Removal of bacteria by filtration in planted and non-planted sand columns.

In order to diminish hygienic hazards from pathogens, the elimination of pathogenic bacteria in a pre-treatment step is important for the use of domestic wastewater for irrigation purposes. Therefore, we analysed the removal of bacteria in laboratory-scale model sand filters simulating vertical flow systems of constructed wetlands (CW). Sand-filled glass columns were planted with Juncus effusus or Phragmites australis and non-planted columns were used as controls. Processes of bacteria removal such as adsorption, lysis, and the biotic effects caused by plants, protozoa, and Bdellovibrio were studied with E. coli as a model bacterium. E. coli suspensions (10(8) cellsml(-1)) were trickled on the columns by intermittent loading under non-water-saturated operation conditions. In non-planted and sterilized sand columns, an initial removal of cells was observed in the range as expected by the adsorption capacity of the sand columns. After loading of the sand with cells, an increasing reduction of the cell concentrations by 3-4 orders of magnitude in the effluent was registered up to volumetric loads of more than 548mll(-1) day(-1) (230mm day(-1)). In planted columns, no higher levels of removal were observed. Predation by protozoa, which were found in concentrations up to 10(4)ml(-1) in the effluent, is considered to be the main reason for the elimination. However, Bdellovibrio bacteriovorus was also found in plaque-forming units of about 10(4)g(-1) sand, suggesting that this bacteriovorous bacterium plays an additional role in the removal process. In a second series of experiments, domestic wastewater was applied and removal efficiencies up to four orders of magnitude of the cfu's of coliform bacteria, faecal streptococci, and salmonellae were observed. Considering the transpiration of the plants, higher removal efficiencies were found in the planted variants. Protozoa and Bdellovibrio were detected in the domestic wastewater in varying concentrations, suggesting that predation and lysis were the major removal mechanisms.

Comparison of API 20NE and Biolog GN identification systems assessed by techniques of multivariate analyses.

The increasing use of commercial multitest systems for identification of environmental bacteria creates the problem of how to compare the identification results obtained from different systems. The limited use of species designations in such comparisons is caused by low usage of environmental bacteria in the development of commercial identification schemes. Two multivariate statistical methods, the Mantel's test and the co-inertia analysis, were applied to analyze data derived from the Biolog GN and the API 20NE systems of identification for 50 environmental bacterial strains. We found these two methods to be useful for revealing the relationship between the two sets of numerical taxonomic traits. Both of these methods showed that the distances according to the Biolog GN results between the studied strains were related to those derived from the API 20NE results, despite the differences in the test sets of the two systems. In addition, the co-inertia analysis allowed us to visualise the relationships between classifications of strains derived from the two identification systems and, simultaneously, to estimate the contribution of particular tests to the differentiation of bacterial strains.

Functional and structural successions in arbitrary samples of heterotrophic bacteria during aerobic treatments of lignite-carbonization wastewater in in situ enclosures.

In situ mesocosm experiments were performed in Lake Schwelvollert (located in the district of Weissenfels, Saxony-Anhalt, Germany), an anaerobic lignite-carbonization effluent lake containing phenolic compounds and their autoxidation products (anthropogenic humic matter). In the aeration enclosure, the anaerobic Schwelvollert wastewater was aerated and in the flocculation enclosure, it was flocculated to precipitate the oxygen-trapping anthropogenic humic matter to enhance the input of oxygen by diffusion. To gain an insight into the metabolic state of the aerobic heterotrophic microbiota during the treatments, arbitrary samples of bacterial isolates were taken from a general agar medium and tested for their abilities to cleave predominant phenolic contaminants by a procedure called the isolate sample assay. In this way, successions of degradation potentials were observed in both mesocosms, with degradation abilities for meta- and para-alkylated phenols appearing before degradation abilities for ortho-substituted phenols as a common phenomenon. To examine the structure of samples, the respective isolates were characterized using the Biolog GN MicroPlate system, the random amplified polymorphic DNA nucleic acid (RAPD) fingerprinting technique, and amplified ribosomal DNA restriction analysis (ARDRA). Although similar functional patterns occurred in both mesocosms, the compositions and diversities of the respective bacterial communities varied significantly, even at different depths from the same enclosure, with members of the Pseudomonas RNA group I being predominant.

Acquisition of a deliberately introduced phenol degradation operon, pheBA, by different indigenous Pseudomonas species.

