[Chronic pulmonary cryptococcosis. A case contribution for diagnosis of cryptococcosis in man from a pathologic-anatomic viewpoint].

Routine use of differential nutrients for isolating cryptococcus neoformans (Staib-Agar) during post-mortem examination revealed previously unsuspected chronic pulmonary cryptococcosis in a 64-year-old woman. Cause of recurrent pleural effusions over three years were probably sub-pleural cryptococcomas. In on-target and specific search for the cryptococcosis pathogen, the lung deserves special attention as a probable gateway.

[The effect of substance P on the regeneration of nerve fibres in vitro (author's transl)].

Explants of the ganglion trigeminale from chick embryos (PNS) and of the hippocampus from fetal rats (CNS) were cultivated in maximow chambers in the presence of synthetic substance P (SP). SP Stimulated the outgrowth of neuronal processes in PNS-culture. In SP-treated CNS cultures a stimulation was not observed. The density of SP-treated medio-dorsal neuroblasts decreased. The possible role of SP as a system in controlling in-vitro-processes of nerve fibre regeneration is discussed.

[Effect of substance P on the ganglion trigeminale in tissue culture (author's transl)].

Explants of the ganglion trigeminale from chick embryos were cultivated in Maximow chambers in the presence of 10-6...10-8 M substance P (SP . 3CH3COOH.4H2O). 1. In SP-treated cultures the index of areas covered by the explants was increased in shorttime tests. 2. The density of cells was related to the type of medio-dorsal (MD) and ventro-lateral (VL) neuroblasts. The density of SP-treated VL cells was not altered. The density of MD cells decreased. 3. The percentage of dark neuroblasts was decreased under the influence of SP. 4. A stimulation of VL neuroblasts did not take place. 5. The diameters of MD pericarya and the areas of MD cell nuclei and the areas of nuclei from nonneuronal cells increased. 6. The possible role of SP as a factor controlling In-vitro-processes is discussed.

[Electron microscopic investigations about the influence of the cyclic N6-monobutyryladenosine-3':5'-monophosphate (mb-cAMP) on the neuron differentiation in the explantate cultures of the hippocampus (author's transl)].

1. The influence of the cyclic N6-monobutyryladenosine-3':5'-monophosphate (mb-cAMP) on the neurogenesis was investigated by means of electron microscopic and morphometric methods at the explantate cultures of the fetal hippocampus of the rats in vitro. 2. After three days of the in vitro cultivation the neuroblasts of the treated cultures show not significantly changes of the total number of ribosomes in comparison to the controls. 3. After 7 and 14 days of the in vitro cultivation the neurogenesis is significantly stimulated under the influence of the mb-cAMP. The drug produces a significant increase of the free ribosomes and polysomes in company with a higher degree of differentiation of the granular endoplasmatic reticulum. Under the influence of the mb-cAMP the total number of ribosomes significantly increases by 32% (after 7 days) respectively 20% (after 14 days). 4. The results might suggest, that mb-cAMP might stimulate the protein synthesis in connection with an increased nerve fibre growth in vitro.

[The development of nervous tissue to experimental manipulation in vitro (author's transl)].

Explants of nervous tissue (CNS: hippocampus from fetal rats, telencephalon and PNS: ganglion trigeminale from chick embryos) were cultivated in maximow chambers. Biological extracts (brain and embryo extract) or chemical agents (aminoacid mixtures and peptides with known sequence of amino acids) were tested. Biological extracts and aminoacid mixtures stimulated the differentiation in PNS and CNS cultures. A stimulating effect of peptides seems to exist only on PNS explants. The fundamental importance of suitable reference systems, parameters, optimal concentrations and periods of application of effective substances for the evaluation of the results is discussed.

[Effect of substance P on nerve fiber regeneration in tissue culture].

Explants of the ganglion trigeminale from chick embryos (PNS) and of the hippocampus from fetal rats (CNS) were cultivated in maximow chambers with growth medium or maintanance medium. Varied concentrations of substance P (SP . 3 CH3COOH . 4 H2O) were added. 1. The effect of substance P (SP) is related to concentration. In the presence of 10(-7)M SP in the growth medium and of 10(-4)M SP in the maintanance medium the cultivation of PNS cultures indicates positive results. These doses are suitable. 2. Within the first 24 hours in vitro SP stimulates the index of area in PNS cultures. The index of characterizes the relation of the outgrowth zone to the explant. In CNS cultures a significant difference of this effect was not observed. 3. The index of growth of nerve fibers may compare the test cultures with the control cultures. SP significantly increases the index of fiber growth in PNS cultures. A stimulation of CNS cultures was observed, significance was not found. 4. From the beginning of the cultivation with SP up to 48 hours in vitro the growth of nerve fibers significantly increases in the treated cultures in comparison with the control cultures. After this time the growth of nerve fibers decreased and a morphological conformity of test cultures and controls was observed. 5. The role of SP is discussed in specific activity on PNS tissue in vitro. The reactive neurons may be from the medio dorsal group of cells of the sensible ganglion.

