Activation of two discrete signaling pathways by erythropoietin.

Erythropoietin stimulation of erythroid cells induces a rapid increase in c-myc and decrease in c-myb mRNA levels. The signal pathway to c-myc requires activation of protein kinase C. We now report that erythropoietin down-regulates expression of c-myb via a discrete, serine/threonine-specific phosphatase-dependent pathway. The protein kinase C-blocker H7 completely prevents the c-myc response to erythropoietin, but has no effect on the c-myb response. In contrast, the phosphatase blocker okadaic acid prevents the c-myb response but not the c-myc response. This effect of okadaic acid on the c-myb response is concentration-dependent. Both the protein kinase C-dependent signal to c-myc and the phosphatase-dependent signal to c-myb regulate gene expression by a transcriptional arrest mechanism operative within the first intron of the respective protooncogenes. In contrast, the chemical inducer of differentiation, dimethyl sulfoxide, regulates expression of c-myc and c-myb without activation of these phosphatase- and kinase-dependent pathways.

Age-associated changes in mitogen-induced protein phosphorylation in murine T lymphocytes.

Since T lymphocyte proliferation declines with age, and since activation of a still only partially defined set of protein kinases is thought to play a critical role in T cell activation, we have carried out a systematic survey of age-related changes in protein phosphorylation in T lymphocytes. We used two-dimensional electrophoretic analysis to examine lysates prepared from T cells after 10 min of exposure to mitogens [anti-CD3, concanavalin A (Con A) and phorbol 12-myristate 13-acetate (PMA) plus ionomycin] and nonmitogenic activators (PMA or ionomycin used separately). Our results show a progressive, life-long decline in the levels of anti-CD3-induced phosphorylation of all 16 phosphoproteins that respond vigorously in T cells of young mice. Mice 10-18 months of age showed levels of response that were clearly below those induced in young mice, while responses of mice 22-24 months of age were even more severely diminished. Responses to Con A, PMA, ionomycin and a combination of PMA plus ionomycin were equally blunted in old mice, except for 2 (of 12) phosphoproteins that continued to respond to PMA even in the old animals. None of the phosphoproteins which were ionomycin responsive in young mice continued to respond in old mice, although 1 (of 3) ionomycin-inhibitable phosphoproteins remained inhibitable in old mice. We also found three phosphoproteins which became phosphorylated in response to anti-CD3, PMA and Con A (but not ionomycin) only in old mice, and a pair of phosphoproteins which were unresponsive to all mitogens but showed a higher "baseline" phosphorylation in T cells from old mice. Comparing phosphoprotein patterns between CD8- and CD8-CD45RB- T cells from young mice allowed us to identify nine phosphoproteins that were strongly responsive to anti-CD3 only when CD45RB+ (i.e. virgin) cells were present. Although the immune system of old mice consists largely of memory T cells, the change in phosphoprotein phosphorylation patterns cannot simply be explained by the accumulation of this cell type. We conclude that aging leads to a global impairment of several distinct protein kinase pathways in both virgin and memory T cell sets.

Alpha-tocopherol deficiency fails to aggravate toxic liver injury but liver injury causes alpha-tocopherol retention.

