Peripheral blood leukocytes and lymphocyte subpopulations as determined by flow cytometric measurements in healthy children.

Peripheral venous blood leukocyte subpopulations were determined in 69 healthy children and compared in two age groups: 8 to 12 years and 13 to 17 years, the period of pubescence and intensive growth of the organism. As compared to work of other authors, more leukocyte subpopulations were recognized in the examined large group of children. Whole blood monocyte and total lymphocyte absolute counts decreased with age as well as the absolute counts of T lymphocytes and CD8+ T lymphocytes. Percentage of CD4+ cells and CD4/CD8 ratio were higher in the older age group. Percentage as well as absolute counts of CD19+ CD5+ cells and CD19+ CD10+ cells are given.

Myoinositol incorporation into lymphocytes of chronic renal failure patients is impaired.

Incorporation of myo-[2-3H]-inositol into peripheral blood mononuclear cells (PBMNC) and T-cell enriched lymphocytes was evaluated in in-vitro experiments in chronic renal failure (CRF) patients and healthy subjects. Incorporation of myo-[2-3H]-inositol into the cells of CRF patients on conservative and haemodialysis treatment was found to be impaired in comparison with that observed in normal cells. Following PHA stimulation of the cells of CRF patients myo-[2-3H]-inositol incorporation decreased even further, while it increased in normal cells. Five-hour haemodialysis session significantly depressed myoinositol incorporation into PBMNC, while its incorporation into T-cell enriched lymphocytes remained unaffected. Myoinositol incorporation into PBMNC and T-cell enriched lymphocytes was inhibited by prostaglandins and leukotrienes and was inversely related to the extent of pertussis toxinsensitive G protein activation. Reduced myoinositol incorporation into uraemic PHA-stimulated PBMNC may depend at least in part on their enhanced PGE2 and LTB4 release accompanied by increased intracellular cAMP production. In CRF impaired myoinositol incorporation into immune cells may prove the disarrangement in the early events of transmembrane signal transduction, which may share the responsibility for the cell-mediated immune defect in these patients.

[Adhesion and aggregation of neutrophils in patients with unstable angina].

The adherence and aggregation abilities of neutrophils (PMNs) were evaluated in thirty three patients with unstable angina, who were qualified for PTCA procedure. The control group consisted of forty one clinically healthy persons. The blood for investigations was obtained from coronary sinus and basilic vein just before the procedure, while in the control group from basilic vein only. The adherence of PMNs to plastic surface (rest and stimulated by PMA) was estimated in vitro according to Oez's et al. method by measuring optical density of generated formazan, whereas the aggregation of PMNs was evaluated using the leukergy test according to the method of Fleck in Berliner's and Aronson's modification. In patients with unstable angina statistically significant higher (p < 0.001) adherence of peripheral blood PMNs, compared with control groups was found (patients: rest-0.525 +/- 0.245, stimulated-0.839 +/- 0.419, control group: rest-0.260 +/- 0.129, stimulated-0.522 +/- 0.377). The aggregation of peripheral blood PMNs was significantly higher (p < 0.05) in the sick than in the control group (the sick-10.98 +/- 4.29%, controls-4.65 +/- 3.01%). No differences in investigated parameters of PMNs obtained from peripheral or coronary sinus blood were found.

The effect of short-term myocardial ischemia on the expression of adhesion molecules and the oxidative burst of coronary sinus blood neutrophils.

We examined the influence of transient myocardial ischemia on the number and function of neutrophils in patients with effort angina (EA). We tested fluorometrically the expression of neutrophil membrane molecules (CD11b, CD11c, CD18) and neutrophil oxidative burst using a chemiluminescence (CL) generation system. The estimations were conducted before, 1 min after and 20 min after percutaneous transluminal coronary angioplasty (PTCA) in 15 patients qualified for the treatment because of single-vessel disease. Eight EA patients subjected to coronary arteriography (CA) comprised a control group. We did not observe any marked changes in leucocytosis or lymphocyte number in peripheral blood (PB) or in coronary sinus blood (CSB) after the procedure. The percentage of granulocytes in coronary blood decreased significantly 20 min after reperfusion. No significant changes in white blood cell count were noted in peripheral blood of PTCA patients or in control CA subjects. Oxidative burst of nonstimulated and fMLP, PMA and zymosan stimulated sinus blood neutrophils was significantly depressed 1 min after inflation, and enhanced 20 min after reperfusion. We found a significant increase in the percentage of the CD11c+ neutrophils from 56.7 +/- 7.4% to 64 +/- 6.5% 20 min after inflation and postischemic decrease in the CD11c molecule expression on CSB neutrophils. Significant positive linear correlation (Rval = 0.71) between inflation time and the CD11c molecule expression on CSB immediately after reperfusion was also noted. The results may reflect local activation of neutrophils in ischemic myocardium as a response to ischemia induced increase of activating stimuli.

