[Artificial lactation induction in 12- to 19-month-old heifers with regard to the behavior of the concentration of sex hormones and lactogenic proteohormones in the blood plasma].

In 24 prepubertal heifers the lactation was artificially induced by two different methods of hormone application. The changes in milk yield and the concentrations of estradiol-17 beta (E), progesterone (P), prolactin, somatotropin and insulin-like growth factor I in blood plasma during the first 55 days of lactation were analyzed. Depending on the age, the animals were divided in three experimental groups (EG) of eight animals each: 12 months old (EG I), 15 months old (EG II) and 19 months (EG III). Four animals of each EG were treated similarly: group E and P, which was injected by estradiol benzoate and P for 7 days, or group E, which received only estradiol benzoate for 10 days. The oldest heifers (EG III) showed an induction rate of 100% with E and P treatment and of 75% with E alone. On the other hand, in heifers of EG I and EG II the induction rate was only 50% irrespectively of the treatment. The animals of EG III had the highest milk yield. The daily milk yield on the 50th day of lactation was 7.0 kg and 4.0 kg in group E and P or group E, respectively. Comparable levels in 15 months old heifers (EG II) were 4.8 kg and 1.4 kg, and in 12 months old heifers (EG I) 2.5 kg and 2.5 kg. The changes of the concentrations of the hormones in blood plasma are demonstrated in diagrams, their effect on the milk production is discussed.

[The composition of milk after artificial lactation induction in 12- to 19-month-old heifers].

In 24 prepubertal heifers the composition of milk after an artificial induction of lactation was investigated. Depending on the age, the animals were divided in 3 experimental groups (EG) of 8 animals each: 12 months old (EG I), 15 months old (EG II), and 19 months old (EG III). Four animals of each EG were treated similarly: either with estradiol benzoate and progesterone for 7 days (group E and P) or only with estradiol benzoate for 10 days (group E). During the first 18 days of lactation the protein and lactose content of the milk showed similar changes compared to the period of milk maturation during natural lactation of same duration. However, a typical colostral period with high concentrations of immunoglobulins in the milk was not observed. After the artificial induction of lactation, in the period until to the 53rd day of lactation no age dependent differences in the composition of milk were found, except whey proteins. Formation of milk substances, which are synthesized in the mammary gland, is much more induced by treatment with E and P than by treatment of E alone, with the exception of caseins.

Follow-up of patients with double stranded DNA antibodies induced by sulfasalazine during the treatment of inflammatory rheumatic diseases.

Twenty-four patients with chronic inflammatory joint diseases developed double stranded DNA (dsDNA) antibodies during sulfasalazine (SAS) therapy and 20 of these 24 patients could be followed over a mean period of 11 months. Only one of the patients showed clinical symptoms of systemic lupus erythematosus (SLE). DsDNA antibodies were transient in two thirds of the patients discontinuing SAS treatment and in the same proportion of the individuals who did not interrupt this medication.

[Methods and results of electrocardiography and arterial blood pressure determination with remote recording and drug administration in cattle].

An account is given in this paper of catheterisation of the common carotid artery and external jugular vein of cattle by means of silicone rubber hoses together with a method of wireconnected remote recording of blood pressure and cardiac potentials, using strong subcutaneously placed electrodes. Also described are techniques for recording of respiration through one simple, intranasally applied thermo-couple as well as for remote application of pharmaceuticals, using infusion pumps or syringes with extension tubes. Electrocardiograms (ECG) and shapes of blood curves recorded from non-medicated animals were analysed. Cattle ECG usually exhibited a small positive P-spike, one ventricular complex usually made up of one small positive R-spike and one large negative S-spike as well as one large positive T-spike. The pericardial arterial blood-pressure curve was found to rise, without preliminary waves, to two main waves separated from one another by a clear incisure, before it dropped steeply, later on more smoothly and oscillating to the enddiastolic pressure point.

bSTH and IGF-I levels in blood and milk of black and white dairy cows before and after application of recombinantly derived bovine somatotropin.

In an 8 week trial period six black and white dairy cows were injected with Optiflex, a slow release formulation of recombinantly derived bovine somatotropin (BST). Three cows each were injected at two-weeks (BST-14) or four weeks (BST-28) intervals. The dosage was 640 mg BST per injection. Average milk yield increased by 18.3% and 13.8% in groups BST-14 and BST-28 respectively. Blood plasma levels of bSTH and IGF-I increased till day 10 post-injection; thereafter they declined, except IGF-I levels in group BST-14 which persisted continuously elevated with minor variations. No bSTH above detection level could be assayed in the milk of treated animals. The IGF-I level in milk was slightly elevated after BST-application but stayed within the normal range.

Results of a multicenter placebo-controlled double-blind randomized phase III clinical study of treatment of rheumatoid arthritis with recombinant interferon-gamma.

