[Role of the initial level of plasminogen activator in changes in fibrinolysis].

Fibrinolytic activity of blood in rats depended on initial level of plasminogen activator. Physical exercises, tissue plasminogen activator and fungii protease (longolytin) infusions, acetylsalicylic acid and brufen significantly increased content of the plasminogen activator with low initial level and had no such effect if the initial level of plasminogen activator was high enough.

Plasminogen activator content in heart tissue and perfusate under various experimental conditions.

In order to elucidate the mechanisms of plasminogen activator release into the circulation during the first minutes of experimental myocardial ischaemia in vitro, isolated rat hearts were perfused under optimal conditions and under conditions inducing myocardial ischaemia. In ventricular tissues the content of plasminogen a activator was measured; in the perfusate the plasminogen activator and anticoagulating activities were determined. Under optimal conditions the plasminogen activator content in contracting heart was found to be essentially the same as under in vivo conditions; in the perfusates the release of plasminogen activator and of substances possessing the anticoagulating activity was insignificant. Under myocardial ischaemia the release of these substances into the circulation increased, while plasminogen activator content decreased.

[Effect of high doses of tissue plasminogen activator on the hemostatic and fibrinolytic systems].

Intravenous administration of tissue activator plasminogen (TAP), isolated from a frozen pig heart using the technique of Bachman et al., to white rats produced considerable fibrinolysis activation in the euglobulin fraction only. The clot lysis time of this fraction was reduced 2.4-fold within 10 minutes after TAP administration, and high levels of the activator were maintained for 60 minutes. The absence of changes in the fibrinolytic activity of undiluted plasma might be due to TAP binding with antiactivator as the latter's level dropped after 10 minutes. Successive administration of heparin after a therapeutic dose of TAP resulted in decreased heparin activity and inhibited TAP which may be due to the formation of a complex between the two whose specific activity is not manifested in the presence of excessive heparin.

[Plasminogen activator from a kidney cell culture].

Plasminogen activator (PA) isolated from kidney's cells culture (doses 10 to 5000 IE/kg) induced lysis of experimental thrombi. Lysis time decreased from 214 to 23 min after PA infusion in doses 10 to 700 IE/kg, further doses decrease from 700 to 5000IE/kg does not accelerate lysis time of thrombi. PA infusion in doses mentioned increases the activator activity of plasma and euglobulin lysis time, but does not change the fibrinolytic activity, antiplasmin content and fibrinogen concentration.

Changes of the fibrinolytic system of animals induced by injection of tissue plasminogen activator.

The changes in the fibrinolytic system of rats induced by repeated injections of tissue plasminogen activator (TPA) have been studied. The euglobulin fraction fibrinolytic activity increases 10 min after the first injection of TPA. Repeated injections of TPA for one or more days do not stimulate the fibrinolytic activity or TPA accumulation in the blood. Apparently in the absence of fibrin clots in the blood flow of healthy animals TPA is incapable of activating plasminogen conversion into plasmin. TPA, in its turn, is rapidly bound to the antiactivator and is excreted from the organism.

[Seasonal changes in the blood coagulating and anticoagulating system indices in men at the preclinical stage of ischemic heart disease].

Examination of 52 males (aged 23 to 40 years) in the preclinical stage of ischemic heart disease revealed seasonal differences in the values of the blood coagulation and anticoagulation systems: in the spring, there was an increase in blood coagulation activity displayed by growth of the concentration of fibrinogen and soluble fibrin and a reduction in the amount of the plasminogen activator. The authors suggest conducting preventive treatment of patients in the spring, the most unfavourable season in respect of the effect of the pathogenetic factors.

[State of the fibrinolytic and kinin systems of the blood after physical loading].

A moderate physical load (a dosed running in a treadban) activates both the fibrinolytic and the kinin blood systems in rats: plasminogen increases for over 30 min whereas the kinin system activation is shorter. Repeated physical exercises exhaust the plasminogen activator in tissues. Response of the fibrinolytic system depends on its clinical state. Only under conditions of decreased fibrinolytic activity the discharge of a large amount of the plasminogen activator into the blood stream occurs.
