[Genetic polymorphism of 3 STR loci on chromosome X and its forensic application in Chinese population].

To investigate the genetic polymorphisms of three short tandem repeats loci of chromosome X in Chinese Han population in Chengdu area and its use in forensic science. Three X-chromosome linked short tandem repeat loci were analyzed by PCR followed by polyacrylamide gel electrophoresis. Hardy-Weinberg equilibrium was tested and forensic interested value was calculated . The power of exclution of DXS6804, DXS9896 and GATA144D04 is 0.5990, 0.6220, 0.4280, respectively. The result showed that all the three STR loci were polymorphic among 100 unrelated females and 120 unrelated males from Chinese Han population. Chi(2) tests demonstrated that genotype frequencies in females did not depart from Hardy-Weinberg equilibrium. Three X-chromosome linked short tandem repeat loci have high polymorphism, they can be applied to forensic medicine and population genetics.

Characterization of eight Y-STR loci and haplotypes in a Chinese Han population.

In this study we analyzed the eight Y-STR loci, DYS443, DYS444, DYS448, DYS453, DYS455, DYS456, DYS457 (DYS437) and DYS458, investigated haplotype distributions of these Y-STR loci in a Chinese Han population, and sequenced alleles of the eight loci for clarifying the structure. Extracted DNA was amplified by PCR and the PCR products were analyzed by non-denaturing horizontal polyacrylamide gel electrophoresis with a discontinuous buffer system. Alleles were sequenced on an ABI 3700 using a Dye Terminator Cycle sequencing kit. DYS443, DYS453, DYS455 and DYS456 were found to be simple repeat systems, while DYS444, DYS448, DYS457 (DYS437) and DYS458 were complex repeat systems. The gene diversities of DYS443, DYS444, DYS448, DYS453, DYS455, DYS456, DYS457 (DYS437) and DYS458 were 0.7742, 0.7671, 0.7453, 0.3545, 0.0549, 0.6988, 0.6148 and 0.8213, respectively. The haplotype diversity for 8 Y-STR loci was 0.9996, and the discrimination capacity was 0.9815. The results indicate that these eight loci are useful Y-linked markers for forensic applications.

Enhanced washout of 99mTc-tetrofosmin in hypertrophic cardiomyopathy: quantitative comparisons with regional 123I-BMIPP uptake and wall thickness determined by MRI.

The diagnostic value of technetium-99m tetrofosmin (TF) washout in hypertrophic cardiomyopathy (HCM) was examined by investigating its relation to the metabolic abnormality depicted by iodine-123 beta-methyl- p-iodophenylpentadecanoic acid (BMIPP) uptake and the left ventricular (LV) myocardial wall thickness as measured by magnetic resonance imaging (MRI). TF washout was evaluated in 31 patients with HCM and 23 normal control subjects using 30-min (early) and 3-h (delayed) TF single-photon emission tomography images. The LV myocardial wall was divided into 19 segments and the percentage TF washout, regional BMIPP uptake and LV wall thickness were measured in each segment. Mean TF washout in the patients with HCM was significantly faster than that in normal control subjects (23.7+/-5.7 vs 13.4+/-4.1, P<0.0001). In the patients with HCM, TF washout showed an excellent correlation with MRI wall thickness ( r=0.82, P<0.0001) and a good inverse correlation with regional BMIPP uptake ( r=-0.72, P<0.0001). In addition, a good linear correlation was observed between TF uptake and MRI wall thickness in the 19 regional segments. In conclusion, the degree of TF washout corresponds well with the severity of myocardial wall thickness and the degree of metabolic abnormality in patients with HCM. These results suggest that enhanced TF washout might provide additional clinical information regarding metabolic alterations in HCM.

[Specifics anti-tumor immunity induced by gene immunization with ectopic hCGbeta encoding gene].

