[Study on preparation of refined Xiong-gui sustained release pills].

OBJECTIVE: To optimize the molding formula of refined Xiong-gui sustained release pills, types and assorted dosages of water-soluble and non water-soluble drug carrier were studied. METHOD: An orthogonal design was performed to sift the pill formulation. The processing conditions were determining in terms of roundness, hardness, viscosity, fluidity, easily drop. RESULT AND CONCLUSION: According to the final optimal formula, the amount of drug release can be detected instantly, while remaining over 80% at 10 h.

[Expression of Skp2 and p27 proteins in laryngeal squamous cell carcinoma and their significances].

OBJECTIVE: To investigate the expression of Skp2 and p27 and their association with clinical, histologic characteristics and prognosis in laryngeal squamous cell carcinoma (LSCC), the correlation between expression of Skp2 and p27 in LSCC. METHOD: Skp2 protein and p27 protein were examined by using immunohistochemical SP staining method in 20 cases of normal laryngeal tissues (NLT), 27 cases of laryngeal precancerous lesion (LPL) and 27 cases of LSCC. RESULT: The positive rate of Skp2 in NLT, LPL and LSCC were 10.00% (2/20), 37.04% (10/27) and 41.43% (29/70), respectively. The positive rates in the last two groups were higher than the first one. The Skp2 protein expression in LSCC was positively associated with the clinical stage and lymph node metastasis. The positive rate of p27 protein in NLT and LSCC were 85.00% (17/20) and 48.57% (34/70), respectively, and significant difference was found between two groups. The p27 protein expression was inversely associated with lymph node metastasis. Skp2 was inversely associated-with expression of p27 in LSCC. The survival rate of patients with both Skp2 positive expression and negative expression of p27 was significantly lower than that of other cases. CONCLUSION: Skp2 expression might play an important role in the development and progression of LSCC via cyclin-depended kinase inhibitor p27 proteolysis, and the combined examination of Skp2 and p27 expression have a great significance in estimating prognosis of patients with LSCC.

[The changing pancreatic cancer mortality in China (1991-2000)].

OBJECTIVE: To describe the mortality rate of pancreatic cancer and its distribution in China during the period of 1991-2000. METHODS: Based on the data of demography and death collected through China's Disease Surveillance Point System (DSPS) over the period of 1991-2000, the distribution of death rate of pancreatic cancer was described in terms age group, gender, calendar year, rural/urban residence and administrative district. RESULTS: A total of 1619 mortal cases attributed to pancreatic cancer (975 men and 644 women) were reported by DSPS from 1991 to 2000. The reported, adjusted and age-standardized mortality rates increased from 1.46, 1.75, and 2.18 per 100,000 populations in 1991 to 2.38, 3.06, and 3.26 per 100,000 population in 2000. The majority (69.62%) of the mortalities of pancreatic cancer was seen in the age group of 60 years and older. The mortality rate was higher in men than in women, but the male to female death rate ratios decreased during the 10 years. Our data also showed that the death rate of pancreatic cancer in urban areas was about 2-4 fold higher than that in rural areas, and in the districts of Northeast and East China, the death rates were higher than those in the other 5 administrative districts. CONCLUSION: The death rate due to pancreatic cancer was rising during the period of 1991-2000 and the peak mortality of pancreatic cancer might arrive in future in China.

[Association of TNFA polymorphisms with the outcomes of HBV infection].

