Establishment and characterization of a human cell strain, KT, with high sensitivity to UV-killing and to cell proliferation inhibition by interferon.

We have established a human cell line, designated KT, with high susceptibility to both cell proliferation inhibition by interferon and UV-killing, from a metastatic breast carcinoma. A tumor marker, a pregnancy-specific glycoprotein (Schwangerschaftsprotein 1; SP1), and carcinoma characteristics compatible with ductal carcinoma of the breast were seen in KT cells by electron microscopic observation. KT cells were slightly more resistant to X-ray-induced toxicity than fibroblastic cells, termed KS, from the scalp of the patient. But, KT cells had lower cloning efficiency after UV irradiation than did KS cells: D0 values of 1.5 J/m2 and 7.2 J/m2, respectively. KT cells also appeared more susceptible to human interferon (HuIFN) preparations (alpha, beta, gamma and natural or recombinant) than did KS cells, as measured by cell colony formation ability, proliferation rates, and [3H]deoxythymidine incorporation levels into acid-insoluble cell materials. The sensitivity of KT cells to UV and HuIFN was greater than that of human RSa cells, a cell line with high sensitivity to both agents. KT cells had more capacity for UV-induced DNA-repair replication synthesis than did RSa cells, the capacity being much the same as that of KS cells. There was no significant difference in levels of antiviral activity induced by HuIFN and binding capacity for 125I-labeled IFN-alpha A between KT and KS cells. KT cells appeared refractory to cell proliferation inhibition by tumor necrosis factor (TNF) preparations.

A scanning electron microscopic study of crescentic Masugi nephritis in the rabbit.

Crescentic glomerulonephritis was induced in the rabbit with two intravenous injections of goat nephrotoxic serum (NTS). Prominent proliferative glomerulonephritis, characterized by intracapillary as well as extracapillary emigration of monocytes and fibrin deposition, developed 7 days after the first injection. The changes rapidly progressed to crescent formation. In order to observe alterations of the glomerular basement membrane (GBM) related to crescent formation, unfixed, isolated glomeruli were treated with Triton X-100. The GBM thus denuded was shown to have a number of microperforations, which subsequently became much larger holes or fissures. It is suggested that monocytes and fibrin deposits may play a role in the induction of the GBM change.

Histopathologic studies on adenocarcinoma of the parotid gland.

Of 683 cases of primary epithelial tumors arising in the parotid gland, 20 cases could be defined as adenocarcinomas, constituting 2.9% of the total number or 10.0% of 201 malignant tumors. These adenocarcinomas were characterized by a marked diversity in the histologic manifestations, presenting tubular, papillary, pseudoadenoid cystic, cribriform, trabecular, sarcomatoid, and other patterns. Basically, however, they could be classified into two major subtypes, i.e., tubular and papillary variants on the basis of their predominant histological patterns. The tumor cells showed a distinctive sign of high grade malignancy such as the occurrence of many mitotic figures and involvements of lymphatic vessels and/or blood vessels. Ultrastructurally, the tumor cells consisted of two basic varieties, resembling myoepithelial cells and duct epithelium. It is thus suggested that immature tumor cells could differentiate into a structure mimicking an intercalated duct. Clinicopathologically, the tumors were seen with a higher frequency in middle-aged males (average 44 years of age).

Epithelioid haemangioendothelioma of the bone tissue.

An epithelioid haemangioendothelioma arising in a 43-year-old Japanese woman is presented. The patient first complained of severe back pain and neurogenic disturbances due to compression fracture of the spinal bone. Radiographically the tumour masses showed osteolytic lesions without reactive new bone formation. Similar lesions were noted in other bones. Biopsy and surgical specimens showed the features of epithelioid haemangioendothelioma. A lymphocyte associated antigen, HLA-DR which has been regarded as a B lymphocyte marker was detected in the tumour cells. The importance of this finding was emphasized with respect to the identification or classification of vascular tumours.

[A case of polymorphous adenocarcinoma in the right parotid gland].

The nature of adenocarcinomas of parotid gland origin seems to be still unclear because of the rarity of this lesion. A 65-year-old man with a "polymorphous adenocarcinoma" of the parotid gland is presented. This tumor is characterized by a marked diversity in the histologic manifestations, presenting dominantly solid, tubular, pseudoadenoid cystic sarcomatoid patterns. The tumor cells showed distinctive signs of high grade malignancy such as the occurrence of mitotic figures and involvements of lymphatic vessels and/or blood vessels. "Polymorphous adenocarcinoma" is a histologically distinctive neoplasm unlike other adenocarcinomas of parotid gland origin.

[A case of adenolipoma of the breast].

A 40-year-old woman was found to have a well-circumscribed mass in the upper outer quadrant of the right breast at a routine physical examination. Mammographically, the tumor was a radiolucent mass, measuring 4.6 X 3.2 X 4 cm, surrounded by a capsule. The tumor consisted of fat and numerous strings with the appearance of ordinary glandular tissue. There was no area of microcalcification. Grossly, the tumor mass was encapsulated by fine connective tissue. On the cut surface, the tumor consisted of fatty tissue containing islands of grayish-white breast tissue. Histologically, the fatty tissue presented (a feature) of mature lipoma. Embedded in the fat were areas of breast tissue simulating fibrocystic disease. In limited areas, the fat cells had direct contact with the ducts without intervening fibrous tissue.

