Chemical modification of the antitumor antibiotic glidobactin.

A variety of glidobactin analogs modified at the fatty acid, L-threonine and nucleus moieties of the molecule were synthesized and their structure-activity relationships examined. The antitumor and antifungal activity was greatly influenced by modification of the fatty acid glidobactin, with the dodecanoyl and tetradecanoyl analogs exhibiting better antitumor activity than the parent antibiotics. Replacement of the L-threonine with other amino acids greatly reduced the activity and reduction of the double bond of the nucleus completely eliminated the biological activity of glidobactin.

BMY-28438 (3,7-dihydroxytropolone), a new antitumor antibiotic active against B16 melanoma. I. Production, isolation, structure and biological activity.

A new antitumor antibiotic BMY-28438 was isolated from the cultured broth of Streptomyces tropolofaciens No. K611-97. The antibiotic showed potent cytotoxicity against cultured B16 melanoma cells and remarkable prolongation of life span of mice bearing B16 melanoma. The structure of BMY-28438 was determined as 3,7-dihydroxytropolone on the basis of spectral analyses and direct comparison with the authentic compound synthesized.

Acetylcholine-receptor-like protein from human thymoma associated with myasthenia gravis.

Cobrotoxin-binding protein was isolated by affinity chromatography from human thymoma which had been surgically removed from patients with myasthenia gravis. The protein was composed of polypeptides with a molecular mass of 40, 51, 65, and 74 kilodaltons as determined by polyacrylamide gel electrophoresis in the presence of sodium dodecylsulphate. Isoelectric focusing of the protein gave pI values of 5.2-5.6 and 11. This is the first report of the isolation of the protein from human thymoma. These findings suggest that the cobrotoxin-binding protein from human thymoma patients with myasthenia gravis has subunits similar to those of fish electric organs or mammalian muscles.

Effects of insulin and retinoic acid on the anchorage-independent growth of transformed BHK21/C13 cells cultured with different lots of fetal bovine serum.

Several lots of fetal bovine serum (FBS) were divided into 2 groups; in one group, retinoic acid (RA) suppressed the anchorage-independent growth (AIG) of spontaneously transformed BHK21/C13 cells (clone Ag8-1) dose-dependently, but in the other group, little suppression occurred. In neither group of FBS was the anchorage-dependent growth (ADG) of the cells suppressed by RA. The addition of insulin enhanced the AIG of the cells in both groups even in the presence of RA. The 2 types of AIG systems in Ag8-1 cells, one susceptible and the other resistant to RA, are discussed.

[A case of adult T-cell leukemia achieving complete remission with epirubicin, vincristine and prednisolone chemotherapy].

A 68-year-old man was referred to our hospital because of generalized lymphadenopathy. His abnormal findings were hepatomegaly, leukocytosis and elevated serum LDH. Anti-ATLA antibody was positive. As about half of the peripheral lymphocytes reacted to the monoclonal antibody CD25, these cells were considered to be compatible with ATL cells having an interleukin-2 receptor. Initially, recombinant beta-interferon was administered, but it was not effective. A combination of vincristine (VCR) and prednisolone (PDN) was partially effective against the lymphadenopathy, but the hepatomegaly and leukocytosis did not improve. 4'-epi-Adriamycin (Epirubicin), at a doses of 20 mg/body, weekly, was subsequently added to VCR + PDN therapy. The patient achieved complete regression 2 months later, and has been in continuous complete remission for more than 3 months.

[Antiemetic effects of combinations of metoclopramide, droperidol and dexamethasone for the prevention of cisplatin-induced gastro-intestinal toxicity: a randomized crossover trial].

Thirty-two patients with primary lung cancer receiving combination chemotherapy including cisplatin at a dosage of 80-120 mg/m2 were entered into an antiemetic randomized crossover trial. Patients received metoclopramide (2 mg/kg i.v. every 2 h X 5), droperidol (0.5 mg/kg i.v.) and dexamethasone (30 mg i.v.) on day 1, and metoclopramide (1mg/kg i.v. every 8 h X 3) (Regimen A) or metoclopramide and droperidol (2.5 mg i.v. every 8 h X 3) (Regimen B) on days 2 to 5. No vomiting occurred within the first 24 hours after cisplatin administration in 75% of patients. Regimen B was found to be more effective than regimen A with respect to the mean duration of vomiting (p less than 0.1), the mean duration of nausea (p less than 0.05), the mean duration of anorexia (p less than 0.05), and the mean score of the patients' opinions (p less than 0.1). When the patients were asked for their opinion of the two regimens, 39% preferred regimen B (p less than 0.05). There were no major side effects with either regimen.

