Involvement of regulatory volume decrease in the migration of nasopharyngeal carcinoma cells.

The transwell chamber migration assay and CCD digital camera imaging techniques were used to investigate the relationship between regulatory volume decrease (RVD) and cell migration in nasopharyngeal carcinoma cells (CNE-2Z cells). Both migrated and non-migrated CNE-2Z cells, when swollen by 47% hypotonic solution, exhibited RVD which was inhibited by extracellular application of chloride channel blockers adenosine 5'-triphosphate (ATP), 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) and tamoxifen. However, RVD rate in migrated CNE-2Z cells was bigger than that of non-migrated cells and the sensitivity of migrated cells to NPPB and tamoxifen was higher than that of non-migrated cells. ATP, NPPB and tamoxifen also inhibited migration of CNE-2Z cells. The inhibition of migration was positively correlated to the blockage of RVD, with a correlation coefficient (r) = 0.99, suggesting a functional relationship between RVD and cell migration. We conclude that RVD is involved in cell migration and RVD may play an important role in migratory process in CNE-2Z cells.

Cloning and tissue expression of chicken heart fatty acid-binding protein and intestine fatty acid-binding protein genes.

Fatty acid-binding proteins (FABPs) are members of a superfamily of lipid-binding proteins, occurring intracellularly in invertebrates and vertebrates. This study was designed to clone and characterize the genes of heart fatty acid-binding protein and intestine fatty acid-binding protein in the chicken. PCR primers were designed according to the chicken EST sequences to amplify cDNA of H-FABP and I-FABP genes from chicken heart and intestinal tissues. Analysis of sequence showed that the cDNA of the chicken H-FABP gene is 75 to 77% homologues to human, mouse, and pig H-FABP genes, and the chicken I-FABP gene is 71 to 72% homologues to human, mouse, and pig I-FABP genes. In addition, Northern blot analysis indicated that of the two genes, similar to the copartner of the mammal, H-FABP gene was expressed in a wide variety of tissues, and I-FABP gene was expressed only in intestinal tissues. The expression levels of the chicken H-FABP mRNA in heart and I-FABP mRNA in intestine had significant differences between the broilers from fat line and Bai'er layers at six weeks of age. The results of this study provided basic molecular information for studying the role of two FABPs in the regulation of fatty acid metabolism in avian species.

Identification of differentially expressed genes in metastatic and non-metastatic nasopharyngeal carcinoma cells by suppression subtractive hybridization.

BACKGROUND & OBJECTIVE: Nasopharyngeal carcinoma (NPC) is an epithelial neoplasm with high occurrence rates in southern China. The disease often metastasizes to regional lymphnodes at a very early stage. Local recurrences and metastasis occur frequently in patients with NPC and are a leading cause of death, despite improvements on treatment modalities. The molecular mechanism underlying the metastasis of nasopharyngeal carcinoma remains poorly understood, however, and requires additional elucidation. The aim of this study was to explore possible NPC gene candidates that may play key roles in NPC metastasis. METHODS: Subtractive suppression hybridization (SSH) was performed to isolate differentially expressed clones between the metastatic 5-8F and non-metastatic 6-10B nasopharyngeal carcinoma cell lines. Differentially expressed clones were screened and confirmed by reverse Northern blotting. The sequences of cDNA fragments were subsequently analyzed and compared to known sequences in Genbank. RESULTS & DISCUSSION: The SSH library contained thousands of positive clones. Random analysis of 300 clones by PCR demonstrated that 269 clones contained inserted fragments. Reverse Northern blot confirmed that 20 out of 192 clones examined were significantly up-regulated in the 5-8F cell line. Among these 20 clones, 16 were previously identified genes (flotilin-2, ezrin, pim-3, fli-1, mel, neugrin, znf216, ASB1, raly, UBE2A, keratin6A, TMED7, EIF3S9, FTL, two ribosomal proteins RPL21 and RPL16), two were predicted genes (c9orf74 and MDS006), and two sequences shared no homology with known genes listed in GenBank and may represent novel genes. The proposed functions of the genes identified in this study include cell signal transduction, cell survival, transcription regulation, cell mobility, protein synthesis, and DNA damage repair. Flotillin-2, fli-1, pim-3 and ezrin have previously been reported to be associated with tumor metastasis and progression. The remaining up-regulated genes identified in this study have not been reported to be markers of metastasis and may represent new candidates of NPC metastasis-related genes. The results of this study may provide novel points of therapeutic intervention for NPC.

