[Hematologic parameters in the cord blood labor complicated by meconium-stained amniotic fluid].

The aim of our study was to compare hematological values of the cord blood in the presence and absence of meconium-stained amniotic fluid (MSAF). MSAF was associated with decreased total platelet count, increased values of platelet distribution width and red blood cell distribution width. The mean nucleated red blood cells was significantly higher in meconium group. The values of red blood cells, white blood cells, reticulocytes counts and their fractions (HFR,MFR,LFR,IFR) were no different in cord blood of neonate exposed to meconium and in those who were not. These findings can suggest that MSAF contribute to fetal infection rather than hypoxia.

Analysis of some peripheral blood lymphocyte subsets in relation to Borrelia burgdorferi antibodies in patients with Lyme disease.

The aim of our study was to evaluate the changes in T (CD3), B(CD19) lymphocytes, CD4 and CD8 subsets, activated CD3+ HLA-DR+ lymphocytes, lymphocytes with receptor for IL-2 (CD3+CD25), NK cells as well as the rate of CD4/CD8 in 30 patients with recognized Lyme disease, before and after antibiotic therapy. Patients were divided into the following groups: Group I n = 9--without detected specific antibodies against B. burgdorferi and with clinical recognition of EM(erythema migrans). Group II n = 10--with increased IgM production, with a clinical form of Lyme arthritis and neuroborreliosis. Group III n = 11--with increased IgG anti B. burgdorferi production, with clinical recognition of Lyme arthritis. The results were compared with the results obtained in the control group consisting of 90 healthy people. The measurements were performed in the flow cytometer COULTER EPI XL with Becton Dickinson antibodies. The antibodies against B. burgdorferi were detected by means of ELISA method using Dako, Biomedica and Biocom kits. The statistic analysis was performed with AnStat Program. The changes in lymphocyte subsets were characterised by the decrease in the percentage of CD4, CD8, NK and CD3+HLA-DR+ lymphocytes in peripheral blood before treatment with later tendency to increase. The results show that the lag phase of antibodies production coincides with high activity of lymphocytes (group I). The significant level of antibodies in IgM class induced by the collaboration of T and B cells was found in the group II. In the group III, in which antibodies in IgG class predominated, the changes in lymphocyte subsets were less intensive. The results of our investigations indicate that the immune response in Lyme disease develops mainly with the participation of both cellular and humoral response which is involved in both the defense against and the pathogenesis of the disease.

[Immune response and diagnostic aspects during Toxoplasma gondii infection].

Toxoplasmosis is a common and generally benign disease in immunocompetent persons caused by Toxoplasma gondii, which is an intestinal coccidian parasite of felines. Diagnosis of toxoplasmosis is mainly based on the results of serological tests detecting anti-T. gondii specific antibodies, but T lymphocytes and cytokines they produce play a crucial role in determining the outcome of parasitic infection in terms of both protective immunity and immunopathology.

Gammadelta T-cells alterations in the peripheral blood of high risk diabetes type 1 subjects with subclinical pancreatic B-cells impairment.

There is increasing evidence that CD3 + cells bearing gammadelta T-cell receptor (represent the minor subpopulation of the T-cells in the peripheral blood in humans) are involved in autoimmunity development. Gammadelta T-cell receptor (TCR)+ /CD8+ T-cells have been recently found to play a critical role in the pathogenesis and prevention of autoimmune diabetes in the animal model. The aim of the present study was the estimation the gammadelta T-cell subpopulation levels in the peripheral blood of subjects with preclinical and overt type 1 diabetes and their possible associations with the humoral immunity, metabolic parameters and pancreatic B-cells function. The study was carried out in three groups of subjects: 26 first degree relatives of type 1 diabetes patients (prediabetics) with the combinations of autoantibodies against pancreatic B-cells (ICA, GADA, IA-2A, IAA), 22 patients with a recent onset of type 1 diabetes and age and sex-matched 24 healthy volunteers (control group). A decrease was observed in the absolute numbers and percentages of gammadelta+ /CD8+ and gammadelta+ /CD8- T-cell subpopulations in peripheral blood in the prediabetics with the impaired first phase of insulin secretion in comparison to relatives with autoantibodies but still with normal B-cells function, patients with clinical diabetes and healthy controls. In conclusion, the study suggests that the gammadelta T-cells play an important role in the development of insulin-dependent diabetes mellitus (IDDM). It is possible that their levels in the peripheral blood could be an additional marker of preclinical detection of the disease, but further prospective studies in high risk of IDDM subjects are needed.

Analysis of recently activated, memory and naive lymphocyte T subsets in the peripheral blood of patients with Graves' disease and insulin-dependent diabetes mellitus.

It has been recently suggested that the expression of two different isofoms of tyrosine phosphatase antigen (CD45RA and CD45RO) could differentiate T cells into autoreactive and immunoregulatory subsets, which play a crucial role in the autoimmunity process. The aim of the present study was to evaluate the differences between the distribution of memory, naive and recently activated T cells (co-expressing both CD45RA and CD45RO antigens) in the peripheral blood of patients with newly diagnosed Graves' disease and insulin-dependent diabetes mellitus in comparison to healthy controls. The study was carried out in 3 groups: 18 patients with Graves' disease, 16 subjects with a recent onset of type 1 diabetes mellitus and 16 healthy, age and sex matched volunteers. At the onset of both autoimmune diseases the percentage of naive CD+ cells were lower than in the control group and in patients with Graves' disease treated with methimazole. The analysis of CD8+ lymphocyte subsets in the peripheral blood revealed higher levels of CD8+CD45RO+ cells in the newly diagnosed Graves' disease in comparison to the controls, and significant decline in the percentage of memory CD8+ and CD8+CD45RA+CD45RO+ lymphocytes after thyreostatic treatment. The number of CD4+ T lymphocytes co-expressing CD45RA and CD45RO antigens were statistically higher in the patients with recently diagnosed insulin-dependent diabetes mellitus. The abnormal distribution of naive, memory and "transient" T cell subsets in the peripheral blood at the onset of Graves' disease and diabetes type 1 suggest their role in the development of autoimmunity. The significant alterations of lymphocyte T subsets in the peripheral blood of patients with Grave's disease after thyreostatic therapy in comparison to the newly diagnosed subjects supports the immunomodulatory effect of methimazol treatment.