Horizontal transfer of genes of selective value in an environment 6 years after their introduction into a watershed has been observed. Expression of the gene pheA, which encodes phenol monooxygenase and is linked to the pheBA operon (A. Nurk, L. Kasak, and M. Kivisaar, Gene 102:13-18, 1991), allows pseudomonads to use phenol as a growth substrate. Pseudomonas putida strains carrying this operon on a plasmid were used for bioremediation after an accidental fire in the Estonia oil shale mine in Estonia in 1988. The water samples used for studying the fate of the genes introduced were collected in 1994. The same gene cluster was also detected in Pseudomonas strains isolated from water samples of a nearby watershed which has been continuously polluted with phenols due to oil shale industry leachate. Together with the more frequently existing counterparts of the dmp genes (V. Shingler, J. Powlowski, and U. Marklund, J. Bacteriol. 174:711-724, 1992), the pheA gene was also represented in the phenol-degrading strains. The area where the strains containing the pheA gene were found was restricted to the regular route of phenolic leachate to the Baltic Sea. Nine Pseudomonas strains belonging to four different species (P. corrugata, P. fragi, P. stutzeri, and P. fluorescens biotypes B, C, and F) and harboring horizontally transferred pheBA operons were investigated. The phe genes were clustered in the same manner in these nine phe operons and were connected to the same promoter as in the case of the original pheBA operon. One 10.6-kb plasmid carrying a pheBA gene cluster was sequenced, and the structure of the rearranged pheBA operon was described. This data indicates that introduced genetic material could, if it encodes a beneficial capability, enrich the natural genetic variety for biodegradation.

Adult Wilms' tumor: case report, management, prognosis.

Wilms' tumor being a rare disease in adults, even large centers have had experience with isolated cases only. In our clinic, we have treated two cases of histologically confirmed Wilms' tumor in the last 3 years. On the basis of two case reports, we describe the staging, treatment and prognosis of this highly malignant tumor. To a large extent, diagnostics and treatment follow the recommendations for Wilms' tumor in children.

[Therapy of prostate abscess by employing pigtail drainage with transrectal ultrasound control].

Prostatic abscess is a rare but often severe disease. In most cases diagnosis is difficult. The best diagnostic tool is transrectal ultrasound, which depicts prostatic abscess very clearly. The therapy of choice to date is surgery, usually transurethral resection. At the Department of Urology of the University of Kiel we have employed another form of treatment, namely perineal drainage of the abscess with a pigtail tube under transrectal ultrasound guidance. This method is expected to be particularly advantageous in younger patients, for whom transurethral resection may cause problems. Five patients aged 24-71 years have been treated in this manner so far.

Enrichment of estradiol receptor from calf uterus.

A three-step procedure to enrich estradiol receptor protein from calf uterus nearly 30,000-fold has been described. Over-all yield is 12%. Control of the single steps has been performed by sodium dodecyl sulfate-electrophoresis, sucrose density gradient centrifugation, gel filtration and determination of receptor quantity. Immunological properties of the preparation obtained have not been controlled.

Bilateral renal angiomyolipoma: involvement of the contralateral kidney after 25 years--an unusual case.

The diagnosis of renal angiomyolipoma often poses problems, both clinically and pathohistologically. With the increasing refinement of imaging techniques it is becoming potentially possible to recognize these benign neoplasms by means of ultrasound, computed tomography and magnetic resonance imaging. Electron microscopy and histochemical methods are often necessary for the correct pathological diagnosis. The problems involved are presented on the basis of the case history of a patient who suffered from renal angiomyolipoma in both kidneys with an interval of 25 years. The first tumor was diagnosed histologically as a liposarcoma 25 years ago.

Xanthogranulomatous pyelonephritis associated with renal cell carcinoma. Report on two cases and review of the literature.

Xanthogranulomatous pyelonephritis is a rare and particularly aggressive variant of chronic destructive pyelonephritis. Even when all modern diagnostic possibilities are exhausted, it is often not possible to distinguish xanthogranulomatous pyelonephritis from a renal cell carcinoma preoperatively. In clinical practice false diagnoses are therefore frequent. The coexistence of xanthogranulomatous pyelonephritis and renal cell carcinoma is extremely rare. We report 2 such cases. In 1 case surgery was performed on the kidney affected by xanthogranulomatous pyelonephritis using a renal sparing technique.

