[New principles in tumor antigens and their significance in future immunotherapies for lymphomas and other malignancies--editorial].

The molecular characterization of self-antigens expressed by human malignancies that are capable of elicitation of anti-tumor immune responses in patients has been an active field in hematology, oncology, and tumor immunology. More than 2000 tumor antigens have been identified. These significant progresses have led to the renaissance of tumor immunology and studies on novel anti-tumor immunotherapies in lymphomas, other hematologic malignancies and tumors. However, despite of the progress in the identification of these self-tumor antigens, current antigen-specific immunotherapies for tumors are far less satisfactory than that expected, which reflects the urgent need to improve our understanding on the basic principles underlying the selection of these self-tumor antigens. In order to develop more effective antigen-specific anti-tumor immunotherapies and to monitor the responses to these immunotherapies in patients with lymphomas and other malignancies, many additional questions need to be addressed. In this brief review, the progress in the identification of tumor antigens in lymphomas and other malignancies was outlined and the new principles of self-tumor antigens and their significance for future immunotherapies to these malignancies were summarized.

[Investigation of calculating the FRET efficiency of donor-acceptor pair].

The research on the method of calculating FRET efficiency is important to organic white LED research. In the present article, a new method of calculating the FRET efficiency of donor-acceptor pair by means of emission and excitation spectra was investigated. This method is easy to use to process data, needs only normal experiment equipments and doesn't need the parameters such as fluorescence quantum yield. However it can only be used to calculate comparative result. With this method, the FRET efficiency of fluorescein sodium, alpha-naphthol, bromocresol purple and riboflavin with different concentrations was studied, and the results match the experimental results completely, so this method has proved to be reasonable.

[Effect of sunlight irradiation on fluorescence properties of dissolved organic matter].

Three-dimensional excitation emission matrix fluorescence spectroscopy (3DEEM) was used to investigate the effect of sunlight irradiation on the fluorescence properties of dissolved organic matter (DOM) from Lake Hongfeng and Nanming River waters and a commercial fluka humic acid (FHA). The results show that the DOM samples and FHA fluorescence properties changed under sunlight irradiation. Interestingly, the photodegradation characteristics were different between aquatic DOM and FHA. The fluorescence intensity of the apparent peaks A, B and C of lake and river water DOM decreased with sunlight irradiation. The initial 3DEEM of Fluka HA had only one fluorescence peak at lamda ex / lamda em = 275/500 nm, while two fluorescence peaks occurred at lamda ex / lamda em = 245/450 nm and 310/450 nm, respectively, after sunlight irradiation. lamda ex and lamda maxima of DOM decreased during 7 days of sunlight irradiation. Changes in r(A, C) of DOM and FHA with sunlight irradiation time suggest that fluorescence peaks A and C had different fluorescence loss rates, while peak C fluorophores were more susceptible to sunlight irradiation. FHA appeared to be less susceptible to photodegradation, and its r(A, C) remained almost the same before and after sunlight irradiation.

[Optical mapping of the membrane potential with voltage-sensitive dyes].

Optical mapping of the membrane potential with voltage-sensitive dyes is an advanced approach that involves in many theories such as molecular photonica, physiology and computer science. Compared with the conventional techniques of membrane patch-clamp and microelectrode, the optical mapping system can measure not only the changes of a single membrane potential from multiple sites simultaneously but also the conduction properties of the cell populations. So this technique provides an important method for studying the electrophysiology of the small cell, the tiny neurite and the cardiac arrhythmia, etc. Because this technique can also avoid the electromagnetic interferences, it uniquely provides an ideal means for studying the mechanisms such as cardiac defibrillation. In this paper the principles and the system structure of the optical mapping are introduced, its applications and future developments are also presented at the same time.

Preliminary analysis of gene expression profiles in HepG2 cell line induced by different genotype core proteins of HCV.