[Serum levels of interleukin 6 (IL-6) and soluble IL-6 receptor (sIL-6R) in children and adolescents with Graves-Basedow disease].

The concentration of interleukin 6 (IL-6), soluble IL-6 receptor (sIL-6R) and intercellular adhesion molecule-1 (sICAM-1) was assayed in 24 children with Graves' disease, aged 6.5-17 years, before treatment with methimazole, during clinical and biochemical remission (8 weeks), and following the eighteen-month therapy with the thyrostatic. ELISA method was used to determine the concentration of interleukin 6, sIL-6R and sICAM-1. An increase was observed in serum concentration of IL-6 and its receptor sIL-6R and soluble intercellular adhesion molecule-1 in the course of Graves' disease. A significant reduction in sIL-6R (8 weeks) and IL-6 (18 months) was noted after therapy with methimazole. A positive correlation was found between microsomal antibodies and the concentration of sICAM-1 and sIL-6R, and between antithyroglobulin antibodies and concentration of sIL-6R in children with fresh hyperthyroidism. No correlation was revealed between the level of thyroid hormones and the concentration of cytokines and adhesion molecules.

English Abstract↗

[The adenoid as lymphoid tissue].

The adenoid is part of the mucosa-associated lymphoid tissue and is organized in distinct compartments. The lymphoid follicles and the interfollicular zone are areas of B lymphocytes and of T lymphocytes, respectively. The adenoid may thus function both as an immunologic inductive and effector organ.

[Clinical consequences and diagnosis of congenital alpha 1-antitrypsin deficiency].

alpha 1-Antitrypsin deficiency is associated with predisposition to developing liver cirrhosis in early childhood and emphysema in adult life. One of the common variants PiZ leads to accumulation of the abnormal protein in hepatocytes and cell damage. Emphysema is a consequence of the protease-antiprotease imbalance. Understanding of the mechanism of disease allows early diagnostics and therapeutic intervention.

The interaction of language and thought in children's language acquisition: a crosslinguistic study.

The purpose of this research was to investigate the potential interaction of conceptual representations and linguistic systems in the process of language acquisition. Language-thought interactions were studied in 80 American, 48 Finnish and 48 Polish preschool children. The research focused on the conceptual and linguistic development of space and time. The spatial and temporal conceptual tasks were designed to measure the transition from experimental to inferential knowledge of space/time representations. In the linguistic domain, comprehension and production tests were used to evaluate the children's capacity to understand mono- and bi-referential location in space and time, where mono-referential location involves a single referent object/event with intrinsic properties (e.g. in/on or past/non-past), and bi-referential location requires two or more referent objects/events and relative perspective (e.g. deictic front/back or before/after). The conceptual and linguistic tests revealed significant changes during the period from two to five years of age, and measures of conceptual development were correlated with measures of linguistic development. As spatial and temporal representations became more structured, children were able to move from mono- to bi-referential location. In a comprehension test, we discovered an interaction of language by dimension. Finnish children found spatial distinctions relatively easy and Polish children found temporal distinctions relatively easy. This interaction was expected on the basis of the relative complexity of the morpho-syntactic coding in the spatial and temporal systems of the two languages. However, the argument relating the timing of acquisition to the transparency versus opacity of the linguistic systems was not supported by the English language comparison. Finally, the Finnish children were relatively better able to accomplish the spatial conceptual tasks as compared to the Polish children. This finding is consistent with a developmental concept of linguistic relativity. In general, the research indicates that spatial and temporal linguistic systems and representational knowledge interact during development with the influence occurring in both directions.

[Activation of the antibacterial properties and neutrophil myeloperoxidase and acid phosphatase in patients with unstable angina pectoris].

Neutrophils are very important in pathogenesis of ischemic disease. They take part in the biomorphology of thrombus and also in the damage of myocardium ischemia in a course of unstable angina pectoris. We evaluated the functional status of neutrophils in peripheral blood, by measurement of bactericidal activity and activity of granulocyte's enzymes: myeloperoxidase (MPO) and acid phosphatase in the patients with unstable angina pectoris. We studied a group of 43 people at the age from 34 to 74 years. The blood for investigation was obtained during the first five hours from the moment of hospitalization. The control group were 40 healthy people. The number of granulocytes was significantly higher in patients with unstable angina pectoris and granulocytes were metabolically activated which was shown in the bigger activity of granulocyte's enzymes like MPO and acid phosphatase than in the control group. The activation of neutrophils is developed by many factors in the course of unstable angina pectoris. They take part in the processes of thrombogenesis and thrombolysis and they are a very important origin for active oxygen metabolites, which are responsible for damage of myocardium ischemia.

The in vivo effect of colony stimulating factors (CSFs) on phagocytic functions of granulocytes.

The effect of commercially available colony stimulating factors and mouse post-endotoxin serum on bactericidal activity and phagocytosis of bacteria by blood neutrophils was tested on mice in vivo after intraperitoneal injection. It was found that tested substances are able to induce phagocytosis and bactericidal activity of blood granulocytes. The similar patterns of these actions during 48 hours were observed for mouse IL-3 and human GM-CSF and for GCT-CM and post-endotoxin serum.