[The effect of active substances on nerve fiber regeneration in nerve tissue cultures].

Explants of the ganglion trigeminale from chick embryos and hippocampi from embryonal rats were incubated in maximowchambers with semisynthetic media. The different parts of nervous tissue were influenced experimentally by addition of biological extracts and by substances with known composition. The regeneration of nerve fibers was investigated by the index of growth. The growth index was calculated from the ratio of nerve fibre index of the test cultures to that of the influenced cultures. Under these conditions biological extracts enhanced the growth of nerve fibres. In the same way the growth of nerve fibres was statistically significantly stimulated by substances with known composition of aminoacids, orotic acid, sodium orotate and cyclic monophosphates. A stimulating effect of cyclic guanosinmonophosphate seems to exist only in CNS explants and only in young fetal rats and consists in an increased migration and proliferation of cells as well as in the formation of fibres from neuroblasts. The investigations gave strong evidence for the in vitro testing to be very useful in studies of nerve fibre regeneration. However the choice of suitable reference systems, suitable quantitative parameters, optimal concentration and periods of application of effective substances and the age of the animals are of fundamental importance for the evaluation of the results. Experiments regarding the stimulation of the differentiation of neurons and the growth of nerve fibres are of practical clinical and therapeutical interest.

Pathogenic yeast-like fungi in meat products.

100 samples of sausage and ham (cut in fine slices) were examined to find out whether meat products can harbour yeast-like fungi pathogenic for man. Pieces of the test material were placed in a sterile Petri dish containing 1 ml distilled water and incubated at 26 degrees C for 2 days. The colonies grown on the material were identified by international standard methods, tested for extracellular proteolytic activity and serotypes. Experimentally infected meat products were studied culturally and histologically (PAS stain). 14 out of the 100 samples examined were found to contain Candida parapsilosis and 1 C. tropicalis. In vitro, extracellular proteolytic activity was found in 54.5% of the C. parapsilosis isolates. By serotyping, 2 of the isolates wree found to be related with serotype A and 8 with serotype B of C. albicans. The strains isolated from the blood of 27 patients, in part of them, identity of serotype and proteolysing activity was established. In the experimentally infected meat products, C. albicans showed a scarce growth preferably in the pseudomycelium form. C. tropicalis grew very well on the boiled sausage of salami type but mostly in the pseudomycelium form. C. parapsilosis on the other hand, showed a strong growth preferably as round blastospores on all 3 meat products and Cryptococcus neoformans a moderate growth as round blastospores in all the 3 meat products (Bologna type sausage, boiled sausage of salami type and smoke ham). The findings are discussed with a view of their implications for Medical Mycology, epidemiology and the practice of food control.

[The effect of orotic acid and sodium orotate on organ cultures of the hippocampus of embryonic rats].

Organ cultures of hippocampus from fetal rats were cultivated in Maximow chambers with semisynthetic media up to 12 days in vitro and fixed after 1, 5 and 12 days in culture, cuted 15 micrometer and coloured with the method of Klüver-Barrera. The effect of 5 x 10(-6) M sodiumorotate and 6 x 10(-6) M orotic acid was tested. 1. The density of cells is related to the zone of cells, the drug and time of incubation. 2. The density of cells increased in the zone of matrix after application of sodiumorotate and orotic acid. The density of cells significantly decreased in the migration zone. The density of neurons significantly increased in the region CA 3. 3. Orotic acid and sodiumorotate influenced the migration of cells in vitro. Sodiumorotate stimulated migration and differentiation. Longtime cultured hippocampi degenerated in vitro after influence of orotic acid. 4. The area of nuclei from neurons increased up to 12 days in vitro, when the cultures were tested with sodiumorotate. Only within the first 24 hours orotic acid effected an increase of the area of nuclei from matrix cells. It is discussed, sodiumorotate influenced in vitro metabolic processes in suitable systems.

[Localised aspergillosis and oxalosis of the lung caused by aspergillus niger: soil of ornamental plants as a reservoir of aspergilli (case report) (author's transl)].

