose corticosteroids were necessary in a few dogs to prevent recurrence.

Modulation of carbohydrate residues in regenerative nodules and neoplasms of canine and feline pancreas.

The glycoconjugates of regenerative acinar cells, acinic cell carcinomas, islet cell tumors, and normal canine and feline pancreas were studied. The authors used biotinylated lectins as probes and avidin-biotin-peroxidase complex as visualant to identify and to compare the distribution of carbohydrate residues on paraffin sections from 74 cases. The findings demonstrate a difference in the staining pattern between normal acinar, islet, and ductal cells in each species and small differences in the staining pattern between the species. It is shown that in nodules of regenerative acinar cells and acinic cell carcinomas there is an increased staining intensity with Concanavalia ensiformis agglutinin, Ricinus communis agglutinin-I, and wheat germ agglutinin. The pattern of lectin staining in regenerative cells and malignant acinar cells reflects the degree of cellular differentiation. Intensive apical staining characterizes a higher degree of differentiation, while dispersed staining is a major feature of poor differentiation. These findings suggest that malignant transformation of pancreatic acinar cells is associated with enhanced expression of glycoconjugates, which resembles that seen in a normal immature acinar cells.

Distinctive peripheral lymph node hyperplasia of young cats.

Peripheral lymph node enlargement was found in 14 of a series of 132 feline lymph node biopsy specimens. Six of nine cats tested had antibodies for feline leukemia virus (FeLV). Half of the cats were clinically normal while the remainder had fever, lethargy, anorexia, and hepatosplenomegaly. There was severe distortion of lymph nodal architecture with variable loss of discernible follicles and sinuses. Histiocytes, lymphocytes, immunoblasts, and plasma cells were present in expanded paracortical regions which encroached on, and occasionally effaced, lymphoid follicles. Postcapillary venules were numerous and prominent throughout the paracortex. The lymphadenopathy was most commonly transient (86% of cases) with subsequent development of lymphoma in one cat. Lymph nodes from seven kittens with experimental FeLV infection were compared with spontaneously enlarged lymph nodes; four of seven had B and T lymphocyte hyperplasia with normal nodal architecture. Three had partial loss of nodal architecture as a result of expanded paracortical regions populated largely by histiocytes and lymphocytes. Proliferation of postcapillary venules was not prominent in nodes from FeLV-infected cats. The cause of spontaneous lymph node hyperplasia of young cats was not determined. However, the similarity of lesions to those of kittens with experimental FeLV infection and the association with FeLV by serologic tests in six of nine cats suggest that this retrovirus may be involved in the pathogenesis of the lesion.

Unsuccessful treatment of disseminated protothecosis in a dog.

Systemic protothecosis was diagnosed in a 7-year-old dog that had only ocular manifestations. During the 3-month course of disease, a variety of drugs was administered, including amphotericin B, gentamicin, and ketoconazole. The ocular signs initially abated, but subsequently worsened during this period. The dog was found dead 3 months after initial examination, and systemic protothecosis was confirmed at necropsy.

Thyroglobulin and calcitonin immunoreactivity in canine thyroid carcinomas.

Although thyroid gland neoplasms are well-recognized entities in dogs, the diagnosis and classification of these tumors often is difficult. In contrast to human thyroid carcinomas, which are predominantly of the papillary or follicular types, a relatively high proportion of the canine tumors contain compact cellular areas and resemble, to some extent, medullary thyroid carcinomas. In order to assess the value of immunohistochemical techniques in the identification and classification of these neoplasms, 21 canine thyroid carcinomas were examined for the presence of thyroglobulin and calcitonin using the peroxidase-antiperoxidase technique. Four major patterns of thyroglobulin immunoreactivity were present in the tumors, including diffuse cytoplasmic positive reaction, apical staining in the cells bordering the neoplastic follicular lumens, intracytoplasmic droplet staining, and staining of intrafollicular colloid. All follicular and mixed compact cellular/follicular tumors contained immunoreactive hormone, while only four of six compact cellular carcinomas were thyroglobulin-positive. The extent of thyroglobulin reactivity was consistently greater in tumors of the follicular and mixed patterns than in carcinomas of the purely compact cellular type. Two of four metastases, each of which retained the mixed pattern of the primary tumors, were thyroglobulin-positive. No medullary thyroid carcinomas were identified, but scattered calcitonin-positive cells in one mixed and in one compact cellular tumor were interpreted as entrapped nonneoplastic C cells. Immunohistochemical localization of thyroglobulin should facilitate the diagnosis of canine tumors of suspected thyroid follicular cell origin, particularly those arising in ectopic sites (i.e., heart base) and those presenting as metastases.