Primary malignant fibrous histiocytoma of the small bowel: a report of an additional case in duodenum.

AIMS: We report herein an additional case of primary malignant fibrous histiocytoma (MFH) in the duodenum and provide a review of the existing literature. METHODS AND RESULTS: A 61-yr-old Chinese man was admitted to our hospital with symptoms of melena, anorexia, and weight loss. An abdominal computed tomography (CT) and gastrointestinal barium meal examination demonstrated a tumor of the duodenum suggestive of primary malignancy. The tumor was successfully treated by pancreaticoduodenectomy. It was histopathologically and immunohistochemically diagnosed to be a storiform-type primary MFH of the duodenum. There have been a total of 40 cases of primary malignant fibrous histiocytoma of the small bowel documented in the literature including our Chinese cases. CONCLUSION: Primary malignant fibrous histiocytoma of the small bowel, especially in the duodenum is extremely rare. The final diagnosis is made only after pathological and immunopathological examination of the tumor. The malignant potential of such tumors is high. The prognosis may be mainly dependent on the invasion and metastasis of tumor, while tumor size is irrelevant. The treatment should be surgery if possible. Early surgical intervention may be the best form of management that may offer the patient good result.

Prenatal diagnosis of congenital fetal heart abnormalities and clinical analysis.

OBJECTIVE: To study the value of detecting fetal congenital heart disease (CHD) using the five transverse planes technique of fetal echocardiography. METHODS: Nine hundred and eighty-two high-risk pregnancies for fetal CHD were included in this study, the fetal heart was scanned with the five transverse planes technique of fetal echocardiography described by Yagel, autopsy was conducted when pregnancy was terminated. Blood from fetal heart was collected for fetal chromosome analysis. A close follow-up was given for normal fetal heart pregnancies and neonatal echocardiography was performed to check the accuracy of prenatal diagnosis. RESULTS: (1) Forty-six cases (4.68%) were found to have fetal heart abnormalities in this study, 69.56% of them were diagnosed by single four-chamber view, another 30.43% fetal CHD were found by combining other views; (2) Forty-one parents of prenatal fetuses with CHD chose to terminate pregnancy, thirty-two of them gave consent to conduct autopsy, 93.75% of which yielded unanimous conclusion between prenatal fetal echocardiography and autopsy; (3) Thirty-two of 46 cases underwent fetal chromosome analysis, 8 cases (25%) were found to have abnormal chromosome; (4) Five cases were found to have right ventricle and atrium a little bigger than those on the left side, with the unequal condition being the same after birth, but there were no clinical manifestations and they are healthy for the time being; (5) Nine hundred and thirty-six cases were not found with abnormality in this study, but one case was diagnosed with ventricular septal defect after birth, one case was diagnosed with patent ductus arteriosus, one case had atrial septal defect after birth. CONCLUSIONS: (1) The detected CHD rate was 4.68% by screening fetal heart with five transverse planes according to Yagel's description of high risk population basis for CHD. The coinciding rate of prenatal diagnosis and autopsy was 93.75%; (2) The sensitivity of detecting fetal heart abnormality is 92%, the specificity is 99.6% using the five transverse planes technique of fetal echocardiography; (3) Fetuses with mild or moderate disproportion of right and left side in the heart are potentially healthy babies.

Blockade of diabetic vascular injury by controlling of AGE-RAGE system.

Vascular complications result in disabilities and short life expectancy in diabetic patients. During prolonged hyperglycemic exposure, non-enzymatically glycated protein derivatives termed advanced glycation endproducts (AGE) are formed at an accelerated rate and accumulated in blood and in tissues. Studies performed in vitro and in vivo revealed AGE and their receptor RAGE as the major accounts for vascular cell derangement characteristic of diabetes. The AGE-RAGE system would thus be considered as a candidate molecular target for overcoming diabetic vascular complications. Potential preventive and therapeutic approaches toward it include inhibition of AGE formation, breakage of preformed AGE-proteins crosslinks, blockade of AGE-RAGE interactions with RAGE competitors or antagonists and RAGE-specific signaling inhibition.

