Human SULT1A1 gene: copy number differences and functional implications.

SULT1A1, which catalyzes the sulfate conjugation of a wide variety of natural and synthetic compounds, is genetically polymorphic. Biochemical and pharmacogenetic studies have demonstrated that individual variation in the level of enzyme activity is inherited. Common single-nucleotide polymorphisms (SNPs) located in the open reading frame and in the 5'-flanking region (5'-FR) may account for a portion of this individual variation. In this study, we demonstrate the presence of SULT1A1 gene deletions and duplications, representing an additional source of variability in the metabolic activity of this enzyme. A quantitative multiplex PCR assay was used to measure the extent of copy number differences and the frequency of these events in different populations. An analysis of DNA from 362 Caucasian-American and 99 African-American showed the presence of 1 to approximately 5 copies of SULT1A1 in individual samples: 5% of Caucasian subjects contained a single copy of the gene and 26% had three or more copies, while 63% of African-American subjects had three or more copies. Analysis of the genomic region surrounding the SULT1A1 gene in three separate cases with a deletion demonstrated that the entire SULT1A1 gene was affected. Reporter assays, constructed for each of the various 5'-FR SNP haplotypes, suggest that these may also play a role in SULT1A1 activity. However, the variability in the level of enzyme activity among 23 human platelet and 267 human liver samples was best explained by gene copy number differences when all sources of genetic variability were considered (P < 0.0001). Overall, these observations have obvious implications for the effectiveness of SULT1A1 as a drug and hormone metabolizing enzyme and its potential role as a risk factor for disease.

Electronic structures of acyl nitrites and nitrates.

The gas phase electronic structures of CM(3)C(O)ONO and CM(3)C(O)ONO(2) (M=H, Cl, F) are studied by photoelectron spectroscopy (PES) combined with the outer valence Green's function (OVGF) calculations at 6-311+G(d, p) basis sets. The highest occupied molecular orbital (HOMO) for each compound is the carbonyl oxygen lone pair (n(O)), the ionizations of these orbitals are associated with the vibrational frequency about 1750 and 1820 cm(-1) reflected on the first band, respectively, for acyl nitrites and nitrate. Comparing with the calculated energies, it can be concluded that the syn conformers with Cs overall symmetry, a planar CC(O)ONO skeleton in nitrites, and a planar CC(O)ON skeleton in nitrates, respectively, are the most stable in the gas phase.

Assembly of a tetrathiafulvalene-anthracene dyad on the surfaces of gold nanoparticles: tuning the excited-state properties of the anthracene unit in the dyad.

Due to the unique features of the tetrathiafulvalene (TTF) unit, such as the electron-donating ability and presence of methylthio groups, dyad 1 can be assembled on the surfaces of gold nanoparticles, as indicated by absorption, electrochemical, and fluorescent-spectral studies. Dyad 1 can also be disassembled by the addition of thiols. Assembly of dyad 1 on the surfaces of gold nanoparticles leads to the formation of a triad (A1-D-A2), which in turn modulates the photoinduced electron-transfer process within dyad 1. Accordingly, the fluorescence intensity of dyad 1, after assembly with gold nanoparticles, increases, and the fluorescence lifetime is prolonged. Furthermore, the assembly of dyad 1 on gold nanoparticles facilitates photodimerization of the anthracene units of dyad 1. Both fluorescence and photodimerization are associated with the excited-state behavior of the anthracene unit, thus it may be concluded that the excited-state properties of the anthracene unit can be tuned upon complexation with gold nanoparticles.

Development and validation of pheochromocytoma of the adrenal gland scaled score for predicting malignant pheochromocytomas.

OBJECTIVES: To evaluate the diagnostic performances of the pheochromocytoma of the adrenal gland scaled score (PASS) proposed in a previous report and that of a logistic model developed in this investigation. METHODS: In all 130 patients with malignant or assumed benign pheochromocytomas, 15 predictive variables were observed. Receiver operating characteristic (ROC) curve analysis was performed to evaluate the diagnostic performance of PASS. The logistic model was developed using the 15 predictive variables. Its performance was evaluated by calculating the area under the ROC curve and comparing it with that of the PASS. RESULTS: The PASS had the area under the ROC curve of 0.899 (95% confidence interval 0.844 to 0.954). Of the 15 variables entered in the logistic regression analysis, 9 were retained in the model. The area under the ROC curve for the logistic model was 0.983 (95% confidence interval 0.967 to 0.998). CONCLUSIONS: ROC analysis indicated that the PASS could be used for the diagnosis of malignant pheochromocytomas. The logistic model was able to improve the diagnostic performance of the PASS using a different variable weighting method. We emphasize, however, that a clinical prospective evaluation is needed to confirm their actual value.

