Improvement of the reproductive performance of sows by treatment with a GnRH superactive analogue.

The effect of a Hungarian-made superactive analogue of GnRH (Ovurelin, D-Phe6-GnRH-EA, Reanal, Hungary) on the postpartal sexual function of sows was monitored. GnRH treatment was carried out on day 19 before weaning. The sows were inseminated at the first oestrus after weaning. GnRH treatment markedly increased litter size at weaning, substantially reduced (to 25 and 50%, respectively) the number of sows failing to come into oestrus after weaning, and increased the number of sows coming into oestrus within one week after weaning by 42.5% and 9%, respectively. These beneficial effects were particularly apparent on the far using closed management technology.

Simultaneous occurrence of selective ACTH insensitivity, achalasia and alacrimia accompanied by hyperlipoproteinaemia.

An extremely rare clinical syndrome on a 7-year-old-girl is presented. Besides isolated glucocorticoid insufficiency, achalasia and alacrima disturbance of the lipid metabolism was also detected--being a special feature of this case. The details of the endocrine workup is discussed, providing clues for the possible pathomechanism. The correct diagnosis and specific therapy is of utmost importance in the everyday life of the patient.

[Management of tibial fractures with Ender nailing].

Authors have performed in 1984-1986 46 Ender nailings based on definite indications as treatment of lower leg fractures in the Department of Traumatology, Arpád Hospital, Ujpest. In their clinical study they stress the criteria of the choice of this method, underline the advantages and disadvantages of the stabilization with elastic nails. This treatment used especially for fractures of the distal third lightens the care, the mobilization, shortens the stay in the hospital, diminishes the number of the septic complications. In many cases authors recommend it as a good alternative method, based on own experiences and results, to surgeons forced to accept a compromise.

Correlation between human lactoferrin binding and colicin susceptibility in Escherichia coli.

Escherichia coli H10407 demonstrated low 125I-human lactoferrin (HLf) binding (7%) and was insusceptible to group A (A, E1, E2, E3, E6, and K) and group B (B, D, Ia, Ib, and V) colicins. Conversely, a spontaneous HLf high-binding (44%) variant, H10407(Lf), demonstrated an increase susceptibility to both colicin groups. Colicin-insusceptible E. coli wild-type strains 75ColT, 84ColT, and 981ColT showed a low degree of HLf binding, i.e., 4, 8, and 10%, respectively. The HLf binding capacity was high in the corresponding colicin-susceptible mutants 75ColS (43%), 84ColS (32%), and 981ColS (43%). Furthermore, HLf low- (less than 5%) and high- (greater than 35%) binding E. coli clinical isolates (10 in each category) were tested for susceptibility against 11 colicins. Colicin V susceptibility did not correlate with HLf binding in either categories. However, with the remaining colicins, three distinct HLf-binding, colicin susceptibility patterns were observed; (i) 10 of 10 HLf low-binding strains were colicin insusceptible, (ii) 6 of 10 HLf high-binding strains were also colicin insusceptible, and (iii) the remaining HLf high binders were highly colicin susceptible. Certain proteins in the cell envelope and outer membrane of wild-type H10407 (HLf low binder, colicin insusceptible) showed a lower mobility in sodium dodecyl sulfate-polyacrylamide gel electrophoresis compared to the corresponding proteins of mutant H10407(Lf) (HLf high binder, colicin susceptible). These mobility differences were also associated with HLf-binding proteins in Western blot (ligand blot) analysis. The wild type showed a smooth form of lipopolysaccharide (LPS) with a distinct ladder of O-chains, compared to the rough LPS of the mutant.(ABSTRACT TRUNCATED AT 250 WORDS)

Effect of treatment with Prolan or with a GnRH superactive analog on the sexual function of sows after weaning.

On a large, closed pig farm using artificial insemination (AI), 29 sows were treated with Prolan S-öl injection (Bayer, FRG) and 31 sows with a GnRH superactive analog (Ovurelin, D-Phe6-GnRH-EA, Reanal, Hungary) 48 h after weaning. The effect of treatment on the sows' sexual function was monitored by serum progesterone radioimmunoassay (RIA). The conception rate in the control group (36 sows) was 69.4%. In the groups treated with Prolan and Ovurelin it was 79.3 and 71%, respectively. The use of Prolan S-öl injection markedly reduced the number of acyclic sows and of those having an irregular oestrous cycle. Treatment with the GnRH analog inhibited the manifestation of weaning-induced heat; subsequently, however, it induced a regular cycle in 30 out of the 31 sows treated.

Cloacin tolerance and aspecific colicin insensitivity of human Escherichia coli strains.

Of 182 wild-type human, aerobactin producer Escherichia coli strains 86.3% were insensitive to cloacin. All randomly chosen 51 strains were relatively cloacin tolerant. Cloacin tolerant strains were not considerably more sensitive to hydrophobic drugs than the cloacin sensitive descendant strains. Pathogenicity of the cloacin sensitive strains was significantly lower (p less than 0.05) in intraperitoneal mice infection than that of the cloacin tolerant ones. Suggesting a new aspect of the uptake mechanism of colicins, cloacin tolerance was very frequently associated with an aspecific insensitivity to a broad spectrum of colicins.

