Absence of correlation between the degree of alveolitis and antibody levels to Micropolysporum faeni.

Specific IgG antibody (Ab) levels to Micropolysporum faeni (MF) antigen were measured in three study groups: 29 farmers of whom one had acute, two chronic and 26 with a past history of Farmer's lung disease (FLD), 91 clinically asymptomatic but exposed farmers and 23 unexposed donors. This study confirms the greater sensitivity of enzyme linked immunosorbent assay (ELISA) over double immunodiffusion (DID): 34 (37%) sera negative by DID were positive by ELISA and only two (4%) were positive by DID and negative by ELISA. Antibody levels to MF were higher in the serum of farmers whether symptomatic (113.2 +/- 196.1 u) or not (25.2 +/- 64.7 u) than in that of unexposed donors (1.05 +/- 1.46 u), (P less than 0.001). In a subgroup of farmers, antibody levels were compared in the serum and bronchoalveolar lavage (BAL) fluid; the levels were higher in the serum than in BAL fluids in all farmers except the only patient with acute FLD. A significant correlation was seen between Ab levels of the serum and BAL fluids. Finally, no correlation was seen between the serum antibody levels and the total number of cells or the percentage of lymphocytes found in BAL fluids, either in the subjects with a history of FLD or the asymptomatic group.

[Stripped veins for secondary vascular access in hemodialysis. Pathologic study of the grafts after surgical excision].

The suitability of stripping vein homografts as AV fistulae for intermittent hemodialysis has been evaluated. Microscopic investigation of stored veins indicated the superiority of cryopreservation over refrigeration. A systematic study of 12 venous homograft fistulae surgically excised from hemodialysis patients revealed that the homograft walls are the sites of both connective tissue proliferation and of mural degeneration. It was also noted that the implants failed in the short term (less than 1 yr) mainly due to stenosis, in the medium term (1 yr-3 yrs) mainly due to aneurysmal dilatation and for a variety of reasons beyond 3 years. It was concluded that when suitably selected, handled and stored, stripping vein homografts are a viable alternative to treated human umbilical or bovine heterografts as blood access fistulae if no autograft is available.

Eosinophil-rich human polymorphonuclear leukocyte preparations characteristically release leukotriene C4 on ionophore A23187 challenge.

Blood samples were obtained from a group of 20 patients with hypereosinophilia (greater than or equal to 1500 eosinophils/mm3). The polymorphonuclear leukocytes (PMNLs) were prepared from blood treated with ethylenediaminetetra-acetic acid by successive dextran sedimentation of the red blood cells, separation of mononuclear leukocytes and PMNLs on Ficoll-Paque, and ammonium chloride treatment of the PMNL fraction. The eosinophil content of the final PMNL preparations ranged from 15% to 75%, as assessed by Wright-stained smears, and the remaining leukocytes were predominantly neutrophils with only 3% to 5% mononuclear cells. The eosinophil-rich PMNL preparations as well as PMNL preparations from normal volunteers were incubated under various conditions and the arachidonic acid metabolites were analyzed by reverse-phase high-performance liquid chromatography. The synthesis of 5-lipoxygenase products was strongly stimulated by the ionophore A23187 in both normal and eosinophil-rich PMNL preparations. Whereas the normal PMNL preparations, which were eosinophil poor, produced 10 to 25 times more leukotriene B4 than leukotriene C4, the eosinophil-rich PMNL preparations characteristically released leukotriene C4 in equal or up to 20 times greater amounts than leukotriene B4.

[Retrospective study of mental patients who have committed criminal acts].

The study presented here aims to gather descriptive information which would be useful to the development of research hypotheses, policies, and treatment programmes for persons suffering from mental illness who commit crimes. The project tracked a cohort of 225 persons for the seven to nine years after they had been judged unfit to stand trail or not guilty by reason of insanity. A comparison is made here between the cases treated in a maximum security hospital and those treated in other hospitals. The population studied is characterised principally by its misery. The majority are individuals who have been diagnosed as psychotic, who have few, if any, personal or social resources and who live on the edge of society. Thirty-six percent of these cases were treated in a security hospital. Of all the variables studied, only a few distinguished these cases: the place and type of crime of which they were initially accused, the length of out-patient treatment, the type of residence once released from hospital, and the occupation. Questions are raised with respect to appropriate treatments for such persons.

The role of fibronectin in the cryoprecipitation of monoclonal cryoglobulins.