Cellular aspects of rabbit Masugi nephritis V. Ultrastructural cytochemistry of monocytic cells.

Ultrastructural cytochemistry for nonspecific esterase (NSE) was performed on normal and nephritic rabbit kidneys. In normal glomeruli distinct reaction deposits appearing as electron-dense granules were present in visceral epithelial cells (podocytes) and epithelial cells of Bowman's capsule, but only a few small or minute deposits were seen in some endothelial and mesangial cells. In acute proliferative glomerulonephritis (Masugi nephritis), many reaction deposits occurred in mononuclear cells accumulating in glomerular tufts which presented the characteristic features of monocytic cells. Macrophages which had migrated into subendothelial space as well as epithelioid cells and multinucleated giant cells, all of which are known to be derived from monocytes, also exhibited the reaction product. The NSE granules in mesangial and endothelial cells were much smaller and fewer in number than those in monocytic cells. The present method may contribute to the more precise differentiation of monocytic cells from mesangial and endothelial cells in proliferative glomerulonephritis.

Signet ring cell carcinoma of the breast.

A rare case of signet ring cell (SRC) carcinoma of the breast occurring in a 51-year-old female is presented. She was first admitted under the diagnosis of gastric carcinoma. Incidentally, a right breast tumor was detected and double cancer was thus suspected clinically. Concurrent right mastectomy and partial gastrectomy were performed. Histologic examination on the removed tumors revealed infiltration of numerous SRCs. The signet ring appearance of tumor cells resulted from formation of a large intracytoplasmic globule. From this unique cytologic feature along with the presence of in situ lesion in the breast tumor, it was concluded that the gastric malignancy was a sequela of metastasis of infiltrating lobular carcinoma. Wide-spread metastasis later occurred, especially on the serosal surfaces. The importance for the knowledge about the unique metastatic pattern of SRC carcinoma of the breast was emphasized.

Primary nongestational choriocarcinoma of submental region.

A case of a 47-year-old man with choriocarcinoma is reported. This case is quite unique in that the primary tumor presented in the submental region. Multiple remote metastases developed within a few weeks after the tumor extirpation, resulting in a fatal outcome. The neoplasm revealed the features of pure choriocarcinoma. By the immunoperoxidase method, it was demonstrated that both cyto- and syncytiotrophoblasts produced HCG.

A histopathologic study of benign and malignant lymphoepithelial lesions of the parotid gland.

Twenty-three cases of benign and malignant lymphoepithelial lesions without Sjögren's syndrome were examined: 21 were classical benign lymphoepithelial lesions, and were mostly found in males older than 50 years of age. The other two cases were identified as undifferentiated carcinoma arising from benign lymphoepithelial lesion (8.7%; 2/23). Histologically, this carcinoma is characterized by the formation of cell nests of variable size and by marked lymphoplasmacytic infiltrations into the stromal tissue. In immunofluorescence, an increase of plasma cells containing IgA and IgG was noted in the marginal areas of carcinoma cell clusters. Electron microscopy disclosed that the carcinoma was composed of poorly differentiated cells with occasional squamous differentiation. No dysplastic characteristics were found in the infiltrating lymphocytic components. These two cases of carcinoma were found in middle-aged females.

Cytologic and ultrastructural studies of a rare breast carcinoma with osteoclast-like giant cells.

An unusual case of breast carcinoma with osteoclast-like giant cells (OGCs) was reported, for which the diagnosis was made by aspiration biopsy. The OGCs appeared to derive from large mononuclear cells, probably via their cellular fusion. The mononuclear cells had abundant lysosome-like granules, endoplasmic reticulum, and mitochondria. They were thought to be histiocytic cells, though active phagocytosis could not be demonstrated. Histologically, the tumor showed a pattern of well differentiated ductal adenocarcinoma, of which the stroma was crowded with histiocytic cells, OGCs, and lymphocytes. Marked hemorrhage and fibrosis were also seen. The tumor cells tended to be distorted and greatly diminished in accord with the accumulation of OGCs.

Histopathologic studies of undifferentiated carcinoma of the parotid gland.

Five-hundred-fifty-five primary epithelial tumors of parotid gland origin were examined. Eighteen cases could be defined undifferentiated carcinomas, constituting 3.2% of the total number or 10.2% of 176 malignant tumors. The undifferentiated carcinomas could be divided on the basis of the cell size into two types: (1) Small-cell type (12/18; 66.7%); the tumor cells were as large as, or slightly larger than lymphocytes. The cytoplasm was scanty with dark chromatin. Mitotic figures were numerous. The tumor cells in the periphery of the cell clusters exhibited a palisading arrangement. Electron microscopy revealed that these cells were rich in tonofilaments and connected by desmosomes. (2) Large-cell type (6/18; 33.3%); the carcinoma cells were roughly twice as large as those of the small-cell carcinoma. The tumor cells were spheroidal or spindle in shape with rich and clear cytoplasm. Many of the tumor-cell clusters showed macroalveolar structures, suggesting the possibility that this type occurred as a variant of poorly differentiated adenocarcinoma or epidermoid carcinoma. Clinicopathologically, there was no sex difference in the total number of the undifferentiated carcinomas, but the large-cell carcinomas were frequently encountered in females (83.3%). As to the age distribution, the carcinomas were found frequently in middle-aged patients from 30-49 years (66.7%). Many of the tumors were 2-4.9 cm at their greatest diameter (55.6%). Tumor mass less than 1.9 cm could not be found.

