A new species of strictly anaerobic chytridiomycete was isolated from dried faeces of the Saharian ass that had been stored for up to 150 days. Because of its monocentric thallus and uniflagellate zoospores it belongs to the genus Piromyces. It exhibits a high affinity for P. mae and P. dumbonica but differs from them in its morphological and ultrastructural characteristics. Its flagellar apparatus is similar to that of all previously reported fungi.
A lectin (LDL) has been isolated from carpophores of the edible mushroom, Lactarius deliciosus, using a combination of affinity chromatography on stromas of group O erythrocytes embedded in polyacrylamide gel and hydroxylapatite, and gel filtration chromatography. Its molecular weight, as determined by gel filtration, is about 37,000 and its structure is dimeric, with two distinct types of subunits (about 19,000 and 18,000). It appeared homogeneous on HPLC gel filtration, but exhibited microheterogeneity on isoelectric focusing. Amino acid analysis revealed that it contains a large amount of glycine. Hapten inhibition assaying indicated that the Lactarius lectin is most specific for D-Gal beta 1----3D-GalNAc. The lectin was found in the mycelium and its possible role in the fungus is discussed.

The technique based on fluorescein-linked lectins used to determine the cell wall structure of anaerobic rumen fungi belonging to genera: Neocallimastix, Piromonas and Sphaeromonas, appears to be an interesting tool for distinguishing between strains. Furthermore this technique shows differences of cell wall composition between different parts of the thallus (spores, sporangia, rhizoïds).
Using fluorescein isothiocyanate-labeled lectins of various specificities, differences in the cell wall polysaccharide composition among the different parts of the thallus and during the cycle of S communis were demonstrated.
A new species of strictly anaerobic fungus was isolated from the cow rumen. It is characterized by a polycentric thallus, a polynuclear rhizomycelium, mucronate zoosporangia and uniflagellated zoospores. It is also singular in that the sporocysts do not react to the specific lectins of L-fucose, N-acetyl-D-galactosamine and diacetyl chitobiose. These characteristics justify the creation of a new genus.
The expression of end-chain sugar residues and of oligosaccharidic sequences has been investigated in chick germ cells at critical stages during the migration, proliferation and sexual differentiation of these cells. Fluorescent lectins and indirect immunofluorescence studies using the NC-1/HNK-1 monoclonal antibody indicate a remarkable control of glycosylation during germ cell embryonal life. Besides a retained expression of glucose/mannose residues, it was found that alpha- and beta-galactose residues, N-acetyllactosamine and N-N' diacetylchitobiose sequences as well as the sulfated trisaccharidic NC-1 epitope were detectable in a stage-specific pattern. Present at a very high density in the cytoplasm and on the surface of the early germ cells at premigrative and migratory stages, the staining for these carbohydrate sequences gradually disappeared when the germ cells settled and proliferated in the developing gonadal primordia. The disaccharide Gal beta 1----3 Gal NAc was exclusively detected in migrating PGCs. In sexualized gonads, acetyllactosamine and/or diacetylchitobiose were similarly reexpressed in both oogonia and spermatogonia. Spermatogonia displayed beta-galactose residues and a high immunoreactivity with the NC1 Mab, indicating modulations in PGC glycosylations related to the acquisition of sexual phenotypes. In addition NC-1 was found to be expressed in the somatic component of the undifferentiated gonad and in the testis interstitial gland.

The surface polysaccharides of two strains of lizard leishmanias, of the guinea pig parasite L. enrietti and of nine strains of human leishmanias belonging to the groups mexicana, donovani and tropica were studied by lectin-mediated agglutination. Twenty-three lectins prepared from seeds or from higher fungi carpophores were used. They revealed the presence of L-fucose, N-N'-diacetylchitobiose, alpha-D-glucose, alpha-D-mannose, beta-D-galactose, N-acetyl-D-galactosamine and lactose. We noted a range in the number and variety of lectin receptor sites detectable on the surface of the leishmanias, with the reptiles strains having the fewest sites and the tropica group having the most sites. We were unable to find specific group lectins, but it seems possible to identify a strain within each group by a lectin-binding pattern.
Lectins in the seeds of Ricinus communis L. were separated by affinity chromatography using stromata, and 3 fractions obtained by ion-exchange chromatography. Polyacrylamide-gel electrophoresis showed that 2 fractions were homogeneous and that 1 fraction gave 2 bands. Immunoelectrophoresis with rabbit antisera and polyacrylamide-gel isoelectric focusing showed that the 3 fractions were separate and distinct entities consisting of many proteins with isoelectric points near the isoelectric point of the main protein. The molecular weights of the separated proteins and their subunits were studied by SDS polyacrylamide-gel electrophoresis.
