Static light scattering measurements along a critical decomposition line of a ternary liquid mixture.

In the present paper we publish results of static light scattering measurements in the ternary system N,N-dimetylformamide+n-heptane+toluene. We determined the critical decomposition line in a concentration range of 0 up to 25 mole-% toluene. In seven mixtures along this line we obtained the correlation length of fluctuations and the generalized osmotic susceptibility as a function of the temperature. We apply simple power laws to calculate the critical exponents of both properties. The determined exponents show a systematic growth if the fraction of the third component increases. This growth exceeds the values for critical exponents that were predicted by the critical exponent renormalization.

Visual hallucinations, white matter lesions and disease severity in Parkinson's disease.

OBJECTIVES: To determine if visual hallucinations in patients with Parkinson's disease are associated with an increased prevalence of white matter lesions. PATIENTS AND METHODS: Fifteen patients with (group 1) and 15 patients without (group 2) a history of visual hallucinations were studied. Both groups were matched for age. Magnetic resonance imaging was performed in all patients using standard T2 weighted Fast-Spin-Echo sequences. Assessment of cerebral white matter changes was performed using a modification of established criteria, with semiquantitative evaluation of periventricular and deep white matter changes. RESULTS: There was no significant group difference with regard to the total amount of white matter changes, nor was a group difference found between the amount or extent of periventricular hyperintensities or deep white matter lesions. Group 1 was significantly (P = 0.001) more disabled as evaluated by Hoehn/Yahr stage controlling for age and duration of disease. Mean increases in Hoehn/Yahr stage were not significantly greater in group 1 compared with group 2 at a 2-year follow-up examination (0.6 vs. 0.3, P = 0.166). CONCLUSION: Our data suggest that visual hallucinations are an indicator of a more aggressive course of the disease, but are not associated with a higher prevalence of global or occipital white matter lesions.

Treatment of restless leg syndrome with pergolide--an open clinical trial.

Dopaminergic treatment with levodopa (L-dopa) has been proven as the treatment of first choice in patients with restless leg syndrome (RLS). Augmentation of symptoms and end-of-dose rebound phenomena under L-dopa/decarboxylase inhibitor treatment present major problems in some patients. To evaluate the efficacy of pergolide in RLS, we treated 15 patients suffering from severe RLS, who had previously experienced an augmentation of symptoms under long-term treatment with L-dopa, in an open clinical trial with pergolide. All patients reported an improvement of their RLS symptoms. Our study shows that pergolide, if administered at a mean dose of 0.4 mg in combination with domperidone, is a very effective drug in the treatment of sleep disturbances and daytime symptoms associated with RLS, and does not cause any serious side effects during the observation period of 6 months.

Contrast echocardiography displays increased subendocardial perfusion after nitroglycerin administration.

A mechanism proposed to contribute to the antianginal effect of nitroglycerin is a redistribution of coronary blood flow to the subendocardium. Contrast echocardiography combines ultrasound with echogenic contrast agents to assess regional myocardial perfusion. This study aims to assess the effect of nitroglycerin on myocardial transmural perfusion with contrast echocardiography in humans. Nine patients scheduled for coronary angiography received 300 microg intracoronary nitroglycerin. Contrast echocardiographic studies were performed before and immediately after the administration of intracoronary nitroglycerin. Videodensitometric analysis was performed off-line to measure subendocardial and subepicardial opacification. Subendocardial opacification greater than subepicardial opacification increased from six of 13 patients before nitroglycerin administration to 11 of 13 after nitroglycerin administration (p <0.05). Similarly, these observations increased from nine of 13 patients to 13 of 13 after nitroglycerin administration during diastole (p <0.05). Contrast echocardiography demonstrates increased subendocardial perfusion after the administration of nitroglycerin in these patients.

Retrograde-delivered cardioplegia is not distributed equally to the right ventricular free wall and septum.

