A mathematical model for simulating the bone remodeling process under mechanical stimulus.

OBJECTIVES: Among the current mathematical models for bone remodeling, few can consider bone resorption due to overload. The objective of this paper is to develop a new bone remodeling model which can simulate both underload and overload resorptions that often occur in dental implant treatments. METHODS: Based on the traditional model, a new mathematical equation relating the density change rate with mechanical stimulus has been developed. The new equation contains an additional quadratic term which can produce reduction in bone density at high load levels. In addition, to fully exploit the characteristics of this model, a range of different bone remodeling behaviors were studied under the load cases with both constant and varying stress magnitudes. Finally, the model was applied in conjunction with the finite element method to a practical case of dental implant treatment. RESULTS: The FE analysis results showed that bone resorption at the neck of the implant occurred due to occlusal overload but then resorption stopped after some time before reaching the coarse threads. Meanwhile, the density of the bone deeper into the mandible increased slightly due to the additional mechanical stimulus provided by the occlusal load. This phenomenon is observable in some clinical situations. SIGNIFICANCE: The new model can describe the bone overload resorption, a feature which is absent in most of the current models. And by simulating the dental implant treatment using FE method, the ability of the new mathematical model to simulate overload bone resorption has been clearly demonstrated.

Preparation and pharmacokinetics in rabbits of breviscapine unilamellar vesicles.

The purpose of the study was to prepare the unilamellar liposomal vesicles of breviscapine (Breviscapine-LUVs) and investigate the pharmacokinetics of Breviscapine-LUVs in rabbits. Breviscapine-LUVs were prepared by the film dispersion method and treated further by extrusion. Its size distribution and zeta potential were determined by photon correlation spectroscopy. The encapsulation efficiency (EE) and cumulative release of Breviscapine-LUVs were assayed by the dialysis method. The crossover design (two periods) was used in six rabbits, which were administered Breviscapine-LUVs and reference preparation. Results showed that the particle size of Breviscapine-LUVs was 50.8 nm, and the polydispersity index was 0.287. The zeta potential was -24 mV +/- 9 mV (n = 3), and the EE% was 81.1 +/- 1.1% (n = 3). The cumulative release of vesicles in 0.9% NaCl was 17.2 +/- 0.78%, 26.1 +/- 0.68%, and 29.9 +/- 0.81% in 2, 8, and 24 h, respectively. The mean concentration-time curves of breviscapine liposomes and reference preparation were both fitted to a two-compartment model with the main pharmacokinetic parameters as follows: t(1/2beta) of Breviscapine-LUVs and reference preparation were (42.5 +/- 28.6) min and (6.01 +/- 4.64) min, respectively; CL((s)) were (15.3 +/- 9.03) mL x min(-1) and (84.6 +/- 40.6) mL x min(-1), respectively; AUC(0-300) were (1267 +/- 1083) microg x min x mL(-1) and (196 +/- 107) microg x min x mL(-1), respectively. Compared with the reference preparation, breviscapine liposomes had a much higher concentration in plasma and contained characteristic of sustained-release, which ameliorated the pharmacokinetic properties of scutellarin.

Standardizing global gene expression analysis between laboratories and across platforms.

To facilitate collaborative research efforts between multi-investigator teams using DNA microarrays, we identified sources of error and data variability between laboratories and across microarray platforms, and methods to accommodate this variability. RNA expression data were generated in seven laboratories, which compared two standard RNA samples using 12 microarray platforms. At least two standard microarray types (one spotted, one commercial) were used by all laboratories. Reproducibility for most platforms within any laboratory was typically good, but reproducibility between platforms and across laboratories was generally poor. Reproducibility between laboratories increased markedly when standardized protocols were implemented for RNA labeling, hybridization, microarray processing, data acquisition and data normalization. Reproducibility was highest when analysis was based on biological themes defined by enriched Gene Ontology (GO) categories. These findings indicate that microarray results can be comparable across multiple laboratories, especially when a common platform and set of procedures are used.

[Epidemiological survey of the incidence of bronchial asthma in occupational populations in Beijing area].

OBJECTIVE: To investigate the correlation of asthma incidence with environmental pollution. METHODS: We performed stratified-cluster-disproportional-random-sampling survey in the occupational populations from 10 professional systems by using an uniform protocol, questionnaire and procedure in this multicenter study. Original data were processed and statistically analyzed with SPSS 8.0 for windows. Prevalence rates were compared by chi(2) test. RESULTS: A total of 50,558 people supposed to cover more than 200,000 staff and workers were sampled. Six hundred and thirty asthmatics were identified and the overall prevalence rate in Beijing area was 1.25%. The incidence of asthma in occupational populations from urban commercial and trade circles, common chemical plants, and a certain petrochemical works was 0.51%, 1.06% and 2.81% respectively. The morbidity of asthma in the latter two populations was significantly higher than that in peasants (0.43%) in the remote outskirts of Beijing city (P < 0.01). The relative risk (RR) of asthma incidence in occupational populations from the commercial and trade circles, common chemical plants, the petrochemical works and the students in elementary and middle schools was 1.88, 2.79, 5.19 and 3.05 respectively as compared to peasants, the differences being statistically significant. CONCLUSION: The results suggest that the difference in asthma incidence in occupational populations may be attributable to environmental pollution.
