Changes of 3H-thymidine incorporation into DNA and thymidine kinase activity in rat thyroid lobes, following their exposure to neuropeptide Y.

The effects of neuropeptide Y (NPY) on 3H-thymidine incorporation into DNA of rat thyroid lobes in a 4 h incubation, and on the activity of thymidine kinase (TK) in the homogenates of thyroid lobes in vitro, as well as the influence of thyrotropin (TSH), applied alone or together with NPY, were investigated. The effects of NPY were dependent on the concentration used. Neuropeptide Y, in the lowest and the highest concentrations, decreased both the incorporation of 3H-thymidine to DNA and TK activity, and when used in intermediate concentrations NPY did not significantly affect the processes examined. Thyrotropin decreased TK activity and revealed a tendency towards lowering 3H-thymidine uptake, the latter effect being not statistically significant. NPY (10(-10) M) used together with TSH, produced an increase of 3H-thymidine incorporation into DNA, when compared with controls and the TSH-exposed group. Following the joint exposure to NPY and TSH, TK activity declined.

Reorientation time of DNA molecules in pulsed-field gel electrophoresis.

The precise determination of the influence of an electric field strength E on the resolution of DNA molecules during a pulsed-field gel electrophoresis shows that the maximal molecular size Nmax of still resolved DNA molecules is described by the equation Nmax = k tau E alpha, where k is a coefficient, tau is a pulse time, and alpha is an exponent (calculated as approximately 3/2). We assume that the best estimation of the reorientation time tau R for each DNA fragment is such a pulse time in which this DNA molecule is the largest separated one.

Adenosine deaminase: physical and chemical properties of partially purified mitochondrial and cytosol enzyme from rat liver.

Adenosine deaminase (ADA) was partially purified 486- and 994-fold from rat liver mitochondria and cytosol, respectively. Relative molecular mass of the enzymes from both fractions was 34,000. Km for adenosine and 2'-deoxy-adenosine were 3.08 x 10(-5) M and 3.03 x 10(-5) M for mitochondrial ADA and 3.12 x 10(-5) M and 2.87 x 10(-5) M for cytosolic ADA. The enzyme from both subcellular fractions had the maximum activity at pH 7.5-8.0, and pI 5.2 and 4.2 for mitochondrial and cytosolic enzyme, respectively. The enzyme was inhibited by erythro-9-(2-hydroxy-3-nonyl)adenine and 2'-deoxycoformycin with Ki 4.4 x 10(-7) M and 3.2 x 10(-7) M for mitochondrial ADA and 4.9 x 10(-7) M 2.8 x 10(-7) M for cytosolic ADA. Among the natural nucleoside and deoxynucleotide derivatives tested, deoxy-GTP and UTP inhibited only cytosolic adenosine deaminase by 60% and 40%, respectively.

Nucleotide sequence of satellite I and II DNA from alpaca (Lama pacos) genome.

Agarose and polyacrylamide gel electrophoresis of a total alpaca (Lama pacos) DNA, digested with several restriction enzymes, revealed the presence of two tandemly organized repetitive DNA sequences, named Satellite I and Satellite II. Three Xhol-monomers from Satellite I DNA and two BspRI-monomers from Satellite II DNA were sequenced. As determined by dot hybridization analysis, the total alpaca DNA consists of 3.4% and 1.3% Satellites I and II, respectively. Computer search revealed no homology to any primate, rodent or mammalian sequences published in the Gen Bank Release 48.0 (February 1987) and the EMBL Bank Release 10 (December 1986).

[Emergency psychiatry and crisis intervention in a primary clinic].

The terms Notfall and Krise, and the semantic problems arising, are elucidated; and an analysis of one years work in provision for emergency psychiatry and stress intervention is presented. Psychoreactive syndromes were found to be most common, followed in second place by psychotic conditions. In third place come disorders in connection with alcohol, which display a rising tendency. 40% of the clinic's beds are occupied by acute emergency cases, chiefly of short duration. Emergency provision is thus shown to be a major feature in the work of psychiatric clinics.

Radiation doses from 51Cr-bleomycin.

Extensive studies were made of long-term whole-body retention, plasma concentration, urinary excretion and organ retention of 51Cr in rats after intravenous injection of 51Cr-bleomycin. A similar but less extensive investigation was performed on rabbits, and whole-body retention, plasma concentration and urinary excretion was also followed in patients given 51Cr-bleomycin for diagnostic purposes. From an analysis of the collected information it followed that integrated whole-body and organ retention, corrected for radioactive decay, is similar in man and rabbit, and higher by a factor of about 2 in the rat. Doses to organs and whole body in man were calculated using the MIRD methodology and assuming conservatively that the kinetic data derived in rats are applicable to man. The organ and whole-body MIRD doses after 51Cr-bleomycin administration are comparable to those after 67Ga-citrate and the effective dose equivalent from a diagnostic amount of 740 MBq of the 51Cr complex amounts to about 6 mSv. However, the subcellular distribution in the liver of 51Cr, administered in the form of 51Cr-bleomycin, indicated a significant enrichment of 51Cr in the nuclei and in the DNA over mean concentrations in the liver cells. Also, the quality factor for Auger electrons, emitted by 51Cr, when assessed on the basis of the Q vs. LET relationship, as proposed by ICRP, was substantially higher than unity. Doses calculated for cell nuclei and the DNA in the liver cells were higher than the cell-averaged values by a factor 2.5 and 5, respectively, and the corresponding dose equivalents by a factor of 9 and 24. The effective dose equivalent, estimated on the basis of dose equivalents to cell nuclei and the DNA, amounted to 33 and 83 mSv per examination (740 MBq of 51Cr-bleomycin), respectively.

[Invalid status in neurologic diseases].

Authors analysed causes of invalidity in neuropsychiatric diseases within a single district. The share of neurological complaints amounts to 24%. Multiple sclerosis and epilepsy are the most important classes of syndromes. The results was compared with data known by pertinent literature. Demands appearing relevant for expert valuation of that kind of diseases are referred.

[Experiences with the combination of communication training and dynamic group psychotherapy].

Three psychotherapy groups consisting of altogether 37 patients took part in communication training after L. Schäbisch and M. Sieme in preparation for group-centred dynamic psychotherapy. The results of pretherapy and post-therapy surveys with the MMPI were tested for quantitative changes using Posthoff and H. F. Böttcher's gnoseologically based test. The findings suggest that this combination of methods has a high standard of efficiency, which is finally discussed.

[Development of anxiety arousal in group psychotherapy].

37 patients taken from three psychotherapy groups had completed a course of communication training as preparation for dynamic group psychotherapy. Two anxiety tests were performed at different times before, during and after the therapy. After completion of the communication training, the anxiety diminished as a result of general factors that had come into effect. After completion of the dynamic group psychotherapy they diminished owing to a genuine improvement in social competence. The different modes of action of two psychotherapeutic methods are clearly shown.

Purification and properties of deoxyadenosine kinase from rat liver mitochondria. I. Purification and physical properties.

Deoxyadenosine kinase (ATP: deoxyadenosine 5'-phosphotransferase, EC 2.7.1.76, AdR kinase) from rat liver mitochondria has been partially purified and compared with partially purified AdR kinase from the cytosol of the same biological material. Some physical properties of both enzymes, including molecular weight, gel electrophoresis and gel isoelectric focusing were investigated and considerable differences between these data for mitochondrial and cytosol AdR kinase were found.
