Low expression of human histocompatibility leukocyte antigen-DR is associated with hypermethylation of human histocompatibility leukocyte antigen-DR alpha gene regions in B cells from patients with systemic lupus erythematosus.

The relationship between the expression of HLA-DR antigens and the HLA-DR alpha gene methylation was examined in systemic lupus erythematosus (SLE). Using permanent B cell lines, we found reduced DR expression in SLE. The low DR expression was correlated with high anti-DNA antibody titers in patients' sera. The amounts of DR alpha message were lower in SLE cells than in normal controls, suggesting that the low expression of DR antigens is associated with gene functions. The extent of DNA methylation was examined at five CCGG sites in the HLA-DR alpha locus. DNA from both SLE and normal cells showed variable methylation patterns. Since the DR alpha gene is a single-copy gene, such a variability is the result of assaying a mixture of transformed clones containing methylated DR alpha gene, with other clones containing unmethylated DR alpha gene. A distinctive feature of normal cells was a consistent methylation pattern: 12 normal cell lines showed exactly the same pattern. In contrast, 28 SLE cell lines showed a cell-line-specific methylation, and hypermethylation at the DR alpha locus. The hypermethylation is often associated with transcriptionally inactive genes. Thus, our results suggest that (a) B cells with hypermethylated DR genes might express no or few DR antigens; (b) the ratio of cells with differently methylated DR genes is consistent in normal individuals, while, in SLE patients, cells with hypermethylated DR genes predominate, resulting in apparently reduced DR antigen expression; and (c) the aberrant DR expression could be associated directly with immunoregulatory dysfunctions in SLE disease.

Relationships between red cell cation contents and blood pressure level.

Red cell sodium (R-Na), potassium (R-K) and sodium to potassium ratio (R-Na/K) were studied in 2,542 adults participating in periodic medical examinations. The mean values of R-Na and R-Na/K in untreated borderline (n = 406) and definite hypertensives (n = 485) were higher than those in normotensives (n = 1,651), but that of R-K was not different among the groups. The increased R-Na and R-Na/K in the hypertensives were observed only in those under 50 years, but not in those 50 years and over. In the normotensive subjects, age and sex affected the intraerythrocyte cation contents, but a positive family history of hypertension did not. Although a few characteristics correlated with both R-Na and mean blood pressure, and with both R-Na/K and mean blood pressure, multiple regression analyses revealed that R-Na and R-Na/K independently and significantly contributed to the variation in mean blood pressure. It was also indicated that these relations of R-Na and R-Na/K to mean blood pressure were significant in those under 50 years, but not in those 50 years and over. While several characteristics confound the relationships between red cell cation contents and blood pressure, these results suggest that the contributions of R-Na and R-Na/K to the mechanisms of human hypertension still persist in young to middle-aged people.

Abnormal relationship between dietary sodium intake and red cell sodium transport in salt-sensitive patients with essential hypertension.

The effects of high sodium intake on erythrocyte 22Na efflux rate constants were studied in 25 patients with essential hypertension and 9 normal subjects. With changes in sodium intake from 100 mEq to 300 mEq/day, both total and ouabain sensitive 22Na efflux rate constants decreased significantly (p less than 0.001) in "salt-sensitive" patients (-0.031 +/- 0.005 and -0.035 +/- 0.006 /hr, respectively), but these responses were variable in "nonsalt-sensitive" patients and in normal subjects. The "salt-sensitive" patients showed a significant increase in their body weight, while intraerythrocyte sodium contents remained unchanged in the both groups. These results suggest that the abnormal change in membrane Na-K-ATPase activity may, at least in part, be involved in the mechanism of sodium susceptibility in patients with essential hypertension.

Blood glucose kinetics in whole body and mammary gland of lactating goats exposed to heat.

Whole-body and mammary kinetics of blood glucose were measured in lactating goats exposed to thermoneutral, moderate hot, and severe hot environments for 4 d. Milk yields were reduced by 3 and 13% during moderate and severe heat exposure, but heat production was unchanged during the experiment. Concentrations of plasma glucose and free fatty acids did not change during heat exposure. Concentration of thyroxine tended to decrease and concentration of prolactin increased with increasing temperature. Whole-body turnover of blood glucose decreased significantly during both moderate and severe heat exposure. Blood glucose oxidation rate and contribution of blood glucose to total carbon dioxide production were not influenced by heat exposure. Mammary glucose uptake tended to decrease during heat exposure, and this reduction may account for the decreased whole-body turnover of blood glucose. The lactose concentration in milk was decreased on the 4th d of severe heat exposure. A relatively low production of milk lactose was apparently derived from blood glucose. These results suggest that the whole-body turnover of blood glucose decreases in step with a decrease in mammary glucose uptake during heat exposure in lactating goats.