In present investigation, we constructed recombinants expressing the HCV genotypes 1b, 2a, and 4d core proteins, and established human hepatocellular carcinoma (HepG2) cell line that expressed various genotype core proteins. The gene expression profiles in the cells expressing different HCV genotype core proteins were compared with those in the control by microarray analysis. In data analysis, a threshold was set to eliminate all genes that were not increased or decreased by 2.5-fold change in a comparison between the transfected cells and control cells. The preliminary microarray analysis suggests that the gene expression profiles regulated by three kinds of genotype core proteins are mainly involved in transport, signal transduction, regulation of transcription, protease activity, etc., and that some pathogenesis/oncogenesis gene expressions are up/down- regulated simultaneously in the HepG2 cell line. The data suggest that each core protein has its gene expressions profile and that the profiles are implicated in HCV replication and pathogenesis, which may open up a novel way to understand the function of the HCV variant core proteins biological and their pathogenic mechanism.

[Expression and significance of X-linked inhibitor of apoptosis protein and its antagonized proteins in acute leukemia].

To investigate the expression and significance of X-linked inhibitor of apoptosis protein (XIAP) and XIAP-associated factor 1 (XAF1) in acute leukemia, the expression of XIAP, XAF1, Smac, and HtrA2 mRNA in the bone marrow aspirates from 87 newly diagnosed AL patients, 23 patients in remission, 6 patients in relapse, and 17 normal controls were detected by means of reverse transcriptase polymerase chain reaction (RT-PCR), and their relationship with clinical therapeutic efficiency was analyzed. The results showed that the expression level of XIAP mRNA in newly diagnosed AL patients (0.990 +/- 0.337) was significantly higher than that in normal controls (0.395 +/- 0.148) (P < 0.01); the positive rate and expression level of XAF1 mRNA in newly diagnosed AL patients (56.32%, 0.246 +/- 0.267) were significantly lower than that in normal controls (100%, 0.964 +/- 0.387) (P < 0.01). In 69 out of 87 newly diagnosed AL patients, efficacy remained evaluable. AL patients with high level of XIAP achieved a lower complete remission (CR) rate than patients with low level of XIAP (54.55% and 86.11%, respectively) (P < 0.01). XAF1 positive patients achieved a higher CR rate than XAF1 negative patients (86.84% and 51.61%, respectively) (P < 0.01). It is concluded that the overexpression of XIAP and negativity of XAF1 may be two adverse prognostic factors in AL patients.

[Calculation of soil water erosion modulus based on RUSLE and its assessment under support of artificial neural network].

With Hengshan County of Shanxi Province in the North Loess Plateau as an example, and by using ETM + and remote sensing data and RUSLE module, this paper quantitatively derived the soil and water loss in loess hilly region based on "3S" technology, and assessed the derivation results under the support of artificial neural network. The results showed that the annual average erosion modulus of Hengshan County was 103.23 t x hm(-2), and the gross erosion loss per year was 4. 38 x 10(7) t. The erosion was increased from northwest to southeast, and varied significantly with topographic position. A slight erosion or no erosion happened in walled basin, flat-headed mountain ridges and sandy area, which always suffered from dropping erosion, while strip erosion often happened on the upslope of mountain ridge and mountaintop flat. Moderate rill erosion always occurred on the middle and down slope of mountain ridge and mountaintop flat, and weighty rushing erosion occurred on the steep ravine and brink. The RUSLE model and artificial neural network technique were feasible and could be propagandized for drainage areas control and preserved practice.

[Polymorphisms of hsp 60 gene in Chinese Han people].

OBJECTIVE: To identify the single nucleotide polymorphisms (SNPs) in the regulatory and coding regions of heat shock protein 60 gene and search for its genetic makers in Chinese Han people. METHODS: The 5' flank region, parts of the exons and introns of hsp60 gene were resequenced to identify the SNPs in Chinese Han people, and then the sequenced results to the Japanese, European and African's data in National Center for Biotechnology Information (NCBI) and HapMap databases were compared. RESULTS: One novel SNP was identified in exon 2 resulting in synonymous variant and the G allele frequency was 0.025. There were 11 reported SNPs in the sequenced region. The minor allele frequencies of rs1116734, rs3749095, rs1050347, rs8539 were 0.51, 0.30, 0.29, 0.49. The heterozygosity of the other 7 SNPs was 0. The distributions of rs1116734, rs1050347, rs8539, rs3749095 in Chinese Han people were similar to the Japanese's. The hsp60 rs3749095 which was not found in Japanese people was a high-frequency SNP in Chinese Han people; the distribution of rs8539 in Chinese Han people was quite different from European and African's (P < 0.01). CONCLUSION: The SNPs of hsp60 in Chinese Han people are different from the other peoples. The SNPs of hsp60 gene rs1116734, rs3749095, rs1050347, rs8539 are very common in Chinese Han people and might be used for candidate genetic markers of hsp60 gene.