[Effect of pepstatin A on Candida albicans infection in the mouse].

The intravenous injection of 10(6) to 10(7) Candida albicans cells revealed to be a reliable model in mice produce infections of the kidney. Higher germ contents could be yielded in the kidney after the application of protease positive Candida-strains as compared to protease negative ones. Additionally, after the infection with protease positive strains a proteolytic activity could be found in the kidney homogenates in vitro on casein plates. The modulation of this Candida infection by pepstatin A, an inhibitor of extracellular yeast proteases in vitro, has also been studied in the same mice model in vivo. The growth rate of Candida albicans has been measured in the left kidney by counting the germ content as described by Haenel. Infections could be reduced after single doses of 120 micrograms pepstatin A contrary to 60 micrograms pepstatin A. The same was with three doses of 180 micrograms at 24 h intervals. This protease inhibitory effect could also be found in kidney homogenates in vitro on casein plates and lasted until 48 h post injection. On the basis of this positive effects on the Candida infection in mice pepstatin A should be considered as an adjuvans in the therapy of severe yeast infections.

[Priapism following intracavernous injection of vasoactive substances as a urologic emergency].

From 1986 to 1989 papaverine was used alone or in combination with phentolamine in 165 patients in diagnosis and treatment of erectile dysfunction. In 14 patients a prolonged erection did occur after diagnostical introduction of SKAT therapy. Treatment consisted of intracavernous injection of 2-4 mg metaraminol, partly additional by puncture of cavernous bodies and evacuation of viscous blood. As single complication of metaraminol injection a hypertensive crisis could observed. A glando-cavernous shunt was not necessary in any case. After exact dosage adjustment the danger of drug-induced priapism is low.

[Reducing the side effects of aggressive chemotherapy (cisplatin and epirubicin) with xenogenic peptides (factor AF2) in patients with hormone refractory metastatic prostate cancer. A prospective, randomized study].

The indication of a chemotherapy is advisable with patients who are suffering from a progressively metastasised, secondarily hormone refractory carcinoma of the prostate. In search of efficient chemotherapy protocols we combined cisplatin with epirubicin (PE scheme) in our clinic. Massive side effects of that aggressive chemotherapy scheme like gastro-intestinal trouble and myelotoxicity are the limiting factors of the scheme. With measures like reducing the dosage, delaying the next cycle, or breaking off the therapy the effective dosage can often not be achieved. The anti-emetics which are usually used today exclusively give anti-emetic protection. The additional administration of xenogenic peptides (Factor AF2) had additionally myeloprotective effect in former studies. In this study we examined whether, by additionally giving Factor AF2, the patients' subjective condition, and above all their hemogram, could be stabilised in order to achieve the effective dosage or dosage intensity. For that, the patients were prospectively randomised in two groups by means of a random selection board. The analysis of the data gained in the protocol showed that the additional administration of Factor AF2 improves the patients' subjective conditions significantly. Apart from that, we noticed a considerable reduction of the vomiting frequency. Concerning the objective measured parameters of the leukocytes, thrombocytes, erythrocytes, and the hemoglobin level, the significantly myeloprotective effect of Factor AF2 could be proved. Due to the fact that in the verum group there were considerably fewer cases of breaking off or delays of the treatment than in the control group, the effective dosage intensity could be achieved with a higher number of patients in that group.

[Xanthogranulomatous pyelonephritis with fatal outcome in a 2-month-old infant].

A 2-month-old infant died of xanthogranulomatous pyelonephritis. Preoperatively pyonephrosis was suspected, because the child presented with a number of inflammatory, septic symptoms. Nephrectomy was performed and histopathology showed the kidney to be affected by xanthogranulomatous pyelonephritis. Postoperatively the child developed persistent attacks of fever and bronchopneumonia that led to his death with signs of pulmonary insufficiency.

Studies on assay of estradiol binding proteins by cytochemical and immunocytochemical techniques in human breast tumors and other tissues.

Estradiol binding proteins in frozen sections of human breast tumors, ovarian carcinomas, hypernephromas, rat uteri and cow udders were demonstrated by fluorescent labeled estradiol bovine serum albumin conjugate (cytochemical or direct method). The specificity of the binding of estradiol conjugate to tissue sections was tried to approach by addition of various steroid or non-steroidal competitors. The estradiol conjugate binding sites were practically not present in sections of human spleen, cross muscle and colon muscle.