OBJECTIVE: To explore whether the TNFA promoter single nucleotide polymorphisms (SNPs) are associated with the outcomes of hepatitis B virus(HBV) infection in Chinese Han population. METHODS: One hundred and forty-eight self-limited HBV infection subjects and 207 chronic hepatitis B patients were recruited. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and sequence specific primer-PCR(PCR-SSP) were used to detect the SNPs of five sites in TNFA promoter (-238G/A, -308G/A, -857C/T, -863C/A, -1031T/C). The frequency distributions of genotypes and haplotypes in different groups were analyzed by EPI and EH programs. RESULTS: The frequencies of -238GG genotype in chronic hepatitis B patients were significantly higher than that in self-limited infection subjects (P=0.02). The frequencies of -857TT genotype in chronic hepatitis B patients were clearly lower than that in self-limited infection subjects (P=0.02). Haplotypic frequencies of GGCCT (-238/-308/-857/-863/-1031) in chronic hepatitis B patients was significantly lower than that in self-limited infection subjects (P=0.03), and the frequencies of haplotype GGCAT or GGTAT in chronic hepatitis B patients were clearly higher than those in self-limited infection subjects (P=0.0001; P=0.004). CONCLUSION: TNFA promoter polymorphisms are important host genetic factors affecting the outcomes of HBV infection.

[Determination of trace cobalt in water samples by graphite furnace atomic absorption spectrometry after cloud point].

In buffer solution of citric acid monohydrate-disodium hydrogen phosphate at pH 5-7.5, cobalt water samples were chelated by 1-(2-pyridylazo)-2-naphthol (PAN) to form Co-PAN. After water-bath at 66 degrees C for two hours, Co-PAN is extracted into Triton X-100 nonionic surfactant phase and separated from bulk water. Extracted cobalt content was measured by graphite furnace atomic absorption spectrometry. The surfactant phase separated was treated with 0.5% HNO3-0.1% Pd(NO3)2 to remove background interference at 1200 degrees C temperature. The pre-concentration of cobalt in water samples permitted the detection of 0.003 microg x L(-1) (10sigma) with the enhancement factor of 100. The recoveries were 90.5%-106%. The proposed method was applied to the determination of cobalt in water samples and satisfactory results were obtained.

[Risk factors and preventive steps for nasosinusitis after radiotherapy of nasopharyngeal carcinoma].

OBJECTIVE: To study the risk factors and preventive steps of nasosinusitis after radiotherapy of nasopharyngeal carcinoma (NPC). METHOD: Two hundreds and thirtyone NPC patients were retrospectively analysed. RESULT: Among 231 NPC patients, 98 patients (42.4%) had nasosinusitis before radiotherapy, 165 patients (66.7%) suffered from nasosinusitis after radiotherapy. The rate of nasosinusitis in NPC patients with high T stage and nasalcavity anatomical aberrance was higher than in others (P = 0.014, CR = 1. 973; P = 0. 034, OR = 2.156), and the rate of nasosinusitis in NPC patients with nasopharynx therapy was low (P = 0.008, OR = 0.675), most patients with nasosinusitis before radiotherapy had not any improve after radiotherapy,and effected the effect of radiotherapy. CONCLUSION: To reduce nasosinusitis after radiotherapy and improve patients quality of life, we suggest that nasopharyngeal cavity therapy should be carried out during and after irradiation for NPC patients, especially for those patients with high T stage and nasalcavity anatomical aberrance, and for those who had nasosinusitis before radiotherapy.

[High-level expression of the potential vaccine antigen TSO18 of Taenia solium in Pichia pastoris].

TSO18 gene was subcloned into the Pichia pastoris expression vector pPIC9K. The recombinant plasmid pPIC9K-TSO18 was transformed into P. pastoris GS115 by electroporation so that the plasmid will be integrated with chromosome of P. pastoris. The P. pastoris strains containing multi-copy recombinant were screened by G418 and induced by methanol. The expression product was analyzed by SDS-PAGE, Western blot, deglycosylation, and purified by Sephadex column, and was used to immunize mice. The results indicated that the target protein was efficiently expressed in P. pastoris, and glycosylated moderately, and had immunological activity. In a 5 liter fermentor, the expression level of the target protein was up to 2.54 mg/mL. These results will benefit for the development of genetically engineering vaccine.

[Measurement of CD4+CD25+ T cells in breast cancer patients and its significance].

