[Testicular tumor in Mongolian men (report of 35 cases)].

OBJECTIVE: To improve the diagnosis, therapy and prognosis of testicular tumor in Mongolian men. METHODS: A retrospective review of 35 cases of testicular tumors in Mongolian men from seven medical centers dated from 1990 to 2004 was performed. RESULTS: The usual presentation of a testicular tumor was a nodular or painless swelling of one gonad. The mean delay in diagnosis was 40.03 +/- 53.45 weeks. For 16 patients, delay in diagnosis was more than or equal to six months. The histologic composition of this series was 21 (60%) seminoma, 10 (28.6%) nonseminoma, 2 (5.7%) lymphoma, 1 (2.35%) fibroneuroma and 1 (2.35%) leiomyoma. Regarding stage, 22, 2, and 5 of 29 germ cell tumors were seen initially as stage I, II, and III, respectively. Combined therapy, including radical orchiectomy, radiotherapy and chemotherapy, were taken. 29 cases have been followed for 2 months to 10 years, 4 out of them died of distant metastasis, one died of other disease, one lives with tumor, the others live without relapse and metastasis. Three and 5-year survival rates for Mongolian patients with seminoma and nonseminoma were 95.0%, 95.0%, 57.1% and 42.8%, respectively. CONCLUSION: In this article, the rate of seminoma to germ cell tumors is higher than that of general population. There is an increased mean delay in diagnosis for Mongolian patients. Three and 5-year survival rates for nonseminoma are lower than that for seminoma. Better public awareness regarding testicular tumor in this population, advances in diagnosis and therapy will help to improve therapeutic effectiveness and prognosis.

[Experimental study of cartilage reconstruction using directed inducing MSCs-alginate complex].

OBJECTIVE: To evaluate the practicability of cartilage reconstruction using directed inducing bone marrow stem cells(MSCs)- alginate complex. METHODS: The MSCs were seperated by gradient centrifugation on Percoll, then were induced into chon-drogenic differentiation. After 14 days, immunohistochemical techique was applied to detect the expression of collagen type II. Then, the MSCs and calcium alginate were mixed to form a complex and were planted under the skin of nude mice. Histochemical or immuno-histochemical tests were made at 4 weeks and 8 weeks later. RESULTS: The MSCs showed the positive result of collagen type II. There were chondrcytes and cartilage lacuna at 4 weeks later, and chondrocytes and the collagen secreted by them became more at 8 weeks later. But, the MSCs without directed inducing did not show the character of the chondrocyte. CONCLUSION: The way of using directed inducing MSCs-alginate complex can form cartilage; it is possible to repair the defect of cartilage in clinical practice.

Development and Preliminary Application of a Peptide Mass Fingerprinting Technique in Proteome Research.

Proteome means the total proteins expressed by the genome in a cell or tissue. Two-dimensional electrophoresis (2-DE) is now the most powerful separating technique and the key separation method used in proteome. Peptide mass fingerprinting (PMF) is becoming a widely used and vastly developed technique for protein identification in 2-D gels. In this research, a systematic method to identify the proteins in polyacrylamide gels by PMF was developed. Proteins were digested in-gel by enzyme and the masses of generated peptides were measured by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). The data obtained from PMF were used in protein database search and the protein identification. The proteins from human lung cancer cells isolated by 2-DE were subjected to identification by the PMF method developed in this work. Three spots of proteins in gel were identified as G3P2_HUMAN, UBL1_ HUMAN and TPIS_HUMAN.