Chemical modification of spiramycins. V. Synthesis and antibacterial activity of 3'- or 4''-de-N-methylspiramycin I and their N-substituted derivatives.

The 3'- and 4''-de-N-methylspiramycins were synthesized selectively, and then were converted to various N-substituted derivatives. 4''-De-N-methyl derivatives were more active than 3'-de-N-methyl ones. Among the derivatives, 4''-N-Fmoc-glycyl and 4''-N-benzyl-4''-de-N-methylspiramycin I were the most active in vitro, and were comparable to spiramycin I. 4''-De-N-methylspiramycin I was about half as active as spiramycin I in vivo.

Tac antigen-positive T cells activated in autologous mixed lymphocyte reaction regulate the generation of killer T cells against hapten-modified autologous cells.

T cells that proliferate in the autologous mixed lymphocyte reaction (auto-MLR) have been shown to acquire some suppressor or regulatory activities. In the present study, we examined the suppressive effects of T cells activated in the auto-MLR on the induction of hapten-specific cytotoxic T cells. NRFT (depletion of ARFT from UT) were used as the responder cells of TNP-MLR. After primary and secondary TNP-MLR, the cells were harvested and tested for their cytotoxic activities against TNP-modified autologous cells by 51Cr-release assay. When UT cells cultured for 1 wk in auto-MLR were added to primary TNP-MLR at the beginning of culture, the cytotoxic activity tested at the end of the culture was suppressed from 15.6% +/- 2.7 to 5.8% +/- 1.1 (percent cytotoxicity, mean +/- SE). However, these auto-MLR-activated UT cells had little suppressive activity against cytotoxic T cells when they were added to the final assay of TNP-CTR. Suppressive activities of these cells on the generation of cytotoxic T cells during secondary TNP-MLR were also tested. The addition of auto-MLR-activated UT cells to the secondary TNP-MLR at the beginning of the culture reduced the cytotoxic activities of NRFT from 23.8% +/- 2.3 to 9.7% +/- 1.7 after secondary TNP-MLR. Allo-activated T cells, PHA blasts, and fresh autologous T cells were used as the controls, but none of the cells had suppressive effects on the generation of CTL. Characteristics of these suppressor cells were examined. Auto-MLR-activated cells from ARFT fractions exhibit very powerful suppressor activity. Treatment of the auto-MLR-activated T cells with mitomycin C eliminated their suppressive effects on the generation of CTL; 21.2% +/- 6.3 of UT cells became anti-Tac positive after 1 wk of auto-MLR. Treatment of auto-MLR-activated UT cells with anti-Tac antibody plus complement eliminated their suppressive activities on the induction of CTL. Thus, T cells stimulated in auto-MLR were shown to have suppressive effects on the induction of cytotoxic T cells against TNP-modified autologous cells. These cells were mitomycin C sensitive. Because anti-Tac antibody is reactive to activated T cells, activation of T cells during auto-MLR was thought to be necessary for the acquisition of the suppressive activity.

[Experimental and clinical study of reduction surgery combined with chemotherapy of primary liver cancer].

The effects of reduction surgery combined with chemotherapy used by MMC and ADM for DAB induced hepatoma were investigated. Results are as follows. Reduction surgery prolonged the survival time of rats with DAB hepatoma in which tumor was transplanted either in liver or subcutaneous tissue. Reduction surgery in combination with chemotherapy made survival time more prolonged in these rats with than reduction surgery alone. Reduction surgery combined with intraarterial infusion chemotherapy was carried out in three patients with advanced primary liver cancer. One survived for 9 months and another for 3 years after right trisegmentectomy and postoperative arterial infusion chemotherapy. The other is till alive without any recurrence 7 months after right trisegmentectomy with partial resection of a intrahepatically metastasized tumor in the lateral segment and arterial infusion chemotherapy used by ADM. In summary, it is suggested that reduction surgery in combination with chemotherapy is beneficial as a multidisciplinary treatment for advanced primary liver cancer.

[Laser acupuncture therapy for pain and vague complaints in neurosurgery].

The value of diode laser acupuncture for pain and other vague neurological complaints has been studied in chronic patients of head trauma and of cerebrovascular disease or neuralgia in head and neck regions. Gallium-Aluminium-Arsenide (Ga-Al-As) diode laser pain attenuator PANALAS-4000 manufactured by Japan Medical Laser Laboratory was used in the following specifications; wave length: 830 nm, power: 20-40 mW, frequency: 5-50 c.p.s., pulse duty: 0.9. The laser acupuncture was applied to some of the representative Chinese meridian points, local pain points and related nerves for 1 to 7 times in a week. It was given in 310 patients in our neurosurgical department. 175 of 227 patients who complained of pain and 12 of 39 patients who complained of numbness showed improvement within 24 hours, another 24 patients with pain and 16 patients with numbness showed improvement after 24 hours. No side effect was noticed. The duration of the effectiveness was about 2 or 3 days. It is very useful to apply the laser acupuncture for pain and vague complaints in neurosurgery.