[Development of a gold-immunochromatography test for rapid detecting E. coli O157].

OBJECTIVE: To develop a method for rapid detecting Escherichia coli (E. coli) O157 on site. METHODS: A colloidal gold immunochromatography test based on double-antibody sandwich assay for detecting E. coli O157 was developed. Its sensitivity and specificity were then evaluated, and its feasibility of screening food samples were evaluated by analyzing various samples added with E. coli O157. RESULTS: Typical detecting time is less than 15 minutes per sample. The sensitivity of the test is 1 x 10(5) cfu/ml. No any cross-reaction with 30 strains of 24 species in Enterobacteriaceae (including non-O157 E. coli, Salmonella, Shigella, Proteus, Citrobacter, Enterobacter, Serratia and Yersinia), Staphylococus, Listeria, Aeromonas and Vibrios was found. The test could be used to detect E. coli O157 in various samples such as milk powder, flour, starch, coffee, biscuit, cake, jelly and juice. CONCLUSION: The gold-immunochromatography test appears to be a rapid, convenient, specific and sensitive test for detecting E. coli O157: H7 on site.

Dual regulation of soluble tumor necrosis factor-alpha induced activation of human monocytic cells via modulating transmembrane TNF-alpha-mediated 'reverse signaling'.

Transmembrane tumor necrosis factor-alpha (mTNF-alpha) is known to be the precursor of soluble TNF-alpha (sTNF-alpha). mTNF-alpha can act as a ligand on the TNF receptor- (TNFR)- bearing cell through 'forward signaling' or as a receptor on the TNF producing cell through 'reverse signaling'. In the current study, we investigated the role of mTNF-alpha-mediated reverse signaling in regulating sTNF-alpha-induced activation of human monocytic U937 cells. We demonstrated that pretreatment with sTNFRI, for inducing reverse signaling through mTNF-alpha, sensitized U937 cells to sTNF-alpha stimulation, as evidenced by an increase in reactive oxygen production and mRNA levels of proinflammatory cytokines (TNF-alpha, IL-1beta, and IL-8) in these cells. Further experiments revealed that IkappaB-alpha degradation was increased in the monocytic cells primed with sTNFRI, implying that reverse signaling of mTNF-alpha sensitizes U937 cells via an NF-kappaB-dependent mechanism. On the other hand, binding of sTNFRI to mTNF-alpha after sTNF-alpha-induced activation of U937 cells reduced mRNA stability (half-life) of IL-1beta and IL-8. The involvement of reverse signaling in the process was verified by using a mutated form of mTNF-alpha lacking the majority of the cytoplasmic domain. Our results clearly showed that enhanced mRNA degradation of the cytokines occurred only in U937 cells transfected with a wild-type mTNF-alpha, but not in those cells transfected with the mutant mTNF-alpha. Taken together, these data suggest that reverse signaling through mTNF-alpha may exert a double role in modulating sTNF-alpha bioactivity. It is positive when reverse signaling occurs prior to sTNF-alpha stimulation, while it is negative when reverse signaling occurs after the sTNF-alpha signal. Thus, our findings strengthen a role of mTNF-alpha-mediated reverse signaling in the regulation of immune-inflammatory response and control of inflammatory reaction.

[Determination of trace microcystins in water by ultra performance liquid chromatography/tandem mass spectrometry].

