The changes of the juxtaglomerular index values in the superficial, deep and whole cortical areas of rat kidneys after unilateral papillectomy were analyzed. On the papillectomized side the JGI values showed an increase in the deep, and a decrease in the superficial cortical zone. The changes proved to be significant during the whole experimental period in the juxtamedullary index and between the 4--14 postoperative days in the superficial index values. The tendency of the changes of JGI in the contralateral kidneys was similar to those of the papillectomized kidneys, but significant differences were registered less constantly. These changes are probably the consequences of haemodynamic factors. Papillectomy is considered an incompetent model for studying the regulatory function of macula densa in the synthesis of renin.
AIM: The aim of the study was to examine the effects of testosterone and oestrogen on the ECG parameters and expression of cardiac ion channels in male and female dogs, and to compare the dofetilide-induced lengthening of QTc interval in control, castrated and hormone-treated animals. METHODS: ECG records were taken from male and female anaesthetized dogs (n = 10 in each group) before castration, after castration, and following inverted hormone substitution. The animals were challenged with dofetilide at each stage of the experiment. Finally, the hearts were excised and expression of ion channels was studied using Western blot technique. RESULTS: Heart rate was decreased and PQ interval increased by deprivation of sex hormones in both genders (orchiectomy or ovarectomy), while inverted hormonal substitution restored control values. Orchiectomy significantly increased the duration of QT and QTc intervals, QTc-dispersion and the dofetilide-induced lengthening of QTc, while testosterone treatment of castrated females had opposite effects. Intraventricular conduction (QRS duration) was independent of the endocrine status of the animals. Ovarectomy or oestrogen treatment of castrated males failed to alter significantly these parameters except for QTc-dispersion. Expression of ion channel proteins responsible for mediation of I(K1) and I(to) currents (Kir2.1 and Kv4.3, respectively), was significantly higher in the testosterone-treated castrated females and normal males than in the oestrogen-treated castrated males and normal females. CONCLUSION: Repolarization of canine ventricular myocardium is significantly modified by testosterone, but not oestrogen, in both genders. This effect is likely due to augmentation of expression of K(+)-channel proteins, and thus may provide protection against arrhythmias via increasing the repolarization reserve.
The technique of obtaining pancreatic segments was studied in 60 mongrel dogs by removing the right pancreatic lobe. At the same time comparative surgical-anatomical examinations were performed for the detection of the vascular variations of the right pancreatic segment. In 83% of the cases the arterious and venous systems were suitable for gaining grafts (Types I-II). In 12% the segment was suitable for grafting only if the venous network had an optimal course (Type III), while in 5% the segment was unsuitable because it did not have a suturable arterial stump (Type IV).
BACKGROUND/AIMS: We have studied the effects of insoluble glycogen applied intravenously on the experimental acute pancreatitis of dogs. METHODS: Experimental acute pancreatitis was induced by olive oil injected into the main pancreatic duct. The effects of insoluble glycogen were evaluated histologically and by measurements of levels of amylase, lipase in sera. The binding of endotoxin and secretable phospholipase A2 by insoluble glycogen were tested by isotopically labeled materials. RESULTS: Application of insoluble glycogen had beneficial effects on the experimental acute pancreatitis of dogs. CONCLUSION: The binding capacity on endotoxin and secretable phospholipase A2 could play the main role in the protection offered by insoluble glycogen.