Isolation of neural crest-derived stem cells from rat embryonic mandibular processes.

BACKGROUND INFORMATION: Substantial evidence indicates the existence of NCSCs (neural crest-derived stem cells) in embryonic mandibular processes; however, they have not been fully investigated or isolated. The aim of the present study was to isolate stem cells from mandibular process during embryonic development by MACS (magnetic-activated cell sorting). The findings show that the cells are multipotent and self-renewing. RESULTS: LNGFR (low-affinity nerve-growth-factor receptor)+ cells were isolated from rat embryonic mandibular processes by MACS. The cells were grown in clonal culture by limiting dilution to assess their developmental potential. Clone analysis indicated that, first, LNGFR+ cells are multipotent, being able to generate at least neurons and Schwann cells, similar to peripheral neural crest stem cells. Secondly, multipotent LNGFR+ cells generate multipotent progenies, indicating that they are capable of self-renewal and therefore are stem cells. Thirdly, manipulation of the medium supplementation alters the fate of the isolated LNGFR+ cells. CONCLUSIONS: These results indicate that LNGFR antibodies label NCSCs with high specificity and purity, and suggest that positive selection using these antibodies may become the method of choice for obtaining multipotent cells from rat embryonic mandibular processes for tissue engineering or regenerative therapeutic use.

Relations of caregiving stress and health depend on the health indicators used and gender.

Extensive research has evaluated relations between stress and health. These studies have varied in the type of stress examined (acute vs. chronic) and in the way in which health has been operationalized. Here we examine relations between chronic stress and 25 indicators of various health dimensions (e.g., physiological indexes, medical records, and self-reports of global health; symptoms, functional status, health service utilization, and psychosocial distress/quality of life). We also assessed whether such relations are moderated by gender, an individual difference variable that is important to health and longevity. Samples included 157 community-residing older adults (M age = 69.4 years, 31.8% men), approximately half of whom were caregivers for a spouse with Alzheimer's disease, and half were demographically similar noncaregiver spouses. Principal component analyses on the 25 health measures resulted in 5 factors that met standard criteria for acceptance. In women, caregivers reported worse physical health and psychological health than noncaregivers, but their physiological risk was similar. In men, caregivers had greater physiological risk, but they reported better physical health than did men noncaregivers. Researchers who study chronic stress and health should consider the possibility that the relation between chronic stress and health may vary for men and women depending on the type of health being assessed.

[Preparation and quantitative analysis of methyl phenyl carbonate standard sample].

The preparation and quantitative analytical method for high-purity methyl phenyl carbonate (MPC) was developed. Dimethyl carbonate (DMC) and diphenyl carbonate (DPC) were firstly used as reactants to synthesize MPC catalyzed by TiO2/SiO2 because the disproportionation of MPC is reversible and the reverse reaction is thermodynamically favorable. The high-purity MPC standard sample was obtained by reduced pressure distillation and the removal of minor phenol with dilute sodium hydroxide solution. The qualitative analysis by gas chromatography-mass spectrometry (GC-MS) showed there were minor phenol and diphenyl carbonate in the MPC sample. The mass percent concentration of water in the MPC sample was 0.26%, which was determined by Karl Fischer titration. A gas chromatographic method with an OV-101 capillary column was established for quantifying the minor phenol and DPC in the MPC sample. The quantitative results showed the mass concentrations of phenol and DPC were 2.04% and 1.59%, respectively. The difficulty to analyze MPC, the intermediate product of the transesterification of DMC and phenol, was solved with the self-made MPC standard sample.

[Effects of climate change on winter wheat growth and yield in North China].

With the combination of crop model and climate mode, this paper simulated and predicted the effects of climate change on the winter wheat production of North China in future 100 years. The results showed that from 2000 to 2004, the variation trend of simulated winter wheat production in North China was accordant with the measured one, and the difference of winter wheat's lifecycle and yield was not obvious. In the future 100 years, the lifecycle of winter wheat would be shortened by 8.4 days, and its yield would be decreased to different extent, with 10.1% as the average. If proper measures such as adjusting crop parameters and management measures were adopted, the decline of winter wheat yield could be mitigated effectively.

Suppressed hostility predicted hypertension incidence among middle-aged men: the normative aging study.