The possible aggravation of liver injury by impaired cellular antioxidant function was investigated. A vitamin E-deficient diet (0.5 mg/kg alpha-tocopherol; control 100 mg/kg) significantly reduced rat liver alpha-tocopherol concentrations after 4 weeks (1.8 +/- 1.7 micrograms/g; control 34.4 +/- 2.4 micrograms/g, p < 0.001). The effects of copper loading (Cu, 3 g/kg diet); galactosamine (GalN, 0.85 g/kg i.p.); or carbon tetrachloride (CCl4, 10 mmol/kg i.p.) were examined. Serum aspartate transaminase activity was elevated slightly by vitamin E deficiency but not by hepatic copper accumulation. In vitamin E-replete (E+) and vitamin E-deficient (E-) rats, GalN or CCl4 caused a large and comparable elevation in serum AST and OCT activity. This effect on AST was markedly reduced by copper loading in vitamin E replete (E+) rats, but in E(-) rats copper had significantly less protective effect. Copper also diminished the OCT response to GalN in E+, though not E-, rats. A significant rise in total hepatic alpha-tocopherol content followed administration of GalN or CCl4 in both normocupric and copper-laden E(-) rats. Thus alpha-tocopherol deficiency (a) was not hepatotoxic per se; (b) failed to potentiate the toxicity of copper, GalN or CCL4; but (c) partially abolished the protection by copper against toxin-induced liver injury. Retention of hepatic alpha-tocopherol after liver damage may partly explain low serum vitamin E levels seen in clinical liver disease.

Analysis of protein phosphorylation patterns reveals unanticipated complexity in T lymphocyte activation pathways.

Protein kinases are considered likely to play important roles in the still dimly understood process by which mitogens induce resting T lymphocytes to enter the cell cycle. Using two-dimensional electrophoretic analysis of lysates from orthophosphate-labeled cells, we have compared patterns of phosphorylation in freshly isolated murine splenic T cells exposed to three mitogenic agents: antibody to the epsilon-chain of the TCR CD3 complex, the plant lectin Con A, and a mixture of PMA and ionomycin, which together bypass the signal transduction apparatus to activate intracellular pathways. Of 14 phosphoproteins found whose level of phosphorylation was increased (at least fivefold) by anti-CD3 epsilon antibody, 13 also responded to the mixture of PMA and ionomycin. Surprisingly, however, only 5 of these 14 also responded strongly to Con A exposure. We also identified two substrates that were phosphorylated in response to Con A but not to anti-CD3. Phosphorylation patterns were also studied in T cells exposed to either PMA or ionomycin alone, to gain further insight into the role of protein kinase C and calcium-dependent events in the activation process. Of 16 phosphoproteins that responded to mixtures of PMA and ionomycin, 4 were shown to require the ionomycin signal, 2 to require the PMA signal, and 3 others to respond only when both activators were present; the other 7 responded to either agonist added alone. In addition, we found two PMA-sensitive phosphoproteins in which phosphorylation was inhibited by ionomycin induced calcium signals. Finally, we identified several phosphoproteins which show differential responsiveness in CD4+ and CD8+ T cells. Classification of kinase substrates based on their differential susceptibility to these stimuli should provide new insights into the mode of action of agents and diseases that affect T cell activation.

A quantitative histochemical study of D-amino acid oxidase activity in rat liver in relationship with feeding conditions.

The histochemical method for the demonstration of D-amino acid oxidase activity in rat liver, based on the use of cerium ions and the diaminobenzidine-cobalt-hydrogen peroxide procedure, was improved by the application of unfixed cryostat sections and a semipermeable membrane interposed between section and gelled incubation medium. The amount of final reaction product precipitated in a granular form was about four times higher with this technique in comparison with conventional procedures using fixed sections and aqueous incubation media. The specificity of the reaction was proven by the 70% reduction of the amount of final reaction product when incubating in the presence of substrate and D,L-beta-hydroxybutyrate, a specific inhibitor of D-amino acid oxidase activity. Cytophotometric analysis of liver sections revealed that the specific test minus control reaction was linear with incubation time and section thickness. The Km value of the enzyme of 10.3 +/- 2.7 mM, as determined in periportal areas, is about five times the value found with biochemical methods in liver cell homogenates. The enzyme activity in periportal areas is about five times the activity in pericentral areas. Fasting (24 and 48 hr) induced a significant decrease in D-amino acid activity in periportal and pericentral areas. The possible physiological role of the enzyme in liver is discussed.

Quantitative cytochemical detection of malignant and potentially malignant cells in the colon.

