[Symptoms and functional disorders of the respiratory system caused by exposure to tea dust].

A case report is presented of a female worker of a tee packing factory, with a 5 years history of tea dust exposition and symptoms of reversible airway obstruction and early allergic alveolitis. The results of spirometric and lung mechanics demonstrated reversible airway obstruction with co-existent restrictive changes, lowering of CO diffusing capacity (DLc0). Bronchial provocation tests with tea dust were carried out thrice and all were positive. The authors believe that the etiological factor of above mentioned changes were Aspergillus niger spores found in tea dust. Tea dust and the patient's sputum cultured for fungi revealed massive growth of A. niger. Serological tests revealed additionally, antibodies to A. niger and tea dust extracts. The authors discuss the possible mechanisms of airway disturbances in this patient.

[Ectopic splenic tissue in connection with the testis].

A case of ectopic splenic tissue in the testicle is reported. This congenital anomaly has only ever been seen in the left scrotal compartment. The ectopic splenic tissue is separated from the testicle by a fibrotic capsule. When accessory splenic tissue is suspected on examination of a testicular biopsy a frozen section should be performed; orchiectomy can then often be avoided.

Activating-transcription-factor (ATF) regulates human 7S L RNA transcription by RNA polymerase III in vivo and in vitro.

The gene-external part of the human 7S L promoter was analyzed by transcription in vitro and in vivo. Compared to the wild type promoter (-178), a -66 5'deletion mutant revealed full activity in vitro but was inefficiently transcribed in vivo. Further deletion to -37 reduced template activity to 50% in vitro and to basal level expression in vivo (below 5%). A DNase I footprint observed around position -50 protected an ATF-like binding site ('TGACGT'). With respect to 7S L transcription regulation, the functionality of this ATF-like binding site was confirmed in competition experiments and by mutation analysis. Furthermore, S100 extracts of cells pretreated with forskolin in vivo to induce the cAMP system, revealed significantly increased transcription of 7S L RNA in vitro, with no effect on a 7S K RNA gene, lacking such an ATF binding site. Thus, the 7S L RNA gene too is controlled by a regulatory element originally defined in class II promoters and represents another rare example where a specific type of transcription regulation in vivo can be mimicked with cell-free extracts in vitro.

Metabolism of cysteinyl leukotrienes in monkey and man.

The proinflammatory cysteinyl leukotrienes are inactivated in primates by (a) intravascular degradation, (b) hepatic and renal uptake from the blood circulation, (c) intracellular metabolism of leukotriene E4 (LTE4), and (d) biliary and renal excretion of LTC4 degradation products. We have analyzed cysteinyl leukotriene metabolites excreted into bile and urine of the monkey Macaca fascicularis and of man. In both species, hepatobiliary leukotriene elimination predominated over renal excretion. In a representative healthy human subject at least 25% of the administered radioactivity were recovered from bile and 20% from urine within 24 h. In monkey and man intravenous administration of 14,15-3H2-labeled LTC4 resulted in the biliary and urinary excretion of labeled LTE4, omega-hydroxy-LTE4, omega-carboxy-LTE4, omega-carboxy-dinor-LTE4, and omega-carboxy-tetranor-dihydro-LTE4. Small amounts of N-acetyl-LTE4 were detected in human urine only. Oxidative metabolism of LTE4 proceeded more rapidly in the monkey resulting in the formation of higher relative amounts of omega-oxidized leukotrienes in this species as compared to man. [3H]H2O amounted to less than 2% of the administered dose in monkey and human bile and urine samples. Incubation of isolated human hepatocytes with [3H2]LTC4, [3H2]LTD4, and [3H2]LTE4 showed that only [3H2]LTE4 underwent intracellular oxidative metabolism resulting in the formation of omega- and beta-oxidation products. N-Acetylated LTE4 derivatives were not detected as products formed by human hepatocytes. By a combination of reversed-phase high-performance liquid chromatography and radioimmunoassay, endogenous LTE4 and N-acetyl-LTE4 were detected in human urine in concentrations of 220 +/- 40 and 24 +/- 3 pM, corresponding to 12 +/- 1 and 1.5 +/- 0.2 nmol/mol creatinine, respectively (mean +/- SEM; n = 10). Endogenous LTD4 and LTE4 were detected in human bile (n = 3) in concentrations between 0.2-0.9 nM. Our results demonstrate that LTD4 and LTE4 are major LTC4 metabolites in human bile and/or urine and may serve as index metabolites for the measurement of endogenously generated cysteinyl leukotrienes. Moreover, omega-oxidation and subsequent beta-oxidation from the omega-end contribute to the metabolic degradation of LTE4 not only in monkey but also in man.