This study tested whether suppressed hostility predicted incident hypertension (HTN) in initially nonhypertensive men, using prospective data from the Normative Aging Study. Six hundred twenty-seven men who completed the MMPI and participated in a subsequent laboratory assessment were included in the study. The Cook-Medley Hostility scale (Ho), a suppression factor, and other risk factors were used to predict incident HTN over a 3-year period. Logistic regression showed a significant three-way interaction among Ho, suppression, and age in predicting incident HTN. Among middle-aged men (<or=60 years) with higher suppression, a 1-point decrease in the Ho score was associated with an 18% increase in HTN risk. This relationship remained significant after controlling for relevant risk factors. Among older participants, the HoxSuppression interaction was not predictive of HTN incidence. These results provide support for the Suppressed Hostility hypothesis in middle-aged men.

Phenotyping and genotyping studies of thiopurine S-methyltransferase in Kazaks.

OBJECTIVE: This study was conducted to investigate the thiopurine S-methyltransferase (TPMT) activity distribution and gene mutations in Kazaks, and compared the results with those of other ethnic groups. METHODS: Erythrocyte TPMT activity was measured in Kazaks (n = 327) via a validated high-performance liquid chromatography assay. Polymerase chain reaction-based methods were used to analyze three commonly reporter-inactivating mutations: G238C, G460A, and A719G. RESULTS: Unimodal distribution of TPMT activity was found in Kazaks. Six TPMT*3C heterozygotes and two TPMT*3A heterozygotes were found in 327 Kazaks, with allele frequencies of 0.9 and 0.3%, respectively. The subjects with TPMT*3A and TPMT*3C heterogygotes had substantial TPMT activity over the range of 6.40-11.75 U/ml RBC. CONCLUSION: Unlike in most Caucasians, TPMT*3C is a common mutant allele in Kazaks, whereas TPMT*3A is a rare mutant allele. Further studies are needed to explore the clinical impact of these TPMT mutants to thiopurine therapy in Kazak patients.

Morphological control of ZnO nanostructures by electrodeposition.

We report here an electrodeposition route for the preparation of oriented and well-defined ZnO nanostructures by kinetically controlling the growth rates of various facets of the deposit by appropriate capping agents. We demonstrated that adsorption of Cl(-) takes places preferentially onto the (0001) planes to hinder the crystal growth along the c-axis, and results in the formation of platelet-like crystals. It is also shown that the morphology evolved from hexagonal tapers to hexagonal rods and rhombohedral rods by changing the compositions of the capping agents. Furthermore, strong UV emissions at 380 approximately 390 nm and negligible green bands at around 500 nm were observed, indicating that these ZnO electrodeposits are highly crystallized and of excellent optical quality.

[Effects of simulated acid rain on physiological and biochemical characters of eggplant, the host plant of Tetranychus cinnabarinus].

In a series of laboratory trials, this paper studied the responses of eggplant (Solanum melongena), the host plant of carmine spider mite Tetranychus cinnabarinus, to different pH values simulated acid rain. The results showed that with the increasing acidity of simulated acid rain, the CAT activity and the contents of P and soluble protein in egg plant leaves increased significantly first, reaching the highest at pH 4.0 or 3.0, and then decreased; while the POD activity and soluble sugar content were in adverse. The reduced sugar content and SOD activity of eggplant leaves increased, but the pH value decreased with increasing acidity of acid rain. Acid rain had no effect on leaf water content. Among the test indices, leaf POD was most insensitive to the acid rain, followed by leaf pH, SOD and CAT, while the others were very sensitive. Weak acid rain (pH > 4.0) promoted the protective ability of eggplant leaf and its growth, and the growth of T. cinnabarinus was also promoted because of the changed contents of soluble sugar, P and soluble protein in eggplant leaves being more favorable to its eating; while strong acid rain (pH < 3.0) inhibited the growth of both host plant and mite.

Reaction of AgN3 with SOCl2: evidence for the formation of thionyl azide, SO(N3)2.

Pure thionyl azide SO(N(3))(2), which is the only gaseous reaction product, has been generated in a vacuum by the heterogeneous reaction of SOCl(2) vapor with AgN(3) at room temperature at a SOCl(2) vapor pressure of 1 x 10(-3) Torr. Evidence for the formation of SO(N(3))(2) is given by on line photoelectron spectroscopy (PES) combined with outer valence Green's function (OVGF) calculations with the 6-311++G(2df) basis set. The good agreement between the PES experiment and the OVGF calculation shows that SO(N(3))(2) has C(1) symmetry. The first ionization energy of SO(N(3))(2) is 10.18 eV.

Time-resolved spectroscopic behavior of Fe2O3 and ZnFe2O4 nanocrystals.