Telangiectasia of Pembroke Welsh Corgi dogs.

Multiple vascular lesions involving kidneys and various other organs consisting of cavernous, blood-filled spaces lined by endothelial cells with various amounts of mural collagen are described in eight Pembroke Welsh Corgi dogs. The most common clinical sign associated with this condition is hematuria.

Leucocyte ascorbic acid levels and vitamin C intake in older people.

Leucocyte ascorbic acid (L.A.A.) levels and vitamin C intake were measured in a random sample of men and women aged 62-94 years. L.A.A. distributions are positively skewed but log normal. L.A.A. mean values show no age difference in men but are significantly lower in older women. The mean value for all women (23.88mug/10(8) cells) is significantly higher than that for all men (18.11 mug/10(8) cells). L.A.A. values are significantly higher in both sexes in the six months July to December. Vitamin C intake distributions are positively skewed but not improved by log transformation. No significant age or sex differences were found except that a significantly greater proportion of men over than of those under 70 years have intakes less than 30 mg daily. Mean intake is significantly higher in men but not in women in the six months April to September, though in both sexes a significantly greater proportion have intakes less than 30 mg daily in October to March compared with April to September. Fifty per cent. of men and 58% of women have intakes less than 30 mg daily, 23.6% of men and 28.1% of women have intakes less than 20 mg daily, and 4.7% of men and 3% of women have intakes less than 10 mg daily. These percentages increase during the winter. A moderate correlation is present between vitamin C intake and L.A.A. level. L.A.A. levels increase in parallel with but lag behind seasonal increases in vitamin C intake.

Diagnosis of chlamydial infection in pet birds: comparison of cloacal-swab culture and peroxidase-antiperoxidase methods.

Cloacal swabs were examined using a peroxidase-antiperoxidase (PAP) method that employs a monoclonal antibody to chlamydiae. The specificity and sensitivity of cloacal-swab PAP examination and the prevalence of chlamydiosis were calculated using both culture and tissue PAP evaluation as standards. The prevalence of chlamydial infections was 9.3% as determined by culture and 46.3% as judged by tissue PAP evaluation. Cloacal-swab PAP examination yielded a sensitivity of 71.4% and specificity of 85.5% when compared with culture results and a sensitivity of 81% and specificity of 85.7% compared with tissue PAP results. Culture had a sensitivity of 47.4% and specificity of 100% compared with tissue PAP results. Similarly, the efficiency of cloacal-swab PAP test was about 84% compared with culture results and tissue PAP. The efficiency of culture compared with tissue PAP was 73.1%.

Chlamydia immunoreactivity in birds with psittacosis: localization of chlamydiae by the peroxidase-antiperoxidase method.

Diagnosis of psittacosis (infection with Chlamydia psittaci) has traditionally been dependent upon culture or demonstration of chlamydiae in tissues by cytochemical stains such as the Gimenez or Macchiavello's stain. In this study, a peroxidase-antiperoxidase (PAP) technique that employs a commercially available monoclonal antibody directed against Chlamydia is used to demonstrate chlamydiae in the necropsy tissues of nine birds. Chlamydiae were identified in air sacs (83%), liver (78%), spleen (78%), small intestines (67%), large intestines (44%), and kidneys (33%). Usually chlamydiae were associated with microscopic lesions, but in 75% of large intestines examined, organisms were identified within the lumen of histologically normal intestine. PAP studies revealed numerous chlamydiae that were inapparent in hematoxylin-and-eosin-stained sections. These results demonstrate the utility and sensitivity of the PAP method in the identification of chlamydiae in microscopic sections.