OBJECTIVE: To evaluate the changes of CD(4)(+)CD(25)(+) T cells in peripheral blood from patients with breast cancer. METHODS: Sixty four patients with breast cancer, 15 patients with benign breast tumors and 9 healthy volunteers were included in this study. The proportion of CD(4)(+)CD(25)(+) T cells population in total T cells was evaluated by flow cytometric analysis. The cytokine production (TGF-beta1) was measured by ELISA. RESULTS: The population of CD(4)(+)CD(25)(+) T cells in peripheral blood from patients with breast cancer accounted for (5.1 +/- 2.9)% of the total amount of T lymphocytes, and was significantly higher in comparison with that in patients with benign tumors and in healthy volunteers (P < 0.05). The CD(4)(+)CD(25)(+) T cells population in breast cancer patients was positively correlated with the cancer size and with TGF-beta1 level (r = 0.511 and r = 0.253, respectively), and negatively correlated with CD(8)(+)CD(28)(+) T cells and NK cells (r = -0.243 and r = -0.301, respectively). CONCLUSION: The CD(4)(+)CD(25)(+) regulatory T cells in peripheral blood of patients with breast cancer is significantly increased in comparison with that in patients with benign breast tumor and in healthy subjects. It may be responsible for immune suppression in breast cancer patients.

[The effect of surgical treatment on thyroid-associated ophthalmopathy].

OBJECTIVE: To evaluate the effect of muscle surgery on strabismus caused by extraocular muscle for thyroid-associated ophthalmopathy (TAO), and assess the change of vision acuity, intraocular pressure, visual field and blood flow velocity in central retinal artery that made by surgery treatment. METHODS: Recession and release of the extraocular muscles were used in 17 TAO patients (20 eyes) with strabismus caused by the change of extraocular muscles. In those patients one of them was undergone superior rectus tendon resection and other one was undergone superior rectus suspension. Another patient right eye was undergone inferior rectus recession and the left one with superior rectus recession. For rest of 14 patients with 17 eyes inferior rectus recession was performed. The vision acuity, intraocular pressure, visual field, and blood flow velocity in central retinal artery were measured pre- and post-operatively. RESULTS: In 17 patients (20 eyes), 5 eyes vision acuity improved two or more lines and 15 eyes was unchanged postoperatively. Intraocular pressure and mean defect of visual field showed statistically significance (P < 0.01). The peak systolic velocity (PSV) and the end diastolic velocity (EDV) were significantly increased, and the resistance index (IR) was significantly reduced (P < 0.05). CONCLUSION: Muscle surgery of strabismus caused by extraocular muscle for TAO in the quiescent period can eliminate strabismus, decrease intraocular pressure, improve visual field and blood flow velocity in central retinal artery.

[Observation on effects of hepatitis B vaccine immunization for 12 years in children in Beijing].

OBJECTIVE: To evaluate the long-term protective effects of hepatitis B vaccine after immunizing to the children for 12 years in Beijing. METHODS: The multiple stratified cluster sampling was used in this epidemiological survey. The sampling children's blood serum HBsAg, anti-HBs and anti-HBc were checked and measured by the solid phase radioimmunoassay (SPRIA). The serological level of these index and the causes of the children with HBsAg positive were analyzed. RESULTS: There were 2,419 cases 3-12 years-old children immunized with the hepatitis B vaccine in infant period were surveyed and the total HBsAg positive rate was 0.52%. The vaccine protective rate was 88.45% (95% CI: 65.67%-97.89%). The total anti-HBc positive rate was 2.21%, being no statistical significance among the age groups. The average anti-HBs positive rate of 3-6 years-old children immunized with gene recombining vaccine was 38.79% and descending greatly following the age's dropping. The geometric means of anti-HBs serological titer (GMT) was 52.83 mIU/ml, showing no statistical significance among the age groups. The average anti-HBs positive rate of 6-12 years-old children immunized with the blood rooting vaccine was 50.79%. The geometric means of anti-HBs serological titer (GMT) was 61.51 mIU/ml. There were no statistical significances among the age groups. Among the HBsAg positive children, more than 50% of the children's mothers were HBsAg positive also. CONCLUSIONS: The protective effects given by immunization were significant after the hepatitis B vaccine vaccination for 12 years in Beijing. The booster immunization was not necessary, because the HBsAg positive rate didn't ascend obviously as the immunization time prolonging. As the anti-HBs positive rate of children who were immunized by the gene recombining vaccine might be descending following the age's dropping greatly, we should strengthen the serological surveillance of hepatitis B. The main cause that the children became the HBsAg carrier should be a vertical transmission.

