[Isolation of recombinant interleukin-3 produced by E. coli].

A synthetic gene coding for human interleukin-3 (hIL3) was cloned in the plasmid pTE2IL3, the gene expression being controlled by the phage fd PVIII promotor and the phage T7 gene 10 translational enhancer. Under constitutive biosynthesis conditions in E. coli, the accumulation of recombinant hIL3 (in the inclusion bodies) was up to 30-40% of the total cell protein. An effective procedure of the hIL3 isolation is suggested. The hIL3 was solubilized in 5 M guanidinium chloride, renaturated and purified to homogeneity by a single chromatographic step. The protein's yield was 34 mg/g wet cells. The isolated hIL3 showed a specific biological activity.

Insertion of short hepatitis virus A amino acid sequences into poliovirus antigenic determinants results in viable progeny.

In an infectious poliovirus cDNA construct, the determinant encoding antigenic epitope N-Ag1 (in a loop located between two beta-strands in poly-peptide VP1) was altered by site-directed mutagenesis, to be partially similar with the determinants for presumptive epitopes in polypeptides VP1 or VP3 of hepatitis A virus (HAV). The modified constructs proved to be infectious. However, another construct, in which the same locus encoded a 'nonsense' and a relatively hydrophobic amino acid sequence, exhibited no infectivity. These data showed the feasibility of the insertion of foreign sequences in a specific antigenically active locus of the poliovirus icosahedron, and suggest some limitations with respect to the sequences to be 'transplanted'.

Effective method for obtaining long nucleotide chains on partially complementary templates. Processed bovine gamma-interferon gene obtained from human gamma-interferon gene.

The method of obtaining the bovine gamma-interferon gene by means of simultaneous multidirected mutagenesis of the human gamma-interferon gene is presented. The first strand of the bovine gamma-interferon gene was obtained by ligation of synthetic oligonucleotides, using the cDNA of human gamma-interferon, cloned in the single-stranded phage M13mp19 as a template. The second strand was synthesized using a large fragment of E. coli DNA-polymerase I. The double-stranded gene was then treated by restriction nucleases and cloned in a pUC-18 derived vector. The primary structure was confirmed by sequencing.

[Isolation of the porcine alpha-interferon gene using simultaneous directed multipoint mutagenesis].

A method of obtaining the pig alpha-interferon gene by means of simultaneous multidirected mutagenesis of the human alpha 2-interferon gene is presented. Nucleotide homology between these genes is 80.4%. Fourteen synthetic oligonucleotides forming a pig alpha-interferon gene's strand were ligated on a single-stranded template, carrying cDNA of the human alpha 2-interferon gene. The obtained DNA fragment was cloned in the single-stranded or double-stranded form. It was found that the method does not affect the cloning efficiency. The primary structure of the gene was confirmed by sequencing.

trp operon induction during the expression in E. coli of two IFN-gamma sequences.

Two nucleotide sequences coding for mature human immune interferon (IFN-gamma) and differing from each other by nine N-terminal nucleotides were expressed in E. coli under the control of a trp promoter. The longer gene variant after the ATG initiatory codon contained a TGT TAC TGC sequence, which was absent in the shorter gene. When expressed in E. coli under the direction of identical transcription and translation regulatory elements, these genes showed different susceptibility to induction.

[Histophysiology and histopathology of the adrenals in experimental hypokinesia].

In experiments on male rats in the course of 3-month hypokinesia phasic changes were observed in the relative adrenal gland weight, in the volume of the cell nuclei of the glomerular, fasicular zones and the medulla, in the activity of the succinic dehydrogenase, alkaline and acid phosphatases, in the RNA and catecholamine content. These changes are associated with variations in the secretion and biosynthesis of the hormones of the cortical and the medullary layer of the adrenal glands during the hypokinetic stress development.

[Effect of agents used in the therapy of acute ischemic heart disease on the histomorphological changes in the myocardium in acute dystrophy].

The morphological analysis of the portions of the interventricular septum and left ventricular wall in cats has revealed dystrophic and necrotic changes in the myocardium after administration of caffeine followed by adrenaline: haemorrhages, intravascular thrombogenesis, dystrophic and necrobiotic changes in myocytes. In early periods of acute ischemic heart disease morphine, fibrinolysin, heparin and Inosie F either abolished (fibrinolysin) or considerably decreased (heparin and Inosie F) a tendency toward intravascular thrombogenesis and damage to the myocardial parenchyma.

[Bioelectrical, biochemical and histomorphological changes in the ocular tissues in pituitrin hypertension and the use of euphylline and papaverine].

Experimental study on rabbits of bioelectric and biochemical characteristics of eye tissues during pituitrin hypertension and application of euphylline and papaverine as well as comparison of the data obtained indicate that during pituitrin hypertension, transtissue potential difference and density of short-circuit current in the lens are considerably increased whereas the level of tissue respiration in the eye is reduced. Administration of euphylline or papaverine during pituitrin hypertension aggravates hemodynamic disorders in eye tissues, deteriorates the bioelectric and biochemical characteristics in both hypertensive and control animals.

[Comparative evaluation of the effect of plant polysaccharides on the inflammatory process and regeneration in chronic stomach ulcer].

It has been demonstrated in experiments on animals that new polysaccharides of plant origin exhibit an anti-inflammatory action which compares very favourably to that of butadiene. The polysaccharide isolated from the Asteraceae family plant has proved to be a highly active biological substance that demonstrably stimulated reparative processes in the gastric mucosa of rats with ulcerous-destructive lesions.

[Eye lesion from chromium compounds and its treatment in an experiment].

Experiments on 68 rabbits with degree III burn induced by potassium bichromate were made to study and compare the therapeutic effect of subconjunctival injections of autoplasma with heparin and penicillin and instillations of spermaceti oil. Administration of spermaceti oil for the treatment of burns provides a more remarkable clinical effect, contributing to a milder course of burn process.

[Morphometric analysis of the glomeruli and juxtaglomerular apparatus of rat kidneys during the course of experimental hypokinesia].

In the hypokinetic study a certain correlation between variations in the means of the volume of cortical and juxtamedullary glomerules and the function of the juxtaglomerular apparatus has been established. It is hypothesized that at early hypokinesia renal vascular changes are nonspecific. The phenomenon of renal excretion of fluids and electrolytes is explained in terms of a simultaneous increase in the reninangiotensin activity and 11-hydroxycorticosteroid content in plasma.

[Stress reaction in hypokinesia and its effect on overall body resistance].

Rat experiments have demonstrated that the development of the hypokinetic syndrome involves occurrence of a three-stage stress-reaction characterized by a change in the general health state, body mass, and activity of the hypothalamic-pituitary-adrenal system. The hypokinetic stress affects total resistance and ionizing radiation sensitivity of animals. At a certain stage of development the hypokinetic stress contributes to the state of elevated "cross" resistance.
