Primary adrenal ganglioneuroblastoma in an adult.

We present here a case of adrenal incidentaloma in a 35-year-old male. Imaging technology including magnetic resonance imaging (MRI) clarified that the tumor was heterogeneous, and large in size (9x7 cm), and that it originated from the left adrenal gland. MRI scans on T1-weighted sequence showed the tumor with a low signal intensity followed by rapid contrast enhancement, whereas those on T2-weighted sequence showed markedly increased intensity. The tumor was surgically removed, and pathologic diagnosis was well-differentiated ganglioneuroblastoma. Ganglioneuroblastoma in adults is extremely rare, with only about 30 cases documented in people over the age of 20 years.

Inhibition by Guan-mu-tong (Caulis aristolochiae manshuriensis) of the growth of spontaneous mammary tumors in SHN virgin mice.

Multiparous SHN mice with spontaneous mammary tumors (5-10 mm in size) were given water extract of Guan-mu-tong (Gmt; Caulis aristolochiae manshuriensis) (0.5%) ad libitum as drinking water for 10 days. This treatment retarded significantly the growth of mammary tumors compared with the controls. By contrast, normal mammary gland growth, histology of adrenals and ovaries, and body weight were affected little by the Gmt treatment. Gmt appears to be the first agent inhibiting the growth of spontaneous mouse mammary tumors of palpable size by per os treatment.

[Thermal distribution in the agar phantom by a new intracavitary RF applicator for prostate gland].

Some studies suggest that the prostate gland is one of the most difficult portions of the body to heat up even by radio frequency (RF) capacitive heating, which is well established as a useful means for deep regional hyperthermia because of its non-uniform power absorption. We have developed a new type of intracavitary applicator for RF capacitive heating which can be connected to an 8 MHz RF heating device (Thermotron-RF8). The applicator is a balloon-type catheter which is filled with electrolyte, such as physiological saline, 3% saline or 3% ferrous sulfate solution, and then put between a pair of regular RF capacitive heating electrodes. The heating characteristics of the applicator were examined by using an agar phantom that is electrically equivalent to muscles. When the applicator was connected to the electrode of the RF generator with a copper cord, the balloon filled with electrolyte operated as an inner electrode. The effective hot area around the balloon of the catheter was observed, and the size of the hot area was changed by the kinds of electrolytes used and their concentration. With this adaptive technique, it is possible to maintain an adequate thermal distribution at a tumor in the uterus, urinary bladder and rectum, as well as the prostate gland.

[Chemosensitivity test by subrenal capsule assay (SRCA) in transitional cell carcinomas].

From November, 1986 to October, 1989, the antitumor activities of several chemotherapeutic agents were tested by subrenal capsule assay (SRCA) in cases of 23 transitional cell carcinomas. Overall sensitivities of tested drugs to transitional cell carcinomas were compatible to clinical data and correlated with individual clinical courses. This method has some problems, but can be carried out at any institute and serve as an aid to the selection of chemotherapeutic agents.

Diurnal patterns of serotonin, 5-hydroxyindoleacetic acid, tryptophan and fibrinolytic activity in blood of depressive patients and healthy volunteers.

Diurnal changes of serotonin-related factors in whole blood and fibrinolytic activity were determined in depressed patients and healthy controls. Whole blood serotonin concentration of depressed patients showed marked changes throughout daytime, with maximum values in the evening and lowest values in the morning, whereas its metabolite 5-HIAA followed a contrary pattern. The circadian rhythm of 5-HT and 5-HIAA in the control group was quite different from depressed patients. Plasma levels of tPA decreased from 12:30 to 16:30. Concentrations of free plasminogen activator inhibitor (PAI-1) and complex of tPA-PAI-1 decreased from 8:30 to 16:30. Plasma levels of total PAI-1 decreased from 8:30 to 16:30. Plasma levels of the fibrinolytic parameters may be lower in depressive patients than in normal controls. These results support the changes in the circadian rhythm of serotonin and its related substances in the blood of depressive patients.

Protein kinase C in human pheochromocytoma.

Subtypes of protein kinase C were analyzed in adrenal and extra-adrenal pheochromocytoma of humans. Almost all protein kinase C of the adrenal tumor was type III, while the enzyme of the extra-adrenal tumor was separated into two major fractions corresponding to type II and type III by hydroxyapatite column chromatography. The extra-adrenal tumor but not the adrenal tumor spontaneously produced neurite-like processes when the cells were cultured in vitro. These results suggest that the high proportion of type II enzyme may reflect neuron-directed differentiation in human pheochromocytoma.

[Clinical observations of plasma fibronectin in patients with urological malignant disease].

