[Cooperative study on surgical adjuvant immunochemotherapy for prevention of postoperative recurrence of gastric cancer (II). Cooperative study group on surgical adjuvant immunochemotherapy for prevention of postoperative recurrence of gastric cancer supported by the Ministry of Health and Welfare (Kondo's group])

A cooperative study on surgical adjuvant immunochemotherapy for prevention of postoperative recurrence of gastric cancer was carried out by Kondo's group from July 1974 to December 1977. A total of 848 patients with gastric cancer underwent curative resection were eligible. Of 848 patients, 819 cases with complete description of patient's background were evaluable: Group A (surgery + MMC + 5-FU:chemotherapy) 253 cases, group B (surgery + MMC + 5-FU + OK-432 or PSK:immunochemotherapy) 282 and group C (surgery alone) 284. The 5-year corrected survival rates of total cases using Cox's proportional hazard model were 76.2% with group A, 73.6% with group B and 73.3% with group C. And the 10-year survival rates were 67.6% with group A, 64.3% with group B and 63.9% with group C. The difference in survival rate among groups A, B and C was not statistically significant. In the evaluation of prognostic factors by Cox's proportional hazard model, level of curative resection, extent of resection, histological stage and Kajitani's classification had an influence upon survival rates, in which degrees of metastasis in lymph node and serous infiltration were also involved. The significance of postoperative adjuvant immunochemotherapy has not been clarified.

New biological properties of pyrroloquinoline quinone and its related compounds: inhibition of chemiluminescence, lipid peroxidation and rat paw edema.

Pyrroloquinoline quinone (PQQ) inhibited the chemiluminescence (CL) from mouse peritoneal cells initiated by zymosan, carrageenin and N-formyl-methionyl-leucyl-phenylalanine and CL generated by the xanthine-xanthine oxidase reaction and the lipid peroxidation in the rat brain homogenate. The inhibitory activity of PQQ was more potent than that of idebenone, alpha-tocopherol and ascorbic acid in all the three assay systems. In the xanthine-xanthine oxidase reaction, PQQ had no effect on the formation of uric acid at the concentration of CL inhibition. These results suggest that PQQ might have a radical scavenger-like activity. Structure-activity relationship of PQQ and its six related compounds showed that the 7- and 9-carboxyl groups of PQQ as well as the orthoquinone structure are responsible for the radical scavenger-like activity. In addition, the -NH group in the pyrrole ring of PQQ seemed to be essential for the antilipid peroxidative activity in the rat brain homogenate. When administered i.p., PQQ inhibited the development of 0.1% carrageenin-induced paw edema in rats. These results suggest that PQQ might have therapeutic effects on various diseases, of which development or exacerbation has been known to be associated with radical oxygens.

[The quantitative analysis of electroencephalography in primary hypothyroidism].

The EEG topography was recorded in the patients with primary hypothyroidism, and the relationship between equivalent potential and blood thyroid hormonal levels were investigated. To determine the relationship between the % equivalent potential fraction (%EPF) and the levels of blood free T3, free T4, and TSH in each patient, regression lines were drawn in respective EEG spectral bands. The %EPF of each patient showed a tendency to have a negative correlation with the values of blood free T3 and free T4 in slow frequency spectral bands such as delta, theta, and alpha 1 waves, whereas a positive correlation in fast frequency bands such as alpha 2, beta 1, and beta 2 waves. In particular %EPF had a significant relationship with blood free T3 levels in alpha 1 and beta 2 spectral bands. These results suggest that the thyroid hormones may have influence on the mechanisms of EEG rhythm formation in primary pypothyroidism.

Nuclear granules recognized by some monoclonal antibodies against intermediate filament protein locate on chromosomes during mitosis.

AC54 monoclonal antibody (MAb), an anti-desmin MAb, recognizes both intermediate filaments (IFs) and nuclear granules in BHK21/C13 cells. To investigate nuclear granules, similar MAbs were obtained by using desmin fraction as an antigen. Among them, DSB389 MAb recognized mainly nuclear granules in HeLa and rat liver cells. The nuclear granules in HeLa cells were aligned in arrays, sometimes connected by, or part of, a rope-like structure, and stable against treatment with 0.5% Triton X-100 and 2 M NaCl. They located on or around the chromosomes during mitosis. Essentially the same results were obtained with DSB860 and AC54 MAbs. The distribution of the granules in liver nuclei recognized by DSB389 MAb was similar to that of DNA and was different from that of the nuclear pore complexes. The biological significance of the nuclear granules is discussed.

