Purification and properties of deoxyadenosine kinase from rat liver mitochondria. II. Kinetics and inhibition studies.

Kinetic and inhibition studies of partially purified deoxyadenosine kinase (ATP: deoxyadenosine 5'-phosphotransferase, EC 2.7.1.76, AdR kinase) from rat liver mitochondria were performed, including reaction properties, specificity of phosphate donors, phosphate acceptors, influence of nucleotides, nucleosides and their derivatives. The results were compared with those obtained from partially purified AdR kinase from rat liver cytosol.

Regulation of deoxynucleoside kinase activities in rat liver mitochondria.

The activities of TdR kinase2 (ATP: thymidine 5'-phosphotransferase, EC 2.7.1.21), AdR kinase (ATP: deoxyadenosine 5'-phosphotransferase, EC 2.7.1.76), GdR kinase (ATP: deoxyguanosine 5'-phosphotransferase, without EC number), ATP (Mg2+)-ase (ATP phosphohydrolase, EC 3.6.1.3), nucleoside diphosphatase (nucleoside diphosphate phosphohydrolase, EC 3.6.1.6), nucleoside phosphotransferase (AMP: deoxynucleoside phosphotransferase, EC 2.7.1.77) and ribonucleotide 5'-diphosphate reductase (EC 1.17.4.1) were assayed in mitochondria of normal and regenerating rat liver. The activities of deoxynucleoside kinases are regulated: (a) by feedback inhibition of TdR kinase with dTTP and dCTP, and GdR kinase with dGTP; (b) GdR and AdR kinases by AdR and GdR inhibition, respectively; (c) by stimulation of GdR kinase with dTDP, dTTP and dATP. The stimulatory effects are correlated with changes of ATP (Mg2+)-ase and NDP-ase activities in regenerating liver mitochondria.

[Occurrence of depression with internal diseases].

Depressions belong to the most frequent diseases. They are only partly recognized. The largest number of depressive patients is to be found in the practice of the general practitioner and the internist. The relations between an internal disease and physical complaints, respectively, and a depressive syndrome are varied. It is reported on the results of an examination of 420 patients of a Clinic for Internal Diseases. 57 patients (13.6%) showed a depressive concomitant syndrome. Psychogenic depressions were more frequent than endogenic forms, the latter particularly as masked depression. These and other results are discussed and conclusions are made for the practice, in which cases the authors particularly enter the recognition of depressive conditions and the problems of psychic care as well as the therapy with psychopharmaca.

A distinctive activity of 5'-nucleotidase for dTMP in rat liver mitochondria.

The activities of 5'-nucleotidase (5'-ribonucleoside phosphohydrolase, EC 3.1.3.5); adenosine deaminase (adenosine aminohydrolase, EC 3.5.4.4); AMP deaminase (AMP aminohydrolase, EC 3.5.3.6), and ATP-(Mg2+)-ase (ATP phosphohydrolase, EC 3.6.1.3) were assayed in mitochondria of normal and regenerating rat liver 5'-Nucleotidase (5'Nase) and ATP-(Mg2+)-ase activities were compared with similar enzyme activities in the plasma membrane (PM) fraction, obtained from the same biological material. In the regenerating liver, 5'Nase for dTMP diminished its activity by 56% (24 h after partial hepatectomy) and 35 +/- 4% for all substrates in the PM fraction (48 h after operation). In mitochondria, 5'Nase for dTMP manifests sigmoidal substrate activity curve (in contrast with all substrates in the PM fraction and remaining substrates in mitochondria). In vivo 5-azacytidine (a) administered 1 h after partial hepatectomy, prevented changes of 5'Nase activity: (b) administered 24 or 48 h after partial hepatectomy, stabilized low 5'Nase activity (in mitochondria for dTMP, in the PM fraction for all substrates) and decreased ATP-(Mg2+)-ase activity by 51 and 31% in mitochondria and the PM fraction respectively.

5'-Nucleotidase↗

Relationship between 5'-nucleotidase, adenosine deaminase, AMP deaminase, ATP-(Mg2+)-ase activities and dTMP kinase activity in rat liver mitochondria.

The activities of dTMP kinase (ATP-deoxythymidine monophosphate phosphotransferase, EC 2.7.4.9), 5'-nucleotidase (5'-ribonucleoside phosphohydrolase, EC 3.1.3.5), adenosine deaminase (adenosine aminohydrolase, EC 3.5.4.4), AMP deaminase (AMP aminohydrolase, EC 3.5.3.6) and ATP-(Mg2+)-ase (ATP phosphohydrolase, EC 3.6.1.3) were assayed in mitochondria of normal and regenerating rat liver. In regenerating mitochondria, the dTMP kinase activity increased 20 times, 5'-nucleotidase (5'Nase) activity for dTMP diminished by 65% and its activity for other nucleoside monophosphates did not change; adenosine deaminase activity for adenosine (AR) increased by 40%, but for deoxyadenosine (AdR) decreased by 70%. AMP deaminase and ATP-(Mg2+)-ase activities behaved similarly in mitochondria from regenerating liver, decreasing by 70 and 64% respectively. The changes of the amount of dTMP in mitochondria depend on enzyme activities which regulate the AdR concentration.

[Lithium distribution in organs after lithium poisoning].

This paper describes the lithium distribution in the organs of two manic-depressive women who died following lithium intoxication. In both cases intoxication was observed during and after an acute phase of epidemic influenza accompanied by diarrhoea. The patients died from cerebral failure 5 and 7 days after receiving their last lithium dose. Death could not be prevented although NaCl infusions and haemodialyses were carried out. Only slight differences in lithium content were found when comparing extracerebral organs and serum. However, the brain accumulated considerable amounts of lithium ion. This reached toxic concentration in most of the brain areas of both patients. The highest values were found in the white matter, pointing to a lithium accumulation in lipid-enriched brain cells. The results were interpreted in connection with earlier findings of our group and other authors.

[Indications, adverse effects and complications of lithium therapy].

The application of lithium preparations in the therapy and prophylaxis of recidivations of affective psychoses has essentially gathered in point in psychiatry during the last years. Apart from the discussion of the indication, the contraindication and efficacy especially concomitant appearances, side effects and dangers as well as their influencibility are demonstrated, the knowledge of which is of importance also for the physician not dealing with psychiatry.

Bipolar Disorder↗

[Alcohol-induced lesions of the nerve and muscle].

A review is given on the most important neurologic sequelae after long lasting abuse of alcohol. The paper especially deals with the clinical features of neuropathic and myopathic disorders in alcoholics. Electromyography (including electroneurography) is considered to be a major diagnostic aid in these conditions.

[Psychiatric and neurological problems of the older age (author's transl)].

Pyschical crisis and pathological change do not differ at people in the older age. The two terms do not characterize the normal development of the old age, but extreme spheres are circumscribed. The cerebrovascular insufficience is a difficult diagnostic and therapeutic area, but it is accissible. The psychopathological symptoms to be caused by this disturbance are reversible in many cases. The problems of a comprehensive diagnostic and therapy of sleeping-disturbances at aged people are described.