Right ventricular myocardial protection during cardiac surgery continues to be a challenge. Retrograde delivery of cardioplegia has been shown to perfuse left ventricular regions subtended by critical coronary stenosis and not adequately protected by antegrade delivery. However, the distribution of cardioplegia from the coronary sinus to the right ventricle remains in question. A reliable means for assessing such flow distribution intraoperatively is provided by contrast echocardiography. It was hypothesized that conventional use of coronary sinus catheters for retrograde cardioplegia delivery does not reliably perfuse the myocardial region subtended by the right coronary artery. Six patients scheduled to undergo elective coronary artery bypass surgery were evaluated with contrast echocardiography to determine the distribution of retrograde-delivered cardioplegia into the right ventricle. Sonicated Renografin-76 (Squibb Diagnostics, Princeton, NJ) was injected during retrograde delivery of cold crystalloid cardioplegia solution and continuous two-dimensional ultrasound imaging of the heart. On-line videodensitometric analysis was performed with a digital ultrasound system. The area under the curve and peak pixel intensity were determined for the anterior septum, the posterior septum, and the right ventricular free wall for each contrast injection. Recorded VHS videotape images of contrast-enhanced perfusion patterns were also reviewed and scored. On-line acoustic-densitometric analysis showed that right ventricular posterior and anterior septal peak pixel intensities were 4.8 +/- 3.2 and 7.3 +/- 1.5, respectively, compared with only 1.6 +/- 1.2 (p < or = 0.05) in the right ventricular free wall.(ABSTRACT TRUNCATED AT 250 WORDS)

[Consumption and ruminating behavior in sheep. 5. Continuous registration and recording of consumption and rumination dependent jaw movements in sheep based on acceleration measurements].

A method of the registration of the feed intake and ruminating behaviour of sheep which uses piezoelectric converters was tested. The appliance-technological solution and the feasibility and routine measurings are described. It showed that measurings on the basis of acceleration registration produced similar results as the 'halter method with plate feelers' used so far and that disturbance of the test animals is reduced.

[Drug residues from slurry and their effect on the development of resistance of E. coli isolates from swine].

A study of veterinary drug residues in the environment of pigs and their effects on selection of bacterial resistances via external contact. Slurry containing sulphonamide (sulphadimidine) residues in a similar concentration to that which could be expected after medication of a pig herd was applied to the stable floor. In reaction to that faecal E. coli of non-medicated pigs showed no significant variations of total bacterial resistance. However some variations concerning the rate of multiple resistant E. coli were manifest. They passed away although drug residues were still detectable.

Rosmarinic acid: a new inhibitor of complement C3-convertase with anti-inflammatory activity.

Rosmarinic acid (RA) is a naturally occurring compound, isolated from Rosmarinus officinalis or Melissa officinalis which inhibits the in vitro immunohaemolysis of antibody-coated sheep erythrocytes by guinea pig serum. In further experiments this reduced immunohaemolysis was found to be due to inhibition of the C3-convertase of the classical complement pathway. The threshold concentration for inhibition of C3-convertase was 10(-6) mol/l. RA with an optimal inhibitory concentration between 5 and 10 mumol/l., resulting in about 70% inhibition of haemolysis. However, higher concentrations of RA were less effective at inhibiting C3-convertase. The inhibition may not be specific for C3-convertase, since another serine protease, elastase, was also weakly inhibited by RA in vitro. RA also exhibited inhibitory activity in three in vivo models in which complement activation plays a role. Thus, RA (0.316-3.16 mg/kg i.m.) reduced paw oedema induced by cobra venom factor (CVF) in the rat, and at 1-100 mg/kg p.o. inhibited passive cutaneous anaphylaxis in the rat. In addition, at 10 mg/kg i.m. RA impaired in vivo activation by heat-killed Corynebacterium parvum (i.p.) of mouse macrophages, as measured by the decreased capacity of the activated macrophages to undergo the oxidative burst. RA (0.1-10 mg/kg i.m.) did not inhibit t-butyl hydroperoxide-induced paw oedema in the rat, indicating selectivity for complement-dependent processes.

Improvement of the gastric tolerance of non-steroidal anti-inflammatory drugs by polyene phosphatidylcholine (Phospholipon 100).

