Mitochondrial DNA polymorphism in Calabria (southern Italy).

Restriction Fragment Length Polymorphism (RFLP) of mitochondrial DNA (mtDNA) has been investigated in a sample from Calabria (Southern Italy). Seven restriction endonucleases (HpaI, BamHI, HaeII, MspI, AvaII, HincII and PstI) have been employed on 60 mtDNAs from placentas. Two new morphs have been identified (PstI-3Cal and AvaII-24Cal) and the respective polymorphic sites have been located by double digestion. Two new types (74 and 75) have also been identified and their possible origin from other known mtDNA types has been suggested.

Hyperplasia and metaplasia of ovarian surface epithelium in women with endometrial carcinoma. Suggestion for a hormonal influence in ovarian carcinogenesis.

Surface ovarian epithelium and that of related inclusion cysts were comparatively studied in two groups of patients: one group of 50 non-pregnant women without myometrial, endometrial, or cervical hyperplasia and/or neoplasia; a second group of 50 women surgically treated for endometrial cancer and without evidence of ovarian pathology. Papillomatosis, hyperplasia and tubaric, squamoid, endometrioid and mucinous metaplasia were more frequently present in ovarian surface epithelium or in related inclusion cysts in patients with endometrial carcinoma. These findings may be correlated with a hormonal oncogenic stimulus involved in the genesis of endometrial cancer and suggests a new "hormonal" hypothesis in the pathogenesis of common epithelial tumors of the ovary.

Modification of muscle proteins IEF patterns in a fish Lebistes reticulatus, related to diet, immunization and genetic selection.

The effects of some environmental and genetic factors on the isoelectric focusing (IEF) patterns of muscle proteins from a fish Lebistes reticulatus (Cyprinodonts) were investigated. Of two diets applied the restricted one resulted in a loss of bands in the IEF patterns of the treated fishes muscle homogenate. Instead immunization gave an increase in both number and intensity of IEF bands in (4.0-6.0; 7.0-7.9) pH ranges which are typical of immunoglobulins. Lastly IEF patterns from random and assortative mating animals were compared, resulting in loss of IEF-bands in the inbred lines. Results showed that IEF of muscle proteins could be an excellent method to evaluate the response by fish phenotypes to environmental or genetic factors.

G1m(1) and G1m(2) allotypes in Albanian towns of Calabria.

The G1m(1) and G1m(2) allotype distribution was analyzed in a population sample from 11 Albanian towns of Calabria. The unusually high frequency of the G1m(1) marker already observed in Calabria as well as the presence of the Gm(2) phenotype were shown. The Calabrian and Albanian populations were similar, but significantly different from other Italian populations.

Human polymorphic APO-LDL investigated by high-resolution two-dimensional electrophoresis.

The APO part of a human polymorphic low density lipoprotein (LDL) was investigated by two-dimensional electrophoresis. Since these allotypic LDL were purified from sera by specific immunochromatography, the data presented here refer to only one molecular species of LDL. The two-dimensional electrophoresis revealed the presence of isoforms which were correlated to the serological phenotype. A hypothesis about APO-LDL gene organization is outlined.

Morphometric analytical evaluators of lymphoid populations in nonneoplastic lymph nodes.

A morphometric analysis using an Apple II microcomputer and shape analytical morphometry (SAM) software was performed of the nuclear shape in the lymphoid population of normal (nonneoplastic) lymph nodes to provide baseline parameters useful for distinguishing between different subtypes of lymphomas. Fourier harmonic analysis and two indices, a contour asymmetric evaluator (CAE) and a shape asymmetric evaluator (SAE), are proposed as non-dimensional shape parameters. Results obtained with the CAE and SAE indices indicate that they represent sensitive shape descriptors able to reveal even subtle irregularities of nuclear outline. With these parameters, centrocyte nuclei were easily differentiated from those of all other lymphoid cells while an interesting close relationship was found between lymphocytes and centroblasts. Lymphocyte nuclear shape was shown to be less regular than generally reported, suggesting the existence of subtle differences between stimulated and unstimulated nuclei. Immunoblast nuclei were shown to have the most regular shapes.

[Description of the shape of carcinomatous nodules of the breast by means of high degree polynomial equations and Fourier harmonic analysis].

Breast cancer nodule shape has been investigated by two analytical procedures: upper degree polynomials and harmonic Fourier analysis. An automized method (TV camera interfaced with a computer) has been employed to find the coefficients of kth order equation giving the best fit. Then the difference between real node contour and function curve has been calculated and submitted to harmonic analysis. This gives the sine/cosine coefficients of the two series and the amplitude of the harmonics. The results point out that no difference exists between metastatic and non metastatic tumors and suggest that parametrized descriptors calculated may represent a peculiar behaviour in the growth of the tumor.