Fibronectin (FN) is a high molecular weight glycoprotein widely distributed in the body and has a number of biological activities. Recently, there have been reports on the relationship between plasma fibronectin (pFN) levels and malignant diseases, but the significance of pFN is still unclear. Using immunoturbidimetric assay, we measured pFN levels of 24 healthy controls and 61 patients with urological malignant diseases, and obtained the following results. 1. pFN levels before treatment. 1) pFN level was 379 +/- 60.6 micrograms/ml and 356 +/- 123.7 micrograms/ml in the healthy controls and patients, respectively. There was no significant difference. 2) pFN level in patients with metastases was 320 +/- 92.9 micrograms/ml and had no statistical difference as compared with patients without metastases (371 +/- 132.7 micrograms/ml) and controls, despite the decrease in the mean value. 3) pFN level in patients with poor prognosis (300 +/- 107.8 micrograms/ml) was significantly lower than that in patients with good prognosis (410 +/- 157.2 micrograms/ml) and controls (p less than 0.05). 2. pFN levels during conservative treatments for advanced disease. pFN level in patients with a rapid progressive disease (287 +/- 64.4 micrograms/ml) was significantly lower than that in patients with a slow progressive disease (327 +/- 43.3 micrograms/ml) (p less than 0.01). These data suggest that low pFN level predicts rapid progression of the disease and poor prognosis in patients with a urological malignant disease.

[Radiation therapy combined with hyperthermia in advanced cancer].

Radiation therapy combined with radiofrequency (RF) hyperthermia was performed on 5 advanced cancer patients. Included were one each with urinary bladder cancer, hepatoma with left axillary node metastasis, breast cancer, tongue cancer with left cervical metastasis, and mandibular cancer. All had large tumours, which were judged to be uncontrollable by radiotherapy alone. They were treated with irradiation (Linac: 10 MV X-ray 1.8-2.0 Gy/day, 5 days/week), followed within an hour by RF hyperthermia once or twice a week. Partial response was obtained in the urinary bladder cancer patient. Surface overheating around the margin of electrodes occurred in all but no severe complications were observed.

Effects of simultaneous treatment with hydroxyapatite and coffee cherry, the residue left after the removal of coffee beans, on spontaneous mammary tumourigenesis and related parameters in SHN mice.

We have found that the chronic administration of the diet containing 5% hydroxyapatite (HAP) derived from bovine or swine bone or drinking water containing 0.5% extract of coffee cherry (CC), the residue left after the removal of coffee beans, induced a marked inhibition of spontaneous mammary tumourigenesis in SHN mice, while the effects decreased with age. In the present study, the combined effects of HAP and CC on mammary tumourigenesis and related parameters were examined. The inhibitory effects of HAP or CC alone on the development and/or the growth of mammary tumours were reduced by HAP + CC. Decreased food and water intake and retarded body growth caused by CC were ameliorated by HAP. Enhancement by HAP or CC of the excretion of the urine components was mostly nullified by HAP + CC. Parameters such as normal mammary gland growth and uterine adenomyosis on which neither HAP nor CC had an effect also did not respond to HAP + CC. These findings suggest that the target is important when administering natural products in combination if the agents are to manifest their effects, additive, synergistic, antagonistic or otherwise.

Effects of gold banded lily (Lilium auratum Lindl) or Chinese milk vetch (Astragalus sinicus L) on spontaneous mammary tumourigenesis in SHN mice.

Some guanidino compounds have been found to inhibit spontaneous mammary tumourigenesis in mice. In the present study, the effects of chronic treatment with Gold Banded Lily (Lilium auratum LindI) or Chinese Milk Vetch (Astragalus sinicus L), which contains L-arginine or L-canavanine, on spontaneous mammary tumourigenesis were examined in SHN mice. Free access to drinking water containing hot water extract of each natural product at a concentration of 0.5% significantly inhibited the development, but not growth, of mammary tumour. The mechanism of the protective role of each natural product in mammary tumours remains to be clarified; however, there were no significant long-term deleterious side-effects of chronic treatment with either product estimating from body weight change, food and water intakes and plasma component levels. Thus, the present findings suggest that these natural products containing guanidino compounds may act as prophylactic agents for mammary and other types of tumours.

Further study on the effects of motherwort (Leonurus sibiricus L) on preneoplastic and neoplastic mammary gland growth in multiparous GR/A mice.

To evaluate further the chemopreventive role of motherwort (Leonurus sibiricus L; MW) in lesions of the mammary gland and uterus of GR/A mice, the effects on these lesions of the adsorbed (MW1) and unadsorbed (MW2) fractions of MW separated by ion-exchange resins were studied. The incidence of palpable mammary tumours was suppressed and their growth was retarded by both MW1 and MW2, between whose effects no apparent difference was seen. However, neither of them showed effects on pregnancy-dependent mammary tumours (PDMT), mammary hyperplastic alveolar nodules (HAN) or uterine adenomyosis, whereas MW promoted PDMT and inhibited HAN and adenomyosis. All these findings indicate the importance of the synergistic action of several components, specified and unspecified, for the full manifestation of the effects of Chinese medicine.

