A hairy cell leukaemia (HCL) patient is presented in whom the peripheral blood mononuclear cells (PBMCs) carried suppressor T-cell markers (CD3+, CD2+, CD8+/CD4-, CD38+). Analysis of genomic DNA of PBMNC showed the presence of a monoclonal population of T cells, the T-cell receptor (TCR) beta-chain gene being rearranged on both alleles (DR/DR), while the immunoglobulin (Ig) heavy chain-genes were in germline configuration. The neoplastic cells were found to react with the monoclonal antibody RAB-1 - originally described as belonging to the B lineage-restricted monoclonal antibodies - and to carry RAB-1/CD-8 in a double marker assay. Natural killer activity of PBMNCs against K562 target cells was severely reduced, while the cells were found to exert strong antibody-dependent cellular cytotoxicity.
The percentage of pitted erythrocytes and Howell-Jolly bodies in peripheral blood samples of 51 individuals following posttraumatic splenectomy and 20 patients splenectomized because of various haematological diseases differed significantly from each other (p less than 0.001) and from that of healthy controls (p less than 0.001). The percentage of pitted erythrocytes was significantly higher in pathologically shaped red blood cells (RBCs) (acanthocytes, schizocytes, elliptocytes) than in normal discoid shaped RBCs (p less than 0.001). As the number of pits per RBC showed great individual variations, a scoring system for the evaluation of pitted RBCs is proposed.

Increased numbers of circulating lymphocytes and large granular lymphocytes (LGL) were observed in 115 individuals splenectomized for haematological disease (74 cases) or for trauma (41 cases). LGL lymphocytosis was present in 78.4% of haematologically indicated and in 85.4% of traumatic splenectomies. A 70.5% of these values was above the 97.5 percentile upper tolerance limit of healthy controls (200 cases). In addition, 175 haematological controls were investigated. Forty per cent or more of circulating lymphocytes exhibited LGL morphology in nearly half of repeatedly investigated splenectomized persons. The increase in LGL is not attributable to lymphocytosis. It becomes apparent, and persists after the first postoperative week, irrespective of the cause of surgery. In spite of the two-fold increase in LGL concentration in the blood, in vitro natural killer (NK) and antibody dependent cellular cytotoxic (ADCC) activities did not increase in the investigated 48 (NK) and 31 (ADCC) splenectomized persons, as compared with the appropriate healthy or haematological controls.

Platelet sensitivity to adenosine diphosphate (ADP) and to prostacyclin (PGI2) was studied in normal and diabetic pregnant women. The threshold concentrations of ADP inducing the second phase of aggregation were used to determine the platelet sensitivity to PGI2. The sensitivity of platelets to ADP increased in both groups in the second trimester, thereafter it decreased both in normal and diabetic pregnancies. In contrast, sensitivity to PGI2 increased in the last trimester of pregnancy. No difference could be observed between diabetic and normal groups. The similarity of the results between the two groups could be explained by the normoglycaemic state of well-controlled diabetic pregnant women.
In two open randomized studies carried out in female volunteers the effects of a new low dose oral contraceptive (O.C.) combination desogestrel + ethinylestradiol (150 micrograms/30 micrograms; Marvelon) on blood coagulation were compared with those of either the combination levonorgestrel + ethinylestradiol (150 micrograms/30 micrograms) or a triphasic preparation containing levonorgestrel + ethinylestradiol. A total of 78 women were involved in the studies which covered a period of 4 cycles; a pretreatment cycle followed by 3 treatment cycles, each consisting of 21 days of drug administration, followed by a 7-day tablet-free period. A blood sample was taken before treatment, after 2 weeks, 2 cycles and 3 cycles of treatment. The following parameters were measured: prothrombin; activated partial thromboplastin time; factors VII, VIII, X; fibrinogen; antithrombin III; euglobulin lysis time; platelet count and aggregation ratio; triglycerides and cholesterol and a glucose tolerance test was made. The effects of the different preparations on these parameters were compared statistically using either an analysis of covariance or the Wilcoxon test. No clinically significant differences were observed between the desogestrel containing preparation and the two other preparations. Any changes in treatment versus pretreatment values were slight and in agreement with the literature on other O.C.'s.
In attempt to elucidate whether acetylsalicylic acid (ASA) has an in vivo effect on prostacyclin (PGI2)-like activity released from trophoblast we have evaluated PGI2-like activity in pregnant women scheduled for pregnancy termination after ASA ingestion. Following subjects were studied: Group I: 7 healthy pregnant women who were treated with 1.5 g ASA for two days; Group II: 18 control pregnant women who received placebo for two days. Trophoblast specimens were obtained by legal abortions; PGI2-like activity in trophoblast was measured by the method of Moncada. In normal pregnant women (8-10 weeks gestation) treated with ASA the mean PGI2-like activity of trophoblast significantly decreased compared to the controls. These data indicate that treatment with ASA of early pregnant women might have a harmful effect on trophoblast and the problem should be further explored before allowing the administration of cyclooxygenase inhibiting drugs during early pregnancy.

Two different high performance liquid chromatographic cleanup methods, absorption chromatography and nonaqueous reversed-phase chromatography, were investigated and their respective adventages discussed. The first was performed on 5 cm silica and alumina columsn, while for the latter a 5 cm octadecylilica column was used. Optimum mobile phase moderator concentrations were selected in order to achieve complete separation between the most retained organochlorine pesticide (endosulfan sulfate) and lipids on absorption columns. Separation efficiency was controlled by monitoring the eluate at 205 nm. The specific application fields of the described approaches were indicated and their possibility to preclude extracts from up to mg of lipid substances proved.

Stomach, small intestine contents, blood, liver, kidney and urine of a 28-years old man, were analyzed for residues of Endosulfan (6, 7, 8, 9, 10, 10-hexachloro-1, 5, 5a, 6, 9, 9a-hexahydro-6, 9-methano-2, 4, 3-benzo(e)dioxathiepin 3-oxide). The analysis results showed the presence of high concentrations of the two endosulfan isomers in all samples. Since also alcohol was present in all the tissues analyzed, it was concluded that the victim died of a combined endosulfan-alcohol poisoning. No other drugs were found.
