Comparative analysis of the two-step reaction catalyzed by prokaryotic and eukaryotic phytochelatin synthase by an ion-pair liquid chromatography assay.

Genes encoding phytochelatin (PC) synthase have been found in higher plants, fission yeast and worm. Recently, kinetic and mutagenic analyses of recombinant PC synthase have been revealing the molecular mechanisms underlying PC synthesis, however, a conclusive model has not been established. To clarify the mechanism of PC synthase found in eukaryotes, we have compared the two-step reactions catalyzed by the prokaryotic Nostoc PC synthase (NsPCS) and the eukaryotic Arabidopsis PC synthase (AtPCS1). Comparative analysis shows that in the first step of PC synthesis corresponding to the cleavage of gamma-glutamylcysteine (gamma-EC) from glutathione (GSH), free GSH or PCs acts as a donor molecule to supply a gamma-EC unit for elongation of the PC chain, and heavy metal ions are required to carry out the cleavage. Furthermore, functional analyses of various mutants of NsPCS and AtPCS1, selected by comparing the sequences of NsPCS and AtPCS1, indicate that the N-terminal region (residues 1-221) in AtPCS1 is the catalytic domain, and in this region, the Cys(56) residue is associated with the PC synthesis reaction. These results enable us to propose an advanced model of PC synthesis, describing substrate specificity, heavy metal requirement, and the active site in the enzyme.

Single-chain magnet (NEt4)[Mn2(5-MeOsalen)2Fe(CN)6] Made of Mn(III)-Fe(III)-Mn(III) trinuclear single-molecule magnet with an S(T) = 9/2 spin ground state.

The cyano-bridged trinuclear compound, (NEt(4))[Mn(2)(salmen)(2)(MeOH)(2)Fe(CN)(6)] (1) (salmen(2)(-) = rac-N,N'-(1-methylethylene)bis(salicylideneiminate)), reported previously by Miyasaka et al. (ref 19d) has been reinvestigated using combined ac and dc susceptibility measurements. The strong frequency dependence of the ac susceptibility and the slow relaxation of the magnetization show that 1 behaves as a single-molecule magnet with an S(T) = (9)/(2) spin ground state. Its relaxation time (tau) follows an Arrhenius law with tau(0) = 2.5 x 10(-)(7) s and Delta(eff)/k(B) = 14 K. Moreover, below 0.3 K, tau saturates around 470 s, indicating that quantum tunneling of the magnetization becomes the dominant process of relaxation. (NEt(4))[Mn(2) (5-MeOsalen)(2)Fe(CN)(6)] (2) (5-MeOsalen(2)(-) = N,N'-ethylenebis(5-methoxysalicylideneiminate)) is a heterometallic one-dimensional assembly made of the trinuclear [Mn(III)(SB)-NC-Fe(III)-CN-Mn(III)(SB)] (SB is a salen-type Schiff-base ligand) motif similar to 1. Compound 2 has two types of bridges, a cyano bridge (-NC-) and a biphenolate bridge (-(O)(2)-), connecting Mn(III) and Fe(III) ions and the two Mn(III) ions, respectively. Both bridges mediate ferromagnetic interactions, as shown by modeling the magnetic susceptibility above 10 K with g(av) = 2.03, J(Mn)(-)(Fe)/k(B) = +6.5 K, and J'/k(B) = +0.07 K, where J' is the exchange coupling between the trimer units. The dc magnetic measurements of a single crystal using micro-SQUID and Hall-probe magnetometers revealed a uniaxial anisotropy (D(T)/k(B) = -0.94 K) with an easy axis lying along the chain direction. Frequency dependence of the ac susceptibility and time dependence of the dc magnetization have been performed to study the slow relaxation of the magnetization. A mean relaxation time has been found, and its temperature dependence has been studied. Above 1.4 K, both magnetic susceptibility and relaxation time are in agreement with the dynamics described in the 1960s by R. J. Glauber for one-dimensional systems with ferromagnetically coupled Ising spins (tau(0) = 3.7 x 10(-)(10) s and Delta(1)/k(B) = 31 K). As expected, at lower temperatures below 1.4 K, the relaxation process is dominated by the finite-size chain effects (tau'(0) = 3 x 10(-)(8) s and Delta(2)/k(B) = 25 K). The detailed analysis of this single-chain magnet behavior and its two regimes is consistent with magnetic parameters independently estimated (J'and D(T)) and allows the determination of the average chain length of 60 nm (or 44 trimer units). This work illustrates nicely a new strategy to design single-chain magnets by coupling ferromagnetically single-molecule magnets in one dimension.