1H-NMR spectroscopy of urinary components of SHN mice in the course of spontaneous mammary tumourigenesis.

Proton nuclear magnetic resonance (1H-NMR) spectroscopy of urine was studied before and after the development of spontaneous mammary tumours in SHN mice. Urinary allantoin and lactic acid levels declined from 2 weeks before mammary tumour appearance and increased with the tumour growth after its appearance. The level of component at 3.4 ppm, which reacted with Ninhydrin and was possibly assigned to taurine, continued to decline beginning 2 weeks before tumour appearance. No considerable changes were observed in urinary levels of creatine and creatinine before and after tumour development, however, these components as well as allantoin or lactic acid had positive correlations with mammary tumour size. A negative correlation was observed between the level of 3.4 ppm component and tumour size. Surgical removal of tumours resulted in an apparent decline in urinary levels of allantoin and lactic acid. At all stages, urinary allantoin level was approximately 4 times as high as the levels of lactic acid and 3.4 ppm component. All results suggest that urinary allantoin would be a good indicator for the development of spontaneous mammary tumours of mice.

Protection by tree-peony (Paeonia suffruticosa Andr) of obesity in (SLN x C3H/He) F1 obese mice.

Water extracts of 10 medicinal plants were checked for protection against obesity in female and male (SLN x C3H/He) F1 obese mice. Each extract was given as drinking water at the concentration of 0.5% between 3 and 32 weeks of age. Among agents examined, only the root extract of tree-peony (Paeonia suffruticosa Andr) retarded growth, especially in males. In males, the treatment with tree-peony resulted in a significant decline in food intake and of the Lee index, an index of obesity, and an increase in glucose tolerance. While no significant difference was observed between the experimental and the control groups in serum free fatty acid levels, the range of the levels was much larger in female and male mice given tree-peony. There was little difference between groups in the weights of heart, liver, kidney, lung, spleen and major endocrine organs in both sexes and in the pattern of oestrous cycles in females. All results indicate that tree-peony can protect against obesity, especially in males, at least partly by a decrease in food intake and an increase in glucose metabolism.

Inhibition by lombricine from earthworm (Lumbricus terrestris) of the growth of spontaneous mammary tumours in SHN mice.

The effects of lombricine extracted and purified from earthworm (Lumbricus terrestris) skin on the growth of palpable sizes (approximately 5 mm) of spontaneous mammary tumours were studied in SHN mice. In Experiment 1, daily subcutaneous injections of lombricine (0.3 mg/0.05 ml olive oil) inhibited markedly the growth of tumours associated with the retardation of the growth of preneoplastic mammary hyperplastic alveolar nodules. In 1H-NMR spectra, the experimental mice had lower serum levels of lactic acid and glucose than the control. On the other hand, urine of the former group contained higher levels of allantoin, creatine and creatinine than that of the latter. In Experiment 2, lombricine given as diet at the concentration of 120 mg/kg also inhibited the growth of tumours, though to a lesser degree than the injection. The treatment had little effect on 1H-NMR spectra of either serum or urine and normal and preneoplastic mammary gland growth. All results indicate that the inhibition by lombricine of the growth of mammary tumours is at least partly due to the maintenance of homeostasis of the body including the regulation of the excess uptake of glucose as a source of energy and nutrition.

Effects of motherwort (Leonurus sibiricus L) on preneoplastic and neoplastic mammary gland growth in multiparous GR/A mice.

Chronic ingestion of methanol extract of the above-ground part of motherwort (Leonurus sibiricus L) in drinking water at the concentration of 0.5% enhanced the development of both pregnancy-dependent mammary tumours (PDMT) and mammary cancers originated from PDMT. By contrast, the treatment markedly suppressed the development of mammary cancers that originated from hyperplastic alveolar nodules (HAN) associated with the decreased formation of HAN. The incidence of uterine adenomyosis was also inhibited in mice given motherwort. The urinary excretion of allantoin, creatine and creatinine and glucose tolerance were stimulated by motherwort. The cause of discrepancy of the effects of motherwort on mammary cancers due to their origins is not clear at present. However, the stimulation by the agent of the excretion of any carcinogenic factors may at least partly contribute to its inhibition of mammary cancers originating from HAN.

Protection by coffee cherry against spontaneous mammary tumour development in mice.

Chronic ingestion of an extract of coffee cherry (CC), the residue left after removal of coffee beans, induced a marked suppression of spontaneous mammary tumour development in a high mammary tumour strain of SHN virgin mice. The growth of normal and preneoplastic mammary glands was also apparently inhibited by the treatment. While the treatment resulted to some extent in a decrease of water intake and a retardation of body growth, the excretion of urinary components such as urea, allantoin and creatinine was increased and serum free fatty acid level was decreased by CC. Little modulation of the endocrine system was noted by the treatment. Finally, a promising chemopreventive role of CC in tumours is indicated.