OBJECTIVE: To investigate the specific anti-tumor immunity induced by gene immunization with ectopic hCG encoding gene. METHODS: BALB/c mice were immunized with plasmid TR421-hCGbeta coding for hCGbeta and mock DNA for 3 times at 3 weekly intervals. The level of specific anti-hCGbeta IgG antibody in the serum was determined by ELISA at the indicated time in the two groups. The growth inhibitory activity of the sera against tumor cells was examined in vitro by [(3)H]-Thymidine incorporation assay. Specific lympho-proliferation versus hCGbeta was detected by [(3)H]-Thymidine incorporation assay with hCGbeta protein or inactivated SP2/0-hCGbeta cells as specific stimulating antigen. Cytotoxic T lymphocyte (CTL) activity of the splenocytes derived from the immunized mice was measured by [(3)H]-Thymidine release assay. Protective assay was performed by subcutaneous inoculation of SP2/0-hCGbeta cells into the immunized mice. The weight and formation rate of the tumor were evaluated after challenge. RESULTS: All mice immunized with plasmid TR421-hCGbeta developed high level of anti-hCGbeta antibodies, which could inhibit the growth of Hela cells and SP2/0-hCGbeta cells compared with the serum from animals immunized with mock DNA (P < 0.05). The high-level specific lympho-proliferation against hCGbeta protein or/and inactivated SP2/0-hCGbeta cells were shown in TR421-hCGbeta immunized mice, whereas no significant proliferative activity was found in mock DNA immunized animals (P < 0.01). A strong cytotoxic activity against SP2/0-hCGbeta in TR421-hCGbeta immunized mice was found. Inoculation of SP2/0-hCGbeta cells into the mice immunized with mock DNA developed large tumors within 25 days. But a marked reduction of tumor weight and formation rate was found after the tumor cells challenge in the mice immunized with TR421-hCGbeta plasmid DNA (P < 0.01). CONCLUSION: The gene immunization of ectopic hCGbeta encoding gene, eliciting high-level of specific humoral and cellular immune responses, could inhibit the growth of tumor cells harboring ectopic hCGbeta in vitro and in vivo.

[Application of short tandem repeat DNA in model of human-mouse cord blood transplantation].

OBJECTIVE: To evaluate the significance of applying short tandem repeat DNA in the model of human-mouse cord blood transplantation. METHODS: The techniques of STR-PCR and colony forming cell culture were used to examine the engrafting and influence of hematopoietic growth factor on process in the mice after transplantation of human cord blood (CB). RESULTS: 1. Human DNA expression was detected in the mice that had received tail-vein injection of human CB (1-5 x 10(7) MNC per mouse) but it was not detected in the mice that have received intraperitoneal injection of human CB. 2. Human CB transplanted in SCID mice could reconstitute the hematopoietic function of the mice; the expression level of human cell was not related with the use of hematopoietic growth factor. CONCLUSION: STR-PCR analysis provide a simple and accurate indication of engraftment in the model of human-mouse cord blood transplantation.

Anticancer activity of Hemsleya amabilis extract.

Hemsleya amabilis extract is derived from the medicinal herb Hemsleya amabilis, which has long been used to treat cancer and many other conditions. The underlying mechanism is not clear. To investigate Hemsleya amabili's anticancer activity, we have treated different types of cancer cells including human astrocytoma U87 cells, breast cancer cells MDA-MB-231and Jurkat cells with Hemsleya amabilis extract. This agent significantly inhibited tumor cell growth and colony formation at various concentrations. When astrocytoma cells were seeded in the presence of Hemsleya amabilis extract at very low concentrations, cell spreading was greatly inhibited. Hemsleya amabilis extract also promoted tumor cell death in all the tested cell lines, but with varied sensitivities. Apoptotic assays with Annexin V staining demonstrated that Hemsleya amabilis extract induced astrocytoma cell apoptosis at different concentrations.

Vessel imaging by interferometric phase-contrast X-ray technique.

BACKGROUND: Phase-contrast x-ray imaging using an x-ray interferometer has great potential to reveal the structures inside soft tissues, because the sensitivity of this method to hydrogen, carbon, nitrogen, and oxygen is approximately 1000 times higher than that of the absorption-contrast x-ray method. Imaging of vessels is very important to understand the vascular distribution of organs and tumors, so the possibility of selective angiography based on phase contrast is examined with a physiological material composed of low-atomic-number elements. METHODS AND RESULTS: Phase-contrast x-ray imaging was performed with a synchrotron x-ray source. Differences in refractive index, ddelta, of physiological saline, lactated Ringer's solution, 5% glucose, artificial blood such as pyridoxylated hemoglobin-polyoxyethylene conjugate, and perfluorotributylamine were measured. Because the ddelta of physiological saline has highest contrast, it was used for the phase-contrast x-ray imaging of vessel, and this was compared with absorption-contrast x-ray images. Vessels >0.03 mm in diameter of excised liver from rats and a rabbit were revealed clearly in phase-contrast x-ray imaging, whereas the vessel could not be revealed at all by the absorption-contrast x-ray image. Absorption-contrast x-ray images with iodine microspheres depicted only portal veins >0.1 mm in diameter with nearly the same x-ray dose as the present phase-contrast x-ray imaging. CONCLUSIONS: Phase-contrast x-ray imaging explored clear depiction of the vessels using physiological saline with small doses of x-rays.