Haemoptysis occurred in a 64-year-old man with a history of pulmonary tuberculosis, negative for M. tuberculosis. Precipitating antibodies to Aspergillus niger were found in serum and the fungus found in sputum. Autopsy revealed aspergilloma, colonization of the pulmonary cavity by A. niger and local oxalosis if the cavity wall. Ornamental plant soil containing A. niger conidia, as well as the air in the patient's living-room, was the probable source of infection.

[The effect of a synthetic tripeptide nervous tissue cultured in vitro].

Explants from chick embryo PNS (ganglion trigeminale) and from CNS of embryonal rats (hippocampus) and dissociated cells from chick embryo cerebral hemispheres were cultivated in maximow chambers in the presence of various concentrations of placental serum and of a chemically synthesized tripeptide Gly-His-Lys. 1. The presence of tripeptide in the nutrient medium with a low concentration of serum did not compensate the outgrowth of nerve fibers, that take place in the growth medium. 2. In the presence of tripeptide in the nutrient medium with low concentration of serum the index of growth area increased significantly. 3. Within the first days in cell cultures 0,01 microgram tripeptide pro ml medium stimulated the outgrowth of neuronal processes. 4. The experiments indicated, that the tripeptide did not replace the serum. The possible role of tripeptide as a system in controlling neuron-glial ratio in vitro is discussed.

[Orbital aspergillosis--a contribution to its diagnosis and treatment (author's transl)].

In connection with a case report, the mycological diagnosis and treatment of orbital aspergillosis are reviewed. In a 59-year-old male patient an exophthalmus on the right side could be diagnosed as aspergillosis spreading from the corresponding sinus maxillaris into retro- and peri-orbicular regions, after other causes had been excluded by differential diagnosis. In a biopsy specimen from the retrobulbar tumor, a fungal granuloma was found which, histologically and by isolation of the fungus, could be identified as an infection by A. fumigatus. Because of the good visus of the right eye, no exenteratio orbitae but an enoral revision of the sinus maxillaris, ethmoidal bone, and bottom of orbita was performed.--By infusions of amphotericin B and local washings with pimaricin (natamycin) in the sinus maxillaris and the region of the operation, a complete healing of the mycotic process was achieved.--Since 1 1/2 years, the patient has not had a relaps. This success of therapy was possible by exemplary cooperation of otorhinolaryngologists, ophthalmologists, pathologists and microbiologists.--In view of the prophylaxis of such infections, recent contributions to the epidemiology of aspergillosis are discussed.

[The effect of Solcoseryl on explant cultures of the hippocampus].

Explants of hippocampus from fetal rats were cultivated in Maximow chambers in semisynthetic medium up to 12 days in vitro. The cultures were fixed Bouin, slided 15 micron, coloured with Klüver-Barrera and some morphological parameters were tested. 1. The nerve fiber index increased by influence of 1% Solcoseryl in relation to control cultures, which growed in minimal medium. An essential stimulation was observed by application of placentar serum and embryonal extract into the culture medium. 2. Die decrease of the number of neurons and glial cells per unit of area and a small decrease of the area of neuron nuclei was discussed in relation to the effect of the pharmacon Solcoseryl on O2- consumption. 3. Solcoseryl (firm Solco AG, Base) is an extract of calf blood. It can not substitute other tissue extracts.

[Differentiation of neurons of the cingulate region in explant culture].

The structural changes in explant-cultures of the cingulate region from the brains of 18 day old rat embryos were investigated during several cultivation periods by neurohistological light microscopical methods. The differentiation and the growth of neurons were observed in cultures stained by the method of Klüver-Barrera, and cut perpendicular to the coverslips during the period from the 1st up to the 20th day in vitro. As parameters for the growth of the cells the density of neurons and glial cells per culture area were estimated and the diameters of nuclei and pericarya were measured at different stages of culture. The following results could be obtained. 1. The immature cells differentiated up to the 20th day in vitro to neurons that could be devided into three groups due to histological characteristics: pyramidal cells, multiform neurons and a group of small cells with few cytoplasm. 2. From the 1st to the 20th day in vitro the density of neurons and glial cells markedly decreased. The number of neurons per unit area decreased to 59.7% during this period. However, the most rapid decrease in the number of neurons occurred from the 10th to the 20th day in vitro. 3. The measurements of the diameters showed an increase in both the diameters of nucleus and pericarya from the 1st to the 20th day in vitro. The enlargement of the diameters is especially striking from the 1st up to the 10th day in vitro. The results are discussed in comparison results of ontogenetic investigations in vivo.
