Formation of MTBE-DNA adducts in mice measured with accelerator mass spectrometry.

Methyl tert-butyl ether (MTBE) is a gasoline oxygenate and antiknock additive substituting for lead alkyls currently in use worldwide. Previous studies have shown that MTBE at very high doses induces tumors in rodents. The aim of the present study was to examine directly the binding ability of MTBE onto DNA, demonstrating its potential genotoxicity. MTBE-DNA adducts and their decay kinetics in mice have been measured by using doubly 14C-labeled MTBE with an advanced, ultrasensitive technique: accelerator mass spectrometry (AMS). It was found that MTBE definitely formed adducts with DNA in mouse lung, liver, and kidney in a log/log linear dose-response relationship. The distribution sequence of DNA adducts in these tissues is: lung > liver > kidney. The level of MTBE-DNA adducts peaked at 12 h postadministration in the lung and peaked at 6 h postadministration in the liver. Then the adducts declined rapidly until 5 days postadministration and thereafter declined much more slowly. To our knowledge, this is the first report on DNA adduction with MTBE in vivo. The mechanism of the formation of MTBE-DNA adducts also is discussed.

A novel method for determining the blood/gas partition coefficients of inhalation anesthetics to calculate the percentage of loss at different temperatures.

The loss by blood/gas (lambda) partition of inhalation anesthetics can be estimated by an equation for the percentage of loss. However, because lambdas of inhalation anesthetics at different temperatures have not been fully determined so far, the percentage of loss at varying temperature in various headspace volumes cannot be estimated. Therefore, a novel method was developed for the determination of inhalation anesthetic lambda, in this study. The method was precise, with a relative standard deviation of less than 5%. The average of lambda from seven distinct blood samples at 4 degrees C, 25 degrees C, and 37 degrees C were determined as 6.68, 2.04, and 1.32 of isoflurane; 3.47, 1.10, and 0.65 of sevoflurane; and 2.31, 0.75, and 0.46 of desflurane, respectively. In addition, increasing temperature was found to decrease lambda profoundly by a secondary order mechanism. Using the obtained value of lambda, the percentage of loss of isoflurane, sevoflurane, and desflurane were then predicted using a 5-mL vacuum tube as a collecting container for an example. In conclusion, a novel method was developed here for lambda determination, and lambdas of isoflurane, sevoflurane, and desflurane at various temperatures were given for estimating the loss resulting from liquid/gas partitioning.

Inhibition of nicotine-DNA adduct formation in mice by six dietary constituents.

Nicotine [3-(1-methyl-2-pyrrolidinyl)-pyridine] is a major alkaloid in tobacco products and has proven to be a potential genotoxic compound. Many natural dietary products can suppress the DNA adduction, and hence act as inhibitors of cancer. In this study, we investigated the inhibitory effects of curcumin, garlic squeeze, grapeseed extract, tea polyphenols, vitamin C, and vitamin E on nicotine-DNA adduction in vivo using an ultrasensitive method of accelerator mass spectrometry (AMS). The results demonstrated that all the dietary constituents induced marked dose-dependent decrease in nicotine-DNA adducts as compared with the control. The reduction rate reached about 50% for all agents, except garlic squeeze (40%), even at its highest dose level. Amongst the six agents, grapeseed extract exhibited the strongest inhibition to the DNA adduct formation. Therefore, we may arrive at a point that these dietary constituents are beneficial to prevent the harmful adduct formation, and thus to block the potential carcinogenesis induced by nicotine.

Mizoroki-Heck type reaction of organoboron reagents with alkenes and alkynes. A Pd(II)-catalyzed pathway with Cu(OAc)2 as an oxidant.

[reaction: see text]. In contrast to the Pd(0)-catalyzed mechanism by Uemura, Mizoroki-Heck type reaction of boronic acids is found to proceed under a Pd(II)-mediated pathway using a catalytic amount of Pd(OAc)2 in the presence of Cu(OAc)2 as an oxidant. Treatment of a variety of alkenes with boronic acids, boronates, and sodium tetraphenylborate furnishes beta-arylated and alkenylated products in good to excellent yields. The reactions with norbornene, norbornadiene, and diphenylacetylene are also performed to give 1:2 or 2:1 coupling products.

