The interrelationship between production of reactive oxygen intermediates and inflammatory cytokines by peripheral blood leukocytes in vitro from children with food allergy.

In this study, the activity of neutrophils measured using the chemiluminescence test was estimated in 49 children with food allergy. It was shown that average chemiluminescence values without, as well as after, stimulation by formyl-methionyl-leucine phenylalanine and phorbol myristate acetate were considerably higher in children with allergy in comparison to those in the control group. The correlation between the production of reactive oxygen intermediates and interleukin (IL)-4, IL-5 and IL-10 concentration suggests that an autocrine regulatory mechanism may play a role in allergy in children.

[Increased percentage of "double phenotype" form T lymphocytes in blood of patients with multiple sclerosis].

We studied the percentage of double positive (CD4+CD8+) form of T-cells in a group (total 77) multiple sclerosis (MS) patients, measured by means of monoclonal antibodies anti-CD3, CD4/FITC, CD8/RPE and flow cytometry FACScan (Becton Dickinson). In our study we have shown that the percentage of the double positive T cells is higher (p < 0.05) in the peripheral blood of patients with acute exacerbation of MS (n = 21), and in the course of chronic progressive MS (n = 27) comparing to MS remission (n = 29) and other neurological diseases (n = 25) groups. In the study we have shown that the percentage of double positive T-cells in peripheral blood depends mainly on disease activity.

Chronic beta-adrenergic antagonist treatment affects human T lymphocyte responsiveness "in vitro".

The aim of the present study was the evaluation of the effect of long-term propranolol treatment on some immune parameters in humans. The subjects were 12 duodenal ulcer patients, aged 19-23 years. They were given propranolol in a dose of 40 mg three times a day orally. Blood samples were drawn before and after one-month propranolol treatment. The drug did not significantly affect mitogen (PHA, Con A, PWM)- induced lymphoproliferative response, although a tendency of a slight inhibition of proliferation was observed. In all patients treated with propranolol absolute peripheral blood lymphocyte counts increased. Propranolol did not significantly influence one-way MLR, while it enhanced proliferation in the AMLR test in all patients. The drug increased spontaneous and PHA-induced IL-2R expression as well as IL-2 generation. The study showed that chronic beta-adrenoceptor blockade with propranolol may modify some immune functions in humans.

Involvement of granulocyte factor (GF) in receptor-triggered stimulation of lymphocytes and neutrophils.

Granulocyte factor derived from the specific granules of polymorphonuclear neutrophils affects mitogen--and alloantigen--induced proliferation of human lymphocytes "in vitro". A dual pattern of GF effect was noted: inhibition of mitogen induced lymphocyte proliferation in higher concentrations and costimulatory activity in lower concentrations of GF. GF induced the lymphocyte proliferation in combination with phorbol ester (PMA) or anti-CD2 monoclonal antibodies (mAb) with PMA. Other combinations of anti-CD2 and anti-CD3 mAb with GF do not augment the proliferation. The comitogenic activities of GF are associated with IL-2 production and IL-2R expression. Specific binding of labelled GF to stimulated lymphocytes suggests that it may recognize the specific molecule expressed only an activated cells. GF diminishes the zymosan-induced granulocyte stimulation, but GF acts as a stimulator of granulocyte O2 production. GF may recognize common targets on leukocytes. Leukocyte adhesive glycoproteins may be such putative targets.

Some mechanisms of immunosuppressive action of epinephrine in humans.

The studies were performed on 12 men (9 duodenal ulcer patients and 3 healthy controls) aged 19-23 years. They were given epinephrine in a dose of 0.014 mg/kg subcutaneously. Blood samples were drawn before and 30 and 150 min. after drug injection. The results obtained in the duodenal ulcer patients and controls revealed the same direction of changes. Epinephrine induced a significant increase in the absolute numbers of all investigated peripheral blood mononuclear cell subpopulations (total lymphocytes, T, Th, Ts, NK cells and monocytes) observed at 30 min. following injection, while they nearly normalized at 150 min. A Th/Ts cell number ratio slightly, but significantly, decreased. Epinephrine did not affect spontaneous IL-2 receptor expression and it significantly inhibited PHA-induced IL-2 receptor expression and IL-2 generation at 30 min. following injection. NK cell activity also decreased 30 min. after epinephrine administration and it was still lowered at 150 min. The drug very distinctly inhibited proliferative response in the autologous mixed lymphocyte reaction test.

The stimulatory effect of a single intravenous dose of propranolol of some immune parameters in humans.

Propranolol (PRO) is a beta-adrenergic receptor antagonist. The studies were aimed to evaluate the effect of a single intravenous PRO injection at a dose of 0.1 mg/kg on some immune parameters, and were performed on 12 men, including 9 duodenal ulcer patients and 3 healthy subjects, aged 19-23 years (mean 20.5 +/- 1.2). Blood samples were drawn before, 30 and 150 min following the injection. Results obtained in duodenal ulcer patients and healthy subjects revealed the same type of changes. A single PRO injection did not significantly affect total peripheral blood lymphocyte, Th cell and monocyte counts; however, it slightly but significantly lowered numbers of T and Ts cells, which was accompanied by a rise in Th/Ts cell number ratio and NK cell numbers. PRO significantly augmented spontaneous as well as PHA-stimulated IL-2 receptor expression, IL-2 generation, NK cell activity and AMLR observed at 30 and 150 min after the drug injection. Possible mechanisms of these changes are discussed.