T lymphocyte subpopulations and lymphocyte proliferative activity in normal and pre-eclamptic pregnancy.

The purpose of this study was to assess whether any changes occur in the cellular immunity in normal and pre-eclamptic pregnancy. T lymphocyte subpopulations and the lymphocyte proliferative responses to mitogens (PHA, Con A, PWM) in the fetal calf serum were examined in normal pregnant and pre-eclamptic primiparas in the third trimester of pregnancy. In normal pregnancy the absolute and percentage numbers of CD3+ and CD4+ T cells were significantly lower and the CD4+/CD8+ ratio almost halved, in comparison with non-pregnant subjects. In the pre-eclamptic women a decreased absolute and percentage content of CD8+ T cells and increased percentage of CD3+ and CD4+ lymphocytes were found--in comparison with the normal pregnant women--which led to an almost 2.5-fold increase of the CD4+/CD8+ ratio. No disorders were found in the lymphocyte proliferative responses to mitogens in either normal or pre-eclamptic pregnancy. We conclude that despite the shifts among T lymphocyte subsets, mitogen-induced lymphocyte proliferation presents its functional stability and unchanged reactivity in normal and pre-eclamptic pregnancy.

Immunological status of competitive cyclists before and after the training season.

The total number of leucocytes, T lymphocyte subsets, mitogen induced proliferation of lymphocytes, Il-2 generation, adherence capacity and chemiluminescence of granulocytes were measured and a leukergy test performed in fifteen young cyclists. The investigations were carried out at rest at the beginning of a training season and after six months of intensive training and a racing season, cycling approximately 500 km a week. Baseline values of the tested immune parameters were within the range observed in 16 non-trained healthy controls except significantly increased non stimulated neutrophil chemiluminescence. The second cyclo-ergometer test in August showed a marked improvement in the performance capacity of the cyclists. Significant decrease in absolute numbers of CD3+ and CD4+ cells, diminished IL-2 generation and fMLP and PMA stimulated chemiluminescence of neutrophils were noted. Surprisingly, a marked increase in lymphocyte proliferation induced by PHA and anti-CD3 MoAb and normalisation in non stimulated neutrophil chemiluminescence were also observed at rest after the training season. We conclude that long-lasting intensive training may result in significant alterations in lymphocyte number and composition and in neutrophil oxidative burst capacity, but their actual significance for immunity is seen controversially.

[TNF-alpha (cachectin) as a lymphokine regulator].

TNF alpha gen is localised in macrophages, granulocytes, T lymphocytes, astrocytes, NK cells. Virus and bacteria superantigens are main potent inducers of TNF alpha production. TNF alpha activate nuclear transcription factors and is responsible for production of enzymes, regulatory proteins and cytokins. Consequently this cytokine is main regulatory factor in inflammation and immune reactions.

Essential edema-proteinuria-hypertension (EPH) gestosis and gestosis superimposing pre-existing renal disease: comparison of cellular immunity parameters.

The paper is aimed at evaluation of certain indices of cellular immunity in pregnant women with gestosis superimposing pre-existing renal diseases and comparing them with the immunological condition of women with essential gestosis. T lymphocyte subpopulations and lymphocyte proliferative responses to mitogens (phytohemagglutinin--PHA, concanavalin A--Con A and pokeweed mitogen--PWM) in the fetal calf serum and autological sera were studied. The groups of examined pregnant women in the 3rd trimester did not vary with regard to gestation age, calendar age and to the severity of edema (E), proteinuria (P) and hypertension (H) gestosis symptoms. It has been found that equal changes occur in both groups of women, regarding the examined indices of cellular immunity, when referred to normal pregnant women. In the gestotic women a decreased absolute and percentage content of CD8+ T cells and increased percentage of CD3+ and CD4+ lymphocytes were found in comparison with the normal pregnant women, which led to an almost 2.5-fold increase of CD4+/CD8+ ratio. The sera of gestotic women, in comparison with the healthy pregnant group, strongly increased the proliferation of Con A and PWM-induced lymphocytes. It may then be assumed that the observed immunological changes do not coincide with the primary causes of gestosis. We suggest that immunological factors largely contribute to the development of essential gestosis and gestosis superimposing pre-existing renal disease.