[Effect of Niupo Zhibao Pellet on expression of neuronal nitric oxide synthase in brain of endotoxin-induced shock rats].

OBJECTIVE: To investigate the effect of Niupo Zhibao Pellet (NPZBP) on the expression of neuronal nitric oxide synthase (nNOS) in the brain of endotoxin-induced shock rats. METHODS: SD rats were randomly divided into normal control group, endotoxin-induced shock model group and NPZBP-treated group. Lipopolysaccharide (LPS) (1.5 mg/kg i.v.) and tsD-galactosamine (D-GalN) (100 mg/kg i.p.) were administered to the rats in endotoxin-induced shock model group, as well as to the rats in NPZBP-treated group after seven-day treatment, to induce the shock. The expression of nNOS in the brain of the rats in each of the 3 groups was measured by immunohistochemical methods. RESULTS: In the 3 groups, nNOS immuno-positive cells distributed widely in layer II, III, IV of the cerebral cortex, the molecular layer of hippocampus, the polymorphic layer of the dentate gyrus, the reticular formation of brain stem, and the molecular, granular and Purkinje cell layer of the cerebellar cortex. The number of immuno-positive cells in the NPZBP-treated group was slightly higher than that of the normal control group, and significantly lower than that of the model group (P<0.05) in many regions of the brain, including cerebral cortex, hippocampus, brain stem and cerebellar cortex. CONCLUSION: NPZBP can inhibit the over-expression of nNOS in wide area of the brain in endotoxin-induced shock rats.

[Comparison of the clinical effect of complex transplantation with heterogenetic acellular dermal M(ADM) AND allogeneic ADM].

OBJECTIVE: To explore the short-term clinical effects of complex transplantation among the acellular dermal matrix(ADM) of heterogenic or allogeneic and autogenic split on the burnt wound as to find out a permanent substitution for the treatment on full skin thickness defect without scar. METHODS: Two kinds of ADM were used on the 18 patients with full thickness burn wound through complex transplantation with autogenic splits. The patients with medial-thickness autograft was used as control group. Survival rate was obtained 2 weeks after operation; contraction rate and the scores of Vancouver burn scale were obtained 8 weeks after operation. RESULTS: No significant difference was observed in survival rate among the three groups 2 weeks after operation(P > 0. 05); no significant difference was observed in contraction rate of autografts and scores of Vancouver burn scale among the three groups 8 weeks after operation (P > 0. 05). CONCLUSION ADM of heterogenic and ADM of allogeneic have similar effect on the reconstruction of skin, so the piglet ADM made in this way could be used as a substitution.

[Detection of major gene on litter size in sheep].

The current study was designed to detect SNPs within BMP15 and BMPR-IB gene and investigate the effect of the genes on sheep litter size. Four sheep lines, HU-Yang, Chinese Merino monotocous, Chinese Merino multiparous for wool production and Chinese Merino multiparous for mutton production, were used in this study. Litter sizes were recorded for each ewe in the four lines. Primers for BMP15 and BMPR-IB gene were designed from database sheep sequence and polymorphisms were detected by PCR-RFLP method. The results showed that there was no polymorphism with BMP15 gene among the four lines, and there was an A / G SNP with BMPR-IB gene at base 746 among the four lines. Three types of genotype (BB, B+ and ++), based on A / G locus, were found within each line. The frequencies of genotypes were significantly different among the lines (P<0.001), with BB genotype primarily existing in HU-Yang, ++ genotype in Chinese Merino monotocous line, and B+ genotype in Chinnese Merino multiparous lines. The A / G mutation influence significantly the sheep litter sizes, and the BB and B+ ewes had significant higher litter sizes than ++ ewes. The results of present study showed simultaneously that the genotype of BMPR-IB was a perfect predictor of the sheep litter sizes. These results intensively indicated that BMPR-IB is a major gene to affect litter size in sheep, and could be used as the molecular genetic marker to select litter size in sheep.

[Slide platelet aggregation test used as a monitor for patient treated with anti-pletelet drugs].