Using nanosecond (ns) and femtosecond (fs) time-resolved absorption spectroscopies (pump-probe technique), the carrier dynamics in transition metal oxide nanocrystals of alpha-Fe2O3 and ZnFe2O4 was studied during the photolysis process. For Fe2O3 and ZnFe2O4 nanocrystals, the fs measurements detect similar profiles of a positive nonlinear absorption in their capped nanocrystals, whereas much weak signals in the naked particles. In the nanosecond measurements Fe2O3 and ZnFe2O4 nanocrystals show obvious excitation-power dependent absorption properties and at the low pump power they show weak photobleaching, but at high pump power they produce positive nonlinear absorptions. For Fe2O3 nanocrystals, the threshold power of negative absorption (bleach) to positive absorption increases with reducing size, whereas for the ZnFe2O4 samples, the threshold powers reach minimum at a critical size of 11 nm, grow for both the bigger and the smaller nanocrystals. These results reflect the influences of their microscopic magnetic couplings and carrier correlation on biexciton absorption in Fe2O3 and ZnFe2O4 nanocrystals. All the results indicate that the time resolved photoabsorption techniques are useful to study the microscopic spin interactions and carrier correlations in transition metal oxide nanocrystals.

A study on the fabrication of chemically modified bacteriorhodopsin film and its photochemical properties.

Chemically modified bacteriorhodopsin (BR) films embedded in gelatin matrix were fabricated. It was found that chemically modified BR/gelatin film remained the original conformation of BR and possessed homogeneity. Then, the photochemical conversion from all-trans O state to 9-cis P state in the chemically modified BR film/gelatin was investigated. For comparison purpose, the same conversion in the wild-type BR (BRwt) film was also presented here. To our know, it is the first report to show that the absorption changes of the P state in the chemically modified BR film were larger than in the BRwt film upon illumination with red light at the low actinic power density.

Fabrication and photoelectric response of poly(allylamine hydrochloride)/PM thin films by layer-by-layer deposition technique.

Thin films of poly(allylamine hydrochloride) (PAH) and bacteriorhodopsin (bR) embedded in purple membrane (PM) have been prepared by layer-by-layer (LBL) self-assembly technique. The results obtained by UV-Vis spectroscopy and atomic force microscopy (AFM) analysis showed that the biological activity of bR was preserved and PM fragments could be well oriented onto the ITO substrate. A photo-electrochemical cell with the structure of ITO/(PAH/PM)(n)/electrolyte (0.5M KCl)/Pt was fabricated and studied. The photocurrent peaks of (PAH/PM)(6) corresponding to light-on and light-off were about 200 and 100 nA/cm(2), respectively, with the former enhanced 30% higher than that of the reference films made of (PDAC/PM)(6).

Are metabolic risk factors one unified syndrome? Modeling the structure of the metabolic syndrome X.

The metabolic syndrome, manifested by insulin resistance, obesity, dyslipidemia, and hypertension, is conceived to increase the risk for coronary heart disease and type II diabetes. Several studies have used factor analysis to explore its underlying structure among related risk variables but reported different results. Taking a hypothesis-testing approach, this study used confirmatory factor analysis to specify and test the factor structure of the metabolic syndrome. A hierarchical four-factor model, with an overarching metabolic syndrome factor uniting the insulin resistance, obesity, lipid, and blood pressure factors, was proposed and tested with 847 men who participated in the Normative Aging Study between 1987 and 1991. Simultaneous multi-group analyses were also conducted to test the stability of the proposed model across younger and older participants and across individuals with and without cardiovascular disease. The findings demonstrated that the proposed structure was well supported (comparative fit index = 0.97, root mean square error approximation = 0.06) and stable across subgroups. The metabolic syndrome was represented primarily by the insulin resistance and obesity factors, followed by the lipid factor, and, to a lesser extent, the blood pressure factor. This study provides an empirical foundation for conceptualizing and measuring the metabolic syndrome that unites four related components (insulin resistance, obesity, lipids, and blood pressure).

Is caregiving hazardous to one's physical health? A meta-analysis.

Caring for a family member with dementia is generally regarded as a chronically stressful process, with potentially negative physical health consequences. However, no quantitative analysis has been conducted on this literature. The authors combined the results of 23 studies to compare the physical health of caregivers with demographically similar noncaregivers. When examined across 11 health categories, caregivers exhibited a slightly greater risk for health problems than did noncaregivers. However, sex and the health category assessed moderated this relationship. Stronger relationships occurred with stress hormones, antibodies, and global reported health. The authors argue that a theoretical model is needed that relates caregiver stressors to illness and proffers moderating roles for vulnerabilities and resources and mediating roles for psychosocial distress and health behaviors.