[MnIII2 (5-Rsaltmen)2NiII(pao)2(L)]2+: an S(T)=3 building block for a single-chain magnet that behaves as a single-molecule magnet.

Mn(III)-Ni(II)-Mn(III) linear-type trinuclear complexes bridged by oximate groups were selectively synthesized by the assembly reaction of [Mn2(5-Rsaltmen)2(H2O)2](ClO4)2 (5-Rsaltmen2-=N,N'-(1,1,2,2-tetramethylethylene) bis(5-R-salicylideneiminate); R=Cl, Br) with [Ni(pao)2(phen)] (pao-=pyridine-2-aldoximate; phen=1,10-phenanthroline) in methanol/water: [Mn2(5-Rsaltmen)2Ni(pao)2(phen)](ClO4)2 (R=Cl, 1; R=Br, 2). Structural analysis revealed that the [Mn(III)-ON-Ni(II)-NO-Mn(III)] skeleton of these trimers is in every respect similar to the repeating unit found in the previously reported series of 1D materials [Mn2(saltmen)2Ni(pao)2(L1)2](A)(2) (L(1)=pyridine, 4-picoline, 4-tert-butylpyridine, N-methylimidazole; A=ClO4-, BF4-, PF6-, ReO4-). Recently, these 1D compounds have attracted a great deal of attention for their magnetic properties, since they exhibit slow relaxation of the magnetization (also called single-chain magnet (SCM) behavior). This unique magnetic behavior was explained in the framework of Glauber's theory, generalized for chains of ferromagnetically coupled anisotropic spins. Thus, in these 1D compounds, the [Mn(III)-ON-Ni(II)-NO-Mn(III)] unit was considered as an S(T)=3 anisotropic spin. Direct-current magnetic measurements on 1 and 2 confirm their S(T)=3 ground state and strong uniaxial anisotropy (D/k(B) approximately -2.4 K), in excellent agreement with the magnetic characteristic deduced in the study on the SCM series. The ac magnetic susceptibility of these trimers is strongly frequency-dependent and characteristic of single-molecule magnet (SMM) behavior. The relaxation time tau shows a thermally activated (Arrhenius) behavior with tau0 approximately 1x10(-7) s and Delta(eff)/k(B) approximately 18 K. The effective energy barrier for reversal of the magnetization Delta(eff) is consistent with the theoretical value (21 K) estimated from |D| S2T. The present results reinforce consistently the interpretation of the SCM behavior observed in the [Mn2(saltmen)2Ni(pao)2(L1)2](A)2 series and opens new perspectives to design single-chain magnets.

By utilizing the novel metalloligand l(Cu), [Cu(2,4-pydca)(2)](2)(-) (2,4-pydca(2)(-) = pyridine-2,4-dicarboxylate), which possesses two kinds of coordination groups, selective bond formation with the series of the first-period transition metal ions (Mn(ii), Fe(ii), Co(ii), Cu(ii), and Zn(ii)) has been accomplished. depending on the coordination mode of 4-carboxylate with Co(ii), Cu(ii), and Zn(ii) ions, L(Cu) forms a one-dimensional (1-d) assembly with a repeating motif of [-M-O(2)C-(py)N-Cu-N(py)-Co(2)-]: {[ZnL(Cu)(H(2)O)(3)(DMF)].DMF}(N)() (2), [ZnL(Cu)(H(2)O)(2)(MeOH)(2)](N)() (3), and {[ML(Cu)(H(2)O)(4)].2H(2)O}(N)() (M = Co (4), Cu (5), Zn (6)). the use of a terminal ligand of 2,2'-bipyridine (2,2'-bpy), in addition to the cu(ii) ion, gives a zigzag 1-d assembly with the similar repeating unit as 4-6: {[Cu(2,2'-bpy)L(Cu)].3H(2)O}(N)() (9). on the other hand, for Mn(ii) and Fe(ii) ions, L(Cu) shows a 2-carboxylate bridging mode to form an another 1-d assembly with a repeating motif of [-M-O-C-O-CU-O-C-O-]: [ML(Cu)(H(2)O)(4)](N)() (M = Mn (7), Fe (8)). this selectivity is related to the strength of lewis basicity and the electrostatic effect of L(Cu) and the irving-williams order on the present metal ions. according to their bridging modes, a variety of magnetic properties are obtained: 4, 5, and 9, which have the 4-carboxypyridinate bridge between magnetic centers, have weak antiferromagnetic interaction, whereas 7 and 8 with the carboxylate bridge between magnetic centers reveal 1-d ferromagnetic behavior (Cu(II)-M(II); M(II) = Mn(II), J/k(B) = 0.69 K for 7; M(II) = Fe(II), J/k(B) = 0.71 K for 8).

