Immunocytochemical localization of basic fibroblast growth factor in the rat pineal gland.

The immunocytochemical localization of basic fibroblast growth factor (b-FGF) during the postnatal development of the rat pineal gland was studied using a polyclonal antibody against the fraction 1-24 of bovine recombinant b-FGF. Basic FGF immunoreactivity was evident from day 20 after birth in the endothelial cells and perivascular spaces of the gland. Although b-FGF immunostaining showed its maximal expression at 30-45 days, it was maintained throughout the entire study period (up to 6 months), mainly in the distal zone of the gland. Pinealocytes did not show b-FGF immunoreactivity at any time. There were no differences in the localization patterns or the intensity of b-FGF immunostaining after the prenatal denervation with DSP-4, a neurotoxic amine. The physiological role of b-FGF in the adult pineal gland remains unknown; however, it does not seem to play a major role during the cytodifferentiation period of the parenchymal cells, or during the neovascularization in the early postnatal days. Furthermore, its immunocytochemical expression is not affected by the prenatal sympathetic denervation with DSP-4, in contrast with other neurotropic factors.

Serum bone Gla protein as a marker of bone turnover in acromegaly.

Serum bone Gla protein, a sensitive and specific marker of bone turnover, was measured in 35 acromegalic patients (14 untreated, 8 clinically active, and 13 cured) and 21 controls. We also examined 10 acromegalic patients before and after transsphenoidal surgery. Untreated and clinically active acromegalic patients had significantly higher serum bone Gla protein concentrations than the control subjects. Other nonspecific biochemical markers of bone metabolism, such as urinary hydroxyproline and urinary calcium, were also present in significantly greater amounts in active acromegalic patients. After treatment, a significant decrease in levels was observed, with return to control levels. In acromegalic patients, positive correlations were found among serum bone Gla protein and serum growth hormone and serum insulin-like growth factor I levels, as well as among levels of insulin-like growth factor I and serum phosphorus, serum alkaline phosphatase, and urinary hydroxyproline. These results suggest that serum bone Gla protein is a sensitive marker of the action of growth hormone in bone metabolism in acromegaly, a role that is probably mediated by insulin-like growth factor I.

Immunohistochemical localization of glial fibrillary acidic protein (GFAP) in rat pineal stalk astrocytes.

In the present work, the presence and distribution of astrocytes in the rat pineal stalk is investigated applying an immunohistochemical technique for the demonstration of glial fibrillary acidic protein (GFAP) on Epon-embedded semithin sections (0.5 micron thick). GFAP-immunoreactive cells are evenly and regularly distributed along the entire pineal stalk. The GFAP-immunoreactive cells display a stellate shape showing variable numbers of cell processes that are mainly oriented parallel to the longitudinal stalk axis. Astrocytic processes show a clear tendency to encircle the remaining elements of the pineal stalk; i.e., pinealocytes, nerve fibres and blood vessels. Furthermore, glial processes form a cover layer separating the stalk from surrounding anatomical structures.

[Relationship between various erythrocyte antigens and hematologic disorders].

The antigen pattern pertaining to the ABO (A, B, AB, O (H)), Rh (D, C, c, E, e), Duffy (Fya and b), Kell (K, k) and MNS (M, N) systems were determined in 144 patients between 1 and 74 years of age who had leukemia, lymphoma or myeloma. An association of phenotype Fy (a-b+) with acute lymphatic leukemia and phenotype NN with chronic myeloid leukemia was demonstrated (p less than 0.05, chi sq). Other associations were statistically not significant. Thus, a susceptibility of the aforementioned phenotype patterns to the type of leukemia described is suggested by these findings.

Multiple cerebral hydatid cysts.

A 39-year-old woman was admitted to hospital with headaches, vomiting, psychic impairment and diplopia. Three hydatid cysts of the lung had been previously removed. An avascular mass in the left hemisphere with left-to-right displacement of the anterior cerebral arteries was noted during a brain angioscintigraphy. A computed tomography (CT) brain scan showed two cystic lesions situated in the left-frontal and occipital regions. A CT abdominal scan showed multiple cysts in the liver, spleen and both kidneys. At operation, two brain cysts were totally extirpated without rupture. The definite pathological diagnosis was secondary hydatid cysts. The headaches, vomiting and diplopia were persistent in the post-operative period. Seven days after the operation, a CT brain scan showed an infratentorial cyst. The patient rejected any surgical intervention.

Anticoagulant therapy modifies fibrinolytic dysfunction in chronic atrial fibrillation.

OBJECTIVE: We evaluated the changes in fibrinolytic activation markers before starting anticoagulation, at 1 and 6 months following the introduction of anticoagulant therapy. DESIGN AND METHODS: Tissue plasminogen activator (t-PA), its endothelial inhibitor (PAI-1), plasmin:antiplasmin complexes (PAPc), modified antithrombin III (ATM), D dimer (D-D) and fibrinogen (FIB) were measured in 36 patients with chronic atrial fibrillation. Fifteen of them had rheumatic mitral stenosis and 21 had nonrheumatic atrial fibrillation. Basal levels were compared with a sex- and age-matched healthy control group. RESULTS: At baseline, patients with atrial fibrillation showed significantly higher plasma levels of PAI-1, ATM, D-D and FIB levels (p < 0.05) than controls, and no differences in t-PA and PAPc concentrations. Levels of t-PA, PAI-1, ATM and D-D decreased significantly under anticoagulant therapy, whereas FIB levels were not significantly modified. PAPc levels were significantly increased at 6 months in the rheumatic group but did not differ significantly in the nonrheumatic group. CONCLUSIONS: Patients with chronic atrial fibrillation show a hypercoagulant state and a relatively low fibrinolytic function. After 6 months of anticoagulant therapy, an improvement in fibrinolytic function markers was detected.