[Reproductive conditioning of the partridge (Perdix perdix) with GnRH and an increase in the photoperiod].

Follicular growth and egg deposition were induced in the Grey Partridge (Perdix Perdix) after stimulation with Gn-RH and artificial light. The experiment was carried out from November to December 1983, during the short day period, and during the non egg laying period for this bird-sp. Two groups consisting of ten pairs of Grey Partridge each (female and male), received 3.8 mcg of Gn-RH (pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2) every 8 hours for 10 and 15 days, respectively. With interruption of the Gn-RH treatment we observed follicular regression. For this reason the birds were stimulated with artificial light daily from the 15th day of treatment until egg deposition. Fifty-five days after the start of the Gn-RH treatment the Grey Partridge laid eggs. Forty days of this period the birds were supplemented with artificial light. Twenty pairs of Grey Partridge were used as controls. Periodically two experimental female subjects (treated and control) were sacrificed to observe the development of ovary and oviducts.

Identification of an allotypic IgA in cattle serum.

Alloimmunization of cattle revealed a polymorphism carried on a serum antigen displayed beta-electrophoretic mobility. A genetic test indicated that the antigen is inherited in a Mendelian manner, as product of a dominant allele at an autosomal locus. By immunochemical investigation the allotypic determinant was found to be located on IgA: there were in fact no differences between the peptidic map of the molecule carrying the allotypic determinant and that of IgA from positive sera. Moreover the peptidic map of IgA from sera lacking of the allotype resulted lacking of an anodic peptide, thus suggesting that the polymorphism is indeed due to such a peptide.

Immunochemistry of an allotypic low density lipoprotein in man.

This paper presents data on a human allotypic polymorphism carried on a low density lipoprotein (As-LDL). The As polymorphism was investigated by a rabbit antiserum which was firstly used to analyze the serum level and the isoelectric point of the As lipoprotein in sera of different phenotype. This antiserum was also used to purify As-LDL by immunochromatography. Afterwards the peptidic maps of the APO proteins from different As-LDL were compared. While no correlation did result between the serum level and the serological phenotype of the As lipoprotein, the peptidic distribution of the APO parts from different As-LDL showed differences correlated to the allotypic phenotype.

G1m(a) and G1m(x) allotype distribution in a population from Calabria (Italy).

G1m(a) and G1m(x) allotype distribution is analyzed in a sample population from Calabria (South Italy). The paper presents data showing an unusually high frequency of the G1m(x) marker. Furthermore, results concerning a rare Gm(x) phenotype are also shown. The findings suggest the important contribution yielded to the Calabrian genetic pool by the Albanian population that immigrated to this region in the 15th century.

[Identification of a polymorphic AS marker in human serum. Evidence of a linkage imbalance to the beta thalassemic locus].

Radial Immuno Diffusion performed by including in agar serum from a polytransfused Cooley patient (beta-thalassemic homozygous) allowed to identify a human serum antigen marker of an autosomal polymorphic locus. the antigen and its coding gene were named AS and AS respectively. Genetic tests indicate that the AS1 dominant allele codes for the presence of AS in the serum while the AS0 recessive allele codes for its absence. The frequency of the AS0/AS0 homozygous genotype results to be greater in Cooley than in healthy people thus suggesting that the AS0 and beta-thal alleles may be in linkage disequilibrium. The observation that the frequency of the AS0 beta-thal haplotype is greater than the product of the frequencies of the alleles which form it, confirms such a linkage.

A polymorphic antigen displaying different molecular weights in sheep and cattle sera.

The polymorphic antigen B1 is present, exhibiting different molecular weights, in both sheep and cattle sera. The molecular weight of the antigen, purified from both sera by a specific immunoabsorbent, was analysed by SDS polyacrylamide gel electrophoresis and calibrated gel filtration. Results indicate that only one subunit, 35 000 MW, forms cattle B1 while a further subunit, 60 000 MW, is present in sheep B1. The antigen activity is localized on the 35 000 MW subunit, which appears to be the same in both species.

An Amphioxus lanceolatus agglutinating factor for human red cells.

Humoral proteins from Amphioxus lanceolatus showed a considerable agglutinating activity for human red cells. The haemagglutinating factor eluted in the macroglobulin peak on Sephadex G200 and resulted ConA-1 at affinity chromatography on Sepharose ConA. Immunoelectrophoresis, carried out using a specific antiserum produced in rabbit, revealed alpha mobility for the agglutinating factor, thus suggesting that it was an alpha macroglobulin.