A Mn12 single-molecule magnet [Mn12O12(OAc)12(dpp)4] (dppH = diphenyl phosphate) with no coordinating water molecules.

The preparation and physical characterization are reported for a novel single-molecule magnet [Mn(12)O(12)(OAc)(12)(dpp)(4)] (dppH = diphenyl phosphate) with no coordinating water molecules. The crystal structure analysis reveals that there are four five-coordinate Mn(III) ions with Mn.H approaches. Addition of water in CD(2)Cl(2) solution was monitored by (1)H NMR, which showed that H(2)O could coordinate to a vacant site of a five-coordinate Mn(III) ion in solution. The measurements and analyses of magnetization hysteresis and ac magnetic susceptibility indicate that the title complex is a single-molecule magnet with a quantum tunneling behavior, whose ground state was tentatively assigned to S = 10 with g = 1.78 and D = -0.60 K.

Isolation of a new member of group 3 late embryogenesis abundant protein gene from a halotolerant green alga by a functional expression screening with cyanobacterial cells.

By a functional expression screening with cyanobacterial cells, a new member of group 3 late embryogenesis abundant protein genes (designated cw80lea3) was isolated from the cDNA library of halotolerant green alga Chlamydomonas sp. strain W80. The principle of the screening method was based on the acquisition of NaCl salt-tolerance of the fresh water cyanobacterial cells carrying the halotolerant algal gene. The expression of cw80lea3 gene in the C. W80 cells was induced by salt- and cold-stresses, but not by abscisic acid treatment.

Characterization of phytochelatin synthase-like protein encoded by alr0975 from a prokaryote, Nostoc sp. PCC 7120.

Phytochelatins (PCs) are well known as the heavy metal-detoxifying peptides in higher plants, eukaryotic algae, fungi, and nematode. In contrast, neither PCs nor PC synthase genes have ever been identified in any prokaryotes. The genome sequences for the cyanobacterium Nostoc sp. PCC 7120 were recently completed and allowed us to identify a gene encoding a PC synthase-like protein, termed alr0975. The predicted product of alr0975 contains the conserved N-terminal domain but not the variable C-terminal domain found in eukaryotic PC synthases. The recombinant alr0975 protein strongly catalyzed the first step of PC synthesis, in which glutathione (GSH) is converted to gamma-glutamylcysteine (gamma-EC), although the protein only weakly catalyzed the second step of PC synthesis, namely the transfer of gamma-EC moiety to an acceptor GSH molecule to form PC(2). These results suggest alr0975 protein may be a more primitive form of the PC synthases found in eukaryotes.

Enhancement of stress tolerance in transgenic tobacco plants overexpressing Chlamydomonas glutathione peroxidase in chloroplasts or cytosol.

To evaluate the physiological potential of the defense system against hydroperoxidation of membrane-lipid components caused by environmental stresses in higher plants, we generated transgenic tobacco plants expressing a glutathione peroxidase (GPX)-like protein in the cytosol (TcGPX) or chloroplasts (TpGPX). The activities toward alpha-linolenic acid hydroperoxide in TcGPX and TpGPX plants were 47.5-75.3 and 32.7-42.1 nM min(-1) mg(-1) protein, respectively, while no activity was detected in wild-type plants. The transgenic plants showed increased tolerance to oxidative stress caused by application of methylviologen (MV: 50 microM) under moderate light intensity (200 micro E m(-2) sec(-1)), chilling stress under high light intensity (4 degrees C, 1000 microE m(-2) sec(-1)), or salt stress (250 mM NaCl). Under these stresses, the lipid hydroperoxidation (the production of malondialdehyde (MDA)) of the leaves of TcGPX and TpGPX plants was clearly suppressed compared with that of wild-type plants. Furthermore, the capacity of the photosynthetic and antioxidative systems in the transgenic plants remained higher than those of wild-type plants under chilling or salt stress. These results clearly indicate that a high level of GPX-like protein in tobacco plants functions to remove unsaturated fatty acid hydroperoxides generated in cellular membranes under stress conditions, leading to the maintenance of membrane integrity and increased tolerance to oxidative stress caused by various stress conditions.

The rbcX gene product promotes the production and assembly of ribulose-1,5-bisphosphate carboxylase/oxygenase of Synechococcus sp. PCC7002 in Escherichia coli.