Simultaneous determination of fluorinated inhalation anesthetics in blood by gas chromatography-mass spectrometry combined with a headspace autosampler.

Although the fluorinated inhalation anesthetics, including desflurane, sevoflurane, isoflurane, enflurane, and halothane are commonly used, fatal cases resulting from their abuse or misuse have been reported. To date, gas chromatography (GC) equipped with different kinds of detectors has been utilized to analyze inhalation anesthetics. However, none of them can detect desflurane reliably or analyze all five common anesthetics simultaneously. The purpose of the present work is to further modify the previously developed headspace (HS) GC-MS method for blood isoflurane determination to analyze and distinguish five common clinical inhalation anesthetics, simultaneously. The modified HS-GC-MS method adopts a 60 m x 0.25 mm I.D., 0.25 microm film thickness DB-5 capillary column along with an adequate GC temperature program, which gives the five inhalation anesthetics, including isoflurane and its isomer, enflurane, a high resolution. The method also takes both the volatility and the influence of the top space on the obtained concentration into consideration and therefore keeps the sample loss acceptable even for analyzing the highly volatile desflurane. Within a certain concentration range of the calibration standard (about 20-300 microg/ml), this method shows a good linearity with correlation coefficients greater than 0.999. In addition, both within- and between-run precision and accuracy results meet the validation requirements as well as the tested results of practical blood samples of desflurane. In summary, this is a reliable analytical method to simultaneously determine the concentration of five common inhalation anesthetics in blood. Such a method is very practical for both clinical and occupational monitoring, as well as for analytical toxicology.

Dispersion in poroelastic systems.

We address one key source of the known discrepancies between theory and experiment in poroelasticity, i.e., the fact that Gassmann's equations for the bulk and shear moduli predict that the shear modulus is independent of the saturating fluid properties, whereas it is observed on the contrary that at high enough frequencies the shear modulus can in fact depend on the fluid's elastic properties in many porous materials. One clue to understanding this behavior comes from effective medium theory, which shows that the shear modulus does depend on the fluid properties in many circumstances. In comparison to values predicted by effective medium theory, Gassmann's equations predict different, smaller values for both the effective bulk and shear moduli of porous media. Sorting through these apparent (but not actual) disagreements among theory and theory, and theory and experiment is the main thrust of the paper.

Systemic and local secretions of cytokines and nitric oxide in massive bowel resected rats with or without small bowel segment reversal.

Our previous study suggested that small bowel segment reversal stimulates jejunal hyperplasia, but that the elevated serum interleukin (IL-)6 eliminates whole-body anabolism in massive bowel resected rats. The aim of this study was to investigate systemic and local secretions of cytokines and nitric oxide (NO) in relation to whole-body and tissue responses in rats that underwent massive bowel resection with small bowel segment reversal. Seventy percent small bowel resection was performed in Wistar rats with (REV) or without (CON) a 5 cm reversed small bowel segment. Sham operation (SHAM) was performed on a third group of rats. Twelve days after surgery the REV group had significantly lower weight gain and greater serum levels of NO, IL-2 and IL-6 than the CON and SHAM groups. The weights of the livers in the REV and CON groups were significantly heavier, but these groups had lower levels of protein, tumour-necrosis factor-alpha, IL-2 and IL-6 than the SHAM group. The cecum weights of the REV group were significantly higher with increased protein and NO levels, but decreased IL-6 levels compared with the CON and SHAM groups. In the proximal small intestine the REV group had significantly increased protein levels and mucosal dry weights, but decreased interferon-gamma and IL-2 compared with the CON and SHAM groups. Our results suggested that cytokines and NO may have endocrine and paracrine/autocrine actions in regulating whole-body and tissue responses.

Developmental restriction of the LIM homeodomain transcription factor Islet-1 expression to cholinergic neurons in the rat striatum.

