Factors affecting the activity of immobilized enzymes, I. Diffusional limitation.

The measured (apparent) specific activities of immobilized lactate dehydrogenase, malate dehydrogenase, glucose dehydrogenase, alkaline phosphatase and chymotrypsin decrease with increasing activity loading and increase with decreasing particle diameter and with increasing substrate concentration. The observed inactivation is therefore concluded to be due to diffusional limitation. Real specific activities are not greatly affected by the immobilization, as has been demonstrated after enzymic digestion of the matrix. In the case of lactate dehydrogenase and of malate dehydrogenase real specific activities are not altered by a variation of the number of bonds to the carrier.

Biosynthetic ad morphological evidence for inhibition of aldosterone production following administration of ACTH to sheep.

The effect of ACTH administration for 1-5 days on the morphology and steroidogenic capability of sheep adrenal tissue has been examined. During this period of treatment there was a gradual decline in the in vitro conversion of 3H-labelled precursors to products of solely zona glomerulosa origin (aldosterone and 18-hydroxycorticosterone) while conversion to products of zona fasciculata origin (17-hydroxyprogesterone, 11-deoxycortisol and cortisol) was stimulated throughout. Conversion to DOC, 18-hydroxydeoxycorticosterone and corticosterone (steroids produced by both the zona glomerulosa and the zona fasciculata) declined after initial stimulation. Within 2--3 days of the commencement of treatment, the zona glomerulosa showed a progressive decrease in cell number associated with disruption of cords and cell separation. Ultrastructurally, it was found that typical zona glomerulosa cells had almost disappeared. The majority of residual cells in this area had a structure intermediate between zona glomerulosa and zona fasciculata cells. The similarity in time-course of the alterations in both the morphological and biosynthetic characteristics suggests that the decline in aldosterone output caused by ACTH administration to sheep results from the loss of adrenal zona glomerulosa cells, predominantly due to selective cellular degeneration.

Equine leukocyte antigen system. II. Serological and mixed lymphocyte reactivity studies in families.

Mono- and oligospecific lymphocytotoxic alloantibodies from primiparous mares were tested on cells from horse families of various breeds in the two-step microcytotoxicity assay. The results showed that the detected antigens were inherited co-dominantly and autosomally as simple Mendelian traits. The membrane antigens showed different linkage with one or more other antigens and seem to be coded by a limited number of loci (at least three) from one chromosome. In the families tested one recombinant for the serologically defined antigens was recognized. The mixed leukocyte reactions of cells from horse families compared with the serologically recognized antigens showed that the two systems are inherited with the same chromosome. A homozygote for both antigen systems was recognized in a family.

[Parenteral feeding in outpatients. Indications, technic, experiences].

Home parenteral nutrition is indicated in the short bowel syndrome, chronic inflammatory bowel disease and as an adjunct to cancer therapy. For venous access the cannulation of a collateral of the subclavian vein and the central venous silastic catheter have proved helpful. Our own experience with 19 patients shows, that with careful supervision home parenteral nutrition can be performed with low risk.

[Peripheral venous tolerance of hyperosmolar infusion solutions].

Basic nutrition solutions become ever more important in postoperative infusion therapy. We tested the peripheral venous tolerance of an infusion solution which included aminoacids and carbohydrates (1100 mosm/l, 2400 kJ/l). We compared the peripheral venous tolerance of this nutrition solution with those commonly used in hospitals (Ringer-solution 330 mosm/1, 5% glucose solution 280 mosm/l 800 kJ/l, electrolyte glucose solution 407 mosm/l 800 kJ/l). The infusion thrombophlebitis was examined on 30 patients with the radiojodfibrinogen test. Despite three times higher osmolarity the peripheral nutrition solution shows no difference in peripheral venous tolerance after 48 hours.

[Serum anticonvulsant levels in therapy-resistant epileptics].

The continuous occurrence of particularly severe epileptic attacks represent a great stress for the patients. For this reason, we have conducted anticonvulsant blood-level studies particularly in these patients, this paper being based on 146 first determinations. Among the patients, most of whom are adults and are treated with combinations of drugs, only 24 per cent reached the assumed normal phenytoin range while 69 per cent remained below it. With the administration of phenobarbital, however, 50 per cent reached the therapeutic range and only 14 per cent reached the lower limiting value. It is only i the last-mentioned group that we assumed primarily an insufficient intake of the drugs while with DPH the problems of the determination of the plasma water range had to be discussed. In practice, blood level determinations have their special value for the detection of the intake of a wrong dosage of the drugs as well as bland anticonvulsant intoxications.
