[Videocystometry (VCM)--a method for quantitative and qualitative diagnosis of refluxes (author's transl)].

Videocystometry (VCM) is a combination of videography and cystometry for the investigation of vesico-ureteral refluxes and is an accurate method of reflux classification that registers intravesical pressure and volume simultaneously after X-ray detection. We investigated 86 refluxes detected by micturating cystourethrography; 21 could not be confirmed, 14 disappeared with conservative treatment, and 10 underwent surgery, because their condition became worse during examination. Especially refluxes without primary indications for operation can be verified by comparative data. This facilitates the decision to continue conservative or operative treatment.

[Synthesis and properties of immobilized enzymes. X. Covalent binding of polygalacturonase to insoluble carriers].

The pectinolytic enzymes are of practical interest for the clarification of fruit juice. In the present paper the covalent coupling of polygalacturonase (PG; E. C. 3.2.1.15) is reported. A commercially available enzyme (Rohament P; 5 U/mg) and purified Endo-PG (200 U/mg) are immobilized to the following carriers: BrCN-activated Sepharose, carbodiimide-activated CH-Sepharose, dialdehyde Sepharose, dialdehyde Sephadex, dialdehyde cellulose, CMC-azide, carbodiimide-activated CMC, macroporous glass (isothiocyanate and carbodiimide coupling) and glass beads. The implications of pore diameter (Sephadex- and Sepharose derivatives), of purity of the PG, of protein content of the PG-carrier-complexes as well as the presence of substrate during the coupling reaction, are discused in relation to the relative and specific activity of the bound protein and to the efficiency of the coupling reaction. From the carriers under study derivatives of Sepharose yield the best result (relative activity max. 88%, specific activity max. 5400 U/mg). The immobilization to isothiocyanate glass yields good results, too (relative activity 20%, specific activity 500 U/g). The mechanical instability of the PG-dialdehye Sephadex-complexes and the low relative activity of the bound enzyme are unsatisfactory. Due to their low affinity to PG, the derivatives of cellulose are also inappropriate for covalent coupling of this enzyme. All PG-carrier-complexes are largely stable both during storage at 4 degrees C and repeated activity assays.

[Cellelectrophoretic studies of the effect of methylglyoxal-bis-guanylhydrazone on blood cells of tumour patients (author's transl)].

The effect of methylglyoxal-bis-guanylhydrazone has been studied on the electrophoretic mobility of blood cells of 10 unselected and untreated patients with preferably solid malignant diseases. Prior to the incubation with CH3-G the electrophoretic mobility of lymphocytes and erythrocytes showed increased and of platelets showed decreased rates which extend over a larger range than usual and for which reasons cannot be given till now. The incubation with CH3-G caused a concentration-dependent reduction of the electrophoretic mobility of all three tested types of blood cells. This can be estimated as an impairment of a function of surface membrane and confirms recently obtained results of activity of CH3-G on platelets' surface membrane.

Comparative studies on the behaviour of the plasmatic clotting system and platelet function in surgically treated cancer patients.

By measuring 18 functional parameters of blood coagulation the dynamic pattern of pre- and postoperative blood coagulation was studied in a total of 43 patients with primarily operable solid tumours. The results were compared with those obtained in patients having benign diseases in the same anatomical regions. Results were discussed preferably from the tumourbiological point of view. Changes of blood coagulation occuring in operated cancer patients with benign diseases. Compensatory processes operating in the haemostatic system provide a well balanced coagulation even in cancer patients. Patients with gastric cancer having low adenine nucleotide levels in platelets had a significantly higher risk to die postoperatively than those with normal levels. This indicates metabolic disorder on cellular levels than haemostatic dysfunctions.