LIM homeodomain transcription factors play crucial roles in determining diverse aspects of neuronal development both in vertebrates and invertebrates. In the present study, we studied the expression pattern of Islet-1 (Isl-1), a member of the LIM homeodomain protein family, in the rat striatum during development. The developmental expression of Isl-1 in the striatum is highly dynamic and complex in terms of spatial and temporal regulation. The reverse transcription-polymerase chain reaction and ribonuclease protection assays demonstrated that Isl-1 messenger RNA was expressed in the developing striatum. The immunocytochemical study of Isl-1 protein expression showed that there were prominent mediolateral and caudorostral Isl-1 gradients in the developing striatum. Numerous Isl-1-positive cells appeared in the medial mantle zone of the developing striatal proper, and they co-expressed the postmitotic neuronal marker, microtubule-associated protein 2. The numbers of Isl-1-positive cells were decreased from the medial to the lateral regions, so that there were only a few Isl-1-positive cells scattered in the lateral striatum. These scattered Isl-1-positive cells were doubly labeled with tyrosine kinase receptor A and choline acetyltransferase, which indicated that they were cholinergic neurons. The Isl-1 gradients were most prominent in the embryonic day 18 and 20 striatum. With increases of time, the Isl-1 gradients were gradually reduced, and the gradients disappeared by postnatal day 7. Despite the general down-regulation of striatal Isl-1, a few Isl-1-positive cells were sustained into the adult striatum in which Isl-1 was nearly exclusively expressed by all cholinergic neurons and vice versa. Our study suggests that Isl-1 is likely to be initially expressed by postmitotic cholinergic precursors and some, if not all, non-cholinergic precursors in the developing striatum. During the progression of striatal differentiation, Isl-1 is down-regulated in non-cholinergic cells, but is sustained in cholinergic cells. The developmental restriction of Isl-1 to cholinergic neurons in the striatum may represent a novel mechanism by which LIM homeodomain proteins specify specific cell types in the striatum during development.

Use of isomalto-oligosaccharide in the treatment of lipid profiles and constipation in hemodialysis patients.

Chronic constipation and hyperlipidemia, one of the many atherogenic risk factors, were common complications in hemodialysis (HD) patients. The present trial evaluates the therapeutic efficacy of isomaltose-oligosaccharide (IMO) in the treatment of chronic severe constipation and its effect on lipid profiles in 20 HD patients. After a 2-week basal period, these patients were allocated to receive 30 g of IMO for a 4-week period. After the study period, these patients were observed for another 4 weeks. Bowel frequency, gastrointestinal symptomatology, biochemical parameters, and lipid profiles were assessed. All patients completed this study. IMO induced a significant increase in number of bowel movements and hence improvement of constipation in 76.3% + 30.9% of patients during the 4-week treatment. Some, but well-tolerated gastrointestinal side effects were noted. Statistically significant decreases in total cholesterol and triglycerides (TG) and increases in high density lipoprotein-cholesterol (HDL-C) were noted after IMO treatment (P <.05 compared with baseline and controls). After the study period, those patients receiving IMO had reductions in levels of total cholesterol -17.6%, TG -18.4%, and elevations of levels of HDL-C by +39.1%. In conclusion, IMO once a day is well tolerated and effective in increasing bowel frequency and improving constipation in HD patients. In addition, IMO treatment was effective in lowering total cholesterol and triglycerides and in raising HDL-C in HD patients.

Influence of P450 3A4 SRS-2 residues on cooperativity and/or regioselectivity of aflatoxin B(1) oxidation.

