[Effects of tissue-type plasminogen activator from a culture of calf kidney cells on hemostasis and fibrinolysis in experimental nephritis].

The plasminogen activator 960 IU/mg protein activity isolated from cultured fluid of the calf kidney cells was introduced to albino rats (180-200 g) with experimental Heynmann nephritis every day during 4 days. Nephritis caused activation of haemostasis and inhibition of fibrinolysis in the blood. There was increased excretion of the fibrin, fibrinogen degradation products in urine as a results of the local fibrin deposition in diseased kidneys. The fibrinolytic activity of the cortical zone of kidney was markedly decreased. The plasminogen activator, infused to experimental animals, resulted in normalization of the altered indexes.

[Initial-onset stenocardia: characteristics of the blood coagulation system and fibrinolysis as reaction to physical exertion].

Blood coagulation and fibrinolysis were studied at rest and during bicycle ergometry in 45 patients with angina of new onset (ANO). Fourteen chronic coronary patients and 20 subjects, free of coronary heart disease or coronary atherosclerosis, were taken as controls. Fibrinolysis tended to be depressed in resting ANO patients. Potential fibrinolysis depended on the clinical pattern of the disease and was particularly depressed in patients with severe ANO. Stress did not activate fibrinolysis in patients with vasospastic angina and high basal plasminogen activator level at rest.

[Depression of fibrinolysis in long-term stress in the rat].

A study of fibrinolytic and hemostatic effects of emotional/painful stress induced by Dosiderato's technique in white Wistar rats demonstrated, 4 hours after discontinuation of 6-hour stress exposure, a thrombophilic condition that is characterized by heavily depressed fibrinolysis in combination with increased blood antiplasmin and coagulation activities. It is suggested that depressed fibrinolysis is a result of depletion of fibrinolytic elements, plasminogen activator in particular.

[Characteristics of hemostasis, fibrinolysis and lipid indicators in children and their fathers with a history of myocardial infarction at a young age].

Blood coagulation and fibrinolysis characteristics are described in children whose fathers survived myocardial infarction when fairly young. Hemostatic and fibrinolytic effects of heredity are analysed, as are lipid spectrum disorders. Children with aggravated heredity showed changes in plasma fibrinogen concentration, antithrombin III, total plasma fibrinolytic activity recalcification time. The monitoring of children with aggravated heredity for coronary heart disease, particularly those with attendant dyslipoproteinemia as a specific high-risk group, is proposed.

[Development of research into the physiology and pathophysiology of the fibrinolysis system].

A modern data review on the importance of fibrinolysis system is given. A considerable success has been scored during the study of molecular parameters of fibrinolysis system: the plasminogen, plasmin, its inhibitors, plasminogen activators and the mechanism of activation system have been characterized. The entrance of A, K, C, P and PP vitamins has been established to be necessary for the normal functioning of the fibrinolysis system; the dependence of the blood fibrinolytic activity upon the initial plasminogen content and concentration of its activators in blood has been revealed. The plasminogen activator depletion in tissues has been shown to be one of the reasons of some pathological states development, especially at cardiovascular diseases. The increase of fibrinolysis level by the active fibrinolytic ferment injection in blood has a medical effect at thrombosis. The ferment fibrinolysin received in the laboratory is successfully used in clinical practice. Some other activators of fibrinolytic system: tricholysine and longolytin from the culture of saprophyte fungi, plasminogen activator from the pig heart and the cells culture of the calf kidney have been received and are being studied.

[Effect of the enzyme preparation longolytin on fibrinolysis in animals].

It has been established that fibrinolytically active enzyme longolytine isolated from the culture fluid of the saprophyte fungus Arthrobotrys longa at intravenous injection favours the prolonged increase of the plasma fibrinolytic properties as well as activation of endogenic plasminogen. Maximum values of fibrinolytic activity have been marked in 5 and 30 min after enzyme intravenous injection. The plasminogen activity is high in 120 min. The fibrinolysis indexes--fibrinolytic activity of the euglobulin fraction and amount of plasminogen activator--in 3,5 and 5 times higher than in vitro at intravenous injection. The activation of coagulation system does not occur.

[Fibrinolytic activity of heart tissues and aorta of rats undergoing electroshock].

In the first minute of shock caused by electric current passing through the rats parietal lobe the increase of fibrinolytic activity of euglobulin fraction of blood occurs. During the investigation of the right heart ventriculus tissue and aorta by histochemical method of Todd in our modification the decrease of fibrinolytic activity in wall of aorta (on 80%) and in tissue of heart ventriculus (on 30%) has been revealed in tentative animals as compared with control ones. It is suggested that the entering of plasminogen activator from vessel wall into the blood flow plays an important role in activation of fibrinolytic system in generalized reaction of organism to strong stress action.

[Participation of the kidneys in the reaction of the anticoagulation system to thrombin administered into the blood stream].

Thrombin injection into white rats jugular vein in dose 20 units on 200 g of body mass results in more considerable release of plasminogen activator in blood of renal vein as compared with blood of cava inferior and jugular veins. In this case the fibrinolytic activity of kidney cortical part decreases, but in medullar part it does not alter. The decrease of antiplasmin content in blood of renal vein is due to alpha 2-macroglobulins, in blood of jugular and cava inferior veins--to alpha 1-antithrypsin.

[Changes in the components of the animal fibrinolytic system induced by the action of physiological activators and enzymes of fungal origin].

The rat fibrinolytic system components by means of intravenous injection of physiological activators (tissue plasminogen activator, urokinase) and of enzymes isolated from the Fungi imeprfecti cultures (tricholysin, longolytin) have been investigated. It has been demonstrated that the tissue plasminogen activator and urokinase exert a similar effect on fibrinolysis by inducing a short-term increase of the euglobulin fraction fibrinolytic activity but do not practically change other components of the fibrinolytic system including the antiplasmin content. Tricholysin and longolytin produce a stronger action on the fibrinolytic system by significant affecting almost all the parameters, especially the antiplasmin content. The results obtained show that physiological activators as compared with fungal enzymes possess with better thrombolytic properties.