[Purification and properties of recombinant extremely thermostable and acid-stable amylase].

Extremely thermostable and acid-stable a-amylase produced by Pichia pastoris GS115/pPIC9K-Amy-228 was purified to electrophoretic homogeneity by the steps of ultrafiltration and PAGE. Purification of about 11.7 fold was achieved with an overall yield of 29.8%. Its molecular weight was estimated to be about 55kD by SDS-PAGE. The isoelectric point was 5.0 (room temperature). Michaelis constant of the enzyme for soluble starch was 1.12g/L. The carbohydrate content was 15.4% by the phenol-sulfuric acid method. The optimum temperature and pH of the enzyme activity were 95 degrees C and 4.5 respectively. The enzyme activity was stable under room temperature in the pH rang of 4.0 - 7.0 for 48 hours. About 60% of the initial enzyme activity was measured after 1h of incubation at 110 degrees C. The activity was strongly inhibited by Fe2+, Cr2+ and Cu2+, While Ca2+ had no effect on it. DTT and EDTA had no effect on the activity.

[Interferon-alpha and ribavirin combination therapy for co-infection of hepatitis C virus and human immunodeficiency virus].

OBJECTIVE: To evaluate the clinical effect and side-effect of interferon-alpha (IFN-a) and ribavirin (RBV) combination therapy for Chinese patients with co-infection of hepatitis C virus (HCV) and human immunodeficiency virus (HIV), and to compare them with only HIV infection patients. METHODS: 10 patients with HCV-HIV and 17 patients with only HCV infection received 5 million units of IFNalpha-2b every other day intramuscularly, and 300 mg RBV orally three times a day. Dynamic observations were done for HCV RNA and HIV RNA loads, CD4+ and CD8+ T lymphocyte counts, liver function and blood cell measures, and the side-effects of the medicines. RESULTS: After 12 weeks and 24 weeks of IFNalpha and RBV combination therapy, mean HCV RNA levels reduced 1.14 log (t = 3.843, P < 0.01) and 2.08 log (t =6.564, P < 0.01) from the baseline at week 0 in the HCV-HIV co-infection group, and reduced 1.48 log (t = 6.438, P less than 0.01) and 2.33 log (t = 7.343, P < 0.01) in the HCV infection group. Meanwhile, the HIV RNA levels decreased 1.22 log (t = 3.662, P < 0.01) and 1.73 log (t = 6.119, P < 0.01) from the base line. However, there were no obvious different changes among T lymphocyte counts of HCV-HIV and HCV patients at week 0, week 12 and week 24. All 27 patients showed satisfactory biochemical response to therapy. There were some mild or moderate influenza-like symptoms, intestinal discomfort and decreased blood cell counts in the early stages of the treatments. No neuropsychic and auto-immune disorders were found. CONCLUSIONS: IFNalpha-2b and RBV combination therapy showed similar anti-HCV effects during the 24 week treatment for HCV-HIV and HCV infected patients, and some anti-HIV effect was also observed. No obvious different biochemical responses and side-effects were found between the above two groups.

[Effects of aldose reductase transfection on the proliferation of rat mesangial cells in vitro].

