The role of polymorphonuclear leukocyte (PMNL) granule factors in the control of T cell dependent response in mice.

Factors isolated from human PMNL lysosomes granules enhance the immune response of BALB/c mice immunized with SRBC, augment allogeneic DTH reaction and accelerate skin graft rejection. The immunostimulatory effects does not depend only on proteolytic enzymes. Removal of proteolytic enzymes using DFP as irreversible inactivator did not alter the immunostimulatory effect of PMNL lysosome factors on PFC formation in mice. The PMNL factors have marked potentiating effect on the uptake and retention of labeled CRBC in mouse lymphoid system. This effect suggests an alternative pathway which can provide chronic immunological stimulus and play a significant role in augmentation of immune response. Data presented in this paper suggest that factors responsible for augmentation of the immune response are localized mostly in secondary PMNL granules.

The mitogenic and enzymatic activities of polymorphonuclear leukocyte (PMNL) lysosomal proteins examined in vitro.

The lysosomal granules of human and animal PMNL contain the protein factors which stimulate the lymphocyte transformation in vitro. The performed experiments showed that mitogenic activity is not exclusively associated with proteolytic activity. Using column chromatography fractionation, it has been shown that the mitogenic and proteolytic proteins are localized in different fractions. The irreversible inhibition of proteases by di-isopropyl-phosphofluoridate (DFP) increase the mitogenic activity of PMNL lysosomal proteins. The mitogenic proteins are localized in lysosomal granules of secondary density and they are loosely bound to membranes of lysosomal granules.

Influence of ACTH on activity of some granulocytic proteases in patients with multiple sclerosis.

The effect of treatment with ACTH on activity of granulocytic acid and neutral proteases in patients suffering from multiple sclerosis was studied. Protease activity was distinctly higher in granulocytes from patients. ACTH normalized the activities investigated in patients with exacerbating course multiple sclerosis, but had no influence on hydrolases in the progressive form of the disease.

The role of autoaggressive reactions in recurrent uveitis.

An experimental study in rabbits and clinical study in patients with uveitis was carried out. Experimental uveitis was induced in rabbits sensitized with albumin and antigen was injected intravenously or into the vitrous body of their eyes. Blastic transformation of lymphocytes stimulated with PHA, albumin and extracts of the uvea and cytotoxicity of the serum against normal white blood cells in presence of antigen were determined. In recurrent uveitis blastic transformation induced with uvea antigens, hemagglutinin titers against these antigens and serum cytotoxicity increased. The results indicate a significant role of autoaggression in the pathogenesis of some recurrent eye uveitis cases.

[Role of leukocytes in the healing process in surgical wounds].

In the last period of time there has been stated that in human lysosomes of neutrophilic leukocytes an in ones of some experimental animals occur, which stimulate the growth of lymphocytes and fibroblasts in the study in vitro. With regard to this it was assumed that similar stimulating processes may take place in the surgical wounds. The experiments were carried out on guinea pigs which were wounded and afterwards the wounds were sutured and the allogenic proteins preparations were introduced into its in dose of 100 microgram as follows: lysosomal proteins of granulocytes, mixed and separated on the the DEAE-cellulose columns; lysosomal proteins of macrophages, the proteins isolated from plasma and nuclei both the granulocytes and macrophages. The evaluation of healing was based on the measurement of mechanical resistance of wounds adhesions and was completed with the pathomorphological examinations. It has been shown that the mixture of lysosomal proteins of granulocytes, intruduced into the wounds, caused the acceleration of the healing processes. After giving into the wounds some fractions of proteins which were separated on the colums with DEAE-cellulose, the stimulating processes of healing were seen, while after adding the other ones-the inhibiting processes. The protein with similar evidences were not show inside other structures studied of granulocytes and macrophages. The results obtained seem to support the supposition that in lysosomes of neutrophilic granulocytes there are the factors which regulate the repair processes in wounds.

The stimulatory effect of human PMNI lysosomes on lymphocytes in vitro and in vivo.

The proteins isolated from human PMNL lysosome granules have activated the lymphocytes in vitro. It was found that T cells undergo transformation in response to lysosome mitogens but no LIF production was observed. Transformation induced by PHA, PWM and Con A was depressed in the presence of lysosome mitogens. The in vivo response to T-dependent antigens is amplified by lysosome mitogens. The above experiments demonstrate that lysosome mitogens stimulate T lymphocytes.

The influence of immunization on some functions of polymorphonuclear leucocytes in vitro.

Polymorphonuclear leucocytes (PMNL) obtained from immunized guinea pigs possess an affinity to antigen. The guinea pigs were immunized with human serum albumin (HSA) in complete Freund's adjuvant. The ability of PMNL to bind antigen was tested by means of the functional disorders induced by antigen after incubation with PMNL. The "immune" PMNL appeared to be more responsive to antigen when compared to PMNL obtained from non-immunized animals. The antigen decreases oxygen consumption, the phagocytosis coefficient and 14C-glycine incorporation in the "immune" PMNLs in vitro and also induces leakage of some lysosomal hydrolases from PMNL in the course of an 18 hrs incubation period.

Polymorphonuclear leukocyte lysosome activities and lymphocyte transformation in multiple sclerosis and some other central nervous system chronic diseases.

The role of PMNL lysosomes was studied in some lesions of the central nervous system with speical regard to multiple sclerosis patients. The significant decrease of acid hydrolases was observed in multiple sclerosis (MS) peripheral blood PMNL lysosomal granula and, in parallel, the increase of lysosome proteins mitogenic potency. The other lesions of the central nervous system such as apoplexy, brain tumors, brain trauma and epilepsy have not shown the MS-like activities of PMNLs. The role of PMNLs in pathogenesis of MS and in the stimulation of immunocompetent cells is suggested.

Sport training and some activities of human polymorphonuclear leukocyte lysosomes.

The acid phosphatase and protease, neutral protease, bactericidal and mitogenic activities were determined in the polymorphonuclear leukocyte (PMNL) lysosomes of 16 years old boys. The investigated group consisted of 20 intense sport training persons during the last six years, the control one of the same age boys without any sport training. The significant decrease of hydrolase activity has been observed in the lysosomes of the peripheral blood PMNL's of the sport training boys.

The influence of polymorphonuclear leukocytes and macrophages on the growth of lymphocytes and fibroblasts in vitro.

The influence of polymorphonuclear leukocytes (PMNL) and macrophage plasma, nuclei and lysosome extracts have been tested on lymphocytes and fibroblasts growth in vitro. The stimulatory activity of PMNL lysosomes has been shown on lymphocyte and fibroblast growth. The macrophage lysosomes have shown a similar but lower activity. The role of lysosomes has been suggested in reparation and wound healing processes, presumably in inflammatory cell infiltrated regions.