The effect of co-administration with polyene phosphatidylcholine (Phospholipon 100) on the oral gastrotoxicity of various non-steroidal anti-inflammatory drugs (NSAIDs) was studied in the rat. The highly unsaturated phospholipid reduced gastric mucosal lesions measured 3.5 h after oral administration of aspirin, indomethacin, phenylbutazone, diclofenac, piroxicam and sudoxicam to rats which had received a 3 day bread diet followed by 24 h fasting. The extent of reduction of gastrotoxicity varied amongst the individual NSAIDs. Phospholipon 100 also reduced gastric lesions induced by 3 day oral piroxicam and diclofenac administration. A trend towards reduction of oral diclofenac gastrotoxicity was observed following intravenous Phospholipon 100 administration. Phospholipon 100 H (100% saturated phosphatidylcholine) was less effective than Phospholipon 100 in improving acute gastric tolerance to oral phenylbutazone, diclofenac and piroxicam. Administration of the NSAID-Phospholipon 100 combination produced little change in the anti-inflammatory activities of diclofenac on carrageenan paw oedema and diclofenac and piroxicam on adjuvant arthritis in the rat. Combination with Phospholipon 100 offers a novel means for reducing the gastric side-effects of NSAID therapy.

Chemiluminescence from mouse resident macrophages: characterization and modulation by arachidonate metabolites.

Oxidative burst dependent chemiluminescence (CL), generated by mouse resident peritoneal and alveolar macrophages obtained by peritoneal and lung lavage, has been studied using a lumanol-dependent assay. Alveolar macrophages generated greater CL in response to opsonized zymosan (OpZ, 0.017-0.67 mg/ml) than did peritoneal cells, but the former were less sensitive to inhibition by joint cyclo-oxygenase/lipoxygenase inhibitors and superoxide dismutase. Responses of peritoneal macrophages to OpZ and calcium ionophore A23187 (0.1-0.5 microM) were small in comparison to responses of C. parvum-elicited cells, while FMLP (2-20 microM) was ineffective. Arachidonic acid alone (40-80 microM) generated slight CL from resident peritoneal macrophages. When added with OpZ (0.017 mg/ml), arachidonate (40 microM) initially enhanced, then inhibited CL, the inhibition being indomethacin (10 microM) sensitive. These data indicate that modulation of resident macrophage CL by arachidonate products differs somewhat from that reported for elicited macrophages.

Platelet activating factor (PAF) induces the oxidative burst in macrophages.

The response of guinea pig peritoneal macrophages to platelet activating factor (1-O-octadecyl-2-acetyl-sn-glycero-3-phosphoryl-choline) was examined. In Corynebacterium parvum induced macrophages, platelet activating factor, over a wide dose range (3.8 X 10(-5) to 3.8 X 10(-9)M) triggered the oxidative burst as indicated by increased luminol-dependent chemiluminescence and hydrogen peroxide release into culture supernatants. This effect of PAF was inhibited by superoxide dismutase and catalase. Resident macrophages exhibited only slight respiratory activity in response to platelet activating factor which could be increased by adding 1% gelatine to the medium. Activation of macrophages is a new biological effect exerted by platelet activating factor. In immuno-inflammatory reactions, cells capable of generating platelet activating factor may come into close contact with macrophages and by liberating this mediator cause them to release highly toxic oxygen species known to be microbicidal and cytocidal and able to produce vascular endothelial injury. Our findings lend further support to the view that platelet activating factor is a potent and rapid activator of physiological defence mechanisms.

Macrophage, lymphocyte and chronic inflammatory responses in selenium deficient rodents. Association with decreased glutathione peroxidase activity.

