Isolation and amino acid sequence of bovine platelet factor 4.

Bovine platelet factor 4 was isolated by affinity chromatography using dextran sulfate Sepharose and purified by subsequent gel filtration. The complete amino acid sequence of this 88-residue, 9505-Da protein was determined by isolation and analysis of the overlapping peptides from tryptic and Staphylococcus aureus hydrolysates of reduced, carboxymethylated, and reductive methylated protein. Primary structure comparison was made between bovine platelet factor 4, human platelet factor 4, and human beta-thromboglobulin. The bovine platelet factor 4 amino-terminal region, which contains two unique phenylalanine residues, is extended by 15 residues relative to human platelet factor 4. The bovine carboxy-terminal region is extended by three residues relative to human platelet factor 4 and differed from beta-thromboglobulin in the absence of two additional terminal residues. Bovine platelet factor 4 shares sequence similarities proportionately with both human platelet factor 4 and beta-thromboglobulin. The sequences of the lysine-rich carboxy-terminal putative heparin binding domains are essentially identical for all three proteins. The heparin neutralizing potencies of bovine and human platelet factor 4 are similar: 40 USP units of heparin neutralized per milligram protein, as measured by a modified chromogenic substrate assay. Heparin neutralization was lost by reduction of the disulfide bonds, but only attenuated by tryptic digestion of the intact protein.

Visual-spatial orientation, gaze direction and dichotic listening asymmetries.

The effects of visual-spatial orientation on dichotic listening performance were investigated in a sample of right-handed male and female adults. The 30 pairs of dichotic stimuli (/ba/ka/da/ga/ta/pa) were presented under three conditions: eye gaze fixed toward center, eye gaze directed 20 degrees to the right, and eye gaze directed 20 degrees to the left of center. Although no effect for gender or order was evident, a significant right ear advantage was found under all conditions. However, over all dichotic listening accuracy was significantly increased when visual-spatial orientation was directed to the right or left side of space.

Improvements in glass capillary gas chromatographic polychlorobiphenyl analysis.

Separation of a 1:1:1:1 calibration mixture of Aroclors 1221, 1016, 1254, and 1260 on soda glass capillaries coated with Apolane (C-87) or Apiezon L is described. Polychlorobiphenyl congener structures are assigned to 112 separated and partially separated zones. The quantitative composition of Aroclor 1221 is reported. The performance of the two stationary phases on different lengths of laboratory and commercially prepared capillaries is compared and found to be very similar. Aroclors 1221, 1016, 1254, and 1260 are employed (1:1:1:1) for the primary calibration mixture because they contain all components of the commercial materials which pollute the environment; they are also easily obtained from the U.S. EPA Repository so that the method can be used in any laboratory by employing the calibration data given here.

Anatomicomechanical study for the hydraulic line of a thermal left ventricular assist system.

A thermal left ventricular assist system currently under development consists of two separate major components, i.e., a pump/actuator module and an engine/thermal battery module. The possible implantation site of the engine/battery module is tentatively determined to be the iliac fossa, which requires a flexible interconnecting line to the pump/actuator module. A quantitative and biomechanical study was done on the effect of the implanted hydraulic line on the bendability of the torso. It revealed that the semirigid interconnecting line would not severely restrict the movement of the patient provided it has the proper prebend configuration. The cadaver fitting study proved the anatomical feasibility of the retroperitoneal iliac fossa as the implant site of the Thermal Ventricular Assist System 8 (TVAS 8) engine/battery module. The location and configuration of the interconnecting line of the TVAS 8 were determined using biomechanical analysis so that the restriction of the body movement due to the line could be minimized. The main route of the line was the left side of the trunk, and the line is W shaped in configuration. The line elongation is the most critical resistive factor and accordingly there may still be rigidity during lateral flexion.

Experimental results for chronic left ventricular assist and total artificial heart development.

After more than 20 years of research on the artificial heart, it is now accepted that the pneumatically actuated total artificial heart can maintain near-normal physiology in experimental animals for several months. Such systems are clinically acceptable for short-term application (a few months) in patients waiting for a suitable heart transplant. However, realistically, a cardiac prosthesis that has the capability of being totally implantable for over 2 years must be available. Based on this premise, our group initiated the development of a mechanically actuated total artificial heart, capable of coupling with either a thermal or electrical actuation system. Since 1977, under the National Institutes of Health chronic left ventricular assist program, we have used two different pusher-plate pumps and electrical systems. Taking advantage of technology generated in these programs, we began developing a prosthesis to replace the total cardiac function. This paper is a summary of both the chronic left ventricular assist and total artificial heart development progress in this laboratory.

Hexsyn trileaflet valve: application to temporary blood pumps.

A trileaflet valve, compression molded of Hexsyn rubber, was evaluated as a component in temporary blood pumps. A pair of valves was integrated into an LVAD which was tested in vitro and in vivo. The valve's hemodynamic performance was satisfactory and the blood compatibility was excellent in a three-week, in vivo experiment conducted to demonstrate performance in a short-term application. Hexsyn, which has excellent properties and produces high quality, complex components with ease of fabrication, offers a potential for low-cost blood pump valves.

Volume compensation for left ventricle assist systems (LVAS): 18 month in vivo evaluation.

The smooth surface cycled chambers, both Biomer and silicone rubber, developed thick tissue capsules and exhibited persistent active tissue reactions at the diaphragm-capsule interface. Textured surface chambers (dacron velour), both the cycled and control, developed a thin, stable capsule, with no active tissue reaction. Textured surface chambers have been explanted and examined after 210 and 494 days. One experiment is continuing, approaching 2 yrs in vivo with good performance. Although the number of textured surface compliance chamber studies as yet is limited, the dramatic improvement in performance and tissue response between the smooth and textured surface studies demonstrates the biological feasibility of this method of variable volume compensation for use in completely intracorporeal LVAS.

Protein involvement in structural transition of erythrocyte ghosts. Use of thermal gel analysis to detect protein aggregation.

In this study, it is shown that systematic temperature-induced protein aggregation occurs on the erythrocyte membrane by intermolecular disulfide bond formation. Specific protein bands disappear from acrylamide gel profiles over rather narrow temperature regions. The aggregation appears to be the result of irreversible structural transitions of the membrane, which can be seen in a sensitive scanning calorimeter. When this method of thermal gel analysis is used, the results suggest that spectrin is a participant in the A transition, that bands 2.1, 4.1, and 4.2 and the cytoplasma portion of 3 are involved in the B transition, and that the transmembrane portion of band 3 may undergo changes in the C transition, previously shown to occur in the anion transport domain of the membrane. The aggregation of specific proteins in the narrow temperature region of these transitions persists as the transitions are moved around on the temperature axis by varying solution conditions. The assignment of particular proteins to specific transitions is reinforced by selective extraction of membrane proteins. Large variations in both the calorimetry and the aggregation pattern occur as salt concentration is increased from 77 mosm to 310 mosm, which is manifested in the splitting of the B transition into two separate transitions, B1 and B2. It is speculated that this occurs as the result of a structural change which may involve components of the cytoskeletal network.