Effect of Tołpa Peat Preparation on some immune parameters in healthy volunteers. Preliminary data.

Investigations of some immune parameters were carried out in healthy humans receiving TPP in doses of 50, 100, 300 and 600 mg daily for 14 days. The percent and composition of T lymphocyte subsets were studied along with the ability to generate interleukin-1 (Il-1) and interleukin-2 (Il-2) as well as spontaneous and PHA stimulated expression of receptors for Il-2 (TAC) and transferrin receptor (Tr) on lymphocytes. Lymphocytes proliferation induced with mitogens and alloantigens, generation of both Tumour Necrosis Factor alpha(TNF alpha) by monocytes and superoxide radicals by granulocytes were estimated. A 50 mg dose of TPP changed neither the T-cell percent among mononuclear cells nor the composition of T-cell subsets. A higher dosage of 100 mg of TPP caused a significant increase of the percent of T-cells on the 14th day but did not influence their subset composition. On the 14th and 28th day of the observation a statistically insignificant decrease of IL-2 generation was observed but with no change of both TAC receptor expression on lymphocytes and the ability to generate IL-1 by monocytes. TPP in the doses of 50 and 100 mg exerted a slight immunomodulatory potential in healthy humans, however the immunological parameters assessed were within the standard values. In other immune parameters examined during the administration of higher TPP doses in human groups consisted of four volunteers considerable differences have been found but individual differences in a limited experimental group and the lack of a placebo group did not allow for definite conclusions but for preliminary data only.

Comparison of the functional activities of two different preparations of human recombinant tumor necrosis factor alpha.

The role of tumor necrosis factor (TNF-alpha) in physiological and pathological reactions has resulted in a progressive increase of expensive TNF-alpha consumption for laboratory and clinical purposes. Following this trend, the first chemical synthesis of the gene for rHuTNF-alpha gene in Poland and its subsequent successful expression in E. coli was recently reported. In the present paper, we verify the in vitro biological activities of this TNF-alpha preparation (CMMS/TNF-alpha) in comparison with a commercially available preparation of TNF-alpha. We demonstrate that our TNF-alpha possesses strong cytotoxic activity against WEHI 164 (clone 13) cells, binds the p55 and p75 TNF receptors on cell lines, induces intercellular adhesion molecule-1 (ICAM-1) expression, and interleukin-6 (IL-6) and granulocyte-macrophage colony-stimulating factor (GM-CSF) release from human umbilical vein endothelial cells (HUVEC). We demonstrate its usefulness for further investigations as an effective reagent for in vitro assays.

The effect of serine and thiol protease inhibitors on the chemiluminescence of human neutrophils in investigations in vitro.

We have studied an indirect role of serine and thiol proteases in the activation of human neutrophils in vitro. Stimulation was evaluated using a chemiluminescence (CL) generation system. Receptor-dependent and receptor-independent stimuli were studied, e.g. opsonized zymosan, formyl-methionyl-leucyl-phenylalanine, platelet activating factor, phorbol myristate acetate, and calcium ionophore A23187. The serine protease inhibitors TPCK and TLCK, and thiol protease inhibitor PHMB, diminished the CL with different potencies and in a dose-dependent manner after treatment of cells with the various stimuli. Non-specific serine protease inhibitor, PMSF, and trypsin substrate TAME, showed a low inhibitory potency with respect to CL generation. Synthetic substrates for chymotrypsin (BTEE, ATEE) significantly inhibited CL with the various stimuli used with some differences in susceptibility to their inhibition. Specific chymotrypsin inhibitors diminished both the resting and activator-induced CL. We suggest that cell-bound chymotrypsin-like protease(s) is involved in the activation of signal transduction in human neutrophils after both receptor-dependent and receptor-independent stimulation.

The effect of thymic hormones (TFX) on lymphocyte subpopulations and proliferation after maximal physical effort.

The studies were performed on healthy well-trained cyclists. Maximal physical exercise was performed on a Monark bicycle ergometer according to individual schemes. Heart rate amounting to about 200 bts/min and oxygen consumption stabilization were considered as criteria for maximal physical exercise. In this study we have investigated the effect of short-term stimulation of conditioned sportsmen with thymic hormones and evaluated T cell subsets, DR antigen and transferrin receptor expression as well as mitogen-induced proliferation of lymphocytes before and after maximal physical effort. The results suggest that intensive physical exercise may be responsible for transient decrease of CD4/CD8 ratio and mitogen responsiveness, and increase of mononuclear cells number bearing HLA DR+ and CD71 antigens. These changes were modified by the treatment with thymic hormones.