Cispentacin, a new antifungal antibiotic. II. In vitro and in vivo antifungal activities.

Cispentacin [-)-(1R,2S)-2-aminocyclopentane-1-carboxylic acid) is a new antifungal antibiotic possessing potent anti-Candida activity. The 50% inhibitory concentration (IC50) and IC100 values of cispentacin against clinical isolates of Candida albicans were in the ranges 6.3 approximately 12.5 and 6.3 approximately 50 micrograms/ml, respectively, by turbidimetric measurement in yeast nitrogen base glucose medium. No significant activity was seen against any yeasts and molds when tested by the agar dilution method using three different agar media: KNOPP's agar, yeast extract-glucose-peptone agar and Sabouraud dextrose agar. This antibiotic demonstrated good therapeutic efficacy against a systemic Candida infection in mice by both parenteral and po administrations. The 50% protection dose (PD50) values after single iv and po administrations were 10 and 30 mg/kg, respectively. It was also effective in a systemic infection with Cryptococcus neoformans and in both lung and vaginal infections with C. albicans in mice. Cispentacin did not induce acute lethal toxicity at 1,000 mg/kg by iv injection and 1,500 mg/kg by ip and po administrations in mice.

[Measurement of carcinoembryonic antigen, squamous cell carcinoma related antigen and neuron-specific enolase in the bronchoalveolar lavage fluid in patients with peripheral lung cancer].

Carcinoembryonic antigen (CEA), squamous cell carcinoma related antigen (SCC) and neuron-specific enolase (NSE) in bronchoalveolar lavage fluid were measured in 30 patients with peripheral lung cancer, 11 patients with benign lung disease and 19 healthy controls. The mean levels and positive rates of lavaged fluid CEA were 128.0 +/- 16.9 ng/mg and 33.3% in patients with lung cancer, 68.1 +/- 25.9 ng/mg and 9.1% in patients with benign lung disease, and 68.3 +/- 11.6 ng/mg and 5.2% in healthy controls, respectively. The mean levels and positive rates of lavaged fluid CEA in patients with lung cancer were significantly higher than those in patients with benign lung disease (p less than 0.05) and those in healthy controls (p less than 0.05). The mean levels of lavaged fluid SCC and NSE showed no significant difference between cases of lung cancer and benign lung disease or healthy controls. No lavaged tumor marker level in patients with lung cancer showed any close correlation with histologic types and serum levels. In conclusion, measurement of lavaged fluid CEA was considered to be useful in the differential diagnosis of peripheral lung cancer.

Opioid kappa receptors correlate with the development of conditioned suppression of motility in mice.

Mice exhibited a marked suppression of motility (conditioned suppression) when placed in the same environment in which they had previously received an electric footshock. The conditioned suppression of motility was potentiated by U-69,593, a selective kappa (non-mu) agonist. This effect of U-69,593 was antagonized by MR-2266, a kappa antagonist. Additionally, in contrast to U-69,593, MR-2266 alone attenuated the conditioned suppression of motility. These results suggest that the activation of kappa receptors may be responsible for the development of conditioned suppression of motility.

Effect of age and maturation on sudomotor nerve regeneration in mice.

This study demonstrates that regeneration of unmyelinated sudomotor axons in mice becomes progressively slower during aging. Identical lesions were made in mice aged 0, 2, 4, 6, 7, 24 and 60 weeks. The peroneal, sural and saphenous nerves were cut and tied to prevent regeneration. The sciatic nerve was then frozen at the thigh, leaving the hind paw completely denervated. By 7 days, sweat glands (SGs) of the paw had ceased sweating after pilocarpine injection. Subsequent regeneration of sudomotor axons was judged by the rate of return of pilocarpine sensitivity. SGs in the hind paws of normal newborn mice did not sweat at birth. Cholinergic stimulation first activated sweating at 13 days of life. The number of responsive SGs increased progressively to reach the adult level by 30 days. In one-week-old mice, whose sciatic nerve had been sectioned, the SG response to cholinergic stimulation was very delayed in time and reduced in number. Sweat glands of young mice, between 2 and 4 weeks of age, regained cholinergic sensitivity at a faster rate than mature animals and attained normal SG counts. Throughout a broad intermediate range of age in adulthood (7-24 weeks), the rate of sudomotor nerve regeneration was the same, but in older mice (60 weeks) it was slower and less complete.

