[Effect of opiate receptor agonists on the course of hemorrhagic shock in rats].

The experiments have been performed on 93 male rats, weighing 200-250 g. In acute blood loss various arterial pressure (AP) changes have been demonstrated--the marked hypertension is being changed by gradual AP increase. The injection of m-receptors' agonist DAGO prevents systolic and diastolic AP increase, agonist DADL prevents diastolic AP increase in acute momentary blood loss. In gradual blood loss DAGO (more than DADL) slows down both the decrease and the subsequent AP increase in rats. DAGO is determined to decrease, and DADL--to increase the minute blood volume. The mechanisms of opioids' action and their significance in pathogenesis of hemodynamic disturbances in shock are being discussed.

[Use of synthetic analogs of enkephalins as antiatherogenic means].

The experiments have been performed on 70 male rats weighing 200-250 gr., 53 patients with obliterating atherosclerosis of lower extremities have been examined. Leu-enkephalin analogues (dalargin and DEFN) administration in a dose of 0.1 mg/kg has been determined to decrease cholesterol levels in the blood in acute and chronic experimental hypercholesterolemia. The use of pharmacological preparation -- enkephalin analogue of dalargin in complex treatment of the obliterating atherosclerosis of lower extremities leads to cholesterol concentration decrease, parathormone concentration decrease in the blood of patients and hyperlactatemia decrease in venous blood of the damaged extremity. Leu-enkephalin analogues, action mechanisms and the possibility of their use for atherosclerosis treatment is discussed.

Parathyroid gland function and the possibility of its correction in patients with obliterative vascular disease of the lower limbs.

Parathyroid gland function and the possibility of its correction were studied in 43 patients with obliterative vascular disease of the lower limbs, and in 24 healthy, mostly male, controls. Parathormone (PTH) concentrations in blood plasma samples were determined by radioimmunoassay. Patients with obliterative vascular diseases of the lower limbs were found to have increased activity of parathyroid gland. The role played by hyperparathyroidism in the pathogenesis of obliterative vascular diseases of the lower limbs as well as the possibility of its correction by betablocker and encephalin analogue administration is discussed.

[A simple and informative method of diagnosing myocardial infarct, based on a determination of blood density].

An increased density of blood revealed in myocardial infarction patients served the basis for a simple and highly informative method developed by the authors to early diagnose myocardial infarction when other methods are not sensitive. The high density of blood is registered upon placing a drop of blood into flakes with sodium chloride solution of different concentration.

Effect of intravenous enkephalin administration on kallikrein-kinin system in experimental hemorrhagic shock. Evidence for activation of kallikrein-kinin system by naloxone.

The effects of enkephalin derivates with different opioid receptor subtype specificity and naloxone on cardiovascular responses and kallikrein-kinin system (KKS) were studied in anesthetized rats exposed to 30% hemorrhage. Administration of a mu-receptor agonist (DAGO) in early hemorrhage improved mean arterial blood pressure (MAP) responses to hemorrhage. This effect could be abolished by naloxone pretreatment. Moreover, a delayed MAP recovery after hemorrhage could be observed. Treatment with a delta-agonist (DADL) resulted in transient depression of MAP and heart rate (HR). Hemorrhage by itself caused only a slight activation of KKS as indicated by decreased plasma kallikreinogen concentration and reduced kallikrein inhibitor capacity after 20% blood loss. Enkephalin administration did not exert significant effects on KKS. Naloxone pretreatment, in contrast, induced prehemorrhagic activation of KKS, which was potentiated by subsequent hemorrhage. Naloxone-induced activation of KKS could be confirmed by an in vitro study. Taken together these results suggest that the KKS is not involved in MAP and HR responses to enkephalin administration during hemorrhage, whereas it might be implicated in naloxone-induced delayed posthemorrhagic MAP recovery.

[Participation of leu-enkephalin in regulating carbohydrate metabolism].

The experiments have been performed on 166 white male rats with a mass of 180-220 g. It has been revealed that leucine-enkephalin and its synthetic analogs prevent an increase in glucose blood levels and a decrease in glycogen liver levels caused by adrenaline and parathyroid hormone. At the same time the enkephalins inhibit the secretory activity of pancreatic beta-cells. The mechanisms of opioid peptide effect on carbohydrate metabolism are discussed.

[Effect of enkephalins on the energy potential of the rat liver and cerebral cortex during an early period of traumatic shock].

The experiments were performed on 86 white male rats, weighing 180-220 g. The animals with traumatic shock revealed a decrease in ATP concentration and a rise in ADP and AMP levels in the brain cortex and hepatic tissue, resulting in the energy charge reduction. The injection of synthetic leu-enkephalin analog D-ala2-arg6-leu-enkephalin to animals with traumatic shock led to energy charge normalization in the brain cortex and increase in hepatic tissue. The mechanisms of enkephalin effect on the energy metabolism during shock are discussed.

[Leu-enkephalin levels in the blood of patients with ischemic heart disease].

A total of 32 CHD patients with angina pectoris of effort, functional class I-IV, aged 46 to 68 were examined. Eight patients with cervical osteochondrosis of the same age were controls. In CHD patients with angina pectoris, functional class I-II, the content of leu-enkephalin in the blood plasma did not differ from that in the control group whereas in patients with angina pectoris, functional class II-IV, it was lowered by 42%. In CHD patients aged 46 to 55 the level of leu-enkephalin in the blood plasma was raised, in the group of patients aged 55 to 68 it was considerably lowered.

[Effect of enkephalins on the function of the calcium-regulating endocrine glands in shock].

The experiments on white rats weighing 180-220 g have shown that in traumatic and hemorrhagic shock the initial increase in parathyroid hormone blood concentration is followed by the decrease of functional activity of parathyroid glands. Calcitonin concentration is found to increase during the first hours of shock. The changes in calcium-regulating gland function result in significant disturbances of calcium exchange during shock. The injection of synthetic leu-enkephalin analogs to rats with shock leads to normalization of calcium-regulating endocrine glands function.