[Increased androgen-binding capacity and decreased free androgen index in male alcoholics with liver disease].

Androgen binding capacity in the serum of 30 male patients under 51 years of age suffering from chronic alcoholic liver disease was investigated with tritiated 5 alpha-dihydrotestosterone. The result received by saturation analysis is regarded as an indicator of the SHBG (sex hormone binding globulin) content of the blood. Testosterone was measured simultaneously and the free androgen index (FAI) was calculated. The diagnosis of liver disease was based on case histories, physical symptoms and laboratory abnormalities, in some cases on liver biopsy too. Age-matched control group consisted of 18 healthy males. The results showed markedly elevated androgen binding capacities (SHBG contents), testosterone values in the normal range and significantly decreased FAI, indicating insufficient amounts of biologically active testosterone.

Direct radioimmunoassay of serum progesterone for monitoring the reproductive status of large pig herds.

The reproductive status of sows was monitored by serum progesterone radioimmunoassay (RIA) in a closed pig herd of 1100 sows, producing for the market. The method proved suitable for monitoring reproductive events (progesterone production during lactation, effect of weaning on the resumption of cycling, successful and unsuccessful inseminations, abortion, etc.) and for diagnosing ovarian disturbances (acyclia, irregular cycle, etc.). A total of 813 inseminations were followed up to determine the time of return to oestrus. The serum progesterone values of blood samples taken from the v. cava cranialis on day 22 after insemination indicated that pregnancy was diagnosed with a reliability of 75.5%, while non-pregnant sows failing to return to oestrus were identified with a reliability of 96.1%. The economic benefits to be derived from the use of this method in a pig herd are discussed.

High-dose ketoconazole treatment in advanced prostatic cancer.

For purposes of testosterone suppression, high doses of ketoconazole were administered to 18 patients with advanced cancer of the prostate for an average of 6 months. A well-evaluable response was observed both objectively and subjectively. The blood levels of the steroid hormones were measured during the treatment period and it was found that testosterone biosynthesis was blocked. Mild gastrointestinal symptoms occurred in the minority of the cases, and administration of the drug had to be stopped in one case because of jaundice. The possibility of the use of smaller doses is being investigated, which may lead to the development of an alternative to the endocrine treatment methods.

Determination of phage type, colicin type and plasmid profile in Shigella sonnei.

Complex typing was carried out of 33 Shigella sonnei strains isolated in 1985 and 1986 One to six plasmids were carried by 32 strains typable by phages, whereas plasmid was not demonstrated in a phage-resistant strain. A 1.4 Md plasmid was carried by all strains. By determining the plasmid profile the clonal connection between the strains could be demonstrated. Strains assumed to be in clonal connection on the basis of phenotypic properties, were divided into subclones. Identical plasmid profile was observed among strains belonging to different phage and colicin types.

Curing of plasmid pE194 with novobiocin and coumermycin A1 in Bacillus subtilis and Staphylococcus aureus.

Plasmid pE194 determining macrolide-lincosamid-streptogramin B resistance could be eliminated with novobiocin and coumermycin A1 in Bacillus subtilis and Staphylococcus aureus. In both organisms the curing effect depended on the temperature. In the case of Staphylococcus aureus it increased very much at high temperatures, while in the case of Bacillus subtilis only a moderate increase could be observed. The curing activity of coumermycin was substantially weaker than that of novobiocin in both bacteria. This difference decreased at higher temperatures only in Staphylococcus aureus. In Bacillus subtilis the curing with novobiocin could be antagonized with coumermycin in accordance with the growth experiments reported previously.

Antibiotic resistance and plasmid profiles of Escherichia coli and Klebsiella isolated from in-patients receiving prolonged antibiotic therapy.

Distribution by serogroup, phage type, colicin production, colicin type, sensitivity to antibiotics and plasmid characteristics of 74 Escherichia coli and 11 Klebsiella strains isolated from hospitalized patients receiving prolonged antibiotic therapy indicated that the infections were not associated with the hospital environment. Resistance was tested to 26 antibiotics, some of them being not generally used in therapy; 30 strains were resistant to 4 to 17 antibiotics. There was a significant difference in the antibiotic resistance of strains derived from patients with urinary-tract infections (UTI) and with leukaemia (LP). As compared to the UTI group, among E. coli strains in the LP group the frequency of multiple resistance was significantly higher, the MIC values were higher and R-plasmids were more frequent. Out of 30 multiple resistant E. coli strains 27 were R-plasmid carriers. Three different kinds of plasmid profile were shown in more than one strain (2 out of 10 UTI strains and 3 and 2 out of 10 LP strains). The rest of the isolates differed in plasmid profile from these and from one another; the presence of "epidemic plasmid" was not demonstrated. Plasmid epidemiological examinations may forecast the efficacy of an antibiotic or of a group of antibiotics.