It was found to be possible to distinguish malignant cells from normal cells by using an oxygen-sensitive tetrazolium salt (neotetrazolium) for the histochemical demonstration of glucose-6-phosphate dehydrogenase activity in cryostat sections of human colon. We have studied 12 cases of established adenocarcinoma of the colon in addition to 4 of ulcerative colitis and 4 of adenomatous polyposis (polyposis coli). In a nitrogen atmosphere the activities of malignant and normal cells were similar. However, after incubation in an atmosphere of pure oxygen, only malignant cells gave a positive reaction after 5 min. Three of the four cases of adenomatous polyposis gave a positive reaction for glucose-6-phosphate dehydrogenase activity in oxygen in a manner similar to that of specimens with severe dysplasia. In general, positive foci were histologically indistinguishable from the neighboring tubuli. However, foci of severely dysplastic epithelium usually showed a positive reaction. All three patients eventually developed clear-cut severe dysplasia. The other patient, who showed a negative reaction in oxygen, was diagnosed after 3 years as not suffering from dysplasia. All cases of ulcerative colitis gave a reaction in oxygen comparable with that of normal cells. Therefore, the areas with a positive reaction are considered to be either in the process of malignant transformation or malignant. An explanation for the oxygen insensitivity of cancer cells appeared to be a decrease in the activity of superoxide dismutase (EC 1.15.1.1), as addition of exogenous superoxide dismutase to malignant cells caused a normal reaction. We wish to suggest that this test in combination with the routine histology may be exploited for the diagnosis of polyps in premalignant conditions.

Light microscopical detection of D-amino acid oxidase activity in unfixed cryostat sections of rat kidney and liver using the cerium-DAB-cobalt-H2O2 procedure and a semipermeable membrane.

The two-step method for the light microscopical detection of oxidase activity using cerium ions and a visualization procedure with diaminobenzidine and nickel or cobalt ions as introduced by Anger-müller and Fahimi, has been improved. Inactivation by fixation and leakage of D-amino acid oxidase molecules were avoided by the application of unfixed cryostat sections adhered to a semipermeable membrane separating the sections from a gelled incubation medium. Optimum activity in rat kidney and liver was obtained by increasing the cerium concentration from 3 to 30 mM and the Tris-maleate concentration from 100 to 200 mM in the incubation medium for the first step. D-proline was used as substrate and a concentration of 20 mM gave maximum activity. The second visualization step was performed in a medium consisting of DAB, cobalt and hydrogen peroxide. Final reaction product was found in a granular form in the basal part of the epithelial cells of the proximal tubules and in liver parenchymal cells. The activity in periportal areas was much higher than in pericentral areas. Cytophotometric analysis revealed that a 30 times higher activity could be detected when using the membrane technique in comparison with incubations in aqueous media. The specificity of the reaction was proven by a nearly complete inhibition of the reaction by beta-hydroxybutyrate.

Cellobiose: mannitol differential permeability in small bowel disease.

Cellobiose and mannitol absorption were studied in patients with suspected abnormal function of the small bowel mucosa. The urinary cellobiose:mannitol ratio was increased in subtotal villous atrophy, iron deficiency anaemia, and small intestinal Crohn's disease. The test seems a sensitive indicator of the integrity of small bowel mucosa.

Copper in urine and hair in Indian childhood cirrhosis.

In advanced Indian childhood cirrhosis (ICC) urine copper concentration was higher (range 416-103,448 mg/g creatinine) than in other hepatic diseases (range 67-10,303 mg/g creatinine). In early ICC urine copper concentration was more modestly raised (1188-9470 mg/g creatinine), but rose to high values (2222-42,819 mg/g creatinine) after a single dose of penicillamine 20 mg/kg. A post-penicillamine urinary copper:creatinine ratio greater than 10,000 mg/g supports a diagnosis of ICC. The concentration of copper in the hair, while increased in advanced ICC, is of no diagnostic value in early cases.