The aim was to verify the effectiveness of slide platelet aggregation test (SPAT) to monitor the inhibition effect of anti-platelet drugs. A group of eight healthy volunteers was examined for SPAT value and T(50) (time necessary for reaching 50% of total aggregation) induced by ADP, arachidonic acid (AA) and cationic propyl gallate (c-PG) respectively before and after administration of ASA in dose of 100 mg/day for 3 days. The group of 41 inpatients at the Department of Cardiovascular Disease treated with anti-platelet drugs and the group of 327 healthy blood donors were also examined for SPAT. The SPAT value of healthy volunteer samples stored at room temperature were measured hourly for four hours. The results showed that: (1) no significant difference was detected between the T(50) before and after ASA administration in health volunteer group when ADP was used as inducer, but a significant difference was detected in this group when AA or c-PG was used as inducer. There was significant linear correlation between SPAT value and T(50) induced by c-PG in health volunteer group before and after administration of ASA (r = 0.998, P = 0.000); (2) there was no significant difference between the SPAT value of health volunteer group before administration of ASA and the SPAT value of health blood donors group (P = 0.853), but there was a significant difference between the SPAT values before and after administration of ASA in health volunteer group (P = 0.000). There was significant difference when the SPAT value of the inpatients treated with anti-platelet drugs was compared with that of healthy blood donor group and with that of health volunteer group before and after administration of ASA (P = 0.000). The cut-off value of SPAT in health blood donor group was 44.6 +/- 11.7 seconds, reference value was from 21.1 seconds to 68.0 seconds; (3) there was no significant difference between SPAT values when platelets samples stored at room temperature for 1, 2, 3, 4 hours (P = 0.815). In conclusion, SPAT can rapidly monitor the inhibition effect of anti-platelet drugs and SPAT may have the similar clinic value with T(50) induced by c-PG.

[Study on serum leptin level of urban resident and the relationship between body mass index, blood lipid, insulin and leptin level].

OBJECTIVE: To measure serum leptin concentration of urban residents and investigate the relationship between serum leptin concentration and body mass index(BMI), blood lipid, insulin. METHODS: One hundred fifty-eight men and women ages 18-72 years were initially drawn from a community in Ji'nan City. The parameters measured included height, weight, waist and hip circumference, insulin, blood lipid and leptin concentrations. Statistical analyses were performed with SPSS11.5. RESULTS: The mean serum concentration of leptin was (17.47 +/- 13.52)ng/ml, in male subjects was (8.38 +/- 6.31)ng/ml, in female subjects it was (24.98 +/- 13.29)ng/ml. Serum level of leptin was significantly positively correlated with BMI, waist and hip circumference, not correlated with WHR. Serum leptin level in females was positively correlated with WHR. There was no association between serum level of leptin and insulin. Serum leptin level in males was positively correlated to apo B, in females was positively correlated to TC, LDL-c. CONCLUSION: BMI is an important factor to determine serum leptin level. The relationships between plasma glucose, insulin, blood lipids and serum leptin are affected by gender. In males serum leptin level was positively correlated to apo B, in females was positively correlated to TC, LDL-c. We need further research to explain this difference.

[Identification on Herba Hedyotidis by HPLC fingerprint pattern method].

OBJECTIVE: To establish a new identification and analysis method of Herba Hedyotidis. METHOD: HPLC fingerprint pattern method. RESULT: Experiment and analysis were carried out on seven samples, the standard HPLC fingerprint pattern method and characterisitic diffraction peaks of Herba Hedyotidis were obtained. CONCLUSION: This method can be used for the identification of Herba Hedyotidis.

[Study of Five STR Loci in A Chinese Han Population.].

To understanding the allele structure and genetic polymorphisms at five STR loci in Chinese Han population, and construct a preliminary database, EDTA-blood specimens were collected from unrelated individuals. DNA samples were extracted with Chelex method and were amplified by PCR technique. The PCR products were analyzed using both PAGE horizontal electrophoresis with discontinuous buffer system and automated fluorescence detection approach. As a result, three STRs consist of simple repeat motifs, while two STR contain a complex repeat structure. The STR polymorphisms at all of the five loci have been observed in Chinese Han population. In a word, the obtained data are beneficial to understanding the population genetics of the five STR loci in Chinese Han population. As a simple approach, the PAGE horizontal electrophoresis can be employed for typing the five STR markers.

