Bacterial capacity of blood platelets in patients suffering from pneumonia.

The percent of bacteria engulfed by blood platelets, surviving in them and killed was determined in these cells taken from 22 patients with pneumonia before the treatment and from 14 patients after the treatment. The mean percent of bacteria phagocytosed by platelets collected before the treatment was 6.28 and by those after the treatment 5.01, being considerably higher than in the control group (3.42). The mean percent of bacteria surviving in platelets taken before the treatment equalled 3.10 and after the treatment 2.36 being also higher than in the control group (1.32). The mean percent of bacteria killed by platelets taken before the treatment was 3.18 and by those taken after the treatment 2.65, being also higher than in the control group. These results seem to indicate that in cases of pneumonia the blood platelets are involved into the antibacterial activity.

Serum lysozyme activity in patients with Hodgkin's disease undergoing chemotherapy.

Lysozyme activity was determined in undiluted and diluted sera of 41 patients suffered from Hodgkin's disease and of 40 healthy individuals. Blood was collected before chemotherapy, midway of its course, just after completion of chemotherapy and three weeks and three months later. Lysozyme activity was evidently increased as well in undiluted as in diluted sera in all our tested patients. These findings may be interpreted by the presence of immune complexes in sera which may stimulate the release of lysozyme from viable neutrophils. High activity in diluted sera probably is related to the increased concentration of lysozyme inhibitors. The applied chemotherapy did not significantly change the activity of the lysozyme during its course of even after its completion as found by three-month observation. The enhanced lysozyme activity in sera of patients may be at least in part to compensate decreased antibacterial and anticancer mechanisms.

Does ceftazidime or latamoxef affect the phagocytic capabilities of human granulocytes?

In comparative studies made in patients with pneumonia we have treated 14 cases with Latamoxef and 16 cases with Ceftazidime. We have focused attention on the side effects of these drugs and on the phagocytic capacity of granulocytes. In our patients we have noted a decrease of bactericidal activity of plasma and granulocytes and an elevated NBT reduction capability of resting granulocytes. The other tested function of the granulocytes exhibited only small variations. The applied therapy caused the bactericidal activity to be increased in granulocytes and plasma. Both drugs had no harmful effect on the phagocytic activities of granulocytes.

Nitroblue tetrazolium test in patients with Hodgkin's disease in the course of chemotherapy.

In 41 patients with Hodgkin's disease nitroblue tetrazolium (NBT) dye reduction with resting and stimulated granulocytes, before, in the course and after chemotherapy was examined. Twofold decrease of the capability of NBT reduction test with stimulated granulocytes was found, whereas with resting granulocytes only slight diminution was noted. Chemotherapy did not improve the tested parameters. No relationship was observed between the stage of disease and the presence of systemic symptoms and the NBT dye reduction. It can be assumed that granulocytes from patients with Hodgkin's disease are resistant to stimulation. The observations suggest that NBT test may be useful for evaluation of the function of granulocytes and to some extent for monitoring of the chemotherapy.

Serum lysozyme activity in patients with lung carcinoma subjected to radiotherapy.

Lysozyme activity was determined in undiluted and diluted sera of 30 patients with lung carcinoma and of 40 healthy individuals. Blood was collected before radiotherapy, in half of its course, just after completion of radiotherapy, and three weeks and three months later. In the controls the same time spacing was kept. Lysozyme was found to be increased both in undiluted and diluted sera. Enhancement of the lysozyme activity may be due to the presence of immune complexes (antigen--antibody) which are probably responsible for the release of the enzyme from the viable neutrophils. High activity in diluted sera indicates an increase of lysozyme inhibitors in sera of cancer patients. Radiotherapy has not induced significant changes in the enzyme, either during its course of after its completion as confirmed by three-month observation. Taking into consideration the role of the lysozyme in anticancer mechanisms it appears that determination of its activity may be of value in the course of cancer disease.

[Investigations of thyroid function in children and adolescents after treatment of Hodgkin's disease].

