Histamine uptake by leukocytes in vitro.

Human leukocytes at the concentration of 10(6) cells/ml were incubated for 1h at 37.5 degrees C in a tris-albumin buffer, containing 0, 6.25, 12.5, 25, 50, and 100 micrograms/ml of histamine. Histamine uptake was measured by a photofluorometric method. We found that the leukocytes take up exogenous histamine and attain respective intracellular concentration of 307%, 423%, 438%, 1000% and 645% above controls. It was found that neutrophils and eosinophils possess the greatest capacity to take up exogenous histamine whereas, contrary to the expectation, basophils display poor capacity.

[Mastocytes of the parietal pericardium in the guinea pig].

The present research was performed in the parietal pericardium of guinea pigs aged between 4 and 5 months. After stretching the pericardium on a slide and fixation in Mota's fluid, the following techniques were utilized to study the distribution and the cytochemical characteristics of mastocytes: 1) staining with Toluidine blue or Methylene blue 0,5%; 2) staining with Toluidine blue 10(-4)M in buffered solution at different pH (2,3,4,5,6,7); 3) staining with Alcian blue 8GS pH 1 and pH 2,6. Prior to 1,2,3 a digestion with Ch-ase ABC was performed. The pericardium was also exposed to ortophtaldialdehyde and paraformaldehyde vapours to show the presence of histamine and 5-HT. From our data it results that mastocytes of the parietal pericardium of guinea pig always show an extremely elongated shape. Their granular content is characterized by the presence of condroitinsulphate and ialuronic acid. By the OPT reaction a blue fluorescence becomes evident, showing low histamine levels in mastocytes, while the paraformaldehyde reaction results to be always negative.

Thyroxine and triiodothyronine levels in thyroid vein blood and in thyroid tissue of patients with autonomous adenomas.

Thyroxine (T4) and triiodothyronine (T3) concentrations were measured in peripheral and thyroid vein blood and in nodular and extranodular thyroid tissue from twenty-four patients with autonomous thyroid nodules (AFTN); fifteen of these patients showed clinical signs of hyperthyroidism and nine were euthyroid. Thirteen patients with solitary non-functioning thyroid adenomas who were clinically euthyroid, served as controls; samples of thyroid vein blood and normal thyroid tissue being obtained from the contralateral lobe. T4 (189.4 +/- 27.2 nmol/l) and T3 (7.05 +/- 2.03 nmol/l) concentrations were significantly higher in the thyroid vein blood of patients with AFTN compared with controls (T4 = 119 +/- 9.1 nmol/l, P less than 0.05; T3 = 2.3 +/- 0.21 nmol/l, P less than 0.01) whereas peripheral levels in the two groups were similar. The T3 concentrations (10.56 +/- 4.12 nmol/g wet tissue) in autonomously-functioning thyroid nodular tissue were significantly higher than those of extranodular (1.9 +/- 0.62 nmol/g wet tissue, P less than 0.01) and normal thyroid tissue (2.63 +/- 0.43 nmol/g wet tissue, P less than 0.05). The T4 levels were not different in the tissues examined. The concentrations of T4 and T3 in thyroid vein blood did not show any significant correlation with the hormone levels in thyroid tissue.

[Role of the mastocytes during functional activation of the thyroid].

The behaviour of biogenic amines in mastocytes has been studied in the thyroid glands of rats treated with 2 U.I. of TSH. The histochemical evaluation of histamine and 5-HT in mastocytes with the method of fluorochromization has been carried out at 20 minutes and 120 minutes after TSH administration. After 20 minutes, the thyroid glands do not present evident parenchymal changes, but the positivity for biogenic amines in mastocytes decreases in comparison to the controls, while this positivity begins to be evident in the interstitial tissue. After 120 minutes, the thyroid gland shows a microfollicular arrangement and the fluorescent biogenic amines appear to be released in the interstitium by degranulating mastocytes, nearly entirely negative to fluorochromization.

[Some photometric and fluorimetric characteristics of acridine orange and of its complexes with glycosaminoglycans: proposed method for their spectrophotofluorimetric evaluation. I. Spectrophotometric study].

