[Angiotensin-induced hypertension in pigs].

In pigs receiving up to 14 days daily injections of depot angiotensin, repeated daily bloody measurements of the blood pressure at the carotid artery proved unsuitable because of ensueing septic processes, in contrast to the unbloody measurement at the caudal artery. The depot angiotensin, despite the use of preparations with varying content of angiotensin-II and carrier solution, caused immediately after the injections a very strong maximal rise in systolic and diastolic blood pressure that occurred also with longer periods of the experiment; this rise gradually diminished in the course of the day but remained mostly in the hypertonic range and showed daily initial values above normal. The pig proved to be much more sensitive to angiotensin than the rat, for instance. Despite the proved vasoconstricting action of angiotensin, light-microscopic alterations at the vessels, kidneys, and the myocardium could not be observed. Part of the arterioles showed an increased wall thickness in animals killed on the 8th and 15th day of the experiment. The differences in the pathomorphological and blood pressure behaviour from the rat are discussed. The first experimental results qualify the pig as an experimental animal for comparative hypertension studies.

The substructure of heavy meromyosin. The effect of Ca2+ and Mg2+ on the tryptic fragmentation of heavy meromyosin.

Heavy meromyosin, obtained by tryptic digestion of myosin, containing two main polypeptides whose masses were estimated as 81,000 and 74,000 dlatons from Na dodecyl-SO4 polyacrylamide gel electrophoresis, was further digested with trypsin. The Ca2+-activated ATPase activity remainded unchanged and the K+-EDTA activity increased while various smaller fragments were formed. The formation of some of these fragments is affected by Ca2+ or Mg2+ as first shown by Bálint et al. (Bálint, M., Schaefer, A., Biro, N. A., Menczel, L., AND Fejes, E. (1971) J. Physiol. Chem. Phys. 3, 455). On the basis of the time course of the appearance of fragments the following relationship emerges: see article. The 64K leads to 60K step is inhibited by divalent cations, while the breakdown of the 74K fragment is accelerated. The effect of Ca2+ was maximal at 0 similar to 0.1 muM, that of Mg2+ at 10 muM. The original light chains of myosin are not present in the heavy meromyosin serving as the starting material, but peptide material appears on electrophoresis in positions starting material, but peptide material appears on electrophoresis in positions where the light chains would be found. The fragments marked by an asterisk are considered to ba alpha-helical on the basis of their solubility at low ionic strength after precipitation with ethanol (Bálint et al.). The fact that alpha helical fragments are derived from the 60,000-dalton fragment indicateds that it is adjacent to the light meromyosin in the intact myosin while the 74,000- dalton fragment would be part of heavy meromysoin subfragment 1. Chromatography of Sephadex G-200 separates fractions with ATPase activity corresponding to heavy meromyosin and heavy meromyosin subfragment 1. Electrophoresis of these Sephadex fractions suggests that the main peptide constituting heavy meromysoin subfragment 1 is connected by noncobalent forces to a portion of the rod that is not immediately adjacent to it in the primary sequence. The significance of this finding is discussed in terms of the flexibility of the myosin head.

Functional antagonism by a monoclonal antibody to digoxin in a test system of cultured rat heart myocytes.

A monoclonal antibody with a high affinity for digitoxin (KA = 0.50 nM) and digoxin (KA = 0.55 nM) was produced by somatic cell fusion. This antibody, designated 2A3(47), displayed little cross reactivity with other glycosides. In cultured rat heart myocytes, 2A3(47), antagonized the positive chronotropic effect exerted by digitoxin but did not alter that of ouabain. Our results suggest that this monoclonal antibody may prove to be useful in treating digoxin and digitoxin intoxication.

Borderless teleradiology with CHILI.

Teleradiology is one of the most evolved areas of telemedicine, but one of the basic problems which remains unsolved concerns system compatibility. The DICOM (Digital Imaging and Communications in Medicine) standard is a prerequisite, but it is not sufficient in all aspects. Examples of other currently open issues are security and cooperative work in synchronous teleconferences. Users without a DICOM radiological workstation would benefit from the ability to join a teleradiology network without any special tools. Drawbacks of many teleradiology systems are that they are monolithic in their software design and cannot be adapted to the actual user's environment. Existing radiological systems currently cannot be extended with additional software components. Consequently, every new application usually needs a new workstation with a different look and feel, which must be connected and integrated into the existing infrastructure. This paper introduces the second generation teleradiology system CHILI. The system has been designed to match both the teleradiology requirements of the American College of Radiology (ACR), and the functionality and usability needs of the users. The experiences of software developers and teleradiology users who participated in the first years of the clinical use of CHILI's predecessor MEDICUS have been integrated into a new design. The system has been designed as a component-based architecture. The most powerful communication protocol for data exchange and teleconferencing is the CHILI protocol, which includes a strong data security concept. The system offers, in addition to its own secure protocol, several different communication Methods: DICOM, classic e-mail, Remote Copy functions (RCP), File Transfer Protocol (FTP), the internet protocols HTTP (HyperText Transfer Protocol) and HTTPS (HyperText Transfer Protocol Secure),and CD-ROMs for off-line communication. These transfer METHODS allow the user to send images to nearly anyone with a computer and a network. The drawbacks of the non-CHILI protocols are that teleconferences are not possible, and that the user must take reasonable precautions for data privacy and security. The CHILI PlugIn mechanism enables the users or third parties to extend the system capabilities by adding powerful image postprocessing functions or interfaces to other information systems. Suitable PlugIns can be either existing programs, or dedicated applications programmed with interfaces to the CHILI components. The developer may freely choose programming languages and interface toolkits. The CHILI architecture is a powerful and flexible environment for Picture Archiving and Communications Systems (PACS)and teleradiology. More than 40 systems are currently running in clinical routine in Germany. More than 300,000 images have been distributed among the communication partners in the last two years. Feedback and suggestions from the users influenced the system architecture by a great extent. The proposed and implemented system has been optimized to be as platform independent, open, and secure as possible.