Reinnervation of sweat glands in the mouse: axonal regeneration versus collateral sprouting.

The capacity of sudomotor axons to reinnervate sweat glands by regeneration after freezing was compared with their capacity to reinnervate by the mechanism of collateral sprouting. Two groups of mice were prepared in the same manner, with a single exception. All nerves to the hind paw were cut and tied to prevent regrowth. The exception, the lateral plantar nerve, was left intact in one group and frozen at the ankle in the other group. Reinnervation of sweat glands in the paw was charted frequently for 10 weeks using a silastic mold method. We found that sudomotor axons regenerating from the frozen nerve functionally reinnervated sweat glands of the paw sooner, beginning at 15 days postoperation, and at a higher rate than reinnervation from the collateral branching intact nerve, that began at about 25 days. Full recovery occurred by 41 days in most animals of both groups. We also found that stimulation of sweating by pilocarpine activated more sweat glands during the reinnervation period than stimulation by heating the mice, suggesting that the newly regenerating sudomotor axons have a high threshold.

A role played by sigma receptors in the conditioned suppression of motility in mice.

Mice exhibited marked suppression of motility (conditioned suppression) when placed in the same environment in which they had previously received an electric footshock. The conditioned suppression was attenuated by cyclazocine and N-allylnormetazocine (SKF-10047), sigma agonists. The effect of these drugs was reduced by chronic pretreatment with cyclazocine. This behavioral change was related to the change in binding activity of [3H]-phencyclidine to sigma receptors (defined using non-radioactive SKF-10047). In the chronic vehicle-pretreated conditioned suppression group, the Kd and Bmax values of [3H]-phencyclidine binding at the high affinity site were increased when compared to those in the chronic vehicle-pretreated control group. The increased values were restored to the control levels by acute treatment with cyclazocine and SKF-10047. On the other hand, in the chronic cyclazocine-pretreated conditioned suppression group, acute cyclazocine and SKF-10047 treatment failed to change the increased values of Kd and Bmax at the high affinity site. The present behavioral and receptor-binding experiments suggest that the activation of nervous system mediated by sigma receptors may be responsible for the attenuation of conditioned suppression of motility.

Swelling of neuronal processes in motor neuron disease.

We investigated serial sections of the anterior horns of the lower lumbar cord by a modified Bielschowsky's silver impregnation, in a case of sporadic lower motor neuron disease. We paid special attention to any direct connection between the swellings of neuronal processes and the perikarya. Focal swellings of neuronal processes were occasionally directly connected with the perikarya; some had morphologic peculiarities of axons, and others originated in dendrites.

Importance of cell surface marker to the prognosis of non-Hodgkin's lymphoma.

We studied the correlation between the cell surface markers and prognosis of non-Hodgkin's lymphoma (NHL) patients treated in the Shikoku Cancer Center Hospital from 1980 to 1986. Thirty-one cases were selected on the basis of having a lymphnode as a primary lesion, having been immunophenotyped before chemotherapy, being in the intermediate histologic grade and being in stage II, III or IV. Thirteen cases of the T-cell type (T-lymphomas) and 18 cases of the B-cell type (B-lymphoma) were identified. The complete remission rate was 54% among T-lymphoma patients and 78% among B-lymphoma patients. The median length of survival was 12+ months in T-lymphoma and 26+ months in B-lymphoma. The survival rate of T-lymphoma patients was significantly lower than that of B-lymphoma patients. The importance of making surface marker studies was reappraised in our study.

Porothramycin, a new antibiotic of the anthramycin group: production, isolation, structure and biological activity.

A new antitumor antibiotic porothramycin was produced by a new strain of Streptomyces albus. The antibiotic was isolated in two active forms, the natural free hydroxyl form (porothramycin A) or the crystalline methyl ether form (porothramycin B) depending upon the isolation process used. Structural studies established that porothramycin is a new member of the pyrrolo[1,4]benzodiazepine group antibiotics having only one substituent on the benzene ring. The antibiotic exhibited antimicrobial activity against Gram-positive bacteria and anaerobes and significantly prolonged the survival times of mice implanted with experimental tumors.