We have recently reported that fibronectin is a component of mixed cryoglobulins and is responsible in large part for cryoprecipitate formation. We have now extended our studies to serum cryoprecipitates formed in the presence of monoclonal cryoglobulins in patients with lymphoproliferative diseases in order to investigate for a possibly similar influence of fibronectin on cryoprecipitation. Results have shown that these cryoprecipitates contain fibronectin in proportions varying between 10% and 14% of their total protein content. Furthermore, SDS-polyacrylamide gel electrophoresis revealed that next to the cryoglobulins, fibronectin constitutes the second major component of the cryoprecipitates. Depletion of the sera in fibronectin prior to exposure to 4 degrees C, markedly reduced cryoprecipitation of the cryoglobulins, whereas reconstitution in fibronectin restored it. These studies support and extend our previous findings showing the important influence exerted by fibronectin on the process of immunoglobulin cryoprecipitation.

Immune reactions in the lungs of asymptomatic dairy farmers.

To test the hypothesis that asymptomatic farmers with serum precipitins to farmers' lung disease antigens also have an immune reaction involving the lungs, we did bronchoalveolar lavage (BAL) in 3 groups of dairy farmers. Group 1: 7 patients with acute farmers' lung disease. Group 2: 10 asymptomatic farmers with serum precipitins to Micropolyspora faeni. Group 3: 9 normal farmers without serum precipitins. Group 1 patients had a large number of cells (90.4 +/- 20 X 10(6)) (mean +/- SEM) with 72 +/- 5.8% lymphocytes in their lavage fluid. Their lavage also had high immunoglobulins A and G/albumin ratios. Six subjects in Group 2 had an increased number of cells (54.1 +/- 14.1 X 10(6)) and a high percentage of lymphocytes (52.5 +/- 6.6%) in their BAL. Two subjects in Group 3 had similar alterations in their lavage fluid: 60.0 and 69.6 X 10(6) cells with 20 and 37.5% lymphocytes, respectively. The other subjects in Groups 2 and 3 had normal lavages. Proliferative responses of BAL lymphocytes to phytohemagglutinin was similar in each group. Circulating immune complexes were increased only in subject with acute farmers' lung disease (Group 1). These results show that some normal farmers have signs of an alveolitis.

Detection of immunoglobulin-secreting lymphocytes by the use of a hemolytic plaque assay in liquid phase.

This paper describes a modification of the reverse hemolytic plaque assay allowing the enumeration of immunoglobulin (Ig)-secreting lymphocytes at rest or after in vitro stimulation. Ig-secreting cells are mixed with anti-Ig coated sheep red blood cells, developing anti-Ig antiserum and complement in liquid medium. This mixture is pipetted between two slides and the hemolytic plaques are numerated after incubation at 37 degrees C. The class of Ig secreted can be determined using a monospecific developing antiserum. This technique is easier to perform and more economical than the previously described method using agarose without loss of sensitivity, the number of plaques detected being about the same and the kinetic of their appearance comparable.

Phosphorylation in vitro and in vivo of ribosomal proteins from Saccharomyces cerevisia.

Crude ribosomes from Saccharomyces cerevisiae cultures were phosphorylated in vitro when incubated in the presence of [gamma-32P]ATP. Analysis of the ribosomal proteins with two-dimensional electrophoresis revealed that of the 29 proteins identified in the small subunit, only protein S6 was phosphorylated. Of the 37 proteins identified in the large subunit, one was highly phosphorylated (L3) and two only slightly phosphorylated (L11 and L14). The protein kinase activity associated with the ribosomes was extracted with 1 M KCl and was not dependent on adenosine 3':5'-monophosphate; it preferentially phosphorylated casein and phosvitin, but was less active on histones. Structural ribosomal proteins were also phosphorylated in vivo when the yeast cultures were incubated with [32P]orthophosphate; the radioactivity resistant to hydrolysis by hot perchloric acid was incorporated into the proteins of the two subunits. Radioactive phosphoserine was found by subjecting hydrolysates of ribosomal proteins to high-voltage electrophoresis. After two-dimensional electrophoresis, one poorly phosphorylated protein (S10) was identified in the small subunit. In the large subunit, one protein (L3) was highly labelled, and two proteins (L11 and L24) only slightly labelled.

Decreased drug tolerance following endocrinological manipulation in the rat.

Nine groups of rats of rats were either hypophysectomized, adrenalectomized or left unoperated. These groups were then subjected to daily increasing injections of either morphine HCL, sodium pentobarbital, chloridiazepoxide or saline. Hypophysectomy seemed to lower the lethal dose of the three drugs tested. Adrenalectomy seemed to have only a minimal effect on the lethal dose of morphine HCL or sodium pentobarbital.