In spite of the wide use of AO as basic fluorochrome in cytology, histology and histochemistry, however there still persist some queries on the meaning of fluorescence due to the encounter of a such molecule with anionic polymers. From the spectrophotometric data exposed in the present paper, AO is stressed as a metachromatic colouring and therefore, as such, may be used in histochemical investigation. Besides, because of the reliable identificaton of the single types of GAG, screening may be performed at different pH, since metachromasia appears at different pH for the three different types of GAG. A useful counterproof may be performed by means of selective elutions of the AO/GAG complexes, being solutions with increasing ionic power.

[Some photometric and fluorimetric characteristics of acridine orange and its complexes with glycosaminoglycans: proposed method for their spectrophotofluorimetric evaluation. 2. Spectrophotofluorimetric study].

From the present spectrofluorimetric investigation of AO and its complexes with anionic polymers, derives the possibility of a direct quantitative evaluation of GAG as such, and not through the dosage of one of their molecular components. On the other hand, the method proposed is characterized by its simplicity and rapidity. Besides, the noticeable extent of the range in which there exists a correlation between the response intensity at =640 nm and the doses of GAG considered should be stressed.

Cytochemical detection of histamine in the human granulopoietic series of healthy subjects and of patients affected by chronic myeloid leukaemia. A spectrophotofluorimetric test for checking OPT-induced fluorescence in isolated cells.

Cellular histamine in blood and bone marrow has been identified histochemically using an o-phthaldialdehyde fluorescence reaction. The specificity of the reaction was tested by a spectrofluorometric analysis of cell extracts. In normal blood, the basophils emit a bright yellow fluorescence, whereas neutrophils, eosinophils and platelets react less consistently and when they do, they give off a less intense yellow or blue emission. In normal marrow, basophils react strongly whereas the metamyelocytes and later granular cells show only a weak yellow or blue fluorescence. In chronic myeloid leukaemia, cells of the granular series emit a strong yellow fluorescence at all stages of development, although still less intense than the basophils. During remission, the fluorescence pattern of cells from leukaemic subjects reverts to that of normal cells.

Lectin histochemistry for detection of the oligosaccharides in the testis of the chick embryo.

The oligosaccharidic content, distribution and changes of the glycoconjugates in the testis of the chick embryo, from the 8th day of incubation to hatching, were studied using a battery of seven HRP-conjugated lectins (DBA, SBA, PNA, ConA, WGA, LTA, and UEA I). Our findings showed that ConA and WGA appeared to characterize the spermatogonia during their differentiation, maturation, migration and meiosis. In the Sertoli cells, a change of localization of staining with ConA and WGA was revealed during the differentiation and maturation of these cells. The basal membrane was characterized by the reactivity with ConA and WGA from the early stages of incubation. ConA, WGA and PNA reacted with the endothelial cells of the testis for the whole period of incubation considered. Moreover, the interstitial cells, since their appearance, showed reactivity with ConA, WGA and PNA at the plasma membrane and the cytoplasm.

Lectin histochemistry of enterocytes sugar residues in the gut of the chick embryo and of the newborn.

The glycoconjugates sugar residues content, distribution and changes in the enterocytes of different tracts of the developing intestine of the chick embryo and of 1-day-old chick were investigated, using a battery of seven HRP-conjugated lectins (DBA, SBA, PNA, WGA, ConA, LTA and UEAI). The results of the present research have shown the presence of a large amount of glycoconjugates sugar residues in the enterocytes of duodenal, ileal and colonic anlage, starting from the beginning of the second week of incubation. Differences were detected among the three investigated intestinal segments, as to the time of appearance of the glycoconjugates sugar residues in the enterocytes. The duodenal enterocytes showed the most precocious appearance of lectin-reactive material, followed by the ileal enterocytes and afterwards by colonic enterocytes. The duodenal enterocytes were characterised by the presence of SBA binding sites, which were not detectable in the duodenal enterocytes of the adult animal.