Histopathologic studies of basal cell adenoma of the parotid gland.

Five-hundred-thirty-one cases of primary epithelial tumors of the parotid gland were examined. The incidence of basal cell adenomas was 7.5% (40 cases). The tumors could be classified histologically into these subtypes: basal cell (21/40; 42.5%); tubular, (13/40; 32.5%); trabecular (4/40; 10.0%); and papillary (2/40; 5.0%). The pattern of classic basal cell adenoma predominated in the basal-cell type but was also seen in varying degrees in the latter three types with a reciprocal transition. Accordingly, it could be suggested that tubular, trabecular, and papillary types are variants of basal cell adenoma. The higher incidence of basal cell adenomas in our survey could be accounted for by this categorization. Histologically, basal cell adenomas presented various features; (1) cystic formation (26/40; 65.0%), being most frequently seen in tubular and trabecular types; (2) adenoid cystic pattern (4/40; 10.0%); (3) elastosis in the stromal tissues (2/40; 5.0%). Basal cell adenomas were clinicopathologically compared with 291 cases of pleomorphic adenomas. Basal cell adenomas were seen more often in female patients, (60.0%), as were pleomorphic adenomas (68.4%). They were observed more frequently in patients over 50 years of age and the average was ten years higher than for pleomorphic adenoma. The tumor size tended to be smaller (below 2 cm at the greatest diameter) than pleomorphic adenomas.

Histopathologic studies on carcinoma in pleomorphic adenoma of the parotid gland.

Five-hundred-eighteen cases of primary epithelial tumors of the parotid gland were examined. The incidence of carcinomas in pleomorphic adenoma was 9.3% (48 cases). In addition to cellular atypism, the following histological findings were thought to be important criteria for the diagnosis of carcinomas: (1) capsular invasion, (2) infiltration into adjacent organs, (3) proliferation of atypical cells within fibrous tissues and chondroid matrix in the area of pleomorphic adenoma, (4) vascular involvement, and (5) mitotic figures. Pleomorphic adenomas (282 cases) were found predominantly in females (70.8%), at the age of 30 to 49 (48.2%), whereas carcinomas in pleomorphic adenoma were more often found in males (70.8%) and at the ages over 50 (43.8%). Histologic types were composed mostly of undifferentiated carcinoma (56.2%) and adenocarcinoma (37.5%). The prognosis of the carcinomas, especially the undifferentiated type, was generally poor; about 45% of the cases either experienced recurrence or died within five years. Electron microscopy on this undifferentiated carcinoma revealed characteristic filamentous structure in the carcinoma cells, resembling that observed in myoepithelial cells. It was assumed that the undifferentiated carcinoma cells were of myoepithelial origin.

Immunohistochemical studies on myoepithelial changes in breast tumors.

Specific rabbit antisera directed against human uterine myosin were prepared and used for immunohistochemical studies on myoepithelial cells (MECs) in some breast tumors. In fibroadenomas, MECs were observed confined to the periductal portions but not in the stroma. In infiltrating duct carcinomas, MECs were also present as a distinctive periductal layer. Moreover, they were also seen in the infiltrating area, either associated with a small nest of carcinoma cells or freely in the stroma. Larger nests of medullary carcinomas usually were not accompanied by any layer of MECs. No MECs were detected in lobular carcinomas as far as the infiltrating area was concerned. It is suggested from these results that the behavior of MECs is significantly different, depending upon the types of breast tumors. It also appears that the present method may be useful for further analysis of the origin and growth pattern in various breast tumors.

Tyrosinase-mediated inhibition of in vitro leucine incorporation into mouse melanoma by 4-isopropylcatechol.

The effect of 4-isopropylcatechol (4-IPC), a potent, irreversible cutaneous depigmenting agent, on protein biosynthesis of malignant melanoma cells in mice was studied by examining the in vitro amino acid (leucine) incorporation into a microsome fraction in cell sap. The present study revealed that 4-IPC does not inhibit the protein biosynthesis of the cell-free system in mouse liver, but remarkably inhibits it in mouse melanoma cells, which contain a high level of tyrosinase. The enhanced inhibition was found also in the mouse liver cell-free system when tyrosinase was added. Air oxidation products of 4-IPC were not responsible for such inhibition. These results may indicate that 4-IPC directly inhibits protein biosynthesis, probably by some intermediates that occur in an early stage of enzymatic oxidation of 4-IPC.