The major human liver drug-metabolizing cytochrome P450 enzymes P450 3A4 and P450 3A5 share >85% amino acid sequence identity yet exhibit different regioselectivity toward aflatoxin B(1) (AFB(1)) biotransformation [Gillam et al. (1995) Arch. Biochem. Biophys. 317, 74-384]. P450 3A4 prefers AFB1 3alpha-hydroxylation, which detoxifies and subsequently eliminates the hepatotoxin, over AFB1 exo-8,9-oxidation. P450 3A5, on the other hand, is a relatively sluggish 3alpha-hydroxylase and converts AFB(1) predominantly to the genotoxic exo-8,9-epoxide. Using a combination of approaches (sequence alignment, homology modeling and site-directed mutagenesis), we have previously identified several divergent residues in four of the six putative substrate recognition sites (SRSs) of P450 3A4, which when replaced individually with the corresponding amino acid of P450 3A5, resulted in a significant switch of the characteristic P450 3A4 AFB(1) regioselectivity toward that of P450 3A5 [Wang et al. (1998) Biochemistry 37, 12536-12545]. In particular, residues N206 and L210 in SRS-2 were found to be critical for AFB(1) detoxification via 3alpha-hydroxylation, and the corresponding mutants N206S and L210F most closely mimicked P450 3A5, not only in its regioselectivity of AFB(1) metabolism but also in its overall functional capacity. We have now further explored the plausible reasons for such relative inactivity of the SRS-2 mutants by examining N206S and additional mutants (L210A, L211F, L211A, and N206E) and found that the dramatically lowered activities of the N206S mutant are accompanied by a loss of cooperativity of AFB(1) oxidation. Molecular dynamics analyses with an existing P450 3A4 homology model [Szklarz and Halpert (1997) J. Comput. Aided Mol. Des. 11, 265] suggested that N206 (helix F) interacts with E244 (helix G), creating a salt bridge that stabilizes the protein structure and/or defines the active site cavity. To examine this possibility, several E244 mutants (E244A, V, N, S) were tested, of which E244S was the most notable for its relatively greater impairment of P450 3A4-dependent AFB(1) 3alpha-hydroxylation. However, the results with these E244 mutants failed to validate the N206-E244 interaction predicted from these molecular dynamics analyses. Collectively, our findings to date have led us to reconsider our original interpretations and to reexamine them in the light of AFB(1) molecular modeling analyses with a newly refined P450 3A4 homology model. These analyses predicted that F304 in SRS-4 (I-helix) plays a pivotal role in AFB(1) binding at the active site in either orientation leading to 3alpha- or exo-8,9-oxidation. Consistent with this prediction, conversion of F304 to Ala abolished P450 3A4-dependent AFB(1) 3alpha-hydroxylation and exo-8,9-oxidation.

Activation-induced cell death in peripheral blood mononuclear cells (PBMCs) from patients with chronic hepatitis B may be related to abnormal production of interleukin 12 and 10.

Peripheral blood mononuclear cells (PBMCs) from 70 patients with chronic hepatitis B and 32 normal healthy persons were isolated and cultured with or without Staphylococcus aureus enterotoxin B (SEB; 0.2 mg x l(-1)) and recombinant HBcAg (rHBcAg; 1.0 mg x l(-1)) for 48 h in vitro. After incubation, the cells were harvested by centrifugation and apoptosis of the PBMCs was studied by staining with fluorescent dyes YOPRO-1 and Hoechst 33342. The levels of IL-12 and IL-10 in the serum and the supernatants of cultured PBMCs were assayed by ELISA. The levels of IL-12 heterodimer in the serum and the supernatants of PBMCs cultured with SEB or rHBcAg were lower in patients than controls. The levels of IL-10 in both the serum and supernatants were higher in patients than controls. In addition, the percentage of apoptotic cells in PBMCs from the infected patients was significantly greater than from normal persons in the presence or absence of SEB and rHBcAg. Patients seropositive for HBeAg had much greater percentage of apoptotic cells in the PBMCs cultured with rHBcAg than patients seronegative for HBeAg, reaching 24.08%. We speculate that activation-induced cell death of PBMCs in the patients with hepatitis B may be related to abnormal expression of IL-12 heterodimer and IL-10, which may lead to persistent infection in the patients.

Dysfunction of peripheral blood dendritic cells from patients with chronic hepatitis B virus infection.