[Concept and problems of the early stage of primary liver cancer].

Study of 25 patients with less hepatic tumor than 5 cm in diameter was made on pathohistology and prognosis. In relation of portal vein embolus to tumor size, portal vein embolus (vp1) developed histologically in four (30.8%) to thirteen patients with less than 3 cm tumor in diameter, but in eleven (91.7%) of twelve with more than 3 cm tumor diameter. The accumulative survival rate of patients with less than 3 cm tumor in diameter was 92.3% at one year, 78.0% at three and five years. It is more favorable than survival rate of patients with more than 3 cm tumor in diameter. Therefore, at present it seems to be adequate that tumor size is less than 3 cm, portal vein embolus (vp1) is negative and solitary as concept of early stage of primary liver cancer. Most patients with liver cirrhosis do not meet well functional reserve of the liver and minimized regional resection is often obliged. However we can have favorable results, if the portal vein embolus (vp) and intrahepatic metastasis (im) surrounding the tumor are identified by echogram during operation and resected with tumor. Its accumulative survival rate was 87.9% at one year, 72.5% at three and five years.

Application of high-performance liquid chromatography with synchronized accumulating radioisotope detector to analysis of glyceryl trinitrate and its metabolites in rat plasma.

A new, sensitive, and specific high-performance liquid chromatographic method for the quantitative analysis of [14C]glyceryl trinitrate and its four metabolites in plasma is described. The drugs are extracted from 0.05 ml of plasma with methanol and analyzed by high-performance liquid chromatography using a synchronized accumulating radioisotope detector. The limit of detection is 0.2 ng per injection. The within-day coefficient of variation is 5.9% at a concentration of 27.0 ng per ml of plasma. The method was applied to single-dose pharmacokinetics of glyceryl trinitrate in rat.

Effect of dehydroepiandrosterone sulfate on collagenase production in rabbit uterine cervix culture.

Rabbit uterine cervical explants were found to produce a typical collagenase, latent form, in tissue culture, 4-Aminophenylmercuric acetate and trypsin were potent activators of the enzyme. The enzyme was purified simply in one step of CM-52 cellulose ion-exchange chromatography, and then further characterized. Addition of dehydroepiandrosterone sulfate (DHAS) to culture medium significantly stimulated collagenase production, but DHAS did not directly activate the enzyme. In addition, dehydroepiandrosterone and 17 beta-estradiol, the main metabolites of DHAS in vivo, depressed enzyme production. Our previous result, that increases in cytoplasmic DHAS-binding protein in rabbit uterine cervices parallel the progress of pregnancy, and these results suggest that DHAS might have direct actions toward cervical ripening.

Effect of anti-alpha-fetoprotein serum on human hepatoblastoma.

There is no definitive treatment of hepatoblastoma. Except for radical resection, the prognosis of unresectable cases remains pessimistic. Therefore, the development of new anticancer drugs for this tumor is urgently needed. Using the human hepatoblastoma cell line (c-HB-3) we demonstrated that horse anti-alpha-fetoprotein containing serum suppressed proliferation of the hepatoblastoma cells. This effect was dependent on the duration of exposure and the concentration of anti-alpha-fetoprotein serum.

Loss of chloroplast DNA methylation during dedifferentiation of Chlamydomonas reinhardi gametes.

In Chlamydomonas reinhardi the chloroplast DNA (ch;DNA) of mating type plus cells undergoes cyclical methylation and demethylation during the life cycle. Methylation occurs during gametogenesis, and fully differentiated gametes can be dedifferentiated back to vegetative cells which contain nonmethylated chlDNA by the addition of a nitrogen source for growth. We examined the dedifferentiation process and found that the mating ability of gametes was lost rapidly after the start of dedifferentiation at a time when the chlDNA was still methylated. The enzymatic activity of the 200-kilodalton DNA methyltransferase was lost at a rate consistent with the rate of dilution during cell division. Methylation of chlDNA decreased at a slower rate than was expected from cell division alone but was consistent with the continuing activity of the preexisting methyltransferase so long as it was present. These results support the hypothesis that demethylation of chlDNA occurs by dilution out of enzymatic methylating activity rather than by enzymatic demethylation.