T lymphocyte subsets and NK cell cytotoxicity in chronic hemodialysis patients. The effect of recombinant human erythropoietin (rHu-EPO) treatment.

We investigated subpopulations of T lymphocytes, NK cell number and cytotoxic activity in 14 chronic uremic patients on regular hemodialysis treatment. We observed a significantly decreased absolute lymphocyte number and percentage of CD3 cells. Relative numbers of CD16 cells were significantly elevated, but NK cell cytotoxic activity was within a normal range. Nine patients with chronic renal anemia on maintenance hemodialysis were enrolled in rHu-EPO treatment trial. The treatment was continued till the hematocrit level reached 30%. Each of the patients had corrected anemia and well-being. After 12 weeks of the treatment we observed in these patients decreases in CD3, CD4, CD8 and CD16 cell numbers and elevation of CD4/CD8 ratio. Cytotoxic activity of NK cells did not change significantly. Presented results indicate that chronic hemodialysis patients have significantly diminished lymphocyte number. rHu EPO treatment affects the T lymphocyte subsets inducing a deep decrease of CD8 and CD16 cell percentage leading to normalisation of the CD4/CD8 ratio.

[Certain factors of cellular immunity in clinical evaluation of women with EPH gestosis symptoms].

The applicability of clinical examination was studied in women with symptoms of medium-advanced or advanced primary gestosis of subpopulation of T lymphocytes (CD3+, CD4+, CD8+) in the peripheral blood. Also studied was the usefulness of the proliferative activity of lymphocytes in in vitro cultures: spontaneous and mitogenic (PHA, ConA, PWM)--in the environment of fetal calf serum (FCS) and autologous sera: heat-inactivated and heat-non-inactivated. It has observed that the percentages of particular T Lymphocyte subpopulations and the proportion of CD4+ lymphocytes to CD8+ lymphocytes in the peripheral blood and the mitogenic activity of the heat-non in activated autologous serum in the test of lymphocyte spontaneous blastic transformation reveal a correlation with an enhancement of particular clinical symptoms of EPH gestosis and with the birth condition of newborns.

[Spontaneous proliferative activity of lymphocytes in normal and EPH gestosis complicated pregnancy].

The spontaneous proliferative activity of lymphocytes and CD4+/CD8+ ratio has been estimated in normal and EPH gestosis-affected pregnancy; also the effects of autologous and allogenic sera on this lymphocyte activity was evaluated. Although in normal pregnancy there was a very low CD4+/CD8+ ratio, the spontaneous proliferative activity of lymphocytes and the effects of autologous sera exerted thereupon were the same in either healthy pregnant women or in non-pregnant controls, what indicates a normal cellular immunity in physiological pregnancy. In gestosis an increased proliferative activity of lymphocytes and very high CD4+/CD8+ ratio was observed, when referred to non-pregnant controls. The serum of normal pregnant women did not reveal any immunosuppressive features against the activity of both autologous lymphocytes and gestosis--affected pregnancy, whereas it clearly inhibited the lymphocyte proliferation in non-pregnant controls. A different model of the autologous sera effects on the lymphocyte activity in gestosis women--when compared with the other examined women--is an evidence for altered immuno-modulating parameters of the serum in question.

[Subpopulations of t lymphocytes and lymphocyte proliferative activity in normal pregnancy].

T lymphocyte subpopulations (CD3+, CD4+, CD8+) and the lymphocyte proliferative responses to mitogens (PHA, Con A, PWM), in the environment of fetal calf serum (FCS) were examined in 16 normal primigravidas in the third trimester of pregnancy and in 15 healthy nonpregnant women. In normal pregnant women significantly lower absolute and percentage numbers of CD3+ and CD4+ T cells and almost twice reduced CD4+/CD8+ ratio were found, in comparison with nonpregnant subjects. Despite the shifts among particular T lymphocyte subsets in the peripheral blood, non disorders were found in the lymphocyte proliferative responses to mitogens in normal pregnancy.

[Immunomodulating properties of sera in normal pregnant women].

The purpose of this study was to analyze the immunosuppressive properties of sera of normal pregnant women. Effects of pregnancy sera on the proliferation of mitogen (PHA, Con A, PWM)-induced lymphocytes were examined in healthy primiparas. The sera of normal pregnant women inhibited the proliferation of PHA- and Con A- induced both autologous and allogenic lymphocytes (obtained from nonpregnant and pre-eclamptic women), while remaining neutral towards the proliferation of PWM- induced lymphocytes in comparison with fetal calf serum. The basic immunomodulatory activity of the sera in pregnant women was contained in the heat-stable fraction.
