[Differentiated carcinoma of the thyroid].

Experience in differentiated thyroid carcinoma surgery is presented. It is considered that, although these neoplasias are not very aggressive, they should be treated radically with a combination of surgery and radioisotopes.

[Pharmacologic profile of a new compound with antiinflammatory activity: 2-(4-dimethylvinylphenyl)propionic acid (Gx 258)].

Gx 258, a new phenylalkanoic acid derivative, was shown in animal tests to possess potent antiinflammatory, analgesic and antipyretic activity. Its antiinflammatory activity was not mediated via the pituitary-adrenal axis, since it was also effective in adrenalectomized rats. Since large doses of Gx 258, 9-83 times greater than those showing antiinflammatory activity, were required to produce gastrointestinal lesions in rats, Gx 258 appears to have a very good therapeutic index. Its therapeutic index as an antiinflammatory and analgesic agent was 2.3-20.5 times greater than that of the parent compound ibuprofen. Gx 258, like ibuprofen, inhibited the biosynthesis of prostaglandins in vitro.

Some pharmacological studies on a new tripeptide mustard.

The analgesic, spasmolytic, antiinflammatory, C.N.S., cardiovascular, respiratory, and anaphylactogenic activities of PTT-119 were investigated. PTT-119 possesses good peripheral but only slight central analgesic activity. It presents an interesting in vitro spasmolytic activity of non-competitive type against acetylcholine and histamine. PTT-119 is inactive in rat carrageenin oedema, on the C.N.S. of mice, on cardiovascular and respiratory parameters of cats, and in guinea-pig anaphylaxis.

Synthesis, acute toxicity and chemotherapeutic anti-cancer activities of a new tripeptidic mustard.

A new antitumor agent having the chemical composition of 3-(p-fluorophenyl)-Lalanyl-3-[m-bis-(2-chloroethyl)aminophenyl]-L- alanyl-L-methionine ethyl ester hydrochloride (PTT 119) was synthetized. It fundamental toxicological data in mice and rats were assessed; its antitumor activity was investigated by establishing in mice, inoculated with L 1210 leukemia or in AKR mice with spontaneous leukemia of viral origin, a significant increase in Mean Survival Time as well as a percent increase of Increased Life Span and of the number of survivors.