AIM: To identify the property of dendritic cells (DCs) of peripheral blood monocytes (PBMC) in patients with chronic HBV infection. METHODS: Twenty patients with persistent HBV infection were included in this study, 10 healthy subjects being used as a control group. The peripheral blood mononuclear cells (PBMC) of T cell-depleted populations were incubated and induced into mature dendritic cells in the RPMI-1640 medium in the presence of cytokines GM-CSF, IL-4, FLt-3,TNF-alpha and 100mL.L(-1 )of fetal calf serum for a total of 10-12 days. The expressions of surface markers on DCs were evaluated using flow cytometric analysis. ELISA method was used to determine the cytokine levels of interleukin-12 (IL-12) and IL-10 in the supernatant produced by DCs. For detection of the stimulatory capacity of DCs to T cell proliferation, mytomycin C-treated DC were incubated with allogenic T cells. RESULTS: A typical morphology of mature DCs from healthy subjects and HBV-infected patients was induced in in vitro incubation, but the proliferation ability and cellular number of DCs from HBV-infected patients significantly decreased compared with healthy individuals. In particular, the expression levels of HLA-DR, CD80 (B7-1) and CD86 (B7-2) on DC surface from patients were also lower than that from healthy individuals (0.46 vs 0.92 for HLA-DR, 0.44 vs 0.88 for CD80 and 0.44 vs 0.84 for CD86,P<0.05). The stimulatory capacity and production of IL-12 of DCs from patients in allogenic mixed lymphocyte reaction (AMLR) significantly decreased, but the production level of nitric oxide (NO) by DCs simultaneously increased compared with healthy subjects (86 +/- 15 vs 170 +/- 22 micromol.L(-1), P <0.05). CONCLUSION: The patients with chronic HBV infection have the defective function and immature phenotype of dendritic cells, which may be associated with the inability of efficient presentation of HBV antigens to host immune system for the clearance of HBV.

Efficacy trial of Vi polysaccharide vaccine against typhoid fever in south-western China.

OBJECTIVE: To test the efficacy of locally produced Vi vaccine over a time period of longer than one year. METHODS: A double-blinded, randomized field trial was performed in Guangxi Zhuang Autonomous Region in south-western China, using 30 micrograms doses of locally produced Vi. Enrolled subjects were 3-50 years of age, although the majority (92%) were school-aged children, who have the highest rate of typhoid fever in this setting. A total of 131,271 people were systematically allocated a single dose of 30 micrograms of Vi polysaccharide or saline placebo. The study population was followed for 19 months, with passive surveillance conducted in the Ministry of Health and the Regional Health and Anti-epidemic Centre (HAEC). Clinically suspected cases of typhoid fever were confirmed by blood culture, or by serological reaction with O-antigen (Widal tests). FINDINGS: After 19 months, there were 23 culture-confirmed cases of typhoid fever in the placebo group versus 7 cases in the Vi group (Protective efficacy (PE) = 69%; 95% CI = 28%, 87%). Most of the isolates were from school-aged children: 22 cases in the placebo group versus 6 in the Vi group (PE = 72%; 95% CI = 32%, 82%). No serious post-injection reactions were observed. The locally produced Vi polysaccharide vaccine showed levels of protective efficacy similar to those for Vi vaccine produced in industrial countries. CONCLUSION: The slightly higher dose of vaccine did not seem to alter efficacy significantly in China.

Effects of soil acidity on the uptake of trace elements in soybean and tomato plants.

The effects of soil acidity on the uptake of trace elements (Co, Zn, Se, Rb, Sr, Y, Zr, Tc, Ru, Rh and Re) in soybean and tomato were studied by a multitracer technique. The soybean and tomato plants were cultivated on soils at pH 6.4 (normal soil) and 4.2 (acid soil) and administered with a multitracer for 15-60 d. In general, the uptake of cationic elements in the leaves and stems of soybean plants cultivated on acid soil became higher than those of plants cultivated on normal soil during the late period of growth. However, the effect of soil acidification on the uptake of the anionic element. Se, was quite different from that on the cationic elements. The uptake of Se by the plants cultivated on normal soil was higher than that of the plants cultivated on acid soil at all four harvest points. The uptake behavior of these elements in soybean was discussed in relation to their adsorption behavior on the same soil as was used for soybean cultivation. The growth of tomato plants was seriously affected by the soil acidity and lowering of uptake of elements was observed for the plants cultivated on acidified soil.

A dual electrochemical sensor for nitrite and nitric oxide.

Nafion/lead-ruthenate pyrochlore chemically modified electrode (NPyCME) showed a remarkable dual sensing activity toward NO2- oxidation and NO reduction as demonstrated by cyclic voltammetry (CV), ac-impedance spectroscopy and flow injection analysis (FIA). The mechanistic parameters of current function, charge transfer resistance and exchange current for the NPyCME, GCE and Nafion-coated GCE were evaluated and compared. The disproportionation reaction of NIIIO2- into NIVO3- + NIIO in acidic solution was used as a model system for testing the dual sensing ability of the NPyCME. The obtained crossover peak response for NO2- oxidation and NO reduction in pH 1.65 buffer solution gave the direct proof for the applicability of the NPyCME in the dual electrocatalytic action. By flow injection analysis, under optimized conditions, the calibration curve was linear in the range of 100 nM-100 microM and 800 nM-63.3 microM and the detection limit (S/N = 3) was 4.8 nM and 15.6 nM for NO2- and NO, respectively.