OBJECTIVE: To study the effects of aldose reductase (AR) on the proliferation of rat mesangial cells (MsC) in vitro and to investigate its mechanism. METHODS: Cell proliferation was assessed by MTT colorimetric assay. Cell cycle and apoptosis were analyzed by flow cytometry. The growth of normal MsC and AR transfected MsC was compared. The proliferation of PDGF-BB and cellular growth stimulation by 10% NBS were investigated using AR inhibitors (ARI) Sorbinil and Zopolrestat. The effects of PDGF-BB on the expression of AR, p65 and c-Jun were assessed by Western blot. Activation of AP-1 was measured by EMSA. RESULTS: AR expression of transfected MsC was distinctly higher than that of the control. Transfected MsC grew quicker than normal cells. ARI partially inhibited the proliferation of transfected MsC under the stimulation of PDGF-BB and 10% NBS, whereas 10% NBS had no effect on normal MsC. PDGF-BB upregulated the expression of AR and c-Jun, but had no effect on p65. The upregulation of c-Jun and the activation of AP-1 could be attenuated by ARI. CONCLUSION: AR may participate in the pathological proliferation of MsC through the pathway related to the activation of AP-1.

[Study on proliferation effect of extracts of Piper longum on mesenchymal stem cells of rat bone marrow and the relationship to chemical functional groups].

OBJECTIVE: To observe the effect of volatile oil and aqueous soluable part of Piper longum on proliferation of rat mesenchymal stem cell (MSC) and the chemical functional groups. METHODS: Mesenchymal stem cells were dissociated from rat bone marrow and marked by Brdu, and the expression of CD44, CD54 and double label of Brdu and CD44. The growth of rat mesenchymal stem cell under volatile oil and aqueous soluable part of Piper longum was observed by means of cell viability measurement (MTT) and morphological observation and Brdu, PCNA immunohistochemical methods. RESULTS: Volatile oil of Piper longum could promote the cell viability of MSC and the number of Brdu, PCNA positive cell in dose-dependant. There was significant difference in comparision with control groups, C = C(50.66%), -OH(27.02%) and other functional groups in volatile oil of Piper longum were determined by GC-MS, but the aqueous soluable part of Piper longum could not promote the proliferation of MSC. CONCLUSIONS: Volatile oil of Piper longum which consists of C = C, -OH, and other functional groups has strong effect on enhancing proliferation of MSC.

[Impacts of petroleum-containing wastewater irrigation on microbial population and enzyme activities in paddy soil of Shenfu irrigation area].

The study showed that the upper reaches of main petroleum-containing wastewater irrigation channels had the highest accumulation and distribution of total petroleum hydrocarbon (TPH) (5 213.37 mg x kg(-1) dry soil),and the CFU of soil bacteria and fungi was increased with increasing soil TPH concentration, the correlation coefficient being 0.928 (P < 0.001) and 0.772 (P < 0.05), respectively. The activities of soil dehydrogenase, catalase and polyphenoloxidase had a significantly positive correlation with soil TPH concentration, their correlation coefficient being 0.974 (P < 0.001), 0.957 (P < 0.001) and 0.886 (P < 0.001), respectively, while soil urease activity showed a significantly negative correlation (P = 0.002), which could be used as the most sensitive indicator of petroleum contamination. The substrate-induced respiration (SIR) of polluted soil was significantly correlated with soil TPH concentration (P < 0.001), dehydrogenase activity (P < 0.001), and heterotrophic bacterial CFU (P = 0.006).

[Effects of acupuncture and continuous multiple points pulse stimulation on linguistic frequency hearing loss in the patient of sensorineural hearing loss].

