[Isobutylidene-di-urea as a new NPN source for ruminants. 2. Metabolism of 14C-15N-isobutylidene-di-urea in sheep].

2 male sheep (weighing 45 kg and 44 kg) were fitted with a ruminal fistula and a jugular vein catheter and received isobutylidendi-urea for a 42-day period of adjustment. The diet contained 25% starch, 23.8% glucose, 29.0% cellulose, 10.0% straw, 1.7% sunflower seed oil, 4.3% isobutylidendi-urea, 5.6% minerals and vitamins. Each animal received 60 g of isobutylidendi-urea in daily amounts of 1.4 kg of the ration-4.4% of the total dietary N came from the straw. At the begin of the trial each sheep received 30 g of 14C15N isobutylidendi-urea (C1-siobutyl labelling) administered as a suspension. The animals were then placedin respiration cages. The peak of specific 14C activity in the expired air (including ruminal gas) was observed 2 hrs after the beginning of the trial. 18--30 hrs after the beginning of the trial the highest level of 15N incorporation into the TCE (trichloroacetic acid) soluble fraction of the ruminal fluid was noted resulting from the reflow of urea via the rumeno-hepatic circulatory system in the rumen. A high concentration of 15N was shown to be present, for prolonged period, in the TCE soluble fraction of the ruminal fluid (up to the 30 hr of experiment). The 15N concentration in the blood plasma (TCE soluble portion) was found to increase reaching a peak value 23 hrs after administration of the isotope. The highest level of 14C activity in this fraction appeared 1 hr after isotope administration. The 15N incorporation into the protein fraction of blood plasma reached a constant high level between the 29th and 47th hr of experiment. The highest 15N concentrations in urine were noted after 1 day. 3.5% of the administered dose of 14C activity and 23% of the supplied amount of N were excreted in the urine. 20% of the total amount of 15N excreted in the urine could be detected as 14C isobutyl residues. An excess of between 0.05 and 0.17 atom% of the isotopes were found in muscular tissue and in different organs of the sheep when these were slaughtered on the 7th day of experiment (liver: 0.17%, kidneys: 0.14%, muscle: 0.05%, heart: 0.08%). The results obtained in the present trial clearly indicate that ruminants are able to utilize nitrogen from isobutyldi-urea.

Ultracytochemical demonstration of monoamine oxidase activity in nervous and non-nervous tissue of the rat.

A method is described for the ultrastructural localization of the enzyme monoamine oxidase based on the formation of an osmiophilic formazan from the tetrazolium salt (2-(2'-benzothiazolyl)-5-styrl-3-(4'-phthalhydrazidyl) tetrazolium chloride) (BSPT) and using tryptamine as substrate. It was tested in rat heart, liver, adrenal cortex and medulla, kidney, pineal gland, cerebral cortex and cerebellar cortex and was found to provide good ultrastructural preservation, high contrast of the specific, enzyme-dependent precipitate and improved sensitivity. Monoamine oxidase activity was found in the mitochondrial outer compartment, nuclear envelope and endoplasmic reticulum in all organs studied; it was also seen occasionally in plasma membranes of liver and heart. Similar localizations were observed in interstitial cells and capillary endothelial cells. The enzyme-dependent nature of the observed precipitate was verified by control experiments consisting of incubation of overfixed tissue, omission of substrate from the incubation medium, and incubation in the presence of a specific, nonreversible inhibitor, pargyline. These studies confirm and extend the results of cell fractionation studies using liver, demonstrate the widespread distribution of monoamine oxidase in tissues, and suggest that monoamine oxidase plays important functional roles in extraneuronal cells.

[Electron microscopic presentation of immune reactions on Candida cells: asteroid bodies in Candida albicans from the urine of nephritis patients].

Candida albicans cells from the urine of two nephritis patients were concentrated and incubated with ferritin-labeled antihuman grammaglobulin (either anti-IgA, anti-IgG, or anti-IgM). Electron microscopy showed the electron-transparent yeast cell wall to be surrounded by an electron-dense capsule-like substance of remarkable volume. This must be regarded as an antigen-antibody precipitate corresponding to the "asteroid body" of previous authors. The antibodies involved in the formation of the precipitate are mainly those of the IgA and IgG classes. Considering the results of previous authors, the following definition is proposed: "Asteroid Bodies" are light microscopically visible antigen-antibody precipitates on the cell wall of fungi parasitic condition.

[Epilepsy by looking at patterns].

The author, after pointing out the rare occurence and peculiarities of pattern-sensitive forms of epilepsy, reports the case of a seven-year-old boy with infantile cerebral paresis characterized by symptomatic attacks, who was observed to show absence-like conditions when looking at small-checkered patterns of nylon fabric. Grasping and looking at such fabrics assumed the form of an addiction, with a sense of pleasure being believed to be derived therefrom. Although anticonvulsive therapy resulted in attacks of epilepsy being no longer detectable clinically and by electroencephalography, seizures were still triggered by looking at patterns. Also discussed in this article is the problem of transition from real to psychogenic attacks.

[Adrenocortical diagnosis in ambulatory practice].

The determination of plasma cortisol concentrations is a valuable aid in the diagnosis of some important adrenocortical diseases. Since cortisol is secreted episodically, the diagnostic value of diurnal profiles of plasma cortisol is often limited. On the other hand, a simple ACTH test or an overnight dexamethasone suppression test permit, in most instances, definite diagnosis or exclusion of adrenocortical insufficiency and Cushing's syndrome respectively.

[Fungal infection after antibiotic therapy (author's transl)].

The increasing use of antibacterial chemotherapy in the last four decades encourages the commensal colonization of man by candida yeasts. The sites of particular commensal colonization are the oropharyngeal cavity and the intestinal tract. They are the principal reservoirs from which candida infections can settle into other regions and organs. The metastasis of candida yeasts from the commensal reservoir into regions which are free of microorganisms in healthy persons, usually needs a stimulant noxa: an eliciting bacterial infection, a consumptive primary disease, immunosuppressive therapy among other factors. In the event of candida infection being superimposed on a bacterial infection, successful antibacterial chemotherapy may lead to a remission of the mycosis by removal of the primary noxa.