[Preparation and characterization of non-agglutinating mAbs against membrane antigen on human erythrocytes].

AIM: To prepare monoclonal antibodies (mAbs) against membrane antigens on human erythrocytes and characterize their properties. METHODS: BALB/c mice were immunized with the membrane antigens of human type O erythrocytes. The splenocytes of the immunized mice were fused with Sp2/0 myeloma cells by hybridoma technique. The antibodies to common antigen on human erythrocytes were screened by hexadimethrine bromide (polybrene) test tube method and then the agglutinating antibodies (complete antibodies) were weeded out by slide hemagglutination assay. The hybridoma cells secreting non-agglutinating antibodies (incomplete antibodies) were cloned by limiting dilution method. The stability of the obtained hybridoma cells and the properties of the mAbs were identified. RESULTS: One hybridoma cell 2E8 was obtained, which secreted non-agglutinating antibody. The mAb 2E8 belonged to IgG1, could agglutinate H antigen, and had no species cross-agglutination reaction. The titers of culture supernatant and ascites of 2E8 were 1:1,024 and 1:64x10(6), respectively. When the affinity of mAb 2E8 was evaluated by agglutination reaction, erythrocytes began to agglutinate after 7 seconds and the clots exceeded 1 mm(2) in 3 minutes. CONCLUSION: The non-agglutinating mAb against H antigen was prepared successfully. The mAb 2E8 has good titer, affinity and specificity, which lays the foundation for preparation of bispecific antibodies (BsAb).

[The effects of zhubi capsule on adjuvant arthritis in rats].

OBJECTIVE: To explore the therapeutic effects and mechanisms of Zhubi capsule on adjuvant arthritis (AA) in rats. METHOD: The model of rat was induced by intradermal injection of CFA. The perimeter of ankle joint was measured at different time points. Con A-induced splencyte proliferation was examined by MIT assay and the expression of IL-1, IL-6 in synovium was determined by immunohistochemical method. RESULT: After the preventive administration of Zhubi capsule (5.2, 2.6, 1.3 g x kg x d(-1), x 25 d), it was found that Zhubi capsule significantly inhibited the primary and secondary ankle joint swelling. The increased Con A-induced splencyte proliferation reaction and the activated IL1, IL-6 expression of AA rat were suppressed by the treatment with Zhubi capsule. CONCLUSION: Zhubi capsule has a therapeutic effect on AA rats, and its mechanism is related to the immunoregulative function.

[Experimental study on treatment of minimal change nephropathy with Lespedeza michx].

OBJECTIVE: To study the therapeutic effect of Lespedeza Michx in experimental rat model of minimal change nephropathy (MCN). METHOD: The rat MCN model was reproduced by tail-intravenous injection of Adriamycin. The treatment effect of Lespedeza Michx was determined by the detection of urine protein collection for 24 hours, malondialdehyde (MDA), superoxide dismutase (SOD), ATCO, IL-8, TNF-alpha in serum, and renal histopathological changes were observed by microscope. RESULT: Lespedeza Michx could decrease the contents of urine proteinuria, MDA, IL-8, and TNF-alpha in serum, increase SOD level and improved the pathological change of glomeruli. CONCLUSION: Lespedeza Michx was effective on MCN.

[The clinical analysis of with 106 cases aortic dissection].

OBJECTIVE: To explore the risk factors clinical appearance and examination methods of aortic dissection (AD). METHOD: The clinical data of 106 AD patients admitted from the January, 2001 to July, 2004 were retrospectively reviewed. RESULTS: The symptoms and physical signs of the 106 AD patients were diverse. Chest pain was a common initial symptoms but some patients initial symptoms were atypical. The origins of AD was in ascending aorta when patients had clinical appearance mimicking myocardial infarction, and in descending aorta when patients had pain involving shoulders, dorsum, waist, abdomen or lower limbs. Hypertension was the most important cause of AD. The combination use of echocardiography, CT and MRI may help to make rapid diagnosis. CONCLUSIONS: The symptoms and physical signs of AD were diverse. Clinical symptom had some relationship with the origin of AD, the form of AD and hypertension had obvious relations. An obvious correlation exited between the occurrence of AD and hypertension.