The influence of a selenium deficient diet in mice and rats has been studied on glutathione peroxidase (GSH-Px) and secretory activities of peritoneal macrophages, mitogenesis of spleen cells and adjuvant arthritis. Macrophage GSH-Px activity was significantly reduced from 9 weeks on the selenium deficient diet. This reduction was associated with enhanced macrophage H2O2 release on zymosan stimulation after 12 weeks on the diet, a similar trend in chemiluminescence and reduced mitogenesis of spleen cell cultures to T and B cell mitogens after 8 weeks on the diet. Macrophage beta-glucuronidase release was not significantly altered. Phorbol myristic acetate induced macrophage H2O2 generation was reduced by selenium deficiency, possibly due to increased cellular damage. Adjuvant arthritis of rats was significantly enhanced after 6 and 12 weeks on the selenium deficient diet. The enhanced release of H2O2 by macrophages after zymosan stimulation can be directly attributable to loss of GSH-Px activity leading to reduced peroxide breakdown. Peroxide-mediated cell injury would also account for the reduction in lymphocyte mitogenesis and enhancement of adjuvant arthritis. These data provide support for a role of selenium in immune and inflammatory responses.

Animal experiments on the safety pharmacology of lofexidine.

These experiments were carried out to supplement the pharmacodynamic profile of 2[1-(2,6-dichlorophenoxy)-ethyl]-2-imidazoline hydrochloride (lofexidine, Lofetensin and Loxacor), a potent hypotensive imidazoline derivative with a central mode of action. Effects indicative of a central sedative action were seen in various experiments on mice and rats. However, comparative studies with clonidine, particularly those involving the interaction with hexobarbital, the effect on motor coordination on a rotating rod, the effect on body temperature and other experiments, indicated that lofexidine had a 10 to 100 fold less sedative effect than clonidine, depending on the test and the route of administration. In rats there was a dose-related, diuretic and saluretic effect. This effect was not seen in dogs. In rabbits and guinea pigs, lofexidine exhibited local anaesthetic effects and its potency and duration of action were comparable to those of the reference compounds (tetracaine or mepivacaine). Lofexidine did not modify gastric secretion in rats. On the other hand, it inhibited chemically induced (phenylbutazone) and stress-induced (swimming) gastric ulcer formation in rats. Antiinflammatory and analgesic activities in rats and mice were, at most, very weak. Lofexidine increased blood glucose levels of normoglycaemic rats; clonidine is known to have the same effect. In order to investigate the possibility of combining lofexidine with a diuretic drug for the treatment of hypertension, the interaction with hydrochlorothiazide was studied in some experiments. The results obtained from these experiments showed that there was no adverse effect and the diuretic and saluretic effects of hydrochlorothiazide were not impaired. No findings militated against the use of lofexidine as an antihypertensive drug. It is likely that lofexidine, in contrast to clonidine, has less marked central sedative effects.

[Alteration of cartilage by microbial agents and granulocytes].

Severe polyarthritis was induced in 42 SPF piglets by subcutaneous and intraarticular infection in one joint of the bacterium Erysipelothrix rhusiopathiae (Serotype B, strain T 28), which in its chronic stage morphologically resembles human c.P. The light and electron microscopic examination of the articular cartilage and synovial membrane reveals a parallel evolution of hyaline cartilage degeneration, and activation and proliferation of synovial lining cells. The initial cartilage alteration with demasking of collagen fibrils and focal degeneration of chondrocytes in the erysipelas model is caused by direct action of the microbial agent, fibrin and few granulocytes Erysipelothrix bacteria and neutrophilic granulocytes are able to invade the superficial and intermediate cartilage layers. This model is not considered a suppurative infectious arthritis. In chronic villous erysipelas polyarthritis, which develops without the presence of neutrophils in the cartilage, the invasively growing synovial pannus dominates, which deeply destroys the pre-damaged cartilage, resulting in macroscopic focal or wide-spread cartilage erosion. We consider the poorly vascularized cartilage and the particular fibrosis suitable sites for the extremely long (up to three years) persistence of this microbial agent. The persistence of the agent is considered necessary for the persisting immunological reactions and the perpetuation of erysipelas polyarthritis. With longer duration (1-3 years) of experimental erysipelas polyarthritis the number of bacteriologically positive arthritic joint decreases. Microscopically, the causative bacteria may only sporadically identified.