Dry eye and closed eye tears.

PURPOSE: To establish accurate measurement of tear production in the diagnosis and monitoring of dry eye. METHODS: Reexamination of the Schirmer test indicated that the rate of tear secretion decreases with advancing age. We measured the noninvasive tear film break-up time (NIBUT) in normal and dry eye patients with our original apparatus. The NIBUT showed a reasonable level of accuracy with a cut-off value of 5 seconds. The lactoferrin concentration in tear fluids subsequently was found to have a cut-off value of 1.1 mg/mL with an optimal level of accuracy. RESULTS: The present findings suggest that a combination of clinical tests is needed for the diagnosis of dry eye syndrome. The closed eye condition induces subclinical inflammation on the ocular surface. Fibronectin, a high-molecular weight glycoprotein, plays an important role in corneal wound healing, although its concentration in various types of tear has not been investigated. Fibronectin concentration in open eye tears (19 +/- 24 ng/mL) was significantly different (p = 0.004) from that in closed eye tears (4127 +/- 3222 ng/mL). During reflex tearing with nasal stimulus, the concentration increased significantly (p = 0.028) after 100 mL of reflex tears (220 +/- 126 ng/mL). Albumin concentration in the same samples showed a pattern similar to that for fibronectin. Administration of a topical vasoconstrictor eliminated the increase in fibronectin concentration during reflex tearing. CONCLUSION: These findings suggest that fibronectin in the tear fluid was derived from plasma and that the increase in concentration in closed eye and reflex tear fluid was caused by leakage from dilated conjunctival blood vessels.

[Fluorescent mRNA differential display technique].

AIM: To apply fluorescent mRNA differential display technique. METHODS: Total RNA samples were extracted from human monocyte line U937 treated/untreated with IFN and LPS, and were used as templates in differential display PCR. The anchored primers used were labeled with the fluorescent tag. After running on 5.6% denaturing PAGE gel, differentially expressed bands were excised and recovered, and finally reamplified. RESULTS: Three tested samples all showed amplified bands differed from 300 bp to 2.0 kb, the bands were bright and clear, the background was low. Both yes/no changes and upregulated/downregulated happenings were shown simultaneously. The reamplification bands were sharp and pure. CONCLUSION: We have successfully practiced fluorescent differential display technique in our lab. It is a fast, safe and cost-effective method used to sereen unknown expressed genes.

Antagonistic effect of 3,6-dimethamidodibenzopyriodonium gluconate on lipid peroxidation in cerebral cortical neuronal cultures and rat brains during focal cerebral ischemia reperfusion.

AIM: To study 3,6-dimethamidodibenzopyriodonium gluconate (I-93) antagonistic effects on lipid peroxidation in cerebral cortical neuronal cultures and rat brains during focal cerebral ischemia-reperfusion. METHODS: Cerebral cortical neurons were cultured and rat focal cerebral ischemia-reperfusion model was established by reversible middle cerebral artery occlusion (MCAO) without craniectomy. The efflux of lactate dehydrogenase (LDH) from neurons, content of malondialdehyde (MDA) in neurons and brain homogenate, activity of superoxide dismutase (SOD) in brain homogenate, and index of cerebral edema as well as brain morphology were investigated. RESULTS: I-93 10-40 mumol.L-1 concentration-dependently inhibited efflux of LDH and elevated levels of MDA induced by addition of H2O2(10 mumol.L-1) in vitro. I-93 0.5 mg.kg-1 improved the cerebral morphology, reduced brain edema, decreased MDA content, and enhanced SOD activity in brain homogenate. CONCLUSION: I-93 protects neurons from H2O2-induced neurotoxicity and ischemia-reperfusion mediated damage by increasing the activity of antioxidant enzymes and suppressing the generation of lipid peroxides.