Effect of a single intravenous dose of verapamil on some immune parameters in duodenal ulcer patients.

The effect of a single intravenous dose of calcium channel blocking agent verapamil (VER) on immune parameters in humans remains uncertain. In this study the effects of VER on lymphocyte subpopulations, interleukin 1 (IL-1) and 2 (IL-2) production in vitro, autologous mixed lymphocyte reaction (AMLR), IL-2 receptor expression (Tac positive cells) and natural killer (NK) cell activity were assessed. The investigations were undertaken on 12 hospitalized men, aged 19-23 years, with small abdominal complaints. None of them had active duodenal ulcer disease, while in the remaining no signs of organic disease were found. VER was given intravenously in a dose of 0.15 mg/kg and examinations were performed on peripheral blood mononuclear cells (PBL) drawn before and 30 and 150 min following VER injection. The single VER dose induced a significant, but transient increase in T, T helper, T suppressor lymphocytes and monocytes, and decrease in IL-1 and IL-2 generation as well as diminished Tac antigen expression. These data provide evidence that calcium channel blockers may transiently disturb immunoregulation.

[Suppressor cell activity in multiple sclerosis and other nervous system diseases. Preliminary report].

3H-Thymidine incorporation was assessed in concanavalin A-stimulated cultures of peripheral blood lymphocytes of healthy subjects and multiple sclerosis patients. At the same time the counts of T and B lymphocytes were determined in peripheral blood and the activity of suppressor cells was determined in vitro by the method of Shou et all. in these patients and in other neurological diseases. The studied immunological parameters showed some variability in patients with multiple sclerosis: at the time of exacerbations the activity of suppressor cells decreased and the mitogenic response to Con A was impaired. In remissions and in slowly progressive cases of multiple sclerosis the activity of suppressor cells was increased, particularly in elderly patients. The reported investigations demonstrated that disturbances in the functions of suppressor cells occur also in other neurological diseases. The role of the observed disturbances in the pathogenesis of multiple sclerosis is discussed.

The effect of polymorphonuclear leucocyte (PMNL) factor on human natural killer (NK) cell activity in vitro.

In a previous study [20] there was described the inhibitory effect of lysosomal proteins isolated from the guinea pig peritoneal exudate PMNL on the spontaneous cytotoxic activity of splenic cells in vitro. Moreover, in vivo investigations demonstrated these factors to inhibit also the development and activity of the antigen specific cytotoxic cells. The aim of the present study was to explain the role and mechanism of action of the PMNL factors in the regulation of the natural killer (NK) cell activity. The effect of PMNL factors on the NK cells was estimated in the tests based on 51Cr release using chick red blood cells CRBC). It was found that, similarly as in guinea pigs, the lysosomal proteins isolated from human peripheral blood PMNL exerted an inhibitory effect on the NK cells activity of human blood mononuclears against CRBC and this effect was independent of the activity of proteases present in the fractions of the studied lysosomal proteins. The inhibitory effect was not exerted by the factors released from the PMNL during their adherence. The obtained results seem to indicate that factors released from the PMNL during adherence are not identical to the substances released from the PMNL granules during their breakdown.

Subcellular distribution of inflammatory mediators in guinea-pig polymorphonuclear granulocytes (PMNL).

The experiments were performed for localizing the inflammation-inducing factors in the subcellular fractions of human and guinea pig PMNL. The magnitude of the induced inflammation tested on rats and guinea-pigs represented combined effects of all proteins extracted from PMNL. The isolation of PMNL subcellular components did not elevate the potency of the inflammatory response. The inflammation was completely abolished with SH-dependent protease inhibitor. The inflammation induced by PMNL factors is a complex phenomenon which could not be attributed to special subcellular components.

Granulocyte factors as potential haemopoiesis regulators in irradiated mice.

Protein substances stimulating lymphocytes in vivo and in vitro are present in the lysosomes and secondary granules of neutrophil granulocytes. The mechanism of this stimulation is probably dependent on blockade of T suppressor cell functions. These observations stimulated the present study undertaken to clarify the effect of granulocyte factors on the multipotential stem cells of the haemopoietic system in mice subjected to sublethal irradiation. The experiments were carried out on F1 hybrid (Balb/cx NMRI) mice receiving a whole body dose of 3 Gy irradiation. Granulocyte proteins were injected intravenously in doses of 6 micrograms per one mouse 18, 48 and 72 hours after irradiation. The assayed proteins increased the number of macroscopic and microscopic colony forming cells in the mice spleens. The increase of erythrocyte granulocyte index in colonies and decrease of lymphocyte granulocyte index in peripheral blood have been observed. In the light of these results it is concluded that lysosomal granules and secondary granules of neutrophils contain protein factors stimulating the regeneration of haemopoietic cells of the erythrocyte and granulocyte series in the irradiated mice spleens.