A monoclonal antibody to chicken gizzard desmin that recognizes intermediate filaments and nuclear granules in BHK21/C13 cells.

One hybridoma (AC54), which produces monoclonal antibody (MAb) that recognizes both intermediate filaments (IFs) and nuclear granules in BHK21/C13 cells, and two hybridomas (AC19 and AC36) which produce MAbs that recognize IFs only, were obtained by using a crude actin preparation from chicken gizzard as an antigen. In immunoblotting, both the AC54 and AC19 MAbs reacted with the 52 kD protein (desmin) and some other proteins in gizzard and BHK21/C13 cells. Indirect immunofluorescent microscopy of BHK21/C13 cells showed that the cytoplasmic filaments stained by these MAbs were IFs based on their colchicine-induced whorl formation. The ability of AC54 MAb to recognize IFs was more limited than that of AC19 MAb. The nuclear granules recognized by AC54 MAb were in a different location than the cytoplasmic IFs and sometimes were concentrated in the nucleolus. These results indicate that AC54 MAb is an anti-desmin MAb that reacts with some desmin-related proteins; that it recognizes IFs differently than AC19 MAb, another anti-desmin MAb; and that it recognizes nuclear granules in locations where desmin or desmin-related protein has not yet been reported.

Conditioned suppression and opioid kappa receptor in mice.

Mice exhibited a marked suppression of motility (conditioned suppression) when placed in the same environment in which they had previously received an electric footshock. Furthermore, chronic morphine (mu agonist)-pretreated mice, as well as chronic vehicle-pretreated mice, exhibited the conditioned suppression but chronic ethylketocyclazocine (kappa agonist)- and pentazocine (kappa agonist)-pretreated mice did not. On the other hand, in the synaptic membranes of the chronic vehicle- and morphine-pretreated mice showing conditioned suppression, the specific binding of [3H]phencyclidine (sigma agonist) and [3H]naloxone (mu antagonist) significantly increased, while the specific binding of [3H]ethylketocyclazocine did not change compared to those of the corresponding control groups. However, in the chronic pentazocine- and ethylketocyclazocine-pretreated groups showing non-conditioned suppression, the specific binding of [3H]phencyclidine and [3H]naloxone were not altered. Moreover, a decrease of [3H]ethylketocyclazocine binding was observed in the chronic pentazocine-pretreated group. These results suggest that the binding function of different opioid receptor subtypes may be altered differently by stress, and that the kappa receptor may be important for the conditioned suppression of motility in mice.

[Serological study of the antibody to the adult T-cell leukemia-virus-associated antigen in various hematological diseases].

A serological study of antibody to adult T-cell leukemia-virus-associated antigen (anti-HTLV-I antibody) was made in 170 cases of various hematological diseases at Shikoku Cancer Center Hospital. The titer of anti-HTLV-I antibody was determined by indirect immunofluorescence using the MT-2 cell line. Two of two patients with ATL were positive for antibody. In non-Hodgkin's lymphoma, six of 18 cases of T-cell phenotype were positive (33%), while two of 29 cases of B-cell phenotype were antibody-positive (7%). Some cases of leukemia and aplastic anemia, who had received multiple blood transfusions, were found to be seropositive. Our results suggest that anti-HTLV-I antibody may be related to non-Hodgkin's lymphoma with T-cell phenotype as well as ATL.

[Changes in nonspecific suppressor factors in the serum of gastric cancer patients after surgery and immunochemotherapy].

After curative surgery for gastric cancers judged macroscopically to be at stage 2 or 3, patients were divided into 4 groups by randomization. As the basic treatment, patients were given one-shot Mitomycin and Tegafur as maintenance therapy for 8 months (group A). PSK (group B), OK-432 (group C) or both PSK and OK-432 (group D) were added to the treatment of group A for 8 months. Sera were obtained from these groups of patients at 4 weeks and 3, 6, 9 and 12 months after surgery. In all of these groups, values of IAP increased slightly at 4 weeks but decreased at 3 months and were maintained at such a level by 12 months. On the other hand, suppressive effects of such sera on the blastogenesis of murine spleen cells in response to PHA varied among these group. In groups A and C, the suppressive effect of sera increased after surgery and was detected continuously by 12 months. In groups B and D, in contrast, the suppressive effect disappeared from 3 to 12 months. The rise and fall of such a suppressive effect of sera may reflect the mode of action of PSK.