OBJECTIVE: To explore effective therapeutic methods for sensorineural hearing loss (SNHL). METHODS: Thirty-four cases of mild and moderate SNHL who were definitely diagnosed with pure tone test and by otologists, including 24 cases of insufficiency of the liver and kidney, 10 cases of excess of liver-fire. They were treated with acupuncture at main points, Ermen (TE 21) or Tinggong (SI 19) or Tinghui (GB 2), and Yifeng (TE 17), Shenting (GV 24), Baihui (GB 20), Houding (GV19), Touwei (ST 9), Conger 1-3, Congnao 1-2; and adjuvant acupoints: Huangshu (KI 16), Qihai (CV 6), Guanyuan (CV 4), Taixi (KI 3) were added for insufficiency of the liver and kidney; Zhongwan (CV 12), Tianshu (ST 25), Yanglingquan (GB 34) and Taichong (LR 3) were added for excess of liver-fire. Sequential multipoint pulse stimulation was adopted, twice each week, for 20 consecutive weeks (40 sessions) and then pure tone test was carried out repeatedly. RESULTS: Of 34 cases (58 affected ears), 13 cases were cured and 21 improved, and the various linguistic frequency hearing threshold pure hearing values before and after treatment increased significantly (P < 0.05). CONCLUSION: Acupuncture and continuous multipoint pulse stimulation has definite therapeutic effect on linguistic frequency hearing loss in the patient of SNHL.

Association of 4G/5G polymorphism in PAI1 promoter with PAI1 level in deep vein thrombosis.

OBJECTIVE: To reveal the association of 4G/5G polymorphism in the promoter region of the plasminogen activator inhibitor 1 gene (PAI1) with plasma PAI1 level in deep vein thrombosis (DVT) in Chinese Han ethnic group. METHODS: One hundred and twenty Chinese DVT patients and 120 healthy controls were recruited. The PAI1 promoter 4G/5G polymorphism was detected using polymerase chain reaction (PCR). The antigen of tissue-type plasminogen activator (tPA) or PAI1 was quantified by a commercially available enzyme-linked immunosorbent assay (ELISA) in DVT cases and health controlsì respectively. RESULTS: Neither in the distribution of PAI1 promoter 4G/5G polymorphism nor in the frequencies of 4G and 5G allele was there a difference between two groups. The levels of PAI1 antigen in the carriers of the 4G/4G genotype were significantly higher than those either in the 4G/5G genotype or in the 5G/5G genotype; In the 4G/5G genotype or in the 5G/5G genotype the TG levels are an independently determinant factor of PAI1 antigen levels. CONCLUSION: There is a close relationship of the PAI1 4G/5G polymorphism to its plasma level in deep vein thrombosis in Chinese Han ethnic group, although lack of association between this genetic variation and risk of DVT suggest no major cause-effect pathogenic role of this polymorphism by itself.

[Effects of electroacupuncture on CRH, IL-2 and beta-EP contents in the hypothalamus in the rat of adjuvant arthritis].

OBJECTIVE: To explore Mechanisms of electroacupuncture in anti-inflammation and immunoregulation, and to compare differences among "Dazhui" (GV 14), "Mingmen "(GV 4) and non-acupoint in these actions. METHODS: Adjuvant arthritis rat model was made. The rats were divided into 5 groups: normal group, model group, "Dazhui" (GV 14) electroacupuncture group, "Mingmen" (GV 4) electroacupuncture group and non-acupoint electroacupuncture group. Effects of electroacupuncture on local joint inflammation and corticotropin-releasing hormone (CRH), beta-endorphin (betaEP) and interleukin-2 (IL-2) contents in the hypothalamus were observed and differences of the actions among different points were compared. RESULTS: CRH content in the hypothalamus in the "Dazhui" (GV 14) electroacupuncture group decreased as compared with the model group (P < 0.05); there was no significant change in beta-EP and IL-2 contents of the hypothalamus in the 3 electroacupuncture groups as compared with the model group (P > 0.05); there was positive correlation between CRH and IL-2 contents, and between IL-2 and beta-EP contents in the hypothalamus. The swelling rate of the toe in the "Dazhui" (GV 14) group and the "Mingmen" (GV 4) group was lower than that in the non-acupoint group (P < 0 05). CONCLUSION: Electroacupuncture exerts anti-inflammation and immunoregulation possibly via regulation of CRH, IL-2 and beta-EP each other, and the anti-inflammation action of "Dazhui" (GV 14) and "Mingmen" (GV 4) is stronger than that of non-acupoint.