The operon encoding ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) in the cyanobacterium Synechococcus sp. PCC7002 contains three rbc genes, rbcL, rbcX and rbcS, in this order. Introduction of translational frameshift into the rbcX gene resulted in a significant decrease in the production of large (RbcL) and small (RbcS) subunits of the Rubisco protein in Synechococcus sp. PCC7002 and in Escherichia coli. To investigate the function of the rbcX gene product (RbcX), we constructed the expression plasmid for the rbcX gene and examined the effects of RbcX on the recombinant Rubisco production in Escherichia coli. The coexpression experiments revealed that RbcX had marked effects on the production of large and small subunits of Rubisco without any significant influence on the mRNA level of rbc genes and/or the post-translational assembly of the Rubisco protein. The present rbcX coexpression system provides a novel and useful method for investigating the Rubisco maturation pathway.

A series of single-chain magnets, [Mn2(saltmen)2Ni(pao)2(L)2](A)2 (saltmen(2-)=N,N'-(1,1,2,2-tetramethylethylene) bis(salicylideneiminate), pao-=pyridine-2-aldoximate; A-=ClO4- with L=4-picoline; 2, 4-tert-butylpyridine; 3, N-methylimidazole; 4, and L=pyridine with A-=BF4-; 5, PF6-; 6, ReO4-; 7), was prepared by reactions between MnIII dimer units, i.e., [Mn2(saltmen)2(H2O)2](A)2 (A-=ClO4-, BF4-, PF6-) or Mn2(saltmen)2(ReO4)2, and NiII monomeric units, i.e., Ni(pao)2(L)2, in methanol/water media. The crystal structures of 4, 6, and 7 were established by single-crystal X-ray crystallography. These three compounds are isostructural with [Mn2(saltmen)2Ni(pao)2(py)2](ClO4)2 (1) (Clérac, R.; Miyasaka, H.; Yamashita, M.; Coulon, C. J. Am. Chem. Soc. 2002, 124, 12837) and crystallize in monoclinic space group C2/c. The linear arrangement of MnIII dimer units and NiII building blocks leads to an alternating chain having a repeating unit, [-(O)2-Mn-ON-Ni-NO-Mn-]. The chains are well separated with the nearest interchain intermetallic distance of 10.36 A for 4, 10.51 A for 6, and 10.30 A for 7, and there is no significant pi-pi interchain interaction between ligands. The void space between the chains is occupied by counteranions, which control the three-dimensional organization of the chains. The X-ray diffraction analysis (XRD) on a powder sample was also performed for all compounds. The XRD patterns for 1, 2, and 4-7 are very similar, emphasizing the isostructural nature of these materials although they have individually slight different interchain distances. Inversely, the XRD pattern for 3 reveals a completely different shape being indicative of the peculiar crystal packing compared to the others. Nevertheless, the one-dimensional nature of the structure is also kept in 3 as indicated by magnetic measurements. The whole family of compounds exhibits quasi-identical magnetic behavior compared to that described for 1. Above 30 K, the heterometallic chain can be described as an assembly of antiferromagnetically coupled Mn...Ni....Mn trimers (via oximate bridge, -24.2 K<J(Mn-Ni)/kB<-20.8 K) connected through Mn...Mn ferromagnetic interaction (via the phenolate bridge, J(Mn-Mn)/kB approximately +0.7 K), assuming a ferromagnetic chain with S=3 units. In the low temperature region, combined ac and dc magnetic measurements revealed for the whole series the systematic presence of a magnet behavior exhibiting coercivity and slow relaxation of magnetization below 3.5 K. This behavior was analyzed on the basis of Glauber's theory for an Ising one-dimensional system which predicts an activated dependence (Arrhenius law) of the relaxation time: tau=tau0 exp(Delta/kBT). Similar values of tau0 approximately 1x10(-10) s and Delta/kB approximately 70 K have been found along the series. The described compounds constitute the first example of the rational design of a single-chain magnet family and clearly demonstrate the unique behavior of these heterometallic chains independently of their interchain environments.

Carrier doping effect into quasi-one-dimensional bromo-bridged Ni(III) complexes with strong electron-correlation by Cu(II) ions, [Ni1-xCux(chxn)2Br]Br2-x.

This communication will describe the electron doping effect into Ni(III) complexes by Cu(II) ions, [Ni(1-x)Cu(x)(chxn)(2)Br]Br(2-x) (x = 0.038 and 0.101) by using an electrochemical oxidation method. A drastic increase of electrical conductivity as well as a new absorption band around 0.5 eV in single crystal reflectance spectra was observed by doping Cu(II) ions, indicating the electron doping was successfully made. An ESR result shows unpaired electrons locate in the d(x2-y2) orbitals of Cu(II) and have almost no interaction with those of other ions.