Potassium raises cytochrome P-45011 beta mRNA level in zona glomerulosa of rat adrenals.

During potassium repletion of potassium-deficient rats, a marked rise in the adrenal zona glomerulosa content of cytochrome P-45011 beta mRNA preceded an extensive increase in corticosterone methyl oxidation (CMO) 1 and 2. Potassium intake did not substantially affect zona glomerulosa cytochrome P-450SCC or zona fasciculata-reticularis cytochrome P-45011 beta mRNA levels. We had previously shown that the induction of CMO 1 and 2 in the zona glomerulosa of potassium-repleted rats was associated with the appearance of a second form of cytochrome P-45011 beta with a lower molecular weight (49,000) than the major form of the enzyme (51,000). On the basis of the present observations, mRNAs encoding the two forms of the enzyme would be homologous and of similar length. Potassium intake thus controls the biosynthesis of the 49 kDa enzyme most likely at the level of transcription or, alternatively, by influencing mRNA stability.

Monoclonal antibody 43-9F as a sensitive immunohistochemical marker of carcinoma in situ of human testis.

Invasive germ cell cancer can be prevented if the neoplasia is diagnosed at the stage of carcinoma in situ (CIS). In routine histologic examination CIS may be overlooked, particularly in prepubertal individuals. The detection of this early malignancy may be greatly facilitated by use of immunohistochemical staining. The authors investigated the sensitivity of an immunohistochemical staining procedure with monoclonal antibody 43-9F in detection of CIS. Testicular specimens from 19 adult and two prepubertal individuals with CIS were tested. Positive staining reaction on the surface of malignant germ cells was encountered in all 21 specimens with CIS. The epithelial cells of the excretory ducts between testis and epididymis, including rete testis and canaliculi efferentes, reacted also positively with 43-9F. No staining was observed in nonmalignant testicular cells including Sertoli cells or Leydig cells in any of these samples or in testicular biopsy specimens from 27 adult and 11 prepubertal subjects without evidence of testicular neoplasia. Monoclonal antibody 43-9F was also found to react with cells of all four tested invasive testicular germ cell tumors. Thus, the monoclonal antibody 43-9F is a sensitive immunohistochemical marker of CIS germ cells and may also be of potential value in detection of invasive testicular cancer.

An experimental study on the influence of the derived casts on the accuracy of different recording materials. Part I: Plaster, impression compound, and wax.

An experimental study was conducted to analyze the accuracy of transferring jaw relations with recording materials and the derived casts. A specific measuring system was designed to determine three-dimensional deviations of the condyles of an articulator. Four interocclusal recording materials were analyzed after various storage periods. The results indicated that the accuracy of the derived cast had a critical influence on the accuracy of the transferred jaw relations. None of the materials were reliable; the most accurate material generated deviations of approximately 300 microns.

An experimental study on the influence of the derived casts on the accuracy of different recording materials. Part II: Polyether, acrylic resin, and corrected wax wafer.

A specific testing system was used to determine the three-dimensional changes of interocclusal registration materials for a simulated patient and the derived stone casts. The results indicated that the deviations of the derived casts were more pronounced than the original models. There was great heterogeneity with all recording materials and the materials were not consistently accurate after limited storage of 30 minutes.

Study of the accuracy of different recording materials.

An experimental study was conducted to determine the three-dimensional errors in mounting casts affected by the interocclusal recording materials. Eight commonly used materials or combinations were tested after various storage periods. The results indicated that all the materials induced asymmetric deviations of the condyles after each storage period. Impression plaster was the most accurate and dimensionally stable material; polyether was the second most accurate material, but it must be used within 6 hours. Correction of the teeth imprints with a low-viscosity material was beneficial only in combination with wax wafers.