Radiotherapy focused on the neck, upper mediastinum or cranium can raise the risk of thyroid dysfunction. The influence of radio- and chemotherapy on thyroid morphology and function after treatment of Hodgkin's disease in 31 children and acute lymphoblastic leucaemia in 36 children was evaluated. Clinical examination, estimation of blood TSH, FT4 levels as well as ultrasonographic scan were performed. In the group of children with Hodgkin s disease the blood level of ATG Antithyreoglobulins andMAB, microsmal antibodies were estimated. The results of clinical examination were normal in all patients. Thyroid function tests were also normal, except for one case with elevated TSH values (22.76 microIU/ml) and decreased FT4 values (0.65 ng/ml). Three patients (9.7%) with Hodgkin s disease and three (8.5%) with ALL (only one had cranial radiotherapy) had an abnormal image of thyroid in ultrasonography (hypoechogenicity, heterogenous ultrasound scan, solid nodule). The results of our investigations indicate that abnormalities found in the ultrasound scan in children and adolescents with Hodgkin's disease in whom treatment has been terminated, can be a first prodrome of thyroid pathology.

Activation of blood platelets in chronic hepatitis and liver cirrhosis P-selectin expression on blood platelets and secretory activity of beta-thromboglobulin and platelet factor-4.

BACKGROUND/AIMS: Blood platelets are cells that quite often undergo damage in chronic liver diseases. Endotoxemia and hyperkinetic circulation influence platelets in an active manner. The role of platelets in the development of hepatitis and liver fibrosis is speculative. The aim of the study was to determine the influence of chronic liver diseases on platelets morphologic parameters, their secretory activity and P-selectin expression. METHODOLOGY: The examination was completed in the group of 29 patients with chronic hepatitis and 27 with liver cirrhosis of postinflammatory etiology (HBV, HCV). Liver biopsies were carried out in all patients. Thirty-two healthy individuals were the control group. Platelets morphological parameters (number, volume, platelet crit, micro- and macrothrombocyte fraction) were estimated. beta-thromboglobulin concentration and platelet factor 4 in blood serum as well as P-selectin expression on resting platelets and after thrombin activation were also examined. RESULTS: Number, volume, and platelet crit decreased with the advancement of a liver disease. Megathrombocyte fraction increased inversely with the severity of liver damage. The concentration of beta-thromboglobulin and platelet factor 4 alpha-granule contents in blood serum was higher 2- and 7-times, respectively than in healthy controls. P-selectin expression on resting platelets was considerably higher. After stimulation with thrombin, P-selectin expression was equal (chronic hepatitis) or higher (liver cirrhosis) than in the control. CONCLUSIONS: There are changes of platelet morphological parameters, with accompanying megathrombocyte fraction increase that occur in chronic liver diseases. Thrombocytes in chronic liver diseases and liver cirrhosis are more activated. Platelet sensitivity to stimuli in these ailments is higher (liver cirrhosis) than in the healthy controls.

[Renal function in children after treatment of acute lymphoblastic leukaemia (ALL)].

UNLABELLED: The aim of the study was to evaluate the renal function (on the basis of serum: cystatin C, creatinine, beta2-microglobulin(B2M) levels, creatinine and cystatin C clearance, and urine osmolality) in 37 children with ALL (acute lymphoblastic leukaemia) after completing intensive treatment (9 in course of maintenance therapy), depending on nephrotoxic antibiotics use (</=3 times vs > 3 times) and methotrexate dose (1.0 g/m2 vs 5g/m2). The control group was formed by 15 healthy children. RESULTS: 1) Serum cystatin C, creatinine, beta2-microglobulin levels creatinine and cystatin C clearance, and urine osmolality were similar in the analysed and control groups. 2) We did not find any differences between the analyzed parameters in the groups of children: a) in maintenance therapy and after the end of the therapy, b) received methotrexate in the dose of 5.0 g/m2 vs 1.0 g/m2, c) not treated children and treated </= 3 times and gt; 3 times with nephrotoxic antibiotics. CONCLUSION: The renal function is normal after completion of antileukaemic therapy in spite of previous use of nephrotoxic drugs.

[Phagocytic activity of platelets in patients with pneumonia].

In 22 patients with pneumonia the per cent of phagocytizing platelets and the phagocytosis index of the platelets were determined before treatment and in 14 patients this was done after treatment. Before treatment the per cent of phagocytizing platelets was 1.37%, on average, and after treatment it was 1.20, and it was thus much lower than in the control group X 2.33. The phagocytosis index before treatment was 1.39 on average, and after treatment it was 1.28, while in the control group it was 1.54. The obtained results seem to indicate that in pneumonia platelets participate in the antibacterial processes and the lungs as a thrombopoietic organ contribute to this phenomenon.