In a multicenter placebo-controlled double-blind randomized clinical study, 91 patients with rheumatoid arthritis were given 28 days' treatment with recombinant interferon-gamma (50 micrograms daily for 20 days, then 50 micrograms each second day up to day 28, given by subcutaneous injection). The aim of the study was to provide a methodologically clear demonstration of the efficacy of treatment with interferon-gamma, using criteria that could be handled by statistical tests. Evaluatable documentation was available for 79 patients, of whom 40 were treated with the active compound. The principal criterion for the statistical evaluation of the therapeutic success was improvement of the Ritchie "joint pain index" or Lansbury "joint pain index" by at least 30% within 28 days. The chi-square test showed superiority of the interferon arm over the placebo arm with an error probability of alpha less than 1%. In addition, efficacy of interferon-gamma was demonstrated in respect of practically all parameters investigated. The frequency of side-effects, including febrile reactions, was the same for the active compound and the placebo. During interferon treatment the daily maximum body temperature was raised by 0.3 degrees C on average, but was below 37.2 degrees C at all times.

Human dirofilariasis with reactive arthritis--case report and review of the literature.

A 43-year-old German man suffered from an insect bite, which was followed by a severe local inflammation and lymphadenitis. A year later, he developed dry cough, chest pain, and an oligoarthritis of the ankle and knee joints. While other causes of the symptoms could be excluded, serologic findings suggested a dirofilarial infection with reactive arthritis due to the parasite. The observation points out, that in cases of unclassifiable mono- or oligoarthritis, reactive arthritis due to parasites should be considered in the spectrum of differential diagnoses. The disease is rare at present, but its frequency might be underestimated because of the diagnostic difficulties. Dirofilarial infection should be borne in mind after a stay abroad. The present case, however, shows, that the disease also occurs in patients who had not travelled to regions of risk in recent years.

A three-year prospective study of systemic manifestation in rheumatoid arthritis.

Rheumatoid arthritis (RA) as a systemic disease can attack many other organs in addition to the joints. A variety of pathological lesions of the blood vessels are responsible for the extra-articular features (EAF). In the present study, we investigated firstly whether the presence of blood vessel changes in one organ--namely the skin--may indicate blood vessel pathology and, consequently, EAF in other organs. Secondly, we investigated the number of EAF in individual patients with RA, and observed whether this changed during the course of the disease. Fifty-one RA-patients (40 female, 11 male; ages had a mean of 49.5, minimum 19, maximum 73 years; mean duration of RA was 7.3, minimum 0.25, maximum 41 years) were included in the study. Punch biopsies from the posterior calf were examined immunohistologically for vessel wall immune deposits. Further, EAF were determined by means of instrumental clinical methods such as pulmonary function test, echocardiography, electromyography, and nerve conduction velocity measurement. At the first investigation 21/51 patients had skin vessel wall immune deposits (SVWID). Five patients--all showed SVWID at first investigation--died during the three-year investigation period, 10 patients could not be followed-up for unknown reasons; the skin biopsy of one patient could not be assessed. At the final investigation, we found SVWID in 11/35 patients. SVWID-positive patients had more EAF compared to SVWID-negative patients; this was true both, at the first investigation (1.85 EAF/patient vs 1.05 EAF/patient) and at the final investigation (1.91 EAF/patient vs 0.67 EAF/patient).(ABSTRACT TRUNCATED AT 250 WORDS)

The importance of skin vessel wall immune deposits in the course of the systemic and articular features of rheumatoid arthritis. A prospective study.

Fifty-one rheumatoid arthritis patients were examined prospectively during a 3-year investigation period by means of immunohistological, histological, and instrumental clinical methods. Skin vessel wall immune deposits (SVWID) were found in 21/51 patients at the first and in 11/35 patients at the concluding investigation. Patients with SVWID showed more extra-articular features, more rapid progress of joint damage, higher mortality frequency, higher inflammatory activity, and higher levels of circulating immune complexes, compared with patients without SVWID.

Sonography and NMR imaging in rheumatoid gonarthritis.

Sonography and NMR imaging of the knee joint make it possible to obtain a visual representation of the thickened synovial membrane occurring in patients with rheumatoid gonarthritis, even before inflammatory deformation is radiologically detectable. Both methods can expose effusions and Baker's cysts. In NMR imaging, an indication of the inflammatory genesis can be obtained from the extended relaxation time T1. Due to the good representation of soft tissues, prognostic evidence may be gained of whether a mainly exudative or proliferative form is present.

Correlation of inflammatory RA disease activity with laboratory parameters.

Fifty-one patients with rheumatoid arthritis were examined at 6-month intervals during a 2-3-year investigation period. Correlations of an inflammatory activity index calculated from nine criteria with several laboratory parameters were computed. ESR correlated most closely, followed by the IgM concentration of circulating immune complexes, hemoglobin, C3A, and fibrinogen.

Generalized vasculopathy and finger blood flow abnormalities in rheumatoid arthritis.

Resting blood flow, spontaneous variations of resting blood flow and postischemic peak flow of the fingers, measured by venous occlusion plethysmography, were reduced in patients with rheumatoid arthritis (n = 39) as compared to normals (n = 18). Resting blood flow and postischemic peak flow were reduced only in patients without skin vessel wall immune complex (IC) deposits, whereas spontaneous variations of resting blood flow were found to be reduced in patients with IC deposits. However, after inducing sympathicolysis, postischemic peak flow was reduced in both groups. These findings were consistent with the presence of organic lesions of the finger arteries in both groups associated with additional functional vasospasm in patients without complex deposits, that in the other group may have been counteracted by the presence of IC deposits.