Reaction of cultured pulp cells to eight different cements based on glass ionomers.

A comparative study was carried out to determine the cytotoxicity of eight different glass-ionomer cements by means of cell culture. Only fibroblast-like cells from the pulp of the same subcultures were used. Factors for the evaluation of the biological tolerance were, first, adhesion of cells to the slides as well as to the material, second, relative growth rates of cultures, and, in addition, cytomorphology. The cultured cells reacted quite differently to the glass-ionomer cements tested. The results imply that cytotoxic substances are eluted from some products even after a hardening period of 48 hours. In contrast, three of the glass-ionomer cements tested had only slight cytotoxic effects on the cultured pulp cells.

Experimental study on the biocompatibility of lining cements based on glass ionomer as compared with calcium hydroxide.

An in vitro study with cultured fibroblast-like cells of the pulp was used to assess the biological tolerance of three lining cements based on glass ionomers and two hard liners containing calcium hydroxide. The results indicate that strong cytotoxic components of all g.-i. lining cements tested can be eluted even after hardening for 48 hours. In contrast, the two liners based on calcium hydroxide proved to be of greater biocompatibility after the same hardening period. Therefore, use of calcium hydroxide liners is recommended for placement in deep cavities near the pulp.

Preclinical screening and analysis of the immunotoxic and immunomodulatory activity using a multiple immunoassay (MIA) in mice.

Screening and analysis of immunotoxic and immunomodulatory activity has become an integral component in preclinical studies of pharmaceuticals and xenobiotics. In an attempt to replace laborious and expensive batteries of assays used at present we developed a multiple immunoassay (MIA) enabling the determination, in a single mouse, of: the weight of the thymus, spleen and a group of lymph nodes; delayed type hypersensitivity and antibody response to SRBC; phagocytic activity of peritoneal macrophages and the responsiveness of spleen lymphocytes to "T" (PHA, ConA) and "B" (LPS) mitogens in vitro. The MIA responsiveness to two prototype immunostimulators (Thymostimulin and Listeria factor Ei) was tested at two time periods after antigenic stimulation, not only in normal mice, but also in animals with selectively depressed T-systems (anti-Thy1.2 monoclonal antibody) and B-systems (cyclophosphamide); in both dexamethasone-treated and irradiated animals. The findings indicate, that this MIA is capable of reflecting both immunosuppressive and immunostimulatory activities of the tested agents and permits partial insight into the mechanisms underlying these activities.

Duplication of the streptokinase gene in the chromosome of Streptococcus equisimilis H46A.

The erythromycin resistance plasmid pSM752 carrying the cloned streptokinase gene, skc, was introduced by protoplast transformation into Streptococcus equisimilis H46A from which skc was originally cloned. Cells transiently supporting the replication of pSM752 gave rise to an erythromycin-resistant clone designated H46SM which was plasmid free and produced streptokinase at levels approximately twice as high as the wild type. Southern hybridization of total cell DNA with an skc-containing probe provided evidence for the duplication of the skc gene in the H46SM chromosome. The results, which have some bearing on industrial streptokinase production, can be best explained by a single cross-over event between the chromosome and the plasmid in the region of shared homology leading to the integration of pSM752 in a Campbell-like manner.

Genomic analysis of T-cell receptor and immunoglobulin antigen receptor genes and breakpoint cluster regions in gastrointestinal lymphomas.

Sixteen cases of gastrointestinal B- and T-cell lymphoma were investigated for clonal rearrangements of immunoglobulin and T-cell receptor genes. In all cases the immunogenotype data corresponded well with the immunohistochemical findings, thus confirming the biologic heterogeneity of these lymphomas. Some cases could be distinguished by additional heterogeneity in their genotype which could not be detected by immunohistochemical methods. Moreover, the B-cell lymphomas were studied for the presence of the chromosomal translocations t(11;14) and t(14;18) using probes for bcl-1 and bcl-2 genes, respectively. While bcl-1 rearrangement was not present in these gastrointestinal lymphomas, one case of diffuse low-grade lymphoma of mucosa-associated lymphoid tissue and three cases of centroblastic lymphoma (diffuse large cell) could be shown to have detectable bcl-2 rearrangement.