An analytical method for the analysis of trace microcystins (MCYST) in water was developed using solid phase extraction (SPE) for enrichment and ultra performance liquid chromatography/tandem mass spectrometry (UPLC/MS/MS) for detection. One litre of water was passed through SPE columns, the extracted sample was rinsed off by 10 mL methanol, then evaporated to 1.0 mL before being analyzed with UPLC/MS/MS. The effect of formic acid concentration in the mobile phase on the sensitivity was studied and the results showed that 0.1% was the optimum concentration. Four microcystins, MCYST-LR, RR, LW, LF, can be separated and detected in 5 min, which is much shorter than that by the conventional liquid chromatography. The detection limits were 1.3 - 6.0 ng/L, and the recoveries were 91.1% - 111%. The calibration curves showed good linearity in the range of 1.0 microg/L - 1.0 mg/L with correlation coefficients larger than 0.99. The method was also applied to determine MCYST in real water samples from three reservoirs in Zhejiang Province, and the results showed that the concentrations of LR and RR were 0.044 7 - 2.73 microg/L and 0.020 8 - 1.36 microg/L respectively, and LW and LF were not detected.

[Preparation of soybean isoflavone glucosides by reversed-phase high performance liquid chromatography].

A method was established for the isolation of soybean isoflavone glucosides from the total isoflavone extracts of soybean using preparative reversed-phase high performance liquid chromatography (RP-HPLC). The total isoflavone extracts were separated into four parts by solvent extraction, those are the ethyl acetate extract, butanol extract, precipitate (D4), and the remaining aqueous phase. The part D4 containing soybean isoflavone glucosides was acquired and subjected to preparative HPLC for the isolation of target components. A preparative Nova-Pak HR C18 column (100 mm x 25 mm i. d. , 6 microm) was used in the preparation process. By isocratic elution with methanol-0.1% aqueous acetic acid (23:77, v/v) as the mobile phase at a flow rate of 20 mL/min, followed by concentration and desalination, three soybean isoflavone glucosides were obtained and subsequently identified by mass spectrometry as daidzin, glycitin, and genistin. HPLC analysis showed that the purities of the three soybean isoflavone glucosides were all higher than 99%.

[Correlation of P27 expression and localization to phosphorylated AKT in non-small cell lung cancer].

BACKGROUND & OBJECTIVE: Inactivation of tumor suppressor gene plays an important role in tumorigenesis and tumor development. Functional inactivation of the tumor suppressor gene p27 in human cancer occurs either through the loss of expression or through phosphorylation-dependent cytoplasmic mislocalization. AKT, known as protein kinase B, represents a subfamily of serine/threonine protein kinases. Activated AKT (phosphorylated AKT, p-AKT) could promote cell cycle progression by modulating many effectors in cell cycle, including the expression and localization of P27. This study was to investigate the expression and localization of P27 protein in human non-small cell lung cancer (NSCLC), and further to explore its correlation to p-AKT. METHODS: The expression and subcellular localization of P27 and the expression of p-AKT in 80 cases of NSCLC and 35 cases of non-cancerous lung disease were detected by immunohistochemistry, and their correlations to clinicopathologic factors were statistically analyzed. RESULTS: The total and nuclear positive rates of p27 were significantly lower in NSCLC than in non-cancerous lung disease (72.5% vs. 94.3%, 46.3% vs. 94.3%, P<0.01). There were 21 cases of NSCLC with only cytoplasmic expression of P27, while no cytoplasmic expression alone was found in 35 cases of non-cancerous lung disease (P<0.01). The nuclear positive rate of P27 was significantly higher in NSCLC without lymph node metastasis than in NSCLC with lymph node metastasis (P<0.05), and higher in well-and moderately-differentiated NSCLC than in poorly-differentiated NSCLC (P<0.05). The nuclear and cytoplasmic expression of P27 has no correlation to clinicopathologic factors of NSCLC (P>0.05). The positive rate of p-AKT was significantly higher in NSCLC than in non-cancerous lung disease (78.8% vs. 0, P<0.05); it was significantly higher in well-and moderately-differentiated adenocarcinoma than in poorly-differentiated adenocarcinoma (95.0% vs. 50.0%, P<0.05). In NSCLC, the expression of p-AKT was positively correlated to the cytoplasmic expression of P27 (r=0.437, P<0.01), but had no correlation to the nuclear expression of P27 (r=0.175, P>0.05). CONCLUSIONS: Reduced nuclear expression or loss of expression of P27 may be associated to the development of NSCLC. The localization of P27 only in cytoplasm may be one of the characteristics of lung cancer cells. P-AKT might play a role in the cytoplasmic localization of P27, but has no correlation to its nuclear expression.