Oligonucleotide mixture analysis via electrospray and ion/ion reactions in a quadrupole ion trap.

Electrospray ionization combined with ion/ion reactions in a quadrupole ion trap can be used for the direct analysis of oligonucleotide mixtures. Elements to the success of this approach include factors related to ionization, ion/ion reactions, and mass analysis. This paper deals with issues regarding the ion polarity combination, viz., positive oligonucleotides/negative charge-transfer agent versus negative oligonucleotides/positive charge-transfer agent. Anions derived from perfluorocarbons appear to be directly applicable to mixtures of positive ions derived from electrospray of oligonucleotides, in direct analogy with positive protein ions. Conditions for forming positive oligonucleotide ions devoid of adducts were more difficult to establish than for forming relatively clean negative oligonucleotide ions. A new approach for manipulating negative ion charge states in the ion trap is described and is based on use of the electric field of the positive charge-transfer agent for storage of high-mass negative ions formed during the ion/ion reaction period. Oxygen cations are shown to be acceptable for charge-state manipulation of mixed-base oligomers but induce fragmentation in polyadenylate homopolymers. Protonated isobutylene (C4H9+), on the other hand, is shown to induce significantly less fragmentation of polyadenylate homopolymers.

[The relationship between physical distance and genetic distance on chromosome 22].

OBJECTIVE: To construct a genetic map based on data from the Chinese population in northern part of China and to compare relationship between physical distance and genetic distance on chromosome 22. METHODS: PCR amplification was employed to genotype 6 STR loci on chromosome 22, and pedigree analysis was performed. RESULTS: A genetic map of Chinese Han population in the northern part of China was constructed and a preliminary comparison of the physical and genetic distances between 6 STR loci on chromosome 22 was made. CONCLUSION: There is complex relationship between genetic distance and physical distance: the distance between STR loci is related to physical distance but also recombination fraction, and there are differences of the genetic and physical distances on chromosome 22 between Chinese and Caucasian, and between the male and female.

Typing Y chromosome STR haplotypes using redesigned primers.

A panel of Y-specific STR loci, including DYS19, DYS389, DYS390, DYS391, DYS392, and DYS393 was analyzed using horizontal nondenaturing polyacrylamide gel electrophoresis with a discontinuous buffer system (horizontal disk-PAGE). In order to obtain correct results for the larger DYS389 and DYS392 alleles, it was necessary to design new primers that bind closer to the repeat region and lead to a significant reduction of the amplified fragment size. Using the modified primer sets the horizontal disk-PAGE results were consistent with a nondenaturing approach using fluorescent primers and a 377 automated sequencer. The modified procedure also amplifies the second repeat stretch at the duplicated DYS389 locus as a single fragment, which results in an immediate allele identification. The results indicate that horizontal disk-PAGE with silverstaining is a simple approach to type the recommended Y-specific STR markers.

[Genetic map of 10 STR loci on chromosome 9].

To understand the genetic polymorphism of 10 STR loci on chromosome 9 and reveal the difference on the linkage maps of different populations, the author used a set of nine tetranucleotide- and one tritranucleotide-repeat STR markers on chromosome 9 chosen from Genome Data Bank. EDTA-blood was collected from unrelated family in Baiyin City, Gansu province. DNA was extracted by Chelex-100 method and amplified by the polymerase chain reaction (PCR). The PCR products were analyzed using PAGE electrophoresis. We calculated the allele frequencies and heterozygosity and PIC of every ten STR loci. Using the data of Chinese Han pedigree, we constructed a genetic map of chromosome 9 making up with 10 STR loci. Then we compared our map with the map in GDB. Our results indicate that there are some differences on the genetic map between Chinese and GDB. That means, it is essential to do some population genetic evaluations before using the data of GDB into Chinese population.