A family of diruthenium compounds with dianionic tridentate ligands of N-(2-pyridyl)-2-oxy-5-R-benzylaminate (R = H, Me, Cl, Br, NO(2)): isolation, structure determination, and electrochemistry.

The ligand substitution reaction of Ru(2)(O(2)CCH(3))(4)Cl with 5-substituted N-(2-pyridyl)-2-oxy-5-R-benzylaminate (R = H, Me, Cl, Br, NO(2)) resulted in a family of anionic diruthenium species of [Ru(2)(O(2)CCH(3))(2)(R-salpy)(2)](-) that were isolated by using Na(+)- or K(+)-18-crown-6-ether as the countercation: [A(18-crown-6)(S)(x)()][Ru(2)(O(2)CCH(3))(2)(R-salpy)(2)] (A = Na(+), K(+); S = solvent; R = H, 1; Me, 2; Cl, 3; Br, 4; NO(2), 5). All compounds were structurally characterized by X-ray crystallography. The structural features of the anionic parts are very similar among the compounds: two acetate and two R-salpy(2)(-) ligands are, respectively, located around the Ru(2) unit in a trans fashion, where the R-salpy(2)(-) ligand acts as a tridentate ligand having both bridging and chelating characters to form the M-M bridging/axial-chelating mode. Compounds 1 and 5 with K(+)-18-crown-6-ether have one-dimensional chain structures, the K(+)-18-crown-6-ether interacting with phenolate oxygens of the [Ru(2)(O(2)CCH(3))(2)(R-salpy)(2)](-) unit to form a repeating unit, [.K.O-Ru-Ru-O.], whereas 2-4 are discrete. Cyclic voltammetry and differential pulse voltammetry revealed systematic redox activities based on the dimetal center and the substituted ligand, obeying the Hammett law with the reaction constants per substituent, rho, for the redox processes being 127 mV for Ru(2)(5+)/Ru(2)(4+), 185 mV for Ru(2)(6+)/Ru(2)(5+), 92 mV for Ru(2)(7+)/Ru(2)(6+), and 179 mV for R-salpy(-)/R-salpy(2)(-). For 3, the singly oxidized and reduced species, Ru(2)(6+) and Ru(2)(4+), respectively, generated by bulk controlled-potential electrolyses were finally monitored by spectroscopy. The singly oxidized species can also be slowly generated by air oxidation.

Novel ET-coordinated copper(I) complexes: syntheses, structures, and physical properties (ET = BEDT-TTF = Bis(ethylenedithio)tetrathiafulvalene).

New molecular charge-transfer complexes of bis(ethylenedithio)tetrathiafulvalene (ET), (ET)Cu(2)Br(4) (1), (ET)(2)Cu(6)Br(10) (2), (ET)(2)[Cu(4)Br(6)ET] (3), (ET)(2)Cu(2)Br(4) (4), (ET)(2)Cu(3)Br(7)(H(2)O) (5), and (ET)(2)Cu(6)Br(10)(H(2)O)(2) (6), have been synthesized by diffusing reaction of ET and Cu(II)Br(2). Their crystal structures and physical properties have been investigated. X-ray analyses revealed that ET molecules coordinated to the copper ions with the sulfur atoms of the ethylenedithio groups in all compounds. The Cu-S distances are found in the range 2.268(5)-2.417(8) A, being close to the typical Cu(I)-S coordination bond distances. Strong d-pi interactions between d-electrons of the copper ions and pi-electrons of the ET molecules can be expected through the Cu-S coordination bonds. ET molecules behave as trans-bidentate ligands bonding to two different copper ions in 1 and 3, as cis-bidentate ligands in 2, 5, and 6, and as monodentate ligands in 4. In the crystal structure of 3, there are two types of ET molecules in the crystal structure, where the first type is a trans-bidentate ligand and the second forms a stacking structure by itself. Compounds 1, 2, 4, and 6 show semiconducting behavior down to low temperature (sigma(RT) = 1.6 x 10(-2) S cm(-1) and E(a) = 122 meV for 1, sigma(RT) = 2.1 S cm(-1) and E(a) = 21 meV for 2, sigma(RT) = 5.4 x 10(-4) S cm(-1) and E(a) = 239 meV for 4, and sigma(RT) = 5.1 x 10(-2) S cm(-1) and E(a) = 207 meV for 6). On the other hand, in 3, a metal-like region is observed along the b-axis and c-axis, due to the contribution of stacked ET molecules, and a metal-semiconductor transition occurs at 280 and 270 K, respectively.
