Monogene and polygene therapy for the treatment of experimental prostate cancers by use of apoptotic genes bax and bad driven by the prostate-specific promoter ARR(2)PB.

We have shown that adenovirus-mediated manipulation of apoptotic genes such as bax could be a therapeutic option for prostate cancer. Unfortunately, the response of experimental prostate tumors to a single therapeutic gene of the apoptotic pathway is short-lived, and most of these tumors relapse after a short period of time. In this investigation we present data generated with adenovirus AvARR(2)PB-Bad, in which the apoptotic gene bad was placed under the control of the dihydrotestosterone (DHT)-inducible third-generation probasin-derived promoter ARR(2)PB. This therapeutic virus was given alone or in combination with other therapeutic viruses to a variety of in vitro and in vivo experimental models of prostate cancer. On infection with AvARR(2)PB-Bad, DHT-induced Bad overexpression occurred specifically in androgen receptor-positive (AR(+)) cells of prostatic derivation. The apoptotic effect of AvARR(2)PB-Bad (group 1) was compared with that of AvARR(2)PB-Bax (which overexpresses the apoptotic protein Bax) (group 2), with that of the combination AvARR(2)PB-Bad plus AvARR(2)PB-Bax (group 3), and with that of the control virus AvARR(2)PB-CAT (group 4) in the cell line LNCaP. In addition to identifying the modality of apoptosis induction by overexpressed Bad, the results suggested that group 3 contained more apoptotic cells than any other group. In additional studies, AR(+) androgen-dependent LNCaP cells or AR(+) and androgen-independent C4-2 cells were injected subcutaneously into nude mice. Four groups of six LNCaP or C4-2 tumors were treated with the same combinations of viruses discussed above for groups 1, 2, 3, and 4. Treatment resulted in decreased tumor size in groups 1, 2, and 3 compared with group 4. There was a better response in group 3 compared with group 2, and in group 2 compared with group 1. A better response in group 3 was confirmed during a 8-week follow-up period, in which no treatment was administered. Two LNCaP and C4-2 tumors of group 3 disappeared at the end of treatment and did not recur after an 8-week follow-up period. The data suggest that polygene therapy with apoptotic molecules is more effective in experimental models of androgen-dependent or -independent prostate cancer than monogene therapy.

Fluid-fluid correlation through a model charged membrane: analytical results.

We study the properties of electrical double layers separated by a charged plate of finite thickness. The nonlinear Poisson-Boltzmann equation is analytically solved for this system. It is shown that, for an unsymmetrically charged, narrow plate, the charged fluids at both sides of the plate are strongly correlated, and the local electroneutrality condition (LEC) is not satisfied. The LEC is satisfied only for an infinitely thick plate. Analytical expressions for the induced charge and mean electrostatic potential are given and analyzed. These findings could be relevant for the understanding of protein adsorption on membranes.

A prospective study of detection of pancreatic carcinoma by combined plasma K-ras mutations and serum CA19-9 analysis.

INTRODUCTION: Previous studies have demonstrated the possibility of detecting K mutations in the plasma of patients with pancreatic carcinoma. AIM: To investigate the diagnostic value of combined plasma K mutations with serum CA19-9 in patients with pancreatic carcinoma. METHODOLOGY: The study included 58 consecutive patients with suspected pancreatic masses and 21 healthy controls. DNA was extracted from plasma. Point mutation at codon 12 of K gene was analyzed by means of the enriched polymerase chain reaction-restriction fragment length polymorphism technique. RESULTS: No mutated K codon 12 was detected in the plasma DNA from the 21 healthy controls. Mutations of K codon 12 were found in plasma DNA samples from 29 (70.7%) of 41 patients with pancreatic carcinoma. Elevated CA19-9 was found in 30 (73.2%) of 41 pancreatic cancer patients. In total, 37 of 41 (90.2%) patients with pancreatic carcinoma tested positive by either method. Mutated K was also found in 3 patients and elevated CA19-9 in 8 of the 17 patients with other lesions in the pancreatic area. CONCLUSION: The results indicate that plasma K mutation analysis combined with serum CA19-9 determination could detect the majority of cases of pancreatic carcinoma.

[Effect of cyclosporin A on the growth of human first trimester cytotrophoblasts in vitro].

OBJECTIVE: To investigate effect of cyclosporin A (CsA) on the growth of first trimester human cytotrophoblast in vitro. METHODS: To isolate and purificate first trimester human cytotrophoblasts by way of percoll density gradient centrifugation; to evaluate the effect of CsA on in vitro proliferation of cytotrophoblasts using (3)H-thymidine incorporation assay; to analyze of cell cycle of cytotrophoblasts with flow cytometric assay; and to survey morphology of the cells by scanning electro-microscope respectively. RESULTS: CsA could increase in vitro proliferation of first trimester human cytotrophoblasts when concentration of CsA ranged from 1 x 10(-4) micromol/L to 1 micromol/L, but decrease in vitro proliferation of the cells if the concentration reached to 10 micromol/L. G(2)-M phase increased from (2.8 +/- 0.5)% to (6.7 +/- 1.3)% when the cytotrophoblasts were incubated with culture medium containing 1 micromol/L CsA for 24 hours, while S phase increased from (14 +/- 5)% to (28 +/- 8)% and apoptosis decreased from (8.7 +/- 2.2)% to (3.6 +/- 1.0)% when the culture in the same incubating condition lasted for 48 hours. Pseudopodia of the trophoblasts increased in number and length when they were incubated in culture medium containing 1 micromol/L of CsA for 48 hours, but decreased if the concentration reached to 10 micromol/L with the same incubating duration. CONCLUSION: CsA is able to promote growth of first trimester human cytotrophoblasts and change their morphology, which appears to increase in invasive ability.