[A study of forensic DNA databases].

OBJECTIVE: The forensic DNA databases are very important for individual identification. In order to evaluate the genetic markers used for a forensic DNA databases and the compatibility between the manual DNA typing system and the automatic DNA typing system, a testing DNA database should be constructed. Also, constructing a testing DNA database can increase our understanding of the issue for forensic DNA databases. METHODS: A total of 1000 specimens, including samples of blood, blood stains, salvia stains, semen stains, mixture stains and muscle tissues, were collected from the public security bureau of Chengdu. The DNA of each specimen was extracted by Chelex method and analyzed using Amp-FLP technique. A total of 8 STR loci, including D3S1358, D9S1118, vWA, D5S818, D16S539, D8S1179, CSF1PO and D20S161 were chosen and employed for DNA typing. Each STR locus was amplified by the polymerase chain reaction PCR and the PCR products were typed with the polyacryamide gel electrophoresis. Typing DNA was carried out by comparing with a human allele ladder. A total of 8 human allele ladders for D3S1358, D9S1118, vWA, D5S818, D16S539, D8S1179, CSF1PO and D20S161 were made in-house. Managing software of the testing DNA database was designed using Microsoft Access. RESULTS: The results of DNA typing in 1000 specimens showed that the total discrimination power of 8 STR loci was over 0.99999999. CONCLUSION: This study show that a forensic DNA database should be useful for search purpose. The total discrimination power over 0.99999999 imply that in principle there is no identical genotype at whole 8 STR loci between two persons from a population with 10000000 individuals. This means that 8 STR loci used in this study are suitable to construct forensic DNA databases in Chengdu of China. The result of DNA typing can be repeated and the data have compatibility between the manual DNA typing system and the automatic DNA typing system. The data search in our testing DNA database can be carried out using only some loci of the set of 8 STR markers. Also, the volume of our testing DNA databases could be enlarged easily. The implication from this study is that the legislation should not be negligent before establishing a forensic DNA database. This DNA database provides a model for establishing the forensic DNA databases in China.

[Hemodynamics monitoring of liver transplantation with color Doppler ultrasound].

By hemodynamic monitoring of liver transplantation with Color Doppler ultrasound, we observed the hemodynamic changes pre- and post-operatively and the patency of the anastomotic vessels, to detect or exclude the presence of complications, then assess the prognosis. We measured the hepatic blood flow after orthotopic liver transplantation in recent 6 cases, and observed the patency of anastomostic vessels and the hemodynamic improvement of the portal vein and hepatic arteries. The abnormal liquid and solid echoes were not found around the transplantated liver in all 6 cases. The echoes of hepatic parenchyma and tububes were normal. The direction and speed of blood flow were also normal. Five of anastomoses of portal vein in 6 cases were patent. The rest one was a little stenosed, and the speed of local blood flow was a little faster. There were 11 patent ones and 1 mildly stenosed in the 12 anastomoses of inferior venae cavae. The speed index(SI) and resistance index(RI) of hepatic arterial blood flow distal to the anastomoses were all normal. Liver transplantation is the redical therapying of portal hypertension, and the color Doppler ultrasound is the best non-invasived method for monitoring the hemodynamic changes of transplantated liver.

[Purification and characterization of antifungal proteins in triticale seed].

Using Trichoderma as an indicative fungus, three antifungal proteins in Triticale Zhongsi 237 seed were purified and characterized. These protein components were considered to be a new Class II chitinase and two kinds of beta-1, 3-glucanases. Chitinase molecular mass was 30.5 kD and enzyme activity was maximal at pH 6.0 and 37 degrees C. Two beta-glucanases molecular masses were 51 kD and 23 kD. N-terminal amino acid sequences of Triticale chitinase share high homology with barley chitinase. In some conditions, the chitinase and beta-glucanases all had strong antifungal activity and were able to inhibit Trichoderma growth synergistically. Moreover, the chitinase and beta-1, 3-glucanases were able to inhibit powdery mildew growth on detached susceptible wheat leaves.
